Some physicochemical properties and molecular structure of cow pea, mung bean and acorn starches(mook-forming starches) and red bean, wheat and sweat potato starches(mook-unforming starches) were investigated. Amylose contents of cow pea, mung bean and acorn starch were higher than the others. Cow pea starch was similiar to mung bean starch in gelatinization characteristics by Brabender amylogram but cold viscosity of red bean starch and peak viscosity of sweet potato starch were especially high. Whereas viscosity of wheat starch was low in whole temperature range. Amylose molecules of larger molecular size$(above\;5{\times}10^5\;molecular\;weight)$ of three mook-forming starches were more than shoes of red bean and wheat starch. Chain distribution ratios$(DP\;35{\sim}55\;to\;DP\;10{\sim}20)$ of cow pea, mung bean and acorn amylopectin were higher than thoes of red bean, sweet potato and wheat amylopectin.
The relationship between the molecular structure of acid-hydrolyzed rice starch and the retrogradation rate of starch gel was investigated. The molecular structure of starch was modified by acid hydrolysis with 1 N HCl at $35^{\circ}C$. The molecular weight of starch decreased as acid hydrolysis time was increased. At the early stage of hydrolysis up to 3 hr, the branching point of amylopectin was degraded and thereafter both ${\alpha}-1,4\;and\;{\alpha}-1,6$ linkages were hydrolyzed. The starch gel (50%) stored at $20^{\circ}C$ revealed that the rapid retrogradation occurred during 4 hr of storage which was more pronounced as the hydrolysis time increased. The degree of retrogradation of starch gels after 4 hr storage showed a linear relationship with the yield of hydrolyzate. These results suggested that the retrogradation of starch gel was accelerated by degradation of ${\alpha}-1,6$ linkages with acid.
Production of cyclodextrin (CD) directly from raw corn starch without liquefaction using cyclodextrin glucanotransferase (CGTase) was carried out in an agitated bead reaction system. Similar CD yield and production rate comparable with those of conventional method using liquefied starch were obtained. Especially high purity-CD in the reaction mixture without accumulation of malto-oligosaccharides was obtained. The maximum 54g/l of CD was obtained at raw starch concentration of 200g/l. CD yield was inversely proportional to raw starch concentration, and conversion yield was 0.48 at substrate concentration of 100g/l. The optimal amount of enzyme (CGTase unit/g raw starch) was found to be around 6.0. Granular structure of raw starch degraded by CGTase was observed by SEM in order to investigate the enhancing mechanism, along with those of acid or alkali pretreated raw starch, amylose, and amylopectin. Kinetic constants of CGTase on raw starch in an agitated bead reaction system were evaluated, and CGTase was competitively inhibited by CD.
The effects of high pressure homogenization (microfluidization) on physicochemical properties of normal maize and oxidized maize starch film were studied. The molecular dispersibility of amlyose and amylopectin and the disintegration of granular structure had a marked effect on the physicochemical properties of starch films. The high pressure homogenized starch films showed increased solubility and transmittance due to the absence of gelatinized starch granules. The tensile strength of starch film increased significantly with decreasing oxygen permeability after high pressure homogenization, indicating that starch molecules were more uniformly and fully dispersed during the film formation. As a result, a clear starch film with improved mechanical properties was obtained after high pressure homogenization due to the increased interactions between the uniformly dispersed starch molecules.

Pullulanase (PUL), a debranching enzyme, has been utilized in hydrolyzing the a-1,6 glucosidic linkages in starch, amylopectin, pullulan, as well as related oligosaccharides. It has also been indicated that PUL is a novel indicator of inherent RS (Resistant Starch) formation in rice. In this study, we performed haplotype analysis on 320 bred rice accessions, and additional 54 wild accessions were added to study genetic diversity along with other population-based analyses of the PUL gene. Through these investigations, we summarized a total of 10 functional (non-synonymous) SNPs from 7 different exons on chromosome 4. There were 10 haplotypes, of which only six haplotypes were functional, implicating different subpopulations. Diversity reduction was noticed in temperate japonica (0.0005) compared to the highest one (aus, 0.0154), illustrating their higher genetic differentiation by FST-value (0.926). The highest Tajima^ D value was observed in indica (3.6613), indicating PUL gene domestication signature under balancing selection, while the lowest Tajima's D value was found in temperate japonica (-2.2191) which might have undergone under positive selection and purified due to the excess of rare alleles. PCA, population structure, and phylogenetic analyses provide information on the genetic relatedness between and or among the cultivated subpopulations and the wild based on PUL genomic region.

