알톨 대사에 미치는 인삼의 영향 (Effect of Ginseng on the Alcohol Metabolism in Alcohol Treated Rat)

  • After pretreatment with ginseng followed by induction of acute intoxication of alcohol, the activities of alcohol dehydrogenase (ADH), microsomal ethanol-oxidizing system (MEOS) and aldehyde dehydrogenase(Ald DH) increased respectively compared to the groups treated with alcohol alone. In case that ginseng was given to rats fed with 5% alcohol instead of water for 60 days, the activities of ADH and MEOS increased compared to the groups treated. On the contrary, the activity of Ald DH in mitochondrial fraction decreased to an extent of about 35% in chronic alcoholism, but after pretreatment of ginseng the activity was restored to the control level. On the other hand, the catalase activity was not significantly affected by either treatment. Ginseng butanol fraction significantly increased the serum isocitrate dehydrogenase activity which is inhibited by alcohol-treated in rat. Alcohol-induced lactate dehydrogenase activity was decreased to control level in liver by ginseng treatment. And the serum level of lactic acid also decreased by ginseng treatment in alcohol-intoxicated rat. Ginseng butanol fraction markedly decreased the xanthine oxidase activity in the ethanol-treated rat liver. It was also observed that ginseng reduced the blood concentration of uric acid on experimentally reduced hyperuricemia by alcohol treatment. Uricase activity was not affected by either treatment. Ginseng butanol fraction decreased the hepatic aniline hydroxylase activity which was induced by alcohol-treated rat. These results suggest that the treatment with ginseng can be promoted the recovery from alcohol intoxication and some therapeutic effect on alcoholinduced metabolic disease.

Effects of acrylamide in the presence of vitamin E on sperm parameters, chromatin quality, and testosterone levels in mice

  • Objective: The present study investigated sperm chromatin quality and testosterone levels in acrylamide-treated mice and the possible protective effects of vitamin E on the fertility potential of spermatozoa. Methods: Thirty-two adult male mice were divided equally into four groups. Group 1 was the control, group 2 received acrylamide (10 mg/kg, water solution), group 3 received vitamin E (100 mg/kg, intraperitoneal), and group 4 received both acrylamide and vitamin E. After 35 days, spermatozoa from the right cauda epididymis were analyzed in terms of count, motility, morphology, and viability. Sperm DNA integrity and chromatin condensation were assessed by acridine orange (AO), aniline blue (AB), toluidine blue (TB), and chromomycin A3 (CMA3) staining. Results: In acrylamide-treated mice, significantly lower sperm concentration, viability, motility, and testosterone levels were found in comparison with the control and acrylamide+vitamin E groups (p< 0.05). In the vitamin E group, significantly more favorable sperm parameters and testosterone levels were found than in the other groups (p< 0.05). There were also significantly more spermatozoa with less condensed chromatin in the acrylamide-treated mice than in the other groups. Moreover, significantly more spermatozoa with mature nuclei (assessed by AB, CMA3, AO, and TB staining) were present in the vitamin E group than in the control and acrylamide+vitamin E groups. Conclusion: This study revealed the deleterious effects of acrylamide on sperm parameters and sperm chromatin quality. Vitamin E can not only compensate for the toxic effects of acrylamide, but also improve sperm chromatin quality in mice.

고혈당(高血糖) 쥐의 간(肝) 대사효소계(代謝酵素系)에 미치는 생진양혈탕(生津養血湯)의 영향(影響) (Effect of SAENGCHINYANGHYOLTANG on the hepatic metabolic enzyme system in streptozotocin-induced diabetic rats)

