본 연구는 prebiotics와 probiotics가 in vitro 배양조건에서 돼지 장내 미생물 및 발효산물에 미치는 영향에 대하여 검토 하였다. prebiotics로써 이소말토-올리고당(IMO), 부분분해 치커리이눌린(CI), 라피노스(RA), 사이클로덱스트린(CD)을 사용하였으며 probiotics로는 Lactobacillus reuteri를 사용하였다. In vitro 발효시험은 육성돈 사료를 소화효소로 가수분해 시킨 사료와 5%의 돈분 그리고 prebiotics와 probiotics를 첨가 또는 무첨가하여 24시간 동안 배양시켰다. 배양 후 발효액 내의 미생물, 가스, pH, 암모니아, 황화수소, 단쇄지방산을 분석하였다. 엔테로박테리아는 prebiotics와 probiotics 첨가구가 대조구와 비교하여 유의적으로 감소하였으며, 락토바실러스 수는 유의적으로 증가하였다. 발효액의 pH는 대조구에 비하여 첨가구에서 낮았으며 prebiotics보다 prebiotics+probiotics 첨가구에서 더욱 낮았다. 암모니아, 황화수소 및 스카톨의 농도는 prebiotics+probiotics 구보다 prebiotics 첨가시 유의적으로 감소하였다. 단쇄지방산은 prebiotics 보다 prebiotics+probiotics 구에서 유의적으로 많이 생성되었다. 본 시험의 결과 prebiotics 첨가는 암모니아, 황화수소 및 스카톨의 농도를 감소시키는데 효과가 있었으며, prebiotics+probiotics는 유산균과 단쇄지방산의 농도를 증가시키는 효과가 있었다. In vitro에서 얻어진 실험 결과가 실제로 돼지에 급여 시 같은 결과가 얻어질런지에 대한 추가적인 연구가 필요하다.
본 연구는 토양피복 접촉산화공정에서 새로 개발된 생물담체(Bio-rock)와 기존에 이용되어온 쇄석의 처리효율을 비교하기 위해 실시되었다 합성폐수는 $COD_{Cr}$$150{\sim}370\;mg/L$, $BOD_5$$150{\sim}270\;mg/L$, $T-N$$20{\sim}60\;mg/L$, $T-P$$5{\sim}25\;mg/L$, pH 7, 미량원소용액 2 mL/L로 조제되었다. 반응조는 2기를 준비하여 유입수량을 40 L/day로 하여 약 13개월간 운전했다. 초기 바이오락 반응기는 시멘트중의 $Ca(OH)_2$의 용출에 의해 pH 12까지 증가하였으나, 쇄석은 유기물 분해와 질산화에 의해 pH 4까지 감소하였다. pH의 불균형은 유기물 및 질소 분해균의 성장 및 활성에 저해를 초래했다 그러나 바이오락의 높은 pH는 암모니아 탈기와 인의 화학침전에는 유리한 것으로 판단되었다. 정상상태에서 바이오락은 $COD_{Cr}$ 96%, $BOD_5$ 98%, T-N 80%, T-P 85%의 높은 제거율을 나타내었고. 유입농도의 변화에도 매우 안정적이었다. 반면 쇄석의 경우 $COD_{Cr}$ 96%, $BOD_5$ 96%, T-N 42%, T-P 40%의 제거율을 나타내었다. 바이오락은 쇄석에 비해 질소, 인의 처리효율이 2배나 높았다. 또 전자현미경 분석결과에서 바이오락은 미생물의 부착이 쇄석에 비해 양호했고, 미생물 농도는 바이오락이 $5.2{\times}10^6\;CFU/mL$, 쇄석이 $2.6{\times}10^6\;CFU/mL$로 바이오락이 2배 높았다. 따라서 바이오락은 미생물 부착이 용이하고 처리효율이 높으며 유입농도 변동에도 안정적으로, 향후 처리기간 단축 및 부지면적의 감소에 유리하리라 판단된다.
