Effects of 2-Acetylaminofluorene and Choline Deficiency on Lipid Peroxidation, Glucose 6-phosphatase and Glutathione S-transferase Activities in Rats Fed Different Dietary Fats (2-Acctylaminofluorene과 Choline결핍이 서로 다른 지방을 섭취한 쥐 간의 지질 과산화 반응 및 Glucose 6-phosphatase, Glutathione S-transferase활성도에 미치는 영향)

  • This study examines the effects of choline deficiency and 2-acetylaminofluorene(2-AAF) on the lipid peroxide values, glucose 6-phosphatase(G6Pase) and glutathione S-transferase (GST) activities in rats fed different dietary fats. Weanling Sprague Dawley male rats fed the diets containing 15% beef tallow or 15% corn oil with vitamin fortification mixture or choline free vitamin mixture for 10 weeks. At 3th and 5th week, 2-AAF was injected twice each week intraperitoneally. Total 2-AAF injection was four times. 2-AAF and choline deficiency increased lipid peroxidation in corn oil groups, so the role of 2-AAF and choline deficiency in lipid peroxidation was more important in corn oil groups than beef tallow groups. G6Pase activities tended to be decreased by 2-AAF in choline deficient groups, and in corn oil groups, the enzyme activities were decreased significantly in all subgroups compaired to beef tallow groups. GST activities were increased by 2-AAF in beef tallow groups and choline deficiency in corn oil groups, and might defence against carcinogen metabolism and lipid peroxidation.

Influence of soymilk and skim milk on growth and antibacterial activity of lactic acid bacteria (유산균의 증식과 항균 활성에 관한 탈지유 및 두유의 영향)

  • The purpose of this study was to investigate the effect of lactic acid bacteria (LAB) on the growth and biogenic amines (BA) formation of Enterobacter aerogenes CIH05 in skim milk and soymilk. Lactobacillus acidophilus GK20, Lactobacillus paracasei GK74, and Lactobacillus plantarum GK81 isolated from mustard kimchi did not produce BA in the decarboxylation broth. L. paracasei GK74 exhibited the highest cell viability and antimicrobial compounds producing ability in fermented skim milk and soymilk samples, while the lowest producer was L. plantarum GK81. The production yield of lactic acid, hydrogen peroxide, and bacteriocin was dependent on the species of Lactobacillus and the type of culture medium. As LAB the number of viable cells of E. aerogenes CIH05 were higher in skim milk than in soymilk. When mixed culture with L. acidophilus GK20 and L. paracasei GK74 and treated with bacteriocin solution (300 AU/ml) obtained from these strains in milk media, the cell growth and cadaverine and histamine contents of E. aerogenes CIH05 were significantly (P < 0.05) lower than the respective values in control sample.

Changes in BOD, COD, Chlorophyll-a and Solids in Aquaculture Effluent with Various Chemical Treatments

  • Four chemical treatments with hydrogen peroxide (H2O2), copper sulfate (CuSO4), potassium permanganate (KMnO4) and chlorine (Cl2) were applied to the effluent pond water of a hybrid striped bass saltwater recirculating aquaculture system to compare their oxidation power. Four chemicals were applied at concentrations of 0 (control), 1, 5, 10 and 20 mg/l. An additional concentration of 40 mg/l was included in the chlorine treatment. Water samples from four hybrid striped bass ponds were tested with KMnO4 and Cl2. H2O2 did not reduce any of BOD, COD and chlorophyll-a, and copper sulfate was only effective on chlorophyll-a for the effluent pond. Removal efficiencies for chlorophyll-a by copper sulfate were 19.2%, 37.5%, 54.2% and 74.1% dose-dependently. Potassium permanganate effectively removed the BOD, COD and chlorophyll-a. The COD removal rates in four fish ponds varied from 15.9% to 31.6% at the concentration of 10 mg/l. Interestingly, Cl2 did not reduce the BOD and COD at all, but the BOD and COD instead increased drastically with increasing the Cl2 concentration. The pond water with the highest initial BOD and COD values among the fish ponds tested increased by 350% in the BOD and 150% in the COD at 20 mg/l. Furthermore, Cl2 did not significantly reduce any types of solid matter in this study, while KMnO4 seemed to reduce some extent volatile dissolved solid in the fish pond.

