New Device for Addition of Modifier to Supercritical Fluid Carbon Dioxide Mobile Phase

  • A new device to accurately deliver small amount of modifier into supercritical carbon dioxide fluid is described. Carbon dioxide, the most widely used mobile phase in supercritical fluid chromatography, is a relatively non-polar fluid, and hence the addition of small amount of polar modifiers could be necessary to migrate polar solutes. In this work, supercritical CO₂and modifier are delivered from the pump to a 100 μL mixing chamber in which a small magnetic bar is rotating. After passing through the mixing chamber, supercritical CO₂is changed to a new mobile phase with different polarity. The amount of modifier added into supercritical CO₂is measured by an amperometric microsensor, which is prepared from a thin film of perfluorosulfonate ionomer.

Quantitative Analysis of Ergosterol as a Biomarker of Mold-contaminated Foods Using the Enzyme Biosensor (효소 바이오센서를 이용한 식품의 곰팡이 오염 지표물질인 Ergosterol 정량분석)

  • Ergosterol is the significant component of the cell wall of fungi. Its presence is regarded as evidence of fungi contamination in grain and other foods. Many studies on ergosterol detection have been carried out using chemical methods, but those methods required complicated pre-treatments and long analysis times. In this study, an amperometric biosensor was developed for fast and precise ergosterol detection. The biosensor system used the electron transfer of hydrogen peroxide produced from the reaction of ergosterol with cholesterol oxidase. The biosensor system consisted of a peristaltic pump, a syringe loading sample injector, an enzyme reactor, a fabricated flow-through cell containing a working electrode, a reference electrode and a counter electrode, and a potentiostat/recorder. The working electrode was prepared by coating modified multi-wall carbon nanotube (MWNT) on glassy carbon electrode. The $MWNT-NH_2$ coated glassy carbon electrode linearly responded to hydrogen peroxide in the range of $1{\times}10^{-5}{\sim}8{\times}10^{-5}$ M with a detection limit of $10^{-7}$ M in the basic performance test. The currents produced from the ergosterol biosensor showed the linearity in a range from $1.0{\times}10^{-6}$ M to $1.0{\times}10^{-5}$ M ergosterol.

Analytical Studies of $H_2O_2$-Producing Oxidase Systems ($H_2O_2$-생성 산화효소계에 관한 분석 연구)

  • Spectrophotometric enzymatic analysis and amperometric enzymatic analysis for the determinations of glucose and ethanol were studied utilizing glucose oxidase (GO) and alcohol oxidase (AO), respectively, which commonly consume $O_2$ and produce $H_2O_2$. For the determination of glucose, $H_2O_2$ were coupled to $K_4Fe(CN)_6$ via peroxidase producing $K_3Fe(CN)_6$ whose absorbance was measured at 418 nm or whose diffusion current was measured on the glassy carbon electrode at an applied potential of -55 mV vs. Ag/AgCl (sat. KCl) reference electrode. Amperometric analysis was 1000 times more sensitive as well as 10 times better in the linear concentration range than spectrophotometric analysis. For the determination of ethanol, AO only was used for the enzymatic analysis, since $K_3Fe(CN)_6$ was completely disappeared as soon as AO was added. Either rate of $H_2O_2$ produced was amperometrically measured at +0.900 V or rate of $O_2$ consumed was measured at -0.500 V vs. Ag/AgCl(sat. KCl) reference electrode.

Development of a Hydrogen Peroxide Sensor Based on Palladium and Copper Electroplated Laser Induced Graphene Electrode (PdCu를 전기 도금한 레이저 유도 그래핀 전극 기반의 과산화수소 측정 센서 개발)

