Comparative study on Hsp25 expression in Mongolian gerbil and mouse cerebellum

  • The term 'heat shock protein (Hsps)' was derived from the fact that these proteins were initially discovered to be induced by hyperthermic conditions. In response to a range of stressful stimuli, including hyperthermia, immobilization, UV radiation, amino acid analogues, arsenite, various chemicals, and drugs the mammalian brain demonstrates a rapid and intense induction of the heat shock protein. Moreover, Hsps were expressed on the various pathological conditions including trauma, focal or global ischemia, hypoxia, infarction, infections, starvation, and anoxia. Especially, Hsp25 has a protective activity, facilitated by the ability of the protein to decrease the intracellular levels of reactive oxygen species (ROS) as well as its chaperone activity, which favors the degradation of oxidized proteins. Recently, it has clearly demonstrated that Hsp25 is constitutively expressed in the adult mouse cerebellum by parasagittal bands of purkinje cells in three distinct regions, the central zone (lobule VI-VII) and nodular zone (lobule IX-X), and paraflocculus. The Mongolian gerbil has been introduced into stroke study model because of its unique brain vasculature. There are no significant connections between the basilarvertebral system and the carotid system. This anatomy feature renders the mongolian gerbil susceptible to forebrain ischemia-induced seizure. The present study is designed to examine the pattern of Hsp25 expression in the cerebellum of this animal in comparison with that in mouse.

  • Deafness has been considered in recent publications a transient feature in the neurological complications of carbon monoxide poisoning. The inner ear is a sensitive as central nervous system to anoxia. It is well known a similar vulnerability to the toxic action of carbon monoxide for the two neurosensory epithelia of the organ of Corti. There is geographical character in the carbon monoxide poisoning and more frequently experienced in Korea. 10 cases of hearing loss due to carbon monoxide intoxication were reported with the literature review.

A report on the mass summer mortalities of the farmed Pacific oysters, Crassostrea gigas and Bay scallops Argopecten irradians in the local waters of Goseong Bay, Korea

  • Mass mortalities of farmed shellfish, mostly in summer season, thus named mass summer mortalities, have been a global issue in shellfish aquaculture. The 2013 mass summer mortalities in the confined waters of Goseong Bay, Goseong, Korea were quite a unique and intensive for two farmed species, the Pacific oysters, Crassostrea gigas, and bay scallops, Argopecten irradians. The mortalities were progressive from the bottom of the suspended oysters and caged scallops in the waters, reaching up to 80% for the oyster and 95% for the scallop in about 20 days after the first occurrence, early August, 2013. We monitored a wide range of environmental factors, including water temperature, dissolved oxygen (DO), salinity, turbidity, acidity (pH), organic and inorganic matters, chemical oxygen demand (COD), suspected pathogenic agent, and phytoplankton composition throughout the water column where the two species were suspended or caged. Our survey concluded that the hypoxia or anoxia might be a major cause of the mortalities. Here, we detailed the mortalities and ways to arrive at the conclusion.

Neuroprotective Effects of Daebowonjeon on PC12 Cells Exposed to Ischemia (허혈 상태의 PC12 세포에 대한 대보원전(大補元煎)의 신경보호효과)

  • Neuronal ischemia is a pathological process caused by a lack of oxygen (anoxia) and glucose (hypoglycemia), resulting in neuronal death. It is believed that apoptosis is one of the mechanisms involved in ischemic cell death. Neuronal apoptosis is a process characterized by nuclear DNA fragmentation, changes of plasma membrane organization. To elucidate the mechanism of neuronal death following ischemic insult and to develop neuroprotective effects of Daebowonjeon(DBWJ) against ischemic damage, in vitro models are used. In vitro models of cell death have been devloped with pheochromocytoma (PC12) cell, which have become widely used as neuronal models of oxidative stress, trophic factor, serum deprivation and chemical hypoxia. Using a special ischemic device and PC12 cultures, we investigated an in vitro model of ischemia based on combined Oxygen and Glucose Deprivation (OGD) insult, followed by reoxygenation, mimicking the pathological conditions of ischemia. In this study, Daebowonjeon rescued PC12 cells from Oxygen-Glucose Deprivation (OGD)-induced cell death in a dose-dependent manner The nuclear staining of PC12 cells clearly showed that DBWJ attenuated nuclear condensation and fragmentation which represent typical neuronal apoptotic characteristics. DBWJ also prevents the LDH release and induction of Hypoxia Inducing Factor (HIF)-1 by OGD-exposed PC12 cells. Furthermore, DBWJ reduced the activation of polyADP-ribose polymerase (PARP) by OGO-exposed PC12 cells. These results suggest that apoptosis is an important characteristic of OGD-induced neuronal death and that oriental medicine, such as DBWJ, may prevent PC12 cell from OG D-induced neuronal death by inhibiting the apoptotic process.