전자담배는 무니코틴성이 있는 한편, 니코틴이 함유되어 있는 금연도구로서 사용된다. 전자담배의 구성품 중 액상카트리지는 식품첨가물로 사용되는 합성착향료와 유사한 성분으로 구성되어 있어, 이를 합성착향료로 사용하게 되면, 인체에 유해한 니코틴의 잔류 가능성이 존재한다. 따라서 본 연구는 식품첨가물로 사용되고 있는 합성착향료 중 니코틴 검출 분석법을 개발하였다. 분석에 사용된 검체는 시중에 사용되는 합성착향료를 이용하였다. 검체에 증류수와 디클로로메탄을 가하여 산-염기 액-액 추출법을 이용하여 추출 및 정제과정을 거친 후, 이를 질소 농축하고 이동상으로 재용해하여 HPLC/PDA에 주입하였다. 기기분석은 $C_{18}$컬럼을 사용하였고, 20 mM 개미산암모늄 함유 10% 아세토니트릴과 100% 아세토니트릴을 기울기 용리 조건으로 하여 261.9 nm에서 측정하였다. 또한, 액체크로마토그래프-질량분석기를 통해 확인시험을 수행하였으며, 모든 검증은 Codex 가이드라인의 규정에 따라 실시하였다. 그 결과, 니코틴의 정량한계는 0.4 mg/kg 수준이었고, 회수율은 76.40%-96.28%로 나타났다. 또한, 변동계수는 1.74%-5.12%로 Codex 가이드라인 규정에 만족하는 수준이었다. 따라서 개발된 분석법은 식품첨가물에 함유된 니코틴을 분석하기 위한 효율적인 분석법으로 판단된다.
Number of aspects, not only nutritional but social as well as political involved in human starvation pose nowadays global problems. In order to help establish the minimum nutritional requirements in the daily life of a man and to free people as well from either undernourishment, malnutrition or even starvation many workers have devoted themselves so far on the research programs to know what and how number of metabolic events take place in animals in vivo. It is the purpose of the present paper to examine in effect to what extent both of the protein and nucleic acids (DNA & RNA) together with an enzyme, guanine deaminase, which converts guanine into xanthine and in turn ends up to uric acid as an end product, undergo changes, quantitatively during acute starvation, using the mouse as an experimental animal. The mouse was strictly inhibited from taking foods except drinking water ad libitum and was sacriflced 24, 48, and 72 hours following starvation thus acutely induced. The animals consisted of two experimental groups, one control and another starvation groups, each being consisted of 6-24 mice of whose body weights ranged in the vicinity of 10 g. The animals were sacriflced by a blow on the head, followed by immediate excision of their livers into ice-cold distilled water, washing adherent blood and other contaminant tissues. The liver was minced foramin, by an all-glass homogenizer immersing it in an ice-bath, followed by subsequent fractionatin of the homogenate (10% W/V in 0.25M sucrose solution made up with 0.05M phosphate buffer of pH 7.4). For the liver protein and guanine deaminase assay, the 10% homogenate was centrifuged at 600 x g for 10 minutes to eliminate the nuclear fraction; and for the estimation of DNA and RNA, the homogenate was prepared by the addition of 10% trichloroacetic acid in order to free the homogenate from the acid-soluble fraction, the remaining residue being delipidate by the addition of alcohol and dried in vacuo for later KOH (IN) hydrolysis. The changes in body and liver wegihts during acute starvation were checked gravimetrically. Protein contents in the liver were monitored by the method of Lowry et al; and guanine deaminase activities were followed by the assay of liberated ammonia from the substrate utilizing the Caraway's colorimetry. The extraction of both DNA and RNA was performed by the Schmidt-Thannhauser's method, which was followed by Marmur's method of purification for DNA and by Chargaff's method of purification for RNA. The determinations of both DNA and RNA were carried out by the diphenylamine reaction for the former and by the orcinol reaction for the latter. The following resume was the results of the present work. 1. It was observed that the body as well as liver weights fall abruptly during starvation, and that the loss of body weight showed no statistical correlation with the decreases in the content of liver protein. 2. The content of liver protein and activity of liver guanine deaminase activity as well decline dramatically, and the specific activities of the enzyme (activity/protein), however, decreased gradually as starvation proceeded. 3. Both of the nucleic acids, DNA and RNA, showed decrements in the liver of mouse during acute starvation; the latter, however, being more striking in the decline as compared to the former. 4. The decreases in the liver protein content as resulted from the acute starvation had no statistically significant correlation with the decrements of DNA in the same tissue, but had regressed with a significant statistical correlation with the fall of RNA in the tissue. 5. The decrease in the activity of guanine deaminase in the liver of mouse during acute starvation was functionally more proportional to the decrease in RNA than DNA, and moreover correlated with the changes in the content of the liver protein. 6. The possible mechanisms involved during in this acute starvation as bring the decreases in the contents of DNA, protein, and guanine deaminase were discussed briefly.
