일본어 유악센트 방언과 무악센트 방언의 통사적 애매성의 해소와 운율적 특징 (Syntactic Ambiguities and their Resolution in Prosody in Japanese)

  • The prosody can play a crucial role in differentiating ambiguous sentences to correctly reflect their intended syntactic structures. In what way do the speakers in Tokyo and Sendai dialects of Japanese use prosodic elements to differentiate syntactic ambiguities? Acoustic measurement was made of utterances of ambiguous sentences in Japanese to observe prosodic strategies for disambiguation. Materials were sentences of the type ADV-VP1-NP-VP2, ADV-NP1-NP2-VP2, where the ambiguity lies in locative adverbial modification, ADV modifying either VP1 or VP2. For this construction the Japanese create the same ambiguities. After defining the depth of a syntactic boundary, F0 of the phrase before and after the boundary, and duration of the syllable and pause before the boundary were measured. The results show that Tokyo dialects speakers use F0 after syntactic boundary, and Sendai dialects speakers use of the syllable and/or pause before the boundary.

소아 SPECT/CT 검사를 위한 최저조건에서의 피폭선량측정 및 팬텀의 영상평가 (Abosrbed Dose Measurements and Phantom Image Ecaluation at Minimum CT Dose for Pediatric SPECT/CT Scan)

  • SPECT/CT로 소아 복부를 검사 하는데 있어 저선량 CT 조건에 따른 흡수선량을 측정하고 SPECT와 CT 영상의 질을 평가하고, 우수한 영상의 질을 유지하면서 최저의 흡수 선량을 받을 수 있는 tube voltage (kVp)와 tube current (mA)의 설정 방향을 알아보는데 목적을 두었다. 장비는 Discovery NM/CT 670을 사용하였다. PMMA phatom을 이용하여 80, 100 kVp 10, 15, 20, 25 mA의 조건을 설정하여 중심방향과 주변방향(3, 6, 9, 12시 방향의 평균) 의 흡수선량을 측정하였고, 그에 따른 image를 SNRD로 평가 하였다. CT QA performance phantom으로 CT image의 resolution을 MTF로 나타내었고, jaszczak phantom을 hot sphere와 배후방사의 비를 $^{99}mTc$을 1:8로 주입하여 4개의 sphere에 대한 SPECT image를 CNR로 평가하였다. 선량측정에서는 주변방향의 선량이 중심방향 선량보다 평균 7% 높게 측정되었으며, SNRD는 조건에 따라 유의한 차이가 없었으며 Resolution 평가에서는 0.385 lp/mm 기준으로 100 kVp가 80 kVp보다 평균 12% 재현성이 우수하였으며, jaszczak phantom을 이용해서 CT를 기반으로 한 attenuation correction 된 SPECT image를 CNR로 평가한 결과 CT조건의 변화와 무관하게 4개의 sphere 모두 유의한 차이를 보이지 않았다. 본 연구는 SPECT/CT 검사에서 최저의 흡수선량을 유지하고 우수한 영상의 질을 획득하는데 추가적인 연구가 필요할 것으로 사료된다.

소아 환자에서의 뇌실-복강 단락 감염의 역학적 고찰: 15년 간의 단일 기관 연구 (A Fifteen-year Epidemiological Study of Ventriculoperitoneal Shunt Infections in Pediatric Patients: A Single Center Experience)

