Anti-oxidation and Anti-wrinkling Effects of Jeju Horse Leg Bone Hydrolysates

  • This study focused on the anti-oxidative and collagenase- and elastase inhibition effects of low molecular weight peptides (LMP) from commercial Jeju horse leg bone hydrolysates (JHLB) on pancreatin, via enzymatic hydrolysis. Cell viability of dermal fibroblasts exposed to UVB radiation upon treatment with LMP from JHLB was evaluated. Determination of the antioxidant activity of various concentrations of LMP from JHLB were carried out by assessing 1,1-diphenyl-2-picrylhydrazyl (DPPH) and 2,2-azino-bis-3-ethybenzothiazoline-6-sulphonic acid (ABTS) radical scavenging activity, ferric reducing antioxidant power (FRAP), and oxygen radical absorbance capacity (ORAC). The DPPH radical scavenging activity of LMP from JHLB (20 mg/mL) was 92.21% and ABTS radical scavenging activity (15 mg/mL) was 99.50%. FRAP activity (30 mg/mL) was $364.72{\mu}M/TE$ and ORAC activity (1 mg/mL) was $101.85{\mu}M/TE$. The anti-wrinkle potential was assessed by evaluating the elastase- and collagenase inhibition potential of these LMP. We found that 200 mg/mL of LMP from JHLB inhibited elastase activity by 41.32%, and 100 mg/mL of LMP from JHLB inhibited collagenase activity by 91.32%. The cell viability of untreated HS68 human dermal fibroblasts was 45% when exposed to a UVB radiation dose of $100mJ/cm^2$. After 24 h of incubation with $500{\mu}g/mL$ LMP from JHLB, the cell viability increased to 60%. These results indicate that LMP from JHLB has potential utility as an anti-oxidant and anti-wrinkle agent in the food and cosmetic industry. Additional in vivo tests should be carried out to further characterize these potential benefits.

Effects of Sibseonsan as an Anti-Inflammatory, Anti-Wrinkle, and Skin Whitening Treatment

  • Background: The purpose of this study was to investigate whether Sibseonsan (SSS) is an effective antiinflammatory, anti-wrinkling, and whitening agent. Methods: To determine whether SSS had an anti-inflammatory effect, a murine macrophage cell line was used (RAW 264.7) and production of DPPH, NO, TNF-α, and PGE2 were measured. To ascertain potential anti-wrinkle effects of SSS in these cells, collagenase and elastase production were measured. To verify whether SSS had a whitening effect, tyrosinase activity and DOPA staining were performed using a melanoma cell line (B16/F10). Results: There was no significant reduction in survival of SSS-treated RAW 264.7 cells, up to 400 ㎍/mL. Free radical scavenging (23.96 ± 1.85%) was observed in RAW 264.7 cells treated with SSS at a concentration of 400 ㎍/mL. The SSS treatment group (400 ㎍/mL) significantly inhibited NO production compared with the LPS stimulated treatment group. The SSS treatment of macrophage cells appeared to reduce production of TNF-α in a concentration dependent manner. There was a significant reduction in the concentration of PGE2 by about 25% in the SSS treatment (400 ㎍/mL) group (p = 0.05). Compared with the control, the production of collagenase and elastase in B16/F10 cells treated with SSS (400 ㎍/mL) was greater by 26.37% and 45.71%, respectively. The SSS treatment (400 ㎍/mL) group showed a significant reduction by about 17% in tyrosinase production in B16/F10 cells. The SSS treatment group showed little change in DOPA staining. Conclusion: SSS extract may be useful for the treatment and prevention of inflammatory diseases and may have anti-wrinkle and whitening effects. These results may support the use of SSS in clinical practice.

Effects of ethanol extract of Nelumbo nucifera leaves on Anti-oxidation and Type I Procollagen Expression in CCD-986sk Cells (연잎 에탄올 추출물이 피부 주름에 미치는 영향)

  • The natural plants were screened for new cosmetic materials. Many parts of Nelumbo nucifera has been investigated for anti-wrinkling and anti-furrowing effects. The leaves. flowers and stamens of Nelumbo nucifera were extracted with ethanol or distilled water. The effects of cell viability and $H_2O_2-induced$ cytotoxicity were studied on CCD-986sk human fibroblast cell line by MTT assay. We measured various effects related to skin such as the synthesis of MMP-1 and type I procollagen. The ethanol extract of Nelumbo nucifera leaves (ENL) at concentrations of 30 ${\mu}g/ml$ showed maximum cell viability on CCD-986sk cells by 129.70%. and $H_2O_2-induced$ cytotoxicity was blocked by ENL (5. 10. 20. 30. and 40 ${\mu}g/ml$) concentration-dependently. The synthesis of MMP-1 and type I procollagen were positive in ENL. These results suggest that ENL has anti-oxidant and anti-furrowing effects.