A Study of Transient Estuarine Circulation in the Chunsu Bay, Yellow Sea: Impact of Freshwater Discharge by Artificial Dikes

  • This study examined the ef ects of freshwater discharge by artificial dikes from the Kanwol and Bunam lakes on the dynamics in the Chunsu Bay, Yellow Sea, Korea, during the summer season based on three-dimensional numerical modeling experiments. Model performances were evaluated in terms of skill scores for tidal elevation, velocity, temperature, and salinity and these scores mostly exceeded 90 %. The variability in residual currents before and after the freshwater discharge was examined. The large amount of lake water discharge through artificial dikes may result in a dramatically changed density field in the Chunsu Bay, leading to an estuarine circulation system. The density-driven current formed as a result of the freshwater inflow through the artificial dikes (Kanwol/Bunam) caused a partial change in the tidal circulation and a change in the scale and location of paired residual eddies. The stratification formed by strengthened static stability following the freshwater discharge led to a dramatic increase in the Richardson number and lasted for a few weeks. The strong stratification suppressed the vertical flux and inhibited surface aerated water mixing with bottom water. This phenomenon would have direct and indirect impacts on the marine environment such as hypoxia/anoxia formation at the bottom.

Clinically Correlated Anatomical Basis of Cricothyrotomy and Tracheostomy

  • Objective : Cricothyrotomy and tracheostomy are performed by physicians in various disciplines. It is important to know the comprehensive anatomy of the laryngotracheal region. Hemorrhage, esophageal injury, recurrent laryngeal nerve injury, pneumothorax, hemothorax, false passage of the tube and tracheal stenosis after decannulation are well known complications of the cricothyrotomy and tracheostomy. Cricothyrotomy and tracheostomy should be performed without complications and as quickly as possible with regards the patients' clinical condition. Methods : A total of 40 cadaver necks were dissected in this study. The trachea and larynx and the relationship between the trachea and larynx and the surrounding structures was investigated. The tracheal cartilages and annular ligaments were counted and the relationship between tracheal cartilages and the thyroid gland and vascular structures was investigated. We performed cricothyrotomy and tracheostomy in eleven cadavers while simulating intensive care unit conditions to determine the duration of those procedures. Results : There were 11 tracheal cartilages and 10 annular ligaments between the cricoid cartilage and sternal notch. The average length of trachea between the cricoid cartilage and the suprasternal notch was 6.9 to 8.2 cm. The cricothyroid muscle and cricothyroid ligament were observed and dissected and no vital anatomic structure detected. The average length and width of the cricothyroid ligament was 8 to 12 mm and 8 to 10 mm, respectively. There was a statistically significant difference between the surgical time required for cricothyrotomy and tracheostomy (p < 0.0001). Conclusion : Tracheostomy and cricothyrotomy have a low complication rate if the person performing the procedure has thorough knowledge of the neck anatomy. The choice of tracheostomy or cricothyrotomy to establish an airway depends on the patients' clinical condition, for instance; cricothyrotomy should be preferred in patients with cervicothoracal injury or dislocation who suffer from respiratory dysfunction. Furthermore; if a patient is under risk of hypoxia or anoxia due to a difficult airway, cricothyrotomy should be preferred rather than tracheostomy.

Species Composition and Occurrence Patterns of Zooplankton in Jinhae Bay (진해만에 출현하는 동물플랑크톤의 종조성과 계절별 출현양상)

  • The seasonal distribution pattern of zooplankton were studied on the basis of the zooplankton samples collected from 9 stations in Jinhae Bay in May, August, November and February, 2001 to 2002. A total of 31 species in 37 taxa was occurred in Jinhae Bay. Copepods were pyedominant through fall to spring and cladecerans in summer. There aye high spatio-temporal fluctuations in the zooplankton abundance in a range of 27 to 28,221 indiv. $m^{-3}$. Of these, an neritic species, Acartia omorri and Oentropages abdominals were predominent in february; Palaealanus parvus s. 1 in November; Penilia avirostris in August. Species diversity was low in the northwestern regims where anoxia layer is occurred in summer, while in the other seasons there was no a great difference between stations. It indicates that the distribution pattern of zooplankton may be seasonally strongly affected by a mechanism of hypoxia formation in Jinhae Bay.

