Basic Study for Development of Denitrogenation Process by ion Exchange(II) (이온교환법에 의한 탈질소 공정개발의 기초연구(II))

  • Ion exchange performance to remove nitrate in water was studied using commercially available strong base anion exchange resin of Cl- type in the batch and continuous column reactors. The performance was tested using the effluent concentration histories for continuous column or equilibrium conquilibrium between resin and solution. Anion exchange resin used in this study was more effective than activated carbon or zeolite for nitrate removal. With large resin amount or low initial concentration, nitrate removal characteristics for a typical gel-type resin was Increased. On considering the relation between the breakthrough capacity and nitrate concentration of the influent, the use of anion exchange resin were suitable for the hi선or order water treatment. The nitrate removal of above 90% could be possible until the effluent of above 650 BV was passed to the column. Thus, the commercially available strong base anion exchange resin of $Cl^-$ type used in thins study could be effectively used as economic material for treatment of the groundwater. The breakthrough curves showed the sequence of resin selectivity as $SO_4^{2-}$ > $NO_3$ > $NO^{2-}$ > $HCO_3^-$. The results of this study could be scaled up and used as a design tool for the water purification system of the real groundwater and surface water treatment processes.

Purification of Odontoglossum Ringspot Virus by DEAE-Cellulose Chromatography (DEAE 셀루로오즈 컬럼 크로마토그래피 기법에 의한 Odontoglossum 윤문 바이러스의 정제)

  • Odontoglossum ringspot virus (ORSV) was finally purified from ORSV-infected orchid plants by diethylaminoethyl (DEAE) cellulose anion exchange column chromatography. The virus was reliably eluted by potassium chloride at the concentration from 0.1 M to 0.13 M. Partial purification was done by solubilization with Triton X-100 (allkylphenoxypolyethoxy ethanol) and precipitation with polyethylene glycol (PEG; MW 8,000). The finally purified ORSV represented one distinct homogeneous band and the molecular weight of its capsid protein was about 17,500 Dalton in electrophoretic analysis. Electron microscopy showed not only intact particles ranged from 280 nm to 340 nm in length, but also segmented particles that final 140 nm to 220 nm and even disks. Enzyme-linked immunosorbent assay (ELISA) showed that final yield was 12 mg/100 g of the infected leaves. Bioassay demonstrated that the purified ORSV had the normal infectivity to orchid plants and Nicotiana glutionsa. Based on these data, anion exchange column chromatography could be efficiently applied to the purification of ORSV and other viruses similar to ORSV.

  • Phytase(myo-inositol-hexakis phosphate 3-phosphohydrolase, E C 3.1.3.8) sequentially hydrolyzes phytate to myo-inositol and inorganic phosphate. Phytase of Aspergillus ficuum was purified to homogeneity using ultrafiltration, cation exchange column and anion exchange column. It's molecular weight is estimated as around 90,000 by SDS-PAGE. Antibody against the phytase was produced by immunizing mice with the purified phytase. The titer of the antibody was determined to be 1/25,000.

  • A kinetic study for nitrate removal by anion exchange resin was performed using continuous column reactors. Kinetic approach from the packed bed showed the reaction rate constant k$_1$ was 0.07~0.17 $\ell$/mgㆍhr and maximum exchange quantity q$_{o}$ was 27.75~31.81 mg/g. The results from the continuous column well agreed with that from the batch reactor. An economic analysis of the water treatment plant by anion exchange resin with a regenerating system was performed to design plant and process. Based on the treatment of 20 mg/$\ell$ nitrate-contained wastewater of 10,000 gallons per day to 2 mg/$\ell$, total capital cost and total annual cost are estimated to be 836 million wons and 211 million wons, respectively.y.

  • Ion exchange is a chemical reaction between the ions in solution phase and ions in solid phase and is widely used in softening, demineralization, removal and collection of specific ions, and ion migration in the ground water. The ion selectivity depends on the charge and the hydrated radius of ion. The objective of this study was to examine the applicability of anion selectivity obtained from the ion exchange equilibrium OH/sup -/ < F/sup -/ < HCO/sup -/ < Cl/sup -/ < Br/sup -/ ≤ NO₃/sup -/ < SO₄/sup 2-/ to the column ion exchange. The column ion exchange was facilitated in the lower charge of counter-ion in the background electrolyte.