  • SANGNYANGHYOLTANG(SYT) is one of the most important prescription that has been used in oriental medicine for diabetes mellitus. The sudy was done in order to elucidate the anti-diabetic effect of SYT. After pretreatment of SYT(1,000mg/kg) for 6 weeks, the effect of of SYT was prevented on serum liver function test and hepatic lipid peroxide content in rats i.v. injected with streptozotocin(STZ, 50mg/kg, tail vein) 5 weeks after pretreatment of SYT. The hepatic microsomal cytochrome P-450 and aniline hydroxylase were significantly decreased, and aminopyrine N-demethylase activity was significantly increased in SYT-STZ group as compared with control group. Changes in aldehyde oxidase, xanthine oxidase, superoxide dismutase, catalase, epoxide hydrolase, UDP-glucuronyltransferase and sulfotransferase activities were not significantly different in any of the group. The cytosolic glutathione S-transferase activity was significantly decreased in SYT-STZ group as compared with control group. The selenium-independent glutathione peroxidase was significantly increased in SYT-STZ group as compared with control group, but there was no significant difference in selenium-dependent glutathione peroxidase in any of the groups. The hepatic glutathione concentration was significantly increased in SYT-STZ group as compared with control group, and ${\gamma}-glutamylcystein$ synthetase and glutathione reductase activities were not significantly different in any of the groups. The hepatic lipid peroxide content, serum aminotransferase and sorbitol dehydrogenase activities were slightly decreased in significantly in SYT-STZ groups.

간장내 허혈 및 재관류시 약물대사 효소계의 지질 과산화에 관한 연구 (Lipid Peroxidation of Hepatic Microsomal Drug-Metabolizing System in Hepatic Ischemia ands Reperfusion)

  • This study was done to determine whether specific alterations exist in hepatic microsomal function after varying periods of ischemia (IS) and reperfusion (RP) during microsomal lipid peroxidation occurs. Rats were pretreated with $\alpha$-tocopherol to inhibit lipid peroxidation or with vehicle (soybean oil). Control animals were time-matched sham-ischemic animals. Four groups of animals were studied: Group 1 (sham), group 2 (30 mins IS), group 3 (60 mins IS) and group 4 (90 mins IS). After 1, 5 or 24 hr of reperfusion, liver microsomes were isolated and cytochrome P-450s were studied. In all vehicle-treated ischemic rats, serum ALT levels peaked at 5 hr and were significantly reduced by $\alpha$-tocopherol pretreatment. Similarly, microsomal lipid peroxidation was elevated in all vehicle-treated ischemic animal groups, but this elevation was prevented by $\alpha$-tocopherol pretreatment. Cytochrome P-450 content was significantly decreased in both group 3 and group 4. In all vehicle-treated ischemic animal groups, aminopyrine N-demethylase activity was significantly decreased for the entire reperfusion period. $\alpha$-Tocopherol inhibited reductions of cytochrome P-450 content and aminopyrine N-demethylase activity at both 1 hr and 5hr of reperfusion but did not affect the reduced levels of cytochrome P-450 content and aminopyrine N-demethylase activity at 24 hr of reperfusion. Aniline p-hydroxylase activity was significantly decreased in group 4, whereas it was increased in group 3. These decreases and increases were prevented by $\alpha$-tocopherol pretreatment. Our finding suggests that abnormalities in microsomal drug metabolizing function occur during hepatic ischemia and reperfusion in vivo and this is attributed to microsomal lipid peroxidation.

Protective effects of curcumin on chromatin quality, sperm parameters, and apoptosis following testicular torsion-detorsion in mice

  • Objective: The chief outcome of testicular torsion in clinical and experimental contexts is testicular ischemia. Curcumin, a compound with anti-inflammatory and antioxidant properties, has fascinated researchers and clinicians for its promise in the treatment of fertility diseases. Methods: Thirty-five fully grown male mice were randomly classified into five groups: control, sham, testicular torsion, treatment group 1 (testicular torsion+short-term curcumin), and treatment group 2 (testicular torsion+long-term curcumin). Thirty-five days later, spermatozoa from the right cauda epididymis were analyzed with regard to count and motility. Toluidine blue (TB), aniline blue (AB), and chromomycin A3 (CMA3) staining assays were used to evaluate the sperm chromatin integrity. In addition, the terminal deoxynucleotidyl transferase-mediated deoxyuridine triphosphate nick-end labeling (TUNEL) test was used to assess apoptosis. Results: Treatment group 1 exhibited a remarkably elevated sperm count compared to the testicular torsion group. Additionally, notably lower sperm motility was found in the testicular torsion group compared to the control, treatment 1, and treatment 2 groups. Staining (CMA3, AB, and TB) and the TUNEL test indicated significantly greater testicular torsion in the torsion group compared to the control group (p<0.05). The data also revealed notably lower results of all sperm chromatin assays and lower apoptosis in both treatment groups relative to the testicular torsion group (p<0.05). Significantly elevated (p<0.05) AB and TB results were noted in treatment group 1 compared to treatment group 2. Conclusion: Curcumin can compensate for the harmful effects of testicular ischemia and improve sperm chromatin quality in mice.