  • 목 적 : 뇌실-복강 단락 수술은 수두증이 있는 소아 환자에서 중요한 치료 방법 중 하나이다. 뇌실-복강 단락에서 단락 감염은 중요한 합병증이며 수술의 예후를 결정하는 중요한 요인이다. 이 연구는 소아 환자에서의 뇌실-복강 단락 감염의 역학적 특성과 임상 증상, 치료와 예후를 파악하고 진단 및 치료에 도움이 되고자 하였다. 방 법 : 1995년 4월부터 2010년 6월까지 뇌실-복강 단락 수술을 시행 받은 18세 이하의 환자를 대상으로 하였으며 후향적으로 의무 기록을 분석하였다. 결 과 : 총 190명(여자 82명, 남자 107명)의 환자에서 327건의 뇌실-복강 단락 수술을 시행하였다. 중위 연령은 2.4세이며(0.02-17.9세) 뇌실-복강 단락 수술의 가장 흔한 원인은 악성 뇌종양이었다. 수술 1건 당 감염률은 6.7% (22/327건), 환자 1명 당 감염률은 9.5% (18/190명)이며 100건의 수술-년 당 감염은 0.45건 이었다. 가장 흔한 원인 균은 coagulase-negative staphylococcus (7건) 이며 methicillin resistant Staphylococcus aureus에 의한 감염은 1건 이었다. 10건의 감염에서 vancomycin과 beta-lactam antibiotics (cephalosporin or carbapenem)의 복합 정주 치료를 시행하였으며 7건의 감염에서 vancomycin 단독 정주 치료를 시행하였다. 치료 기간의 중앙값은 26일(7-58일)이었으며 수술적 치료는 18건에서 시행하였다(18/22건, 81.8%). 결 론 : 본 연구는 단일 기관에서 15년 동안의 뇌실-복강 단락 감염의 역학을 요약한 연구로 소아 환자에서의 뇌실-복강 단락 감염의 역학적 정보는 적절한 치료를 시행하는데 큰 도움이 될 것이다. 향후 단락 감염의 발생과 관련한 위험 인자에 대한 추가적인 연구가 필요할 것으로 사료된다.

Interaction of Heliothis armigera Nuclear Polyhedrosis Viral Capsid Protein with its Host Actin

  • In order to find the cellular interaction factors of the Heliothis armigera nuclear polyhedrosis virus capsid protein VP39, a Heliothis armigera cell cDNA library was constructed. Then VP39 was used as bait. The host actin gene was isolated from the cDNA library with the yeast two-hybrid system. This demonstrated that VP39 could interact with its host actin in yeast. In order to corroborate this interaction in vivo, the vp39 gene was fused with the green fluorescent protein gene in plasmid pEGFP39. The fusion protein was expressed in the Hz-AM1 cells under the control of the Autographa californica multiple nucleopolyhedrovirus immediate early gene promoter. The host actin was labeled specifically by the red fluorescence substance, tetramethy rhodamine isothicyanete-phalloidin. Observation under a fluorescence microscopy showed that VP39, which was indicated by green fluorescence, began to appear in the cells 6 h after being transfected with pEGFP39. Red actin cables were also formed in the cytoplasm at the same time. Actin was aggregated in the nucleus 9 h after the transfection. The green and red fluorescence always appeared in the same location of the cells, which demonstrated that VP39 could combine with the host actin. Such a combination would result in the actin skeleton rearrangement.

일반촬영에서 kVp와 mAs의 변화에 따른 농도와 환자 선량 비교 (A Comparison of Density and Patient Doses According to kVp and mAs Changes in General Radiography)

  • 일반촬영의 저관전압 촬영에서 발생되는 저 에너지 X-선은 신체에 흡수가 많고 영상 품질 향상에는 도움을 주지 못한다. 본 연구에서는 일반 촬영에서 적정 농도를 유지하면서 환자의 피폭 선량을 줄이기 위해 농도에 따른 관전압 15%법칙과 농도에 비례하는 관전류량을 이용하여 면적 선량과 입사표면선량을 측정하여 환자의 피폭선량을 비교하였다. Hand, Knee, Abdomen, Skull 촬영에서 kVp를 115%까지 증가하면서 mAs를 50%까지 감소시키고, kVp를 85%까지 감소시키고 mAs를 200%까지 증가시키면서 면적선량과 입사표면선량을 측정하여 각각의 선량을 비교하였다. 그리고 각 영상의 5군데를 정하여 농도를 측정하고 Kruskal wallis H 검증을 하여 집단-간의 유의확률을 알아보았다. 농도를 일정하게하기 위해 관전압을 115%로 증가하고, 관전류를 50%로 감소시킨 조건에서 각 부위별 평균 면적선량과 입사표면선량을 측정한 결과 기준 선량을 100%로 할 때 각각 58.68%, 59.85%로 감소하고, 관전압을 85%로 감소하고 관전류를 200%로 증가시킨 조건에서 각각 147.28%, 159.9%로 증가하였다. 농도 변화를 비교한 결과 Hand, Knee, Abdomen, Skull 촬영 모두 유의확률 >0.05 나타나 농도 변화는 없는 것으로 나타났다. 해상력과 대조도에 영향을 주지 않는 범위에서 적정한 계산을 통해 관전압을 증가시키고 관전류를 낮게 해서 촬영하는 것이 적정농도를 유지하면서 환자의 피폭 선량을 줄이는 간단한 방법으로 사료된다.

