본 연구에서는 물사과 가지 추출물 및 용매 분획물의 항산화 활성과 세포보호효과를 검색하고 유효성분을 분리하여 화학구조를 규명하였다. 물사과 가지 추출물 및 용매 분획물의 DPPH 및 $ABTS^+$ 라디칼 소거 활성을 측정한 결과, 에틸아세테이트 및 부탄올 분획물에서 우수한 라디칼 소거 활성이 있음을 확인하였다. 또한 $H_2O_2$로 유도된 세포 손상에 대한 세포보호효과를 확인한 결과, $20{\mu}g/mL$의 농도에서 추출물 및 에틸아세테이트 분획물에서 보호효과가 나타남을 확인하였다. 항산화 활성이 좋은 에틸아세테이트 분획물에서 유효성분을 찾고자 medium pressure liquid chromatography (MPLC)를 실시하여 2개의 화합물을 분리하였으며 $^1H$ 및 $^{13}C$ NMR 데이터 분석 및 문헌 비교를 통하여 화학구조를 동정하였다; pinocembrin (1), desmethoxymatteucinol (2). 분리된 화합물에 대한 DPPH 및 $ABTS^+$ 라디칼 소거 활성을 측정한 결과, 화합물 1, 2 모두 농도의존적으로 우수한 라디칼 소거 활성이 나타났다. $100{\mu}M$의 농도에서 화합물 1, 2는 $H_2O_2$로 유도된 세포손상에 대하여 세포보호효과를 나타냈다. 이러한 연구 결과를 바탕으로 물사과 가지 추출물은 천연 항산화 소재로써 활용 가능할 것이라 사료된다.

BACKGROUND/OBJECTIVES: Sonchus asper is used extensively as an herbal anti-inflammatory for treatment of bronchitis, asthma, wounds, burns, and cough; however, further investigation is needed in order to understand the underlying mechanism. To determine its mechanism of action, we examined the effects of an ethyl acetate fraction (EAF) of S. asper on nitric oxide (NO) production and prostaglandin-E2 levels in lipopolysaccharide (LPS)-stimulated RAW264.7 macrophages. MATERIALS/METHODS: An in vitro culture of RAW264.7 macrophages was treated with LPS to induce inflammation. RESULTS: Treatment with EAF resulted in significant suppression of oxidative stress in RAW264.7 macrophages as demonstrated by increased endogenous superoxide dismutase (SOD) activity and intracellular glutathione levels, decreased generation of reactive oxygen species and lipid peroxidation, and restoration of the mitochondrial membrane potential. To confirm its anti-inflammatory effects, analysis of expression of inducible NO synthase, cyclooxygenase-2, tumor necrosis factor-${\alpha}$, and the anti-inflammatory cytokines IL-$1{\beta}$ and IL-6 was performed using semi-quantitative RT-PCR. EAF treatment resulted in significantly reduced dose-dependent expression of all of these factors, and enhanced expression of the antioxidants MnSOD and heme oxygenase-1. In addition, HPLC fingerprint results suggest that rutin, caffeic acid, and quercetin may be the active ingredients in EAF. CONCLUSIONS: Taken together, findings of this study imply that the anti-inflammatory effect of EAF on LPS-stimulated RAW264.7 cells is mediated by suppression of oxidative stress.
Objective : The purpose of this study is to investigate the anti-oxidative effect of Jeokbaekhaogwanjoong-tang (JGT) distillate in spleen cells of aged rats.Methods : This experiment was performed using spleen cells of 10w, 52w, 72w old sprague dawley (SD) rats. The spleen cells of 10, 52, 72 weeks old rats were divided into three groups; DW group, Vit. C group, and JGT group. The cells of DW group were treated with distilled water. The cells of Vit.C group were treated with Vitamin C. And the cells of JGT group were treated with JGT distillate. After the treatment of DW, Vit.C, and JGT, the levels of superoxide dismutase (SOD), glutathione (GSH), nitric oxide (NO) and lipid peroxidation (MDA) were measured.Results : In the spleen cells of 72 weeks old rats, SOD activity was significantly increased in the JGT group compared to those in the DW group and the Vit.C group. In the spleen cells of 52 weeks old rats, GSH activity was significantly increased in the JGT group compared to the Vit.C group, and NO concentration was significantly decreased in the JGT group compared to the DW group and the Vit.C group. MDA concentration was not changed significantly in any cell groups by the treatment of JGT distillate.Conclusions : Jeokbaekhaogwanjoong-tang (JGT) distillate may have an anti-oxidative effects in the spleen cells of aged rats. Further investigation is needed for the anti-aging effect of JGT.