HSP27 is Commonly Expressed in Cervical Intraepithelial Lesions of Brazilian Women

  • Heat shock proteins are molecular chaperones that may be constitutively present in cells protecting them from various stresses, such as extreme temperature, anoxia or chemical agents. Cervical cancer is the second most prevalent malignancy of women. In this study, we analyzed the expression of Hsp27 by immunohistochemistry in cervical intraepithelial lesions of Brazilian women, along with samples from non neoplasic lesions (NN). Cervical intraepithelial neoplasia I (CIN I), II (CIN II) and III (CIN III)/in situ carcinoma and squamous cell carcinoma (SCC) were included. Immunostaining was observed in 30 (100%) samples of NN, 46 (92%) in CIN I, 50 (100%) in CIN II, 52 (98.11%) in CIN III/CIS, and 46 (98.11%) in SCC. In group NN Hsp27 immunostaining was heterogeneous, more intense in basal and parabasal layers of the epithelium and less or absent in the intermediate and superficial layer. The majority of the samples of CIS and SCC presented strong staining in all epithelial layers. Metaplasic cells, when present, were strongly stained. In this study, Hsp27 protein was found to be commonly expressed in cervical epithelial cells.

Pharmacokinetics and Blood-Brain Barrier Permeability of Taurine in Spontaneously Hypertensive Rats and Normotensive Rats (자연발생 고혈압 흰쥐와 정상흰쥐 데서 타우린의 체내동태 및 뇌투과성)

  • Taurine, 2-aminoethanesulfonic acid is widely distributed in animal tissues and has a variety of bio-logical activities. A recent worldwide study demonstrated beneficial effects of taurine on aging and age-associated disorders. In general, taurine levels in the brain decease when an animal is subjected to pathologic conditions such as ischemia-anoxia and seizure. But the taurine levles tend to increase in the brain in hypertensive state. In the present study, the blood-brain barrier (BBB) transport of [$^3$H]taurine was compared between spontaneously hypertensive rats (SHR) and normotensive Sprague-Dawley rats (SD) using intravenous injection technique in vivo. We also obtained pharmacokinetic parameters of plasma volume maker, [$^{14}$ C] sucrose and [$^3$H]taurine after inject to rats simulatenously. BBB permeability surface area product (PS) value of [$^3$H]taurine in SHR (16$\pm$2.9$\times$10$^{-3}$ ml/min/g) was significantly higher than that in SD (7.4$\pm$0.8$\times$10$^{-3}$ ml/min/g). There is also significant difference for brain uptake of [$^3$H]taurine between SHR (0.195$\pm$0.031%ID/g) and SD (0.058$\pm$0.003% ID/g). This is due to difference of area under the plasma concentration-time curve (AUC) and that of total clearance (Class) between SHR and SD. No significant difference was indicated from other organ uptakes such as lung, heart, liver SHR and SD. But also kidney uptake was much higher in SHR. In conclusion, [$^3$H]taurine in plasma was slowly eliminated in SHR than in SD and uptake of [$^3$H]taurine in SHR is much higher than that of SD. This results suggest increased taurine level in the brain in hypertension state have an any effect on the brain uptake of taurine.

Calculated V$\_$D/, volume of distribution in brain, and PS, the permeability surface area product of [$^3$H]taurine through the BBB in SHR was a little lower than that in SD. PS for 15s is more higher than that of other seconds in both of them. It could be followed by taurine efflux back into blood after 15s. We also obtained pharmacokinetic parameters using intravenous injection of plasma volume marker, [$\^$14/C]sucrose and [$^3$H] taurine. PS value of [$^3$H]taurine in SHR (16.1 ${\pm}$ 2.9 ${\times}$ 10$\^$-3/ $m\ell$/min/g) was significantly higher than that in SD (7.4 ${\pm}$ 0.8 ${\times}$ 10$\^$-3/ $m\ell$/min/g). There is also significant difference for %ID/g in brain between SHR (0.195 ${\pm}$ 0.031) and SD (0.058 ${\pm}$ 0.003).
