The anaerobic oxidation of methane (AOM) in anaerobic sewage sludge was characterized. The net methane oxidation was observed in samples amended with methane plus sulfate or with methane alone, whereas methane formation was observed in the samples without methane, indicating that methane oxidation and formation occurred simultaneously. The ratio of the net methane oxidation rate to $H_2S$ formation was 100:1, suggesting that the AOM was not closely associated with sulfate reduction in the anaerobic sludge. The net AOM was positively associated with the methane concentration and sludge dilution ratio. However, the rate of AOM was negatively correlated with organic substrate (acetate) concentration. Therefore, the production and oxidation of methane could be controlled by environmental conditions and dissolved organic compounds in the bulk solution.
가스 하이드레이트는 높은 지구 온난화 잠재력을 가지고 있는 메탄가스를 해수 또는 대기 중으로 유입시킬 수 있어 전 지구적 탄소순환과정과 기후 변화에 중요한 역할을 한다. 따라서 해양 또는 대기로 방출되는 메탄의 90% 이상을 미생물 반응을 통해 산화시킬 수 있는 혐기적 메탄산화 과정이 매우 중요하다. 본 연구에서는 동해 울릉분지내 메탄 가스 하이드레이트 퇴적토에 서식하는 미생물 군집의 mcrA 유전자와 16S rRNA 유전자를 분석하였다. 혐기적 메탄산화 고세균(Anaerobic methane oxidizer: ANME) 군집의 수직적 분포를 조사한 결과, 표층과 황산염 메탄전이대(Sulfate methane transition zone: SMTZ)에서는 ANME-1 그룹이, high methane 층에서는 ANME-2c 그룹이 우점하였다. 16S rRNA 유전자를 이용한 고세균의 군집분석 결과, 혐기적 메탄산화가 일어나는 지역에서 주로 발견되는 marine benthic group-B가 50% 이상의 비율로 우점하였다. 세균의 경우 질산염을 환원시킬 수 있는 세균이 SMTZ (Halomonas 속: 56.5%)와 high methane 층(Achromobacter 속: 52.6%)에서 우점하였으며 황산염 환원 세균 군집은 확인되지 않았다. 동해 울릉분지 메탄가스 하이드레이트의 혐기적 메탄산화과정은 일반적으로 해양 퇴적토에서 알려진 혐기적 메탄산화 고세균과 황산염 환원 세균과의 공생에 의한 반응이 아닌 혐기적 메탄산화 고세균과 질산염 환원세균에 의한 반응이 주도할 것이라 생각된다.
This study aims to observe the inhibition of methane generation, the decomposition of organic matter, and the trend of outflowing leachate, using the simulated column of the anaerobic sanitary landfill structure of sulfate addition type which is made by adding sulfate to a current anaerobic landfill structure, and the simulated column of semi-aerobic landfill structure in the laboratory which is used in the country like Japan in order to inhibit methane from a landfill site among the gases caused by a global warming these days, and at the same time to promote the decomposition of organic matter, the index of stabilization of landfill site. As a result of this study, it is thought that the ORP(Oxidation Reduction Potential) of the column of semi-aerobic landfill structure gradually represents a weak aerobic condition as time goes by, and that the inside of landfill site is likely to by in progress into anaerobic condition, unless air effectively comes into a semi-aerobic landfill structure in reality as time goes by. In addition, it can be seen that the decomposition of organic matter is promoted according to sulfate reduction in case of $R_1$, a sulfate-added anaerobic sanitary landfill structure, and that the stable decomposition of organic matter in $R_1$ makes a faster progess than $R_2$. Moreover it can be estimated that $R_1$, a sulfate-added anaerobic sanitary landfill structure has an inhibition efficiency of 55% or so, compared with $R_2$, a semi-aerobic landfill structure, in the efficiency of inhibiting methane.