In vitro Screening of Jeju Medicinal Plants for Cosmeceutical Materials

  • One of the important functions of skin is protection from harmful environments. Many studies have explored how to prevent skin from wrinkling and the occurrence of pigmentation changes. Skin wrinkling and pigmentation changes could be caused by unusual disruption of connective tissue, the formation of free radicals and ultraviolet radiation. In this study, extracts obtained from 254 different kinds of Jeju medicinal plants were screened for inhibitory effects on tyrosinase and elastase, and for free radical scavenging effects. Four herbs, Phormium tenax, Morus bombycis, Morus alba, and Cudrania tricuspidata, were potent inhibitors of tyrosinase ($IC_{50}$ values 4.62, 5.46, 8.17, and 64.17 ${\mu}g$/mL, respectively). Aleurites fordii [$IC_{50}$: 5.29 ${\mu}g$/mL, 1,1-diphenyl-2-picrylhydrazyl (DPPH)], Distylium racemosum ($IC_{50}$: 6.14 ${\mu}g$/mL), Acer palmatum ($IC_{50}$: 5.44 ${\mu}g$/mL), and Spiraea salicifolia ($IC_{50}$: 5.25 ${\mu}g$/mL) showed good antioxidative effects. Furthermore, Distylium racemosum ($IC_{50}$: 7.51 ${\mu}g$/mL), Diospyros kaki ($IC_{50}$: 15.1 ${\mu}g$/mL), Cornus macrophylla ($IC_{50}:$ 16.59 ${\mu}g$/mL), and Psidium guajava ($IC_{50}$: 40.25 ${\mu}g$/mL) exhibited potent inhibitory effects on elastase. These results suggest that medicinal plants possessing several biological activities may be potent inhibitors of the processes involved in pigmentation increases and aging. Further investigations will focus on in vivo assays and on the chemical identification of the major active components responsible for whitening and anti-aging activity in the screened efficacious extracts.

Enhancement of Anti-wrinkle Activities of Abeliophyllum distichum Nakai through Low Temperature Extraction Process (미선나무의 저온 추출 공정에 의한 항주름 활성 증진)

  • Skin anti-wrinkle activities of the stems and leaves of Abeliophyllum distichum Nakai were evaluated by the extracts obtained from various extraction processes such as using hot water at $100^{\circ}C$, 70% ethanol at $85^{\circ}C$, and 70% ethanol with ultrasonication at $60^{\circ}C$ The ultrasonicated extract showed 95.62% of the highest cell viability in addition of $0.3mg/m{\ell}$ of the extracts into the normal human fibroblast cell, CCD-986sk. For antioxidant activities, the extracts using ultrasonicated extract showed the highest DPPH free radical scavenging as 80.27%, followed by 75.88% and 62.44% for the extracts using ethanol extract and water extract. The ultrasonicated extract also showed the highest elastase inhibition activity as 25.32%, compared to ethanol extract and water extract based method at 22.01% and 12.88%, respectively. MMP-1 production was most effectively decreased down to $2908.1pg/m{\ell}$ with ultrasonicated extract while $6640.8pg/m{\ell}$ with water extract and $3609.3pg/m{\ell}$ with ethanol extract, in addition of $0.3mg/m{\ell}$. Collagen production was increased up to $154.7ng/m{\ell}$ in addition of ultrasonicated extract, and followed by $121.4ng/m{\ell}$ and $31.2ng/m{\ell}$ for ethanol extract and water extract, respectively. These results indicate that the ethanol extract should have skin anti-wrinkling activities and can be improved by the ultrasonication process that high energy input elute more amounts of bioactive substances eluting more amounts of bioactive substances from the high energy input of ultrasonication.

Role of Shinorine derived from Microalgae in skin protection (미세조류 유래 Shinorine의 피부세포에서의 효능)

  • In the present study, mycosporine-like amino acids (MAAs)were extracted from in Chlamydomonas hedleyi, and their function was investigated regarding to protective capacity against UV radiation and a possibility to be developed into functional suncream including MAAs by using natural compounds. In particulr, we assessed UV protective ability and anti-inflammation of shinorine in human skin cells. As a result, shinorine can protect the skin against damage by the absorption of energy from UV radiation and functions as an anti-wrinkling substrate through a increase of collagen synthesis. These data suggest that shinorine can be utilized not only as a substrate to protect UV radiation, but anti-aging material in cosmetic products.