  • In this paper, we describe the fabrication and characterization of a hydrogen peroxide ($H_2O_2$) sensor based on palladium and copper (PdCu) electroplated laser induced graphene (LIG) electrodes. $CO_2$ laser was used to form LIG electrodes on a PI film. This fabrication method allows simple control of the LIG electrode size and shape. The PdCu was electrochemically deposited on the LIG electrodes to improve the electrocatalytic reaction with $H_2O_2$. The electrochemical performance of this sensor was evaluated in terms of selectivity, sensitivity, and linearity. The physical characterization of this sensor was conducted using scanning electron microscopy (SEM) and energy-dispersive X-ray spectroscopy (EDS), which confirmed that PdCu was formed on the laser induced graphene electrode. In order to increase the sensor sensitivity, the Pd:Cu ratio of the electroplated PdCu was varied to five different values and the condition of highest amperometric current at an identical of $H_2O_2$ concentration was chosen among them. The resulting amperometric current was highest when the ratio of Pd:Cu was 7:3 and this Pd;Cu ratio was employed in the sensor fabrication. The fabricated PdCu/LIG electrode based $H_2O_2$ sensor exhibited a sensitivity of $139.4{\mu}A/mM{\cdot}cm^2$, a broad linear range between 0 mM and 16 mM of $H_2O_2$ concentrations at applied potential of -0.15 V, and high reproducibility (RSD = 2.6%). The selectivity of the fabricated sensors was also evaluated by applying ascorbic acid, glucose, and lactose separately onto the sensor in order to see if the sensor ourput is affected by one of them and the sensor output was not affected. In conclusion, the proposed PdCu/LIG electrode based $H_2O_2$ sensor seems to be suitable $H_2O_2$ sensor in various applications.

  • 모세관 전기영동 및 전기화학적 검출 시스템을 마이크로 시스템에 적용하여 ITO 유리기판과 polydimethylsiloxane (PDMS)로 제작하였다. 제작된 모세관 전기영동 및 전기화학적 검출 시스템은 일회용으로 사용가능하며 전기화학적 검출에 아주 적합한 특성을 보인다. 모세관 전기영동 및 전기화학적 검출 시스템은 주입과 분리 채널 (80 ${\mu}m$${\sim}$ 40 ${\mu}m$ 깊이)을 가진 PDMS 층과 유리기판 위에 검출 전극으로 사용되는 ITO가 형성된 층으로 구성된다. PDMS 층과 ITO 유리 기판은 UV-$O_3$ cleaner를 사용하여 접합하였다. 완충용액은 10 mM 2-(N-morpholino)ethanesulfonic acid (MES)를 사용하였고 분석물질은 1 mM 농도의 dopamine과 1 mM 농도의 catechol을 사용하였다. 60 V/cm 전계로 주입 및 분리를 하였으며 작업전극과 기준전극 간의 전위는 +600 mV로 유지하며 분석물질의 농도에 비례하는 전류량법으로 측정하였다. 전기화학적 검출 회로는 천기영동 전계의 간섭으로부터 분리하였다. 10 mM MES 완충용액에서 바탕 전류의 크기가 ${\sim}$10 pA 일 때 측정전류 값은 10 nA이다. 측정된 피크 값은 기존의 Au 전극과 비교하여 선택성, 감도, 분해능이 유사한 특성을 보여준다.

Voltammetric Studies of Anion Transfer Reactions Across a Microhole Array-Water/PVC-NPOE Gel Interface

  • Voltammetric characterization of hydrophilic anion transfer processes across a 66 microhole array interface between the water and polyvinylchloride-2-nitrophenyloctylether gel layer is demonstrated. Since the transfer of hydrophilic anions including $Br^-$, $NO_3{^-}$, $I^-$, $SCN^-$ and $ClO_4{^-}$ across the liquid/gel interface usually sets the potential window within a negative potential region, a highly hydrophobic organic electrolyte, tetraoctylammonium tetrakis(pentafluorophenyl)borate, providing a wider potential window was incorporated into the gel phase. The transfer reaction of perchlorate anions across the microhole-water/gel interface was first studied using cyclic voltammetry and differential pulse voltammetry. The full voltammetric response of perchlorate anion transfer was then used as a reference for evaluating the half-wave transfer potentials, the formal transfer potentials and the formal Gibbs transfer energies of more hydrophilic anions such as $Br^-$, $NO_3{^-}$, $I^-$, and $SCN^-$. The current response associated with the perchlorate anion transfer across the micro-water/gel interface versus the perchlorate concentration was also demonstrated for sensing applications.