Effect of Treatment with Selected Plant Extracts on the Physiological and Biochemical Parameters of Rice Plants under Salt Stress

  • High soil salinity is the most severe threat to global rice production as it causes a significant decline in rice yield. Here, we investigated the effects of various plant extracts on rice plant stress associated with high salinity. Additionally, we examined various physiological and biochemical parameters such as growth, photosynthetic activity, chlorophyll content, and lipid peroxidation - in rice plants after treatment with selected plant extracts under salt stress conditions. Of the 11 extracts tested, four - soybean leaf, soybean stem, moringa (Moringa oleifera), and Undaria pinnatifida extracts - were found to effectively reduce salt stress. A reduction of only 3-23% in shoot fresh weight was observed in rice plants under salt stress that were treated with these extracts, compared to the 43% reduction observed in plants that were exposed to stress but not given plant extract treatments (control plants). The effectiveness varied with the concentration of the plant extracts. Water content was higher in rice plants treated with the extracts than in the control plants after 6 d of salt stress, but not after 4 d of salt stress. Although photosynthetic efficiency (Fv/Fm), electron transport rate (ETR), and the content of pigments (chlorophyll and carotenoid) varied based on the types and levels of stress and the extracts that the rice plants were treated with, generally, photosynthetic efficiency and pigment content were higher in the treated rice compared to control plants. Reactive oxygen species (ROS), such as superoxide radicals, hydrogen peroxide (H2O2), and malondialdehyde (MDA), increased as the duration of stress increased. ROS and MDA levels were lower in the treated rice than in the control plants. Proline and soluble sugar accumulation also increased with the duration of the stress period. However, proline and soluble sugar accumulation were lower in the treated rice than in the control plants. Generally, the values of all the parameters investigated in this study were similar, regardless of the plant extract used to treat the rice plants. Thus, the extracts found to be effective can be used to alleviate the adverse effects of stress on rice crops associated with high-salinity soils.

Effect of Carrot Powder on Anti-Oxidative and Quality Characteristics of Perilla-Dasik (당근가루를 첨가한 들깨다식의 품질 특성 연구)

  • Carrot powder was added at different levels (0, 5, 10, 15, and 20%) to Perilla-Dasik, and its effects on the physical and antioxidant properties of Dasik were measured. For texture characteristics, hardness, cohesiveness, and chewiness of Dasik slightly increased as carrot powder content increased. For color properties, both $a^*$ (redness) and $b^*$ (yellowness) values increased as carrot powder content increased. Anti-oxidative effects, as measured by acid value, peroxide value, and free radical scavenging activity, were greater in samples containing carrot powder compared to control. In the sensory test, Dasik with carrot powder was more preferred than control (without carrot powder). Based on these results, 20% addition of carrot powder in the formulation was the most acceptable for Dasik.

  • Imaging technique or computer vision (CV) technology has received huge attention as a rapid and non-destructive technique throughout the world for measuring quality attributes of agricultural products including meat and meat products. This study was conducted to test the ability of CV technology to predict the quality attributes of beef. Images were captured from longissimus dorsi muscle in beef at 24 h post-mortem. Traits evaluated were color value (L*, a*, b*), pH, drip loss, cooking loss, dry matter, moisture, crude protein, fat, ash, thiobarbituric acid reactive substance (TBARS), peroxide value (POV), free fatty acid (FFA), total coliform count (TCC), total viable count (TVC) and total yeast-mould count (TYMC). Images were analyzed using the Matlab software (R2015a). Different reference values were determined by physicochemical, proximate, biochemical and microbiological test. All determination were done in triplicate and the mean value was reported. Data analysis was carried out using the programme Statgraphics Centurion XVI. Calibration and validation model were fitted using the software Unscrambler X version 9.7. A higher correlation found in a* (r=0.65) and moisture (r=0.56) with 'a*' value obtained from image analysis and the highest calibration and prediction accuracy was found in lightness (r2c=0.73, r2p=0.69) in beef. Results of this work show that CV technology may be a useful tool for predicting meat quality traits in the laboratory and meat processing industries.

Purification and Characterization of Glutathione Peroxidase Isolated from Rat Erythrocyte and Histochemical Study of its Localization in Liver of White Rat (흰쥐 적혈구에 있는 Glutathione Peroxidase의 순화 및 성질과 간에서의 용작부위에 대한 조직화학적 연구)

  • A glutathione peroxidase from white rat (Wistar strain)erythrocytes was partially purified and characterized. In addition, localization of this enzyme in the liver was studied by histochemical method. A glutathione peroxidase was purified approximately 33.5-folds by ammonium sulfate precipitation, Sephadex filtration column and DEAE-Sephadex column chromatography. The optimum temperature of the crude glutathione peroxidase was $40^\\circC$, and the optimum pH was 7.5. This crude glutathione peroxidase was most stable at $30^\\circC$ and the values of Km and Vmax were calculated to be 8.5mM and 15.6 $\\mu$moles/min for glutathione, and 40 $\\mu$M and 10.5 $\\mu$moles/min for hydrogen peroxide, respectively. The molecular weight of this enzyme was estimated by Sephadex G-200 gel filtration to be approximately 90, 000. By electron microscopic examination, histochemical reaction products were microbodies that were prominent in the peripheral parts of the lobule. The reaction products exhibited round shapes, the diameter of which varied $0.2\\sim0.7 \\muM$ and their boundary membranes were not distint.