동해 울릉 분지 내 메탄 하이드레이트가 매장된 지역에서 혐기적 메탄 산화와 연관된 미생물 군집 특성을 이해하기 위해 메탄 누출이 있는 대륙사면 정점(UBGH2-3)과 메탄 누출이 없는 분지 정점(UBGH2-10)에서, (1) 퇴적물의 지화학적 성분 및 황산염 환원율을 측정하였으며, (2) 기능성 유전자 분석을 통해 혐기적 메탄 산화 미생물 및 황산염 환원 미생물 군집의 정량 및 다양성 분석을 수행하여 그 결과를 비교하였다. 황산염-메탄 전이지대(sulfate and methane transition zone, SMTZ)는 UBGH2-3에서 0.5~1.5 mbsf (meters below seafloor), 그리고 UBGH2-10에서는 6~7 mbsf에 분포하는 것으로 나타났다. 두 지역의 SMTZ에서 측정된 황산염 환원율은 UBGH2-3의 1.15 mbsf에서 $1.82nmol\;cm^{-3}d^{-1}$으로 나타났고, UBGH2-10의 SMTZ에서 황산염 환원율은 $4.29nmol\;cm^{-3}d^{-1}$으로 높은 값을 보였다. 총 미생물 16S rRNA gene과 기능성 유전자인 mcrA (methyl coenzyme M reductase subunit A) 및 dsrA (dissimilatory sulfite reductase subunit A)의 정량 PCR 결과 두 정점의 SMTZ 부근에서 상대적으로 높게 검출되었다. 그러나 UBGH2-10지역에서 mcrA 유전자는 SMTZ 아래인 9.8 bmsf에서 가장 높게 검출되었다. mcrA 유전자의 다양성 분석 결과 두 지역의 SMTZ와 그 아래 퇴적층에서 혐기성 메탄산화 고세균(ANME: Anaerobic MEthanothoph) 군집인 ANME-1이 우점하였다. 한편, ANME-2 군집은 메탄 누출이 발생하는 UBGH2-3지역의 2.2 mbsf 층에서만 관찰되었고, 더불어 dsr 유전자 다양성 분석 결과 ANME-2와 컨소시엄을 이루는 Desulfosarcina-Desulfococcus (DSS) 이 나타났다. 본 연구 결과, ANME-1과 ANME-2은 동해 심부 퇴적 환경에서 혐기적 메탄 산화 및 생성 과정에 관여하는 중요한 고세균 군집으로 사료되며, 또한 ANME-2/DSS 컨소시엄은 메탄이 누출되는 지역인 UBGH2-3과 같이 혐기적 메탄 산화가 활발한 곳에서 메탄 거동을 조절하는 중요한 미생물 그룹으로 인식된다.
식생이 없는 갯벌에서 메탄($CH_4$) 플럭스는 배출과 흡수 양상을 보인다. 특히 메탄영양세균 개체군은 메탄 흡수에 상당한 영향을 준다. 본 예비연구는 거대 무척추 동물의 서식활동(bioturbation) 영향을 이해하기 위해 원형 밀폐 챔버 방법을 사용하여 갯벌에서 메탄 플럭스를 조사하였다. 챔버는 십각목(decapoda)의 mud shrimp(Laomedia astacina)와 crab(Macrophthalmus japonicas) 서식굴에 약 2시간 동안 배치되었고, 메탄과 이산화탄소($CO_2$) 농도는 밀폐된 $CH_4/CO_2$ 확산 플럭스 관측 장비를 사용하여 지속적으로 모니터링 되었다. 서식굴 길이가 긴 Laomedia astacina 위치에서는 깊은 퇴적층의 메탄 방출 때문에 상대적인 높은 수준의 메탄 발생을 일으킨 것으로 나타났다. 또한, 서식굴 퇴적물에서 발견되는 메탄영양세균 개체군은 메탄 산화(oxidation) 잠재력을 나타냈다. 특히 서식굴 내 아질산염 관련 혐기성 산화(anaerobic oxidation of methane, AOM)가 확인되었다. 이러한 메탄 산화 발생은 챔버 실험 동안 이산화탄소의 탄소 안정동위원소비(${\delta}^{13}C$) 감소로 뒷받침되었다. 따라서 갯벌에서 무척추 동물의 서식활동은 대기 중 메탄 농도를 제어할 수 있는 중요한 생태계 환경으로 보인다.