Anti-Melanogenesis and Anti-Wrinkle Effects of Sargassum micracanthum Extracts (잔가시 모자반 추출물의 주름 개선 및 미백 효과)

  • Sargassum micracanthum (SM) is a member of the family Sargassaceae and commonly occurs along the coast of Korea. Extracts from SM were evaluated for their antioxidant and collagenase and tyrosinase inhibitory activities based on their total phenolic concentration (TPC), 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging effect, and metal-chelating effect. The TPC of SM ethanol and water extracts used in the study was 11.20 and 11.70 mg/g of dry sample, respectively. Both SM ethanol and water extracts had high DPPH radical-scavenging effect. The metal-chelating effect of SM ethanol extract (40.47% at 0.5 mg/ml) was higher than that of water extract (23.28% at 0.5 mg/ml). With regard to the anti-wrinkling effect, SM ethanol extract showed collagenase inhibitory activity with an $IC_{50}$ value of $488.20{\mu}g/ml$. Lastly, regarding the anti-melanogenesis effect, SM ethanol extract showed higher tyrosinase inhibitory activity (45.08% at 5 mg/ml) than that shown by the water extract (21.29% at 5 mg/ml). These results suggest that SM has the potential to be a resource with natural anti-melanogenesis, anti-wrinkle, and anti-oxidant effects.

Development of wrinkled skin-on-a-chip (WSOC) by cyclic uniaxial stretching

  • The skin experiences constant physical stimuli, such as stretching. Exposure to excessive physical stimuli stresses the skin and can accelerate aging. In this study, we applied a method that allowed human fibroblasts and keratinocytes to be perfused with media to form 3D skin equivalents that were then uniaxially 10%-stretched for 12 h per day (at either 0.01 or 0.05 Hz) for up to 7 days to form wrinkled skin-on-a-chip (WSOC). There was more wrinkling seen in skin equivalents under 0.01 Hz uniaxial stretching than there was for non-stretched skin equivalents. At 0.05 Hz, the stratum corneum almost disappeared from the skin equivalents, indicating that stretching was harmful for the epidermis. At both frequencies, the production of collagen and related proteins in the skin equivalents, such as fibronectin 10 and keratin, decreased more than those in the non-stretched equivalents, indicating that the dermis also suffered from the repeated tensile stress. These results suggest that WSOCs can be used to examine skin aging and as an in vitro tool to evaluate the efficacy of anti-wrinkle cosmetics and medicines.

Effects of Ehwa Makgeolli Containing Oriental Herbs on Skin Whitening and Wrinkles (한방 이화주의 미백 및 피부 주름 개선 효과)

  • Extrinsic skin aging is characterized by the loss of skin tone and resilience, irregular pigmentation, and deep wrinkles. The aim of this study was to investigate the effects of Ehwa Makgeolli containing oriental herbs (Glycyrrhiza uralensis Fisch., Lycium chinense MILL., Morus alba L., and Saururus chinensis Baill) on skin whitening and wrinkling in human skin cells. We prepared Makgeolli extracts (HEE) with 70% ethanol. HEE significantly inhibited in vitro mushroom tyrosinase activity and reduced the cellular and secreted melanin content of mouse melanoma melanocytes (B16F1 cells). HEE down-regulated the protein expression of tyrosinase related protein (TRP)-1/-2, a key player in melanogenesis. Treatment with HEE in human keratinoctyes (HaCaT cells) inhibited the proteolytic activities of matrix metalloproteinase (MMP)-2/-9 in a dose-dependent manner and dramatically reduced the expression of MMP-2/-9. In addition, HEE attenuated lipopolysaccharide (LPS)-induced nitric oxide production in murine macrophages (RAW264.7 cells). These results indicate that HEE may be a great cosmeceutical ingredient for its whitening, anti-wrinkle, and anti-inflammatory effects.

Reduction of Photodamage by TopicaI Application of a Novel Anti-Wrinkle Agent Containing Growth Factors

  • DA-3711 is a novel anti-wrinkle agent containing growth factors derived from culture medium of artificial human skin. Photoprotective effect by DA-3711 against chronic UVB (ultraviolet B)-induced skin damage was investigated in hairless mice model. Methods: After hairless mice were irradiated to induce photodamage for 8 weeks with UVB, grouped mice were treated topically once a day with lotion base, DA-3711 (30% or l5%), Cylasphere retinol$^{\circed{R}}$ (2500 I.U.), NouriCel$^{\circed{R}}$ along with concomitant exposure to UVB for further 8 weeks. Then mice were sacrificed to assess photodamage-protective effect by replica analysis, biochemistry and histology. DA-3711 of 30% lotion significantly reduced UVB radiation-induced wrinkling, histological alterations and increased collagen contents. Whereas DA-3711 of l5% lotion and NouriCel$^{\circed{R}}$ treatment showed a partial protective effect on skin wrinkle, epidermal and dermal thickness, and collagen content, Cylasphere retinol$^{\circed{R}}$ showed no protective effects. These results demonstrate that topical application of DA-3711 can alleviate UVB-induced photodamage and potentially be used for reduction of UVB-induced photodamage.