Physico-chemical properties of starches from potato cultivars with different colors were investigated. White Atlantic potato had 10% higher starch yield than violet Bora Valley potato. It turned out that the shape and structure of Atlantic and Bora Valley potato starch were the same by mechanical analysis using X-ray and SEM. The ratio of $50\;{\mu}m$ particle in starches from Atlantic and Bora Valley potato was $45.44\pm2.79%$ and $42.37\pm1.03%$ respectively. The particle size of Atlantic potato starch was less than that of Bora Valley; however, there was no significant difference (p<0.05). As for moisture coupling, there was no difference (p<0.05) between the two potatoes. Swelling power showed a high increase from $65^{\circ}C$ to $80^{\circ}C$. The swelling power of Atlantic starch was higher by about 0.3% than that of Bora Valley at $90^{\circ}C$. Since Atlantic has smaller starch particles than Bora Valley, more starch particles are contained in the same size, and hence a difference in swelling power. As a result of measuring the gelatinization of potato starches from Atlantic and Bora Valley, a higher gelatinization start, climax, and complete temperatures occurred at Bora Valley than Atlantic. As for gelatinization enthalpy, Bora Valley starch with a higher gelatinization temperature consumed more energy for gelatinization.
Sensory properties of cooked rice were affected by the varieties and growing environments of the rice. Moistness, cohesiveness, and adhesiveness of cooked rice were highly and positively correlated each other, whereas firmness was negatively correlated with these attributes. Sixty rice samples which differed in varieties and/or growing environments were divided into four groups based on their textural properties through principal component analysis and cluster analysis. Quality type I showed the highest values for moistness, cohesiveness, and adhesiveness, and the lowest values for firmness of cooked rice. On the other hand, quality type IV showed just the opposite values. There was no significant difference among rice starches in amylose content (P<0.05). A17 (type III) and A09 (type IV) had higher blue values for starch and amylopectin than the other samples (type I and II). On the amylogram, these samples showed lower values for breakdown and higher values for setback than the other samples. Average degree of polymerization, average chain length, and average number of chains for amylose and amylopectin were 597-878 and 2660-3140, 140-230 and 17-19, and 3.1-4.9 and 140-170, respectively.

The compositions of fatty acid and amino acid of specialty rice which includes colored rice (Heugjinju, Jeugjinju, Josangheugchalbyeo), flavored rice (Heughyangmi, Hyangmi1), and giant embryo rice (Keunnun) were determined and compared to those of regular rice (Ilpumbyeo, Whaseonchalbyeo). Major fatty acids were linoleic acid (C18:2) and oleic acid (C18:1), which were composed of 75~80% of total fatty acids. Major amino acids were glutamic acid and aspartic acid in most cultivars but Jeugjinju in which cysteine (169.61 nmol) and GABA (129.32 nmol) were the most abundant amino acids. Thermal properties measured by differential scanning calorimeter (DSC) revealed that the enthalpy (${\Delta}H$) for starch gelatinization was the highest in Josangheugchalbyeo and Whaseonchalbyeo. It suggests that the starch structure of waxy rice could be more crystallized compared to non-waxy rice, and also that amylopectin could have more impact on starch gelatinization than amylose. The on-set and complete temperature for starch gelatinization were higher in colored rice of Heugjinju and Jeugjinju, and regular rice of Whateonchalbyeo.
Fully or nearly fully 6- [4- (4'- (nitrophenylazo)phenoxycarbonyl)]pentanoated polysaccharide derivatives were synthesized by reacting cellulose, amylose, chitosan, chitin, alginic acid, pullulan or amylopectin with 6- [4- (4'- (nitrophenylazo)phenoxy) ] pentanoyl chloride (NA6C) and their thermotropic liquid crystalline behaviors were investigated. Like in the case of NA6C, all the polysaccharide derivatives formed monotropic nematic phases, suggesting that the mesophase structure of the polysaccharide derivatives is dertermined by the mesogenic side groups and not by the polysaccharide backbone. This is the first report of polysaccharide derivatives, except cellulose derivative, that form thermotropic nematic phases. The thermal stability and degree of order of the nematic phases observed for poly saccharide derivatives were significantly different from those reported for the polymers in which the azobenzene groups are attached to flexible or rigid backbones through flexible spacers. The results were discussed in terms of the difference in the arrangement of the main and side chains and the flexibility of the main chain.
Based on its α-amylase activity at pH 5.0 and optimal pH of the crude enzyme, a strain (named B-5) with acid α-amylase production was screened. The B-5 strain was identified as Bacillus amyloliquefaciens through morphological, physiological, and biochemical characteristics analysis, as well as 16S rDNA phylogenetic analysis. Its α-amylase gene of GenBank Accession No. GU318401 was cloned and expressed in Escherichia coli. The purified recombinant α-amylase AMY-Ba showed the optimal pH of 5.0, and was stable at a pH range of 4.0-6.0. When hydrolyzing soluble starch, amylose, and amylopectin, AMY-Ba released glucose and maltose as major end products. The α-amylase AMY-Ba in this work was different from the well-investigated J01542-type α-amylase which also came from B. amyloliquefaciens. AMY-Ba exhibited notable adsorption and hydrolysis ability towards various raw starches. Structure analysis of AMY-Ba suggested the presence of a new starch-binding domain at its C-terminal region.