Trolox C Ameliorates Hepatic Drug Metabolizing Dysfunction After Ischemia/Reperfusion

  • The present study was done to determine the effect of trolox C, a hydrophilic analogue of vitamin E, on hepatic injury, especially the alteration in cytochrome P-450 (CYP)-dependent drug metabolism during ischemia and reperfusion (I/R). Rats were subjected to 60 min of hepatic ischemia and 5 h of reperfusion. Rats were treated intravenously with trolox C (2.5 mg/kg) or vehicle (PBS, pH 7.4), 5 min before reperfusion. Serum alanine aminotransferase and lipid peroxidation levels were markedly increased after I/R. This increase was significantly suppressed by trolox C. Cytochrome P-450 content was decreased after I/R but was restored by trolox C. There were no significant differences in ethoxyresorufin O-dealkylase (CYP 1A1) and methoxyresorufin O-dealkylase (CYP 1A2) activities among any of the experimental groups. Pentoxyresorufin O-dealkylase (CYP 2B1) activity was decreased and aniline p-hydroxylase (CYP 2E1) activity was increased after I/R. Both these changes were prevented by trolox C. Our findings suggest that trolox C reduces hepatocellular damage as indicated by abnormalities in microsomal drug-metabolizing function during I/R, and that this protection is, in part, caused by decreased lipid peroxidation.

Synthesis and characterization of star-shaped imide compounds

  • Novel star-shaped imide compounds containing electron-donating triphenylamine and/or electron-withdrawing bis(trifluoromethyl)phenyl side groups were synthesized via a two-step process. First, 3,6-dibromo-benzene-1,2,4,5-tetracarboxylic acid (2B4BA) was reacted with 4-aminophenyl (diphenylamine) (ATPA) or 3,5-bis(trifluoromethyl)aniline (6FA) by imide reaction. Then, Suzuki coupling reaction was carried out on these compounds with 4-(N,N-diphenylamino)-1-phenyl boronic acid (BTPA) or 3,5-bis(trifluoromethyl)phenyl boronic acid (6FBB), resulting in 3,6-bis[4-(diphenylamino)phenyl]-N,N'-bis[4-(diphenylamino) phenyl]-pyromellitimide (TPTPPI), 3,6-bis[3,5-bis(trifluoro methyl) phenyl]-N,N'-bis[3,5-bis(trifluoromethyl) phenyl]-pyro mellitimide (6F6FPI) or 3,6-bis[4-(diphenylamino)phenyl]-N,N'-bis[3,5-bistrifluoromethyl)phenyl]-pyromellitimide (6FTPPI). The imide compounds obtained were characterized by NMR, FT-IR, DSC, TGA, melting point analyzer, EA, and solubility measurements. In addition, their optical and electrical properties were evaluated by fluorescence spectroscopy, UV-vis spectroscopy, and cyclic voltammetry (CV). 6F6FPI exhibited deep blue emission (443 nm), along with high $T_m$ ($382^{\circ}C$) and relatively high $T_g$ ($148^{\circ}C$).