Effects of Gas Composition in the Modified Atmosphere Packaging on the Shelf-life of Longissimus dorsi of Korean Native Black Pigs-Duroc Crossbred during Refrigerated Storage

  • This study was conducted to observe the effects of gas composition in modified atmosphere packaging (MAP) on the shelf-life of Longissimus dorsi of Korean Native Black Pigs-Duroc Crossbred ($KNP{\times}D$) during refrigerated storage. Muscle sample was obtained from the left side of carcass of seven months old of $KNP{\times}D$ barrow. The sample was sliced into 1 cm in thickness, placed on trays (two slices/tray) and filled with different gas composition, i.e. 0:20:80/$O_2:CO_2:N_2$ (MAP1), 30:20:50/$O_2:CO_2:N_2$ (MAP2) and 70:20:10/$O_2:CO_2:N_2$ (MAP3). Other slices of sample were vacuum packed (VP) as a control. All packs were stored at $5{\pm}1^{\circ}C$. At 12 d of storage, pH value of MAP2 and MAP3 were higher (p<0.05) than that of MAP1 and pH value of MAP1 was higher (p<0.05) than that of VP. At 6 d of storage, redness ($a^*$) value of MAP2 and MAP3 were higher (p<0.05) than that of VP and MAP1 and, at 9 and 12 d of storage, redness value of MAP3 was higher (p<0.05) than that of VP, MAP1, and MAP2. At 3, 6, 9, and 12 d of storage, the 2- thiobarbituric acid reactive substances (TBARS) value of MAP3 was higher than that of MAP2 and TBARS value of MAP2 was higher than that of VP and MAP1. At 3, 6, 9, and 12 d of storage, volatile basic nitrogen values of MAP2 and MAP3 were higher (p<0.05) than those of VP and MAP1. At 3 d of storage, total aerobic plate counts of MAP2 and MAP3 were higher (p<0.05) than those of VP and MAP1 and, at 6 d of storage, total aerobic plate counts of MAP3 was higher (p<0.05) than that of MAP1 and MAP2. However, there was no significant different total aerobic plate count among MAP1, MAP2, and MAP3 at 9 and 12 d of storage. There was no significant different total anaerobic plate count among MAP1, MAP2, and MAP3 during storage. It is concluded that the MAP containing 30:20:50/$O_2:CO_2:N_2$ gas composition (MAP2) might be ideal for better meat quality for $KNP{\times}D$ meat.

Recombinant DNA and Protein Vaccines for Foot-and-mouth Disease Induce Humoral and Cellular Immune Responses in Mice

  • Foot-and-mouth disease virus (FMDV) is a small single-stranded RNA virus which belongs to the family Picornaviridae, genus Apthovirus. It is a principal cause of FMD which is highly contagious in livestock. In a wild type virus infection, infected animals usually elicit antibodies against structural and non-structural protein of FMDV. A structural protein, VP1, is involved in neutralization of virus particle, and has both B and T cell epitopes. A RNA-dependent RNA polymerase, 3D, is highly conserved among other serotypes and strongly immunogenic, therefore, we selected VP1 and 3D as vaccine targets. VP1 and 3D genes were codon-optimized to enhance protein expression level and cloned into mammalian expression vector. To produce recombinant protein, VP1 and 3D genes were also cloned into pET vector. The VP1 and 3D DNA or proteins were co-immunized into 5 weeks old BALB/C mice. Antigen-specific serum antibody (Ab) responses were detected by Ab ELISA. Cellular immune response against VP1 and 3D was confirmed by ELISpot assay. The results showed that all DNA- and protein-immunized groups induced cellular immune responses, suggesting that both DNA and recombinant protein vaccine administration efficiently induced Ag-specific humoral and cellular immune responses.