Fabrication of Pt-MWNT/Nafion Electrodes by Low-Temperature Decal Transfer Technique for Amperometric Hydrogen Detection

  • A Pt nanoparticle-decorated multiwall carbon nanotube (Pt-MWNT) electrode was prepared on Nafion by a hot-pressing at relatively low temperature. This electrode exhibited an intricate entangled, nanoporous structure as a result of gathering highly anisotropic Pt-MWNTs. Individual Pt nanoparticles were confirmed to have a polycrystalline face-centered cubic structure with an average crystal size of around 3.5 nm. From the cyclic voltammograms for hydrogen redox reactions, the Pt-MWNT electrode was found to have a similar electrochemical behavior to polycrystalline Pt, and a specific electrochemical surface area of $2170cm^2mg^{-1}$. Upon exposure to hydrogen analyte, the Pt-MWNT/Nafion electrode demon-strated a very high sensitivity of $3.60{\mu}A\;ppm^{-1}$ and an excellent linear response over the concentration range of 100-1000 ppm. Moreover, this electrode was also evaluated in terms of response and recovery times, reproducibility, and long-term stability. Obtained results revealed good sensing performance in hydrogen detection.

Electrodeposition of Graphene-Zn/Al Layered Double Hydroxide (LDH) Composite for Selective Determination of Hydroquinone

  • A graphene-Zn/Al layered double hydroxide composite film was simultaneously prepared by electrochemical deposition on the surface of a glassy carbon electrode (G-LDH/GCE) from the mixture solution containing GO and nitrate salts of $Zn^{2+}$ and $Al^{3+}$. The modified electrode showed good electrochemical performances toward the simultaneous electrochemical detection of hydroquinone (HQ), catechol (CA) and resorcinol (RE) due to the unique properties of graphene (G) and LDH such as large active surface area, facile electronic transport and high electrocatalytic activity. The redox characteristics of G-LDH/GCE were investigated with cyclic voltammetry and differential pulse voltammetry. The well-separated oxidation peak potentials, corresponding to the oxidation of HQ, CA and RE, were observed at 0.126 V, 0.228 V and 0.620 V respectively. The amperometric response of the modified electrode exhibited that HQ can be detected without interference of CA and RE. Under the optimized conditions, the oxidation peak current of HQ is linear with the concentration of HQ from 6.0 ${\mu}M$ to 325.0 ${\mu}M$ with the detection limit of 0.077 ${\mu}M$ (S/N=3). The modified electrode was successfully applied to the direct determination of HQ in a local tap water, showing reliable recovery data.

A Facile Electrochemical Fabrication of Reduced Graphene Oxide-Modified Glassy Carbon Electrode for Simultaneous Detection of Dopamine, Ascorbic Acid, and Uric Acid

  • This paper describes the simple fabrication of an electrode modified with electrochemically reduced graphene oxide (ERGO) for the simultaneous electrocatalytic detection of dopamine (DA), ascorbic acid (AA), and uric acid (UA). ERGO was formed on a glassy carbon (GC) electrode by the reduction of graphene oxide (GO) using linear sweep voltammetry. The ERGO/GC electrode was formed by subjecting a GO solution ($1mg\;mL^{-1}$ in 0.25 M NaCl) to a linear scan from 0 V to -1.4 V at a scan rate of $20mVs^{-1}$. The ERGO/GC electrode was characterized by Raman spectroscopy, Fourier transform infrared spectroscopy, contact angle measurements, electrochemical impedance spectroscopy, and cyclic voltammetry. The electrochemical performance of the ERGO/GC electrode with respect to the detection of DA, AA, and UA in 0.1 M PBS (pH 7.4) was investigated by differential pulse voltammetry (DPV) and amperometry. The ERGO/GC electrode exhibited three well-separated voltammetric peaks and increased oxidation currents during the DPV measurements, thus allowing for the simultaneous and individual detection of DA, AA, and UA. The detection limits for DA, AA, and UA were found to be 0.46, 77, and $0.31{\mu}M$ respectively, using the amperometric i-t curve technique, with the S/N ratio being 3.

Investigation of the Flow Dependence of a FET-Type Dissolved Oxygen Sensor and Its Reducing Method (FET형 용존 산소 센서의 유속에 의한 영향 조사와 감쇄 기법)

  • Recently, FET type dissolved oxygen sensor was proposed to overcome the disadvantages of the amperometric Clark-type sensor. The inherent problem of the proposed sensor, however, is the flow dependence of the sensor performances since the proposed sensor detects the pH change in close proximity to the working electrode. In this study, we decided the direction which minimize the flow effect in FIA(flow injection analysis) system. And a hydrodynamic buffer layer which can reduce the flow dependence were proposed. The suggested buffer-layers were formed onto sensing area and working electrode with mixed polymer matrix of TEOS(tetraethylorthosilicate) and DEDMS(diethoxydimethylsilane).