The effect of fluoride and casein phosphopeptide-amorphous calcium phosphate (CPP-ACP) pplication on the color and microhardness of bleached enamel (치아미백 후 불소와 CPP-ACP 처리가 치아의 색과 미세경도에 미치는 영향)

  • Objectives : To evaluate the effect of fluoride application on the color and microhardness of bleached enamel and compare it to that of casein phosphopeptide-amorphous calcium phosphate (CPP-ACP) application. Methods : Twenty freshly extracted human adult molar were each sectioned into halves, the specimens divided and treated according to five experimental groups: Group 1, treatment with 10% carbamide peroxide (CP) bleaching agent; Group 2, treatment with 10% CP followed by a 1.23% fluoride gel application; Group 3, treatment with 10% CP followed by a 2.23% sodium fluoride varnish application; Group 4, treatment with 10% CP followed by a 0.11% sodium fluoride gel application; Group 5, treatment with 10% CP followed by a CPP-ACP gel application. All groups were treated 6 h per day for 14 days then immersed in distilled water for 2 weeks. Changes in enamel color were evaluated on Baseline and Day 14. Microhardness were evaluated on Baseline, Days 7 and 14. Statistical analysis was performed using one-way ANOVA and post-hoc Tukey tests. Results : All the bleached enamel specimens revealed increased whiteness and overall color value. Group 1 showed the lowest microhardness values than that of Groups 2, 3, 4 and 5. In all groups, the hardness of tooth after bleaching showed a significant decrease in the microhardness as compared with the one prior to tooth bleaching. The specimens treated with remineralizing agents showed relatively less reduction in enamel microhardness than control group. Conclusions : The addition of fluoride and CPP-ACP did not impede the whitening effect. The use of remineralizing agents during bleaching treatment can significantly enhance the microhardness of bleached enamel.

The Effects of Smoking on Antioxidative Enzyme Activities in Male Adolescents (흡연이 남자 청소년들의 일부 항산화 관련 효소체계에 미치는 영향)

  • Smoking can increase oxidative stress and thereby change the antioxidant defense system in the body. To investigate the relationship between male adolescent smoking and antioxidant status, we surveyed the eating habits and dietary intake of 82 smokers and 44 nonsmokers recruited from a male technical high school. In addition, antioxidant enzyme activity and lipid peroxide values were determined in both the plasma and the erythrocytes. Although the frequency of food intake was not significantly different, most nutrient intake was unexpectedly higher in smokers than in nonsmokers. In comparison with the Korean RDA, especially the average intake of Ca, Fe and vitamin $B_2$ didn t reach 75% of the Korean RDA in either smokers or nonsmokers. An analysis of antioxidant enzyme activity showed that plasma catalase. superoxide dismutase (SOD), glutathione peroxidase (GSH-px), erythrocyte catalase and GSH-px activities showed no significant difference between smokers and nonsmokers. However, the erythrocyte SOD activity of smokers (1.57 unit/mgHb) was significantly lower than that of nonsmokers (2.00 unit/mg Hb). In addition, the plasma ceruloplasmin concentration of smokers (28.68 mg/$d\ell$) was significantly higher than that of nonsmokers (26.30 mg/$d\ell$), whereas the specific ceruloplasmin ferroxidase activity of smokers (0.31 unit/mg) was lower than that of nonsmokers (0.35 unit/mg). The plasma and erythrocyte thlobarbituric acid reactive substance (TBARS) of smokers (2.57 $\mu$mol/L, 0.32 $\mu$mol/gHb) were also significantly higher than those of nonsmokers (2.25 $\mu$mol/L, 0.27 $\mu$mol/gHb). The overall data indicate that adolescent smoking might decrease the antioxidant capacity of the body, in part, by lowering the erythrocyte SOD activity and the specific ceruloplasmin ferroxidase activity.

Effects of Seed Germination on Oil Oxidation and Tocopherol Stability of Perilla Oil (들깨의 발아가 들깨유의 산화 및 토코페롤 안정성에 미치는 영향)

  • Auto- and photo-oxidative stability of oil extracted from germinated perilla seeds during storage at $60^{\circ}C$ for 4 days was studied by determining peroxide and conjugated dienoic acid values. Tocopherol contents during oil oxidation were also monitored by high performance liquid chromatography. Perilla oil was oxidized and tocopherols were degraded during storage at $60^{\circ}C$ regardless of the presence of light. Light increased oil oxidation and tocopherol degradation. Seeds germinated for 12 h had increased tocopherol contents in the oil and improved the auto- and photo-oxidative stability of the perilla oil. Tocopherol played a more important role as an antioxidant in the presence of light than in the absence of light.