Development of a One-Step Duplex RT-PCR Method for the Simultaneous Detection of VP3/VP1 and VP1/P2B Regions of the Hepatitis A Virus

  • The simultaneous detection and accurate identification of hepatitis A virus (HAV) is critical in food safety and epidemiological studies to prevent the spread of HAV outbreaks. Towards this goal, a one-step duplex reverse-transcription (RT)-PCR method was developed targeting the VP1/P2B and VP3/VP1 regions of the HAV genome for the qualitative detection of HAV. An HAV RT-qPCR standard curve was produced for the quantification of HAV RNA. The detection limit of the duplex RT-PCR method was 2.8 × 101 copies of HAV. The PCR products enabled HAV genotyping analysis through DNA sequencing, which can be applied for epidemiological investigations. The ability of this duplex RT-PCR method to detect HAV was evaluated with HAV-spiked samples of fresh lettuce, frozen strawberries, and oysters. The limit of detection of the one-step duplex RT-PCR for each food model was 9.4 × 102 copies/20 g fresh lettuce, 9.7 × 103 copies/20 g frozen strawberries, and 4.1 × 103 copies/1.5 g oysters. Use of a one-step duplex RT-PCR method has advantages such as shorter time, decreased cost, and decreased labor owing to the single amplification reaction instead of four amplifications necessary for nested RT-PCR.

일반촬영 시 거리역자승법칙에 따른 산란선 감약에 관한 연구 (The Study About Attenuation of Scatter Ray According to Distance Inverse Square Law at General Projection)

  • 거리 역자승 법칙에 관한 엑스선 감약의 정도를 파악하고 이를 산란선 발생에서 적용하여 방사선사의 피폭을 저감할 수 있는 공간을 찾도록 알아보고자 하였다. 관전류량 10 mAs, 관전압 60 kVp, 70 kVp, 81 kVp, 90 kVp, 각각의 거리 60 cm, 120 cm, 180 cm에서 1차 선량을 측정하고, 산란선은 관전류량 20 mAs, 관전압 70 kVp, phantom의 중심부로부터 전면과 후면으로 42.5 cm, 52.5 cm, 62.5 cm 떨어진 곳에서 음 양극(좌우측)으로 각각 10 cm씩 60 cm까지 측정하였고, 거리역자승법칙과 비교하기 위해 전 후방 각각 42.5 cm, 85 cm, 127.5 cm에서 산란선을 측정하였다. 1차선은 거리가 2배에서는 20.52 mR(27.20%), 28.58 mR(25.20%), 38.82 mR(26.32%), 48.20 mR(26.27%)로 감약되고 거리가 3배에서는 7.06 mR(8.91%), 9.90 mR(8.73%), 13.64 mR(9.25%), 16.60 mR(9.05%)로 감약되는 것을 알 수 있었고, 산란선은 전 후방 각각 거리가 2배에서는 0.15 mR(23.09%), 0.15 mR(22.08%) 3배에서는 0.07 mR(10.43%), 0.06 mR(8.83%)로 감약되었다. 산란선의 발생량이 평균적으로 3사분면이 적게 발생하기에 환자를 붙잡고 촬영할 시에는 3사분면의 피사체에서 가능하면 거리를 두고 환자를 잡는 것이 피폭선량을 줄일 수 있다.

위조영촬영시(胃造影撮影時) 촬영조건(撮影條件)에 관(關)한 연구(硏究) (A Study on Exposure Technics in Roentgenography of the Stomach)

  • In examinations of the stomach roentgenography, it is imperative to obtain adequate film density throughout all its different regions. Therefore, it is necessary to use more sophiscated exposure techniques. In order to achieve these purpose, the radiologic technologists must be measured abdominal thickness in variations with patient positions. In consideration of these problem, the author was made an experiment on correction method of kVp and mAs by abdominal thickness in roentgenography of the stomach. The results were summarized as follws: 1. When the patient in erect position, abdominal thickness was the most thickened at the level of 3cm inferior to umbilicus without regard to body habitus and it was the most thickened at the level of 3cm superior to umbilicus in prone and supine position. 2. As a result of measuring film density for stomach, the adequate film density was represented from 0.70 to 2.49 in erect position and $0.28{\sim}1.18$ in supine position, $0.5{\sim}2.45$ in prone position. 3. In order to obtain uniform film density in 1.25, the correction factor for kVp by abdominal thickness was represented average ${\pm}4.5kVp\;per\;{\pm}1cm$ in a fixed 50 mAs, and average ${\pm}3.9kVp\;per\;{\pm}1cm$ in a fixed 100mAs. 4. In order to obtain uniform film density in 1.25, the correction factor for mAs by abdominal thickness was represented average ${\pm}30.9%\;per\;{\pm}1cm$ in a fixed 80 kvp and ${\pm}26.9%\;per\;{\pm}1cm$ in a fixed 100kVp.
