GABA tea which was increased of GABA by anaerobic conditions in green tea leaves) was fed in the diet to spontaneously hypertensive rats(SHR). The mean systolic blood pressure of SHR fed a diet containing 1.5% GABA tea after 3 weeks was not different with zero day as 177.3mmHg. However, 1.5% green tea and control after 3 weeks were more increased 4.2 percents and 10 percents than in zero day. The plasma levels of aldosterone of SHR fed a diat containing 1.5% GABA green tea after 3 weeks was more decreased 2.5 times than in water treatment.

As common pollutants in surface and groundwater, nitrate nitrogen($NO_3-N$) and trichloroethylene(TCE) can be chemically and biologically reduced by zero valent iron(ZVI) and peat soil. In batch microcosm experiments, chemical reduction of TCE and nitrate was supported by hydrogen from ZVI. For biological degradation of TCE and denitrification peat soil was introduced. ZVI reduced TCE, while peat provided TCE sorption site and microbes performing biological degradation. Nitrate reduction was also achieved by hydrogen from ZVI. In addition, indirect evidence of denitrification was observed. More reduction of TCE and nitrate was achieved by ZVI+peat treatment however nitrated reduction was hindered in the presence of TCE in the system due to the competition for hydrogen. TCE reduction mechanism was more dependent on ZVI, while nitrate was peat-dependent. Hydrogen and methane concentration showed that peat had various anaerobic denitryfing and halorespiring bacteria.

The microcosm type wetland systems were constructed in order to treat wastewater contaminated with parathion. The microcosm reactor consisted of marsh and pond type. The experiment was carried out using batch (marsh or pond) and continuous (marsh-pond and pond-marsh type) systems. In the batch reactor, marsh-type wetland completely removed parathion in water within 8 days, while pond reactor removed 97% of parathion during the same period. During parathion degradation, the amount of 4-nitrophenol production, one of the metabolites from parathion degradation, was higher in marsh-type batch reactor. In the continuous systems, both marsh-pond and pond-marsh combination systems effectively removed parathion from water, and the production of 4-nitrophenol was also minimal. In the extraction experiment, the parathion and its metabolite were not found in the wetland soil and the plant. In order to achieve both aerobic and anaerobic conditions, the continuous wetland system combining marsh and pond type can be the alternative for the non-point source pollutants such as parathion pesticide.
Periodontitis is generally a chronic disorder characterized by breakdown of tooth-supporting tissues, producing dentition loss. Porphyromonas gingivalis (P. gingivalis), a Gramnegative anaerobic rod, is one of the major pathogens associated with periodontitis. Neutrophils are first line defense cells in the oral cavity that play a significant role in inflammatory response. Xylitol is a known anti-caries agent and has anti-inflammatory effects. In this study, we conducted experiments to evaluate anti-inflammatory effects of xylitol on P. gingivalis infected neutrophils for possible usage in prevention and treatment of periodontal infections. P. gingivalis was intraperitoneally injected and peritoneal lavage was collected for cytokine determination. For in vitro study, neutrophils were collected from mouse peritoneal cells after zymosan injection or bone marrow cells. Neutrophils were stimulated with live P. gingivalis and ELISA was used to determine the effect of xylitol on P. gingivalis induced cytokine production. $IL-1{\beta}$, IL-6, $TNF-{\alpha}$ concentration and neutrophil population in the peritoneal lavage was increased in P. gingivalis-infected mouse. Peritoneal cells infected with live P. gingivalis revealed significantly increased production of $IL-1{\beta}$, IL-6 and $TNF-{\alpha}$ at multiplicity of infection of 10. Neutrophils from bone marrow and peritoneal lavage revealed increased production of $IL-1{\beta}$, IL-6 and $TNF-{\alpha}$. Xylitol significantly mitigated P. gingivalis induced cytokine production in neutrophils. Findings indicate that xylitol is an anti-inflammatory agent in neutrophils infected with live P. gingivalis, that suggests its use in periodontitis management.