흰쥐에 있어서 간손상 정도에 따른 Bromobenzene 대사 (Study on Bromobenzene Metabolism in Rats with Liver Damage)

  • To compare the severe liver damage with the slight one on the bromobeazene metabolism in rats, the animal group described as B7 group was induced the stage of slight liver damage with 7 times bromobenzene injection every other day (400 mg/Kg body wt. i.p.), whereas B40 group was induced that of more severe liver damage with bromobeazene 40 times injection as identified with determination of serum levels of alanine aminotransferase(ALT) activity and the histopathological findings. In the present experimental animal model, the decreasing rate of glutathione(GSH) and the increasing rate of glutathione S-transferase activity to the control group were higher in B7 group than B40 group. Furthermore the single dose of bromobenzene was injected to the two groups and sacrificed at 8hr and the hepatic aniline hydroxylase(AH) activity, GSH content and GST activity were determined. The increasing rate of AH activity to the control was lower in B40 group than B7 group and the decreasing rate of GSH to the control was also lower in B40 than B7 group. Moreover, B7 group showed the increased activity of hepatic GST to the control whereas B40 group showed the decrease activity of the enzyme. And Vmax value in GST was more decreased in B40 group than B7 group.

누룩 또는 밀기울 첨가식이로 성장시킨 흰쥐의 혈중 Cholesterol 및 간조직 유해산소 대사효소 활성 변동 (Effects of Nuruk or Wheat Bran Supplemented Diet on the Serum Levels of Cholesterol and Activities of Hepatic Oxygen Free Radical Metabolizing Enzymes in Rats)

  • The effects of nuruk and wheat bran on cholesterol level in serum and activities of free radical metabolizing enzymes were investigated in rats. The rats were fed a diet containing nuruk or wheat bran for one month. Body weight and food intake were measured. Animals were sacrificed after one month. The increased food efficiency ratio throughout whole growth period was observed in the rats fed with either nuruk containing Aspergillus terreus or wheat bran compared with control group on normal diet. In the rats fed with nuruk, hepatic GSH content, glutathione S transferase activity, hepatic cytochrome P 450 content, and aniline hydroxylase activities were generally increased. In the rats fed with nuruk containing other fungi except Aspergillus terreus, xanthine oxidase activity was decreased. The decreased cholesterol level in serum was observed in rats fed with nuruk prepared from Aspergillus terreus and wheat bran. LDL cholesterol level was decreased in rats fed with nuruk prepared with other fungi such as Penicillium sp. and Rhizopus sp. But HDL cholesterol level was increased in all groups fed with nuruk from any fungi and wheat bran. These results suggested that nuruk or wheat bran supplemented diet might exert their effect by decreasing cholesterol level in serum and amount of oxygen free radical level.

The Roles of Kupffer Cells in Hepatocellular Dysfunction after Femur Fracture Trauma in Rats

  • The aim of this study was to investigate the effects of trauma on alterations in cytochrome P450 (CYP 450)-dependent drug metabolizing function and to determine the role of Kupffer cells in hepatocellular dysfunction. Rats underwent closed femur fracture (FFx) with associated soft-tissue injury under anesthesia, while control animals received only anesthesia. To deplete Kupffer cells in vivo, gadolinium chloride (GdCl$_3$) was injected intravenously via the tail vein at 7.5 mg/kg body wt., 1 and 2 days prior to FFx surgery. At 72 h after FFx, serum alanine aminotransferase (ALT) activity was increased, and this increase was attenuated by GdCl$_3$ pretreatment. Serum aspartate aminotransferase (AST) and lipid peroxidation levels were not changed by FFx. Hepatic microsomal CYP 450 content and aniline p-hydroxylase (CYP 2E1) activity were significantly decreased; decreases that were not prevented by GdC1$_3$. The level of CYP 2B1 activity was decreased by Kupffer cell inactivation, but not by FFx. There were no significant differences in the activities of CYP 1A1, CYP 1A2 and NADPH-CYP 450 reductase among any of the experimental groups. Our findings suggest that FFx trauma causes mild alterations of hepatic CYP 450-dependent drug metabolism, and that Kupffer cells are not essential for the initiation of such injury.