Phytochemical examination of the fruits of Actinidia arguta has led to the isolation three organic acids, one coumarin, and three flavonoids. Structures of these compounds were elucidated as protocatechuic acid (1), caffeic acid (2), $caffeoyl-\beta-D-glucopyranoside$ (3), esculetin (4), quercetin (5), quercetin $3-O-\beta-D-galactopyranoside$ (6), and quercetin $3-O-\alpha-L-rhamnopyranosyl$$(1\rightarrow6)-O-\beta-D-glucopyranoside$ (7) by comparisons with previously reported spectral data. To investigate the anti-inflammatory and anti-oxidative effects of these compounds, nitric oxide production inhibitory activity in LPS-stimulated RAW 264.7 cells and DPPH radical scavenging activities were examined. Nitric oxide productions were reduced significantly by the addition of compounds $[1(IC_{50}=59.27{\mu}g/ml),\;2(IC_{50}=27.95{\mu}g/ml),\;3(IC_{50}=73.09{\mu}g/ml),\;4(IC_{50}=67.44{\mu}g/ml),\;5(IC_{50}=17.40{\mu}g/ml),\;6(IC_{50}=41.99{\mu}g/ml),\;7(IC_{50}=54.46{\mu}g/ml)]$, and extracts $(IC_{50}=56.21{\mu}g/ml)$ compared with positive control, L-NMMA $(IC_{50}=14.48{\mu}g/ml)$. The phenolic compounds also showed anti-oxidative activities. Especially, Compounds $1(IC_{50}=8.87{\mu}g/ml),\;4(IC_{50}=3.41{\mu}g/ml),\;and\;5(IC_{50}=6.06{\mu}g/ml),\;$, showed potent anti-oxidative activities similar to L-ascorbic acid $(IC_{50}=5.89{\mu}g/ml)$.
Dandelions have been reported to have medicinal properties and bioactive components that impact human health. However, the precise biological properties of dandelions and the parts of the plants possessing bioactive components remain uncertain. In this study, we evaluated 3 different types of dandelions based on their cultivation origin (Songpa, Uiryung, and native Uiryung types) as well as their 4 different plant parts (leaf, flower, root, skin). Each sample was extracted with $80\%$ methanol and then compared for the biological activities (anti-oxidative, immune cell proliferative and tumor cell growth inhibitory activities). All 3 types of dandelions possessed a degree of biological functions including the hydroxyl radical scavenger activity, immune cell proliferative activity and tumor cell growth inhibitory activity. However, there was no significant difference in these activities between the 3 dandelion types. Leaves of all three dandelion types showed the highest levels of all biological activities. To a lesser degree, the flower and root parts displayed biological activities. In the skin parts, anti-oxidative activity was also detected only at higher doses of dandelion extracts. Heating the dandelion leaf extract did not affect the biological activity, suggesting a heat-stable nature of the biological compounds. Taken together, these collective data suggest that dandelions, in particular their leaves, possess a high concentration of heat-resistant biological compounds, which are responsible for anti-oxidative, immune cell proliferative and tumor cell growth-inhibitory activities.
Objectives : Recent etiological studies show that oxidative stress might play a major role in the diabetes and its complications. Mori Ramulus (MR) has been known to have antioxidative, anti-inflammatory and antidiabetic effects. The methanol extract of MR was tested for its effectiveness in LLC-PK1 cells exposed to high glucose. Methods : The cytoprotective effect of MR was evaluated by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay. The antioxidative effect was measured in terms of generation amount of ${\cdot}O_2^-$ by 2',7'-dichlorodihydrofluorescein diacetate (DCFDA), NO by 4,5-diaminofluorescein (DAF-2), $ONOO^-$ by dihydrorhodamine 123 (DHR 123) in the high glucose -treated LLC-$PK_1$ cells. Western blotting was performed using anti-AGE, anti-RAGE, anti-MAPKs(ERK1/2, JNK, p38), anti-PI3K, anti-Akt, and anti-NF-${\kappa}$B (p50, p65) respectively. Results : MR extract reduced cell death and inhibited the generation of ${\cdot}O_2^-$, NO, $ONOO^-$ in the high glucose-treated LLC-$PK_1$ cells. MR inhibited the expression of AGE, RAGE, MAPKs, PI3K, and Akt by means of decreasing NF-${\kappa}$B activation. MR also inhibited NF-${\kappa}$B activation itself. Conclusions : These results indicate MR has cytoprotective, antioxidative, and anti-inflammatory effects. Therefore it is suggested that MR might prevent and cure diabetes and its complications.