Several experiments have been done to investigate the removal of hydrogen sulfide ($H_2S$) synthetic gas from biogas streams by means of chemical absorption and chemical reaction with 0.1-1 M Fe/EDTA solution. The roles of Fe/EDTA were studied to enhance the removal efficiency of hydrogen sulfide because of oxidizing by chelate. The motivation of this investigation is first to explore the feasibility of enhancing the toxic gas treatment in the biogas facility. The biogas purification strategy affords many advantages. For instance, the process can be performed under mild environmental conditions and at low temperature, and it removes hydrogen sulfide selectively. The end product of separation is elemental sulfur, which is a stable material that can be easily disposed with minor potential for further pollution. As the Fe-EDTA concentration increased, the conversion rate of hydrogen sulfide increased because of the high stability of Fe-EDTA complex. pH as an important environmental factor was 9.0 for the stability of chemical complex in the oxidation of hydrogen sulfide.

Homeostatic pH is very important for various cellular processes, including metabolism, survival, and death. An imbalanced-pH might induce cellular acidosis, which is involved in many abnormal events such as apoptosis and malignancy. One of several factors contributing to the onset of metabolic acidosis is the production of lactate and protons by lactate dehydrogenase (LDH) in anaerobic glycolysis. LDH is an important enzyme that catalyzes the reversible conversion of pyruvate to lactate. This study sought to examine whether decreases in extracellular pH induce apoptosis of CHO cells, and to elucidate the role of mitogen-activated protein kinases (MAPKs) in acidification-induced apoptosis. To test apoptotic signaling by acidification we used CHO dhfr cells that were sensitive to acidification, and CHO/anti-LDH cells that are resistant to acidification-induced apoptosis and have reduced LDH activity by stable LDH antisense mRNA expression. In the present study, cellular lactic acid-induced acidification and the role of MAPKs signaling in acidification-induced apoptosis were investigated. Acidification, which is caused by $HCO{_3}^-$-free conditions, induced apoptosis and MAPKs (ERK, JNK, and p38) activation. However, MAPKs were slightly activated in acidic conditions in the CHO/anti-LDH cells, indicating that lactic acid-induced acidification induces activation of MAPKs. Treatment with a p38 inhibitor, PD169316, increased acidification-induced apoptosis but apoptosis was not affected by inhibitors for ERK (U0126) or JNK (SP600125). Thus, these data support the hypothesis that activation of the p38 MAPK during acidification-induced apoptosis contributes to cell survival.

Background: Periodontitis is generally a chronic disorder characterized by the breakdown of tooth-supporting tissues. P. gingivalis, a Gram-negative anaerobic rod, is one of the major pathogens associated with periodontitis. Frequently, P. gingivalis infection leads to cell death. However, the correlation between P. gingivalis-induced cell death and periodontal inflammation remains to be elucidated. Among cell deaths, the death of immune cells appears to play a significant role in inflammatory response. Thus, the aim of this study was to examine P. gingivalis-induced cell death, focusing on autophagy and apoptosis in THP-1 cells. Methods: Human acute monocytic leukemia cell line (THP-1) was used for all experiments. Autophagy induced by P. gingivalis in THP-1 cells was examined by Cyto ID staining. Intracellular autophagic vacuoles were observed by fluorescence microscopy using staining Acridine orange (AO); and 3-methyladenine (3-MA) was used to inhibit autophagy. Total cell death was measured by LDH assay. Cytokine production was measured by an ELISA method. Results: P. gingivalis induced autophagy in an MOI-dependent manner in THP-1 cells, but 3-MA treatment decreased autophagy and increased the apoptotic blebs. P. gingivalis infection did not increase apoptosis compared to the control cells, whereas inhibition of autophagy by 3-MA significantly increased apoptosis in P. gingivalis-infected THP-1 cells. Inhibition of autophagy by 3-MA also increased total cell deaths and inflammatory cytokine production, including $IL-1{\beta}$ and $TNF-{\alpha}$. Conclusion: P. gingivalis induced autophagy in THP-1 cells, but the inhibition of autophagy by 3-MA stimulated apoptosis, leading to increased cell deaths and pro-inflammatory cytokines production. Hence, the modulation of cell deaths may provide a mechanism to fight against invading microorganisms in host cells and could be a promising way to control inflammation.
