Identification and Characteristics of a Purple, Non-Sulfur Bacterium, Rhodobacter sp. EGH-24 from Korea Coast (한국 해안으로부터 Purple, Non-Sulfur Photosynthetic Bacterium, Rhodobacter sp. EGH-24의 분리 및 특성)

  • A species of facultative photo-organotrophic, purple, non-sulfur bacterium was isolated from the 47 point at west and south coast of Korea in September 2001. Separated 13 samples of changes with red color under 28-32$^{\circ}C$, 3000 lux, anaerobe conditions for 7 days cultivated in basal medium. For pure isolation from 13 samples, we used agar-shake tube method (0.4 % agar) and separated 5 strains through 13-repetition test. EGH-24 and EGH-30 was identified as the same strain through the RAPD(Random Amplified Polymorphic DNA)-PCR of strain EGH-9, EGH-13, EGH-23, EGH-24, EGH-30. Four isolates cultivated in synthesis wastewater for wastewater biodegradation test. EGH-24 was selected with efficient wastwater treating strain. Based on the results obtained from morphology, nutrient requirements, major bacteriochlorophyll content, 16S-rDNA phylogenetic analysis, EGH-24 strain may be identified as a new strain of the genus Rhodobacter and named Rhodobacter sp. EGH-24.

Characterization of Binding of Treponema denticola to Immobilized Fibrinogen using the Fluorescent Fatty Acid Labeling Method

  • Treponema denticola is a gram-negative anaerobe that can cause periodontal disease. The adhesion of this bacterium to host tissues is considered to be the primary event in the colonization and infection of a host. Fibrinogen is generally found in damaged tissues resulting from periodontitis. The binding ability of T. denticola to fibrinogen may therefore be an important virulence factor in inducing periodontal diseases. It has been reported recently that oral spirochetes can be labeled with fluorescent fatty acids and we speculated that this labeling method could be used in an oral spirochete binding assay. The binding of several different strains of T. denticola to immobilized human fibrinogen was therefore tested using the fluorescent fatty acid labeling method. In the case of immobilized fibrinogen, the T. denticola ATCC 35405 strain showed saturable binding to immobilized fibrinogen. Indeed, all four different T. denticola strains tested in this experiment, T. denticola ATCC 35405, T. denticola ATCC 33520, T. denticola ATCC 35404 and T. denticola OTK showed binding to fibrinogen. The fluorescent fatty acid labeling method thus shows utility in binding assays for T. denticola, different strains of which can generally bind to immobilized fibrinogen.

  • In order to search for new antibiotics from anaerobic bacteria, a large number of samples from domestic soil were collected and processed by apropriate methods. A potential strain, Streptococcus sp. An-21-1, was found to produce antimicrobial compounds. The Results were as follows; 1. During fermentation, the bacteria grew rapidly up to 20hr, thereafter entered the death phase. The optimal temperature and pH for the bacterial growth were $37^{\circ}C$ and pH 7.0, respectively. 2. Antibiotics were purified from culture broth by solvent extraction, silica gel column chromatography and Sepadex L.H 20 column. 3. Physicochemical properties of Ap-1 and Ap-2 were similar ; Their melting points were between $234-237^{\circ}C$. Color reactions of ninhydrin, 2,7-dichlorofluorescein, 4-dimethylaminobenzaldehyde, Dragendroffs reagent and 20% $H_2SO_4$, were positive. Therefore, we assumed that these antibiotics have amine group, immine group, alkaloid, and lipid components. These were stable to heat. UV spectrophotometry showed two peaks at 210 nm and 260 nm. From above results, we assumed these antibiotics are belong to the peptide antibiotic family.

  • Anaerobic bacteria are suggested to be potential source for new antibiotics. In order to search for antibiotics from domestic origin, we collected 800 soil samples across Korean locations and could isolate as many as 989 anaerobic strains. Among them 10, strains were found to have good producing capacity of antibiotics. An anaerobe was finally selected due to secreting antibiotics having high antimicrobial activity towards multiple resistant microorganism(E coli JM 83) transformed by genetic engineering technique. Its morphological, physiological and biochemical charateristics were investigated, together with antimicrobial spectrum therefrom. On antimicrobial spectrum study, substance secreted from this strain, had no activities to fungus and yeast. The selected strain showed G(+) and coccal shape, on Gram, staining and electron scanning microscopy, respectively. Biochemically this strain utilized glucose, fructose lactose, sucrose, but did not arabinose, cellulose, rhamnose, sorbitol, trehalose, mannitol. Catalase test showed negative property. Optimal growth temperature was $37^{\circ}C$. The results obtained above suggest this strain Streptococcus faecium subspp. and we named it Streptococcus sp. An-21-1.