  • A coupled column liquid chromatography system was applied for the separation of the burnup monitors in spent nuclear fuel sample solutions. A reversed phase column was studied for the adsorption behavior of uranyl ions using alpha-hydroxyisobutyric acid as an eluent and used for the separation of plutonium and uranium. A cation exchange column prepared by coating 1-eicosylsulfate onto the reversed phase column was used for the separation of the lanthanides. In addition, retention of Np was checked with the reversed phase column and cation exchange column, respectively, according to the oxidation states to observe the interference effect for the separation of burnup monitors. This chromatography system showed a great reduction in separation time compared to a conventional anion exchange method. A good agreement from the burnup data was obtained between for this method and a conventional anion exchange method to within 1% of a difference for the spent nuclear fuel samples of about 40 GWD/MTU.

  • Expression of Bacillus subtilis glutamyl-tRNA synthetase (GluRS) in Escherichia coli is lethal for the host, probably because this enzyme misaminoacylates ${tRNA_l}^{Gln}$ with glutamate in vivo. In order to overexpress B. subtilis GluRS, encoded by the gltX gene, in E. coli, this gene was amplified from B. subtilis 168 chromosomal DNA using PCR method and the entire coding region was cloned into a pET11a expression vector so that it was expressed under the control or the T7 Promoter. The resulting recombinant pEBER plasmid was transformed into E. coli Novablue (DE3) bearing the T7 RNA polymerase gene for expression. After IPTG treatment, the overproduced enzyme was purified using ammonium sulfate fractionation, Source Q anion exchange chromatography, Superdex-200 gel filtration, and Mono Q anion exchange chromatography. The purified enzyme yielded 18-fold increase in specific activity over the crude cell extract and its molecular weight was approximately 55 kDa on SDS-PAGE.

Decolorization of Aqueous Caprolactam Solution by Anion-exchange Resins

  • Caprolactam is the most important raw material for making Nylon 6 fibers and its quality directly determines the quality of Nylon. So it is necessary to study the techniques and methods to remove the colorful impurities from caprolactam. In this paper, the decolorization of caprolactam aqueous solution by anion exchange resins was studied and the decoloring abilities of five commercial resins were investigated. The regeneration of the resins was also studied, too. This study shows that the resin AMTX202 have excellent decoloring ability in the column experiment and that the decoloring efficiency is correlated with the volume of resins packed and is slightly affected by the flow rate and regenerating times. The fact that the resins can be regenerated and reused without affecting the efficiency of decolorization will decrease the cost of the treatment and operation in the industry. The adsorption of colored compounds with anion exchange resins in the packed columns seems to be technically feasible.

Purification and Characterization of Polyphenol Oxidase from Lotus Root (Nelumbo nucifera G.) (연근의 polyphenol oxidase 정제 및 특성조사)

  • Polyphenol oxidase isoforms were purified from the lotus roots using 50% acetone precipitation, conventional chromatographies of Q-Sepharose and hydrophobic interaction, and high performance liquid chromatographies of Mono-Q and Superdex 75 gel-filtration. Molecular mass of a purified PPO isoform (LPIII-2) was determined to be 56 kDa using gel-filtration chromatography. The active form of LPIII-2 appeared to bea heterodimer, as purified LPIII-2 on SDS-PAGE gel showed two bands that were determined to be 28 kDa and 26 kDa. To further characterize PPO, partially purified PPO isoforms (LP-II, LP-III) were obtained from Q-Sepharose anion-exchange chromatography. In substrate specificity, the partially purified PPO isoform LP-II showed a high affinity to catechol, while LP-III showed a high affinity to pyrogallol. The optimum pH of LP-II and LP-III was pH 7.0. Interestingly, the partially purified PPO isoforms showed high activities at low temperatures $(0{\sim}5^{\circ}C)$, and as temperatures rose, the activities decreased. Both PPO isoforms were stable at $40^{\circ}C$ and were inactivated by incubation at $60^{\circ}C$ for 40 min.

  • An accurate and simple analytical technique for low levels of fallout Pu in the environment was developed using an anion exchange resin. To develop the reliable determination of Pu isotopes in soil samples, decomposition of soil matrix, plutonium oxidation state adjustment on the anion exchange column and source preparation of Pu isotopes have been carried out. The optimum method of Pu isotopes with an anion exchange has been validated by application to IAEA-Reference soils.