A response surface method based on a central composite design experiment was used to determine the optimum conditions for pretreatment of persimmon peel. It was mathematically predicted that the maximum amount of reducing sugars would be obtained at an $H_2SO_4$ concentration of 1.77% (w/v) and a heat treatment time of 26.4 min. A reducing sugar concentration of 63.23 g/l was obtained under the optimum pretreatment conditions determined by RSM. Under anaerobic growth conditions, Saccharomyces cerevisiae NK28 produced 15.52 g/l of ethanol with a yield of 0.34 g ethanol/g glucose from pretreated persimmon peel, which corresponded to 14% and 26% enhancements in ethanol productivity and ethanol yield, respectively, compared with those obtained in aerobic growth conditions. This study suggests that persimmon peel might be a useful substrate for bioethanol production by yeast fermentation.
Tetanus is a specific infectious disease, which is often associated with catastrophic events such as earthquakes, traumas, and war wounds. The obligate anaerobe Clostridium tetani is the pathogen that causes tetanus. Once the infection of tetanus progresses to an advanced stage within the wounds of limbs, the rates of amputation and mortality increase manifold. Therefore, it is necessary to devise a rapid and sensitive point-of-care detection method for C. tetani so as to ensure an early diagnosis and clinical treatment of tetanus. In this study, we developed a detection method for C. tetani using loop-mediated isothermal amplification (LAMP) assay, wherein the C. tetani tetanus toxin gene was used as the target gene. The method was highly specific and sensitive, with a detection limit of 10 colony forming units (CFU)/ml, and allowed quantitative analysis. While detecting C. tetani in clinical samples, it was found that the LAMP results completely agreed with those of the traditional API 20A anaerobic bacteria identification test. As compared with the traditional API test and PCR assay, LAMP detection of C. tetani is simple and rapid, and the results can be identified through naked-eye observation. Therefore, it is an ideal and rapid point-of-care testing method for tetanus.
UASB reactor was operated for treating wastewater containing high sulfate to assess their performance, competition between sulfate-reducing bacteria(SRB) and methane-producing bacteria(MPB), and the change in the characteristics of microbial granules according to change of hydraulic retention time(HRT) in the reactor. The reactor was fed with a synthetic moderate strength wastes(glucose, 2000 mgCOD/l) containing high sulfate($2400mgSO_4{^{2-}}/l$). The organic loading rate(OLR) ranged from 1.5 to 3.0 gCOD/l.d as HRT maintained 15 to 30 hrs in the stage I. The COD removal efficiency was between 80 to 92%. During this period, methane yield rapidly decreased from 0.3 to 0.1 1 $CH_4$/gCODremoved. While sulfide concentration in the effluent increased from 80 to 200 mgS/l. This indicates that SRB becomes dominant over MPB at a relatively long HRT in the excess sulfate. When OLR of reactor maintained from 5 to 8 gCOD/l.d in the stage II, methane yield increased from 0.1 to 0.17 1 $CH_4$/gCODremoved regardless of decrease of COD removal efficiency. This indicates that SRB is more sensitive to the change of a short HRT than MPB. In the competition between SRB and MPB, about 30% of the removed COD was utilized by SRB at HRT of 30 hrs during the start-up period, while about 73% was used by SRB at HRT of 15hrs at the final step of second experimental stage. Whereas after shock exposure of OLR about 62% was utilized by SRB at HRT of 5hrs. It indicates that SRB is strongly suppressed by the wash-out of significant dispersed SRB since a large electron flow is distributed to the MPB. In addition, the granulation in the presence of high sulfate is unfavoured at a long HRT because of substrate transport limitations into MPB like Methanothrix spp. which is an important factor in the composition of the granules. Accordingly, granule sizes in the UASB reactor decreased with time due to weak network frame of granules by the decreased activity of MPB.