  • Eubacterium limosum KIST612 was cultured on phosphate-buffered basal medium (PBBM) with carbon monoxide (CO) as the sole energy and carbon source. The initial growth rate of this strain was approximately 0.17~0.25 $h^-1$/ and the $K_s$ value for dissolved substrate was 0.14 mM. CO was limiting during the growth of the bacterium when the CO partial pressure was less than 0.6 atm (0.5 mM dissolved CO). The bacterial growth rate was reduced in the presence of acetate. When sufficient CO was supplied using a gas-lift reactor, the acetate concentration went up to 90 mM in 116 h. Based on these findings, it is suggested that a pressurized reactor be used to develop a process to convert CO-rich gases into multi-carbon compounds.

Genomic Analysis of Actinomyces sp. Strain CtC72, a Novel Fibrolytic Anaerobic Bacterium Isolated from Cattle Rumen

  • A xylanolytic and cellulolytic anaerobic bacterium strain CtC72 was isolated from cattle rumen liquor. Phylogenetic analysis of 16S rRNA gene sequences revealed that strain CtC72 shared only 97.78% homology with its nearest phylogenetic affiliate Actinomyces ruminicola, showing its novelty. The strain could grow on medium containing xylan, carboxymethyl cellulose and avicel producing $CO_2$, acetate, and ethanol as major fermentation products. The whole genome analysis of the strain CtC72 exhibited a broad range of carbohydrate-active enzymes required for the breakdown and utilization of lignocellulosic biomass. Genes related to the production of ethanol and stress tolerance were also detected. Further there were several unique genes in CtC72 for chitin degradation, pectin utilization, sugar utilization, and stress response in comparison with Actinomyces ruminicola. The results show that the strain CtC72, a putative novel bacterium can be used for lignocellulosic biomass based biotechnological applications.

Isolation and Culture Conditions of Hydrogen-producing Bacterium from Sewage Sludge (하수슬러지에서 수소생산 균주의 분리 및 배양조건)

  • Hydrogen producing bacterium, strain AS12 was isolated from the sludge of the anaerobic wastewater treatment process of south sewage treatment plant, Busan city. Phylogenetic analysis based on 16S rRNA sequence studies indicated that AS12 belonged to the genus Escherichia coli sp.. The optimum temperature and pH for hydrogen production were $35^{\circ}C$ and 8.0, respectively. The impact of the types and concentrations of carbon and nitrogen sources in the media on hydrogen production was investigated. The optimum carbon and nitrogen concentrations were 10 g/L of galactose and 5 g/L of peptone, respectively.

  • Hydrogen is considered as an energy source for the future due to its environmentally friendly use in fuel cells. A promising way is the biological production of hydrogen by fermentation. In this study, the optimization of medium conditions which maximize hydrogen production from Enterobacter aerogenes KCCM 40146 were determined. As a result, the maximum attainable cumulative volume of hydrogen was 431 $m{\ell}$ under the conditions of 0.5M potassium phosphate buffer, pH 6.5 medium containing 30 g/L glucose. The best nitrogen sources were peptone and tryptone for the cell growth as well as hydrogen production. The control of cell growth rate was found to be a important experimental parameter for effective hydrogen production

AN in vitro STUDY OF THE CORONAL LEAKAGE OF TWO ROOT CANAL FILLING TECHNIQUE USING ANAEROBIC BACTERIAL LEAKAGE MODEL (근관 충전된 치아에서의 혐기성세균을 이용한 치관부 미세누출측정에 관한 연구)

  • The aim of this in vitro study was to investigate the coronal leakage of obligate anaerobes into root canals obturated with two different techniques. 48 extracted human teeth with straight, single root canals were prepared with crown-down technique with Profile under copious irrigation until the master apical file was size 40. The teeth were divided randomly into experimental groups (40 teeth) and control groups (8 teeth). In the experimental groups, 20 teeth were obturated with lateral condensation and other 20 teeth were obturated with continuous wave technique with System B. Coronal leakage of two root canal filing technique was evaluated using anaerobic bacterial leakage model with Fusobacterium nucleatum(ATCC 25586) for 60 days. The results were as follows 1. The incidence of bacterial leakage of experimental groups was 65% in group 1 (lateral condensation) and 60% in group 2 (continuous wave technique with System B). This difference was not statistically significant (P>0.05). 2. There was no statistically significant difference(P>0.05) in leakage score between group 1 (lateral condensation) and group2 (continuous wave technique with System B).

Effect of Rice Bran and Wood Charcoal on Soil Properties and Yield of Continuous Cropping of Red Pepper

  • To improve the soil properties of physical and microbial community rice bran and wood charcoal were applied in the continuously cultivated plastic film house soil. Soil physical properties were improved by application of rice bran and charcoal compared to chemical fertilizer application (control) by 8~14% in bulk density and 5~9% in soil porosity. Changes in the biological ratio indexes of fatty acids in the soils were detected depending on the inputted materials. Especially in application of rice bran including mixture with charcoal, much more fungi and less bacteria were detected and the ratio of fungi to bacteria was increased, suggesting the more organic carbon metabolically active in these treatments. The high ratio of aerobe to anaerobe suggested the better aerobic conditions were in the soil inputted wood charcoal. From these results, it is important and possible to select some materials for the organic pepper cultivation, which may improve the poor condition soil.