본 연구에서는 잣피 열수, 20% 주정, 50% 주정 추출물들이 항염증과 항산화에 미치는 영향을 확인하기 위하여 ABTS 항산화능과 SOD, CAT, GPx 항산화 효소, NO의 분비량, 염증성 cytokine의 분비량을 살펴보고자 하였다. 잣피 열수, 20% 주정, 50% 주정 추출물 중 20% 주정 추출물이 항산화능, 항산화 효소 활성 증가, NO의 분비량 감소, 염증성 cytokine 분비량을 감소시켜 염증반응과 항산화 반응에 도움을 주어 내부에서 생성된 활성산소종과 외부로부터 침입한 미생물, 감염된 세포나 종양세포 등을 효과적으로 제거할 수 있을 것이라 예상할 수 있었다. 이는 염증뿐만 아니라 면역반응에서도 영향을 미칠 것이라 생각되며 염증조절 및 항산화 반응에 긍정적인 변화를 보였으므로 추후 염증 조절제로서 기능성 식품의 상업화에 기초 자료가 되어 국내 기능성 소재로서의 개발 가능성을 기대할 수 있다.
This study was performed to investigate the effects of treatment for dermatitis using the herbal gel preparations. Scutellariae Radix(SR) and Zingiberis Rhizoma(ZR) were used for the purpose of herbal preparations. Baicalin, baicalein are the ingredients of SR, having biological effects like anti-inflammatory, anti-oxidative, anti-bacterial and antiallergic action. 6-Gingerol is one of the ingredients of ZR having biologicai effects like anti-inflammatory and analgesic action. The three types of hydrogels(SRE, SRH, SZH) were formulated with Carbopol 940, Labrasol, Triethanolamine etc. Baicalin was hydrolysed to baicalein by $\beta$-glucuronidase for the purpose to increase rate of skin permeation. Content of ingredients by HPLC determination, rate of skin permeation using Franz type diffusion cell, anti-oxidative activity for the free radical, hydroxyl radical, superoxide, anti-inflammatory by using carrageenan injection, efficacy on the dermatitis induced by 2,4-dinitro-chlorobenzene(DNCB) were experimented. Baicalein showed higher permeability than baicalin, so it is considered that baicalein was more suitable form than baicalin for transdermal absorption by its lipophilic property. In the anti-oxidative experiments, SZH gel was the most effective scavenging activity than the other gels in all experiments. In anti-inflammatory test, SRM and SZH gel more decreased edma rapidly than SRE gel. In case of using SZH gel, treatment period for the dermatitis was more declined than that of other gel groups. These results suggests that the SZH hydrogel could be suitable preparations for the treatment of dermatitis.
There is currently increased interest in the identification of antioxidant compounds that are pharmacologically potent and have low or no side effects. Plants produce significant amounts of antioxidants to prevent the oxidative stress caused by photons and oxygen, therefore they represent a potential source of new compounds with antioxidant activity. FARFARE FLOS has been frequently used on the respiratory system including bronchitis, phthisis. In this study, the antioxidant activity of extract from FF was studied in vitro methods by measuring the antioxidant activity by TEAC, measuring the scavenging effects on reactive oxygen species (ROS) [superoxide anion, hydroxyl radical] and on reactive nitrogen species (RNS) [nitric oxide and peroxynitrite] as well as measuring the inhibitory effect on Cu2+-induced human LDL oxidation. The FF extracts were found to have a potent scavenging activity, as well as an inhibitory effect on LDL oxidation. And this study was designed to evaluate whether FFEA may ameliorate oxidative stress and inflammatory status through the antioxidative mechanism in LPS-stimulated RAW 264.7 murine macrophage cell line. Treatment of RAW 264.7 cells with FFEA significantly reduced LPS-stimulated inflammatory response in a dose-dependent manner. In conclusion, the FF extracts have anti-oxidative and anti-inflammatory effects in vitro system, which can be used for developing pharmaceutical drug against oxidative stress and atherosclerosis.
Portulaca oleracea L. is known to have many biological benefits such as anti-oxidant, anti-inflammatory, anti-allergic and anti-tumor. The objective of this study is to explore the neuroprotective effect of P. oleracea L. against glutamate-induced oxidative stress in mouse hippocampal HT22 cells. P. oleracea L. 70% ethanol extract and solvent fractions have the potent neroprotective effects on glutamate-induced nerotoxicity by induced the expression of heme oxygenase (HO)-1 in HT22 cells. Especially, ethyl acetate fraction showed higher protective effect. In HT22 cell, P. oleracea L. treatment with ERK inhibitor (PD98059) and c-JUN N-terminal kinase (JNK) inhibitor (SP600125) reduced P. oleracea L. ethyl acetate fraction induced HO-1 expression and P. oleracea L. ethyl acetate fraction also increased ERK and JNK phosphorylation. Furthermore, we found that treatment of P. oleracea L. caused the nuclear accumulation of Nrf2. In conclusion, the ethyl acetate fraction of 70% ethanol extract of P. oleracea L. significantly protect glutamate-induced oxidative damage by induction of HO-1 via Nrf2, ERK and JNK pathway in mouse hippocampal HT22. Taken together these finding suggest that P. oleracea L. ethyl acetate fraction is good source for taking active compounds and may be a potential therapeutic agent for brain disorder that induced by oxidative stress and neuronal damage.