The wastewater treatment with a two-phase expanded granular sludge bed (EGSB) system for anaerobic degradation of acetate, benzoate, terephtalate and p-toluate from purified terephtalic acid (PTA) production was studied. The feasibility and effectiveness of the system was evaluated in terms of organic oxidation by chemical oxygen demand (COD), gas production, bacterial adaptability and stability in the granular sludge. Average removal efficiencies 93.5% and 72.7% were achieved in the EGSB reactors under volumetric loading rates of $1.0-15kg-COD/m^3/day$ and terephtalate and p-toluate of 351-526 mg/L, respectively. Gas production reached total methane production rate of 0.30 L/g-COD under these conditions in the sequential EGSB reactor system. Higher strength influent COD concentration above 4.8 g-COD/L related to field conditions was fed to observe the disturbance of the EGSB reactors.
During the rhizoremediation of diesel-contaminated soil, methane (CH4), a representative greenhouse gas, is emitted as a result of anaerobic metabolism of diesel. The application of methantrophs is one of solutions for the mitigation CH4 emissions during the rhizoremediation of diesel-contaminated soil. In this study, CH4-oxidizing rhizobacteria, Methylocystis sp. JHTF4 and Methyloversatilis sp. JHM8, were isolated from rhizosphere soils of tall fescue and maize, respectively. The maximum CH4 oxidation rates for the strains JHTF4 and JHM8 were 65.8 and 33.8 mmol·g-DCW-1·h-1, respectively. The isolates JHTF4 and JHM8 couldn't degrade diesel. The inoculation of the isolate JHTF4 or JHM8 significantly enhanced diesel removal during rhizoremediation of diesel-contaminated soil planted with maize for 63 days. Diesel removal in the tall fescue-planting soil was enhanced by inoculating the isolates until 50 days, while there was no significant difference in removal efficiency regardless of inoculation at day 63. In both the maize and tall fescue planting soils, the CH4 oxidation potentials of the inoculated soils were significantly higher than the potentials of the non-inoculated soils. In addition, the gene copy numbers of pmoA, responsible for CH4 oxidation, in the inoculated soils were significantly higher than those in the non-inoculated soils. The gene copy numbers ratio of pmoA to 16S rDNA (the ratio of methanotrophs to total bacteria) in soil increased during rhizoremediation. These results indicate that the inoculation of Methylocystis sp. JHTF4 and Methyloversatilis sp. JHM8, is a promising strategy to minimize CH4 emissions during the rhizoremediation of diesel-contaminated soil using maize or tall fescue.
The process of biological treatment of organic wastewater is principally associated with those of self-purification in the natural water environment. The treatment system has e intensive function of stabilizing wastewater more effectively than in natural water, which is like natural water concentrated in a small space. Biological treatment of wastewater involves activated sludge and various modified process, trickling filter, rotating disk, oxidation ditch, etc. for aerobic decomposition and anaerobic processes such as anaerobic decomposition and methane fermentation. The basic characteristic of these processes is the use of mixed culture for the conversion of pollutants. This review forcuses on the various kinds of microorganisms related to each treatment processes. Kinetic analysis of the activated sludge process is discussed in order to understand the basis of control and maintenance of the biological treatment process.
In this study a system for the treatment or recyling of organic wastes from both urban and rural area was recommended. It was developed based on the resource recovery system regarding human being by four tectnologies; forage, methane production, high-grade composting and complete decomposition. High quality compost can be produced by combining several kind of wastes produced from urban and agricultural areas. High quality compost must possess not only general characteristics of ordinary compost, but also a superior ability to improve the soil properties and must contain more nutrients for plant. Cedar chips were recommended as the main bulking agent to adjust moisture contents and air permeability. Charcoal and zeolite can be used not only as the second bulking agent but also as fertilizer for improve the soil amendment. Complete decomposition of organic wastes is defined by organic matter being completely converted to $CO_2$ and water. All the input water was evaporated by the heat produced through the oxidation of organic matter, In the present study, the complete treatments were successfully achieved for Shochu wastewater, swine wastes, thickened excess sewage sludge, wastes produced by Chinese restaurant and anaerobic digested sludge. First of all, recycling center of organic wastes should be established for the protect the environments and effective recovery of organic resources. This may means the way to derive the recovery of human value.