본 시험에서 건초(티머시, 알팔파 및 클라인)와 짚류(톨페스큐 및 볏짚)의 buffer 용해도와 단백질 분획이 실시되었으며, 조사료 자원의 buffer 추출이 $In$$vitro$ 발효 성상, 분해율 및 가스($CO_2$ 및 $CH_4$) 생성량에 미치는 효과를 조사하였다. 다른 조사료에 비해 총 단백질 중 buffer 가용성 조단백질과 A fraction은 알팔파 건초에서 각각 61% 및 41.77%로 가장 높았으며 볏짚에서 가장 낮았다(각각 42.8% 및 19.78%). 총단백질 중 B1 fraction은 조사된 조사료간 비교적 큰 차이를 보이지 않았으나 B2 fraction에서는 다른 조사료(6.34~8.85%)에 비하여 톨페스큐짚(10.05%) 및 클라인 건초(12.34)%에서 다소 높은 수준을 보였다. 총 단백질 중 B3 fraction이 차지하는 비율은 톨페스큐짚에서 38.49%로 가장 높았으나 다른 조사료 자원 간에는 큰 차이가 없었으며, C fraction의 경우 볏짚에서 가장 높은 비율(15.05%)을 보였다. 모든 사료에서 배양 개시 후 3시간(P<0.01) 및 6시간(P<0.05)에서 buffer 추출 전에 비해 추출 후 배양액의 pH가 증가되었으며, 배양 6시간(P<0.05) 및 12시간(P<0.001)에서 다른 사료에 비해 티모시 건초 및 알팔파 건초로부터의 pH가 낮았다. 배양액의 암모니아 농도는 모든 배양시간에서 가용성 물질의 추출 전 후에 다른조사료에 비해 알팔파 건초에서 가장 높았으나 모든 사료의 추출효과는 배양 3시간(P<0.01)에서만 나타났다. 배양액의 총 VFA 농도는 배양 24시간까지 알팔파 건초에서 가장 높았던 반면 톨페스큐짚과 볏짚에서 가장 낮았다. 또한 모든 조사료에서 buffer 추출 전에 비하여 추출후에 총 VFA 농도가 감소되었다(P<0.01~P<0.001). Acetic acid ($C_2$)의 조성 비율에서는 배양 6시간까지 추출 전에 더 높았으나(P<0.001) 사료 간 차이는 없었다. Propionic acid ($C_3$) 조성 비율 역시 배양 개시 후 3, 24 및 48시간(P<0.001)에서 추출 전에 더 높았으며, 6 및 12 시간의 배양액에서 대부분 건초(티모시, 알팔파 및 클라인)와 짚류(톨페스큐짚 및 볏짚) 간 차이가 있는 것으로 조사되었다(P<0.05). 그러나 butyric acid ($C_4$) 조성비율의 경우 대부분의 배양시간에서 사료 간 차이는 없었다. 건물에서의 분해율 관련 parameter 중 a 값은 조사된 전체 조사료에서 buffer 추출 전이 추출 후에 비해서 높았으며(P<0.001), 다른 조사료에 비해 톨페스큐짚과 볏짚에서 크게 낮았다(P<0.05). 또한 b 값의 경우 역시 추출 전에 비해 추출 후에서 현저히 낮았으나(P<0.001) 사료 간 차이는 없었다. 볏짚을 제외한 조사료에서 추출 후에 비해 추출 전의 건물 유효분해율(EDDM)이 더 높았다(P<0.001). 조단백질에서의 a, b 및 c 값은 추출 전에 비해 추출 후에서 현저히 낮았으나(P<0.05) 사료 간 차이는 없었다. 조단백질 유효분해율(EDCP)에서는 다른 조사료 종류에 비해 톨페스큐짚과 볏짚에서 낮았다(P<0.05). 한편, NDF의 경우 a 값과 b 값(P<0.01) 및 NDF 유효분해율(EDNDF, P<0.001)은 추출 후에 비해 추출 전에 더 높았으나(P<0.01) 사료 간 차이는 보이지 않았다. 반추위미생물에 의해 사료분해과정 중 생성되는 $CO_2$ 량도 24시간 배양까지는 추출 전에 더 많았으며(P<0.05~P<0.001), 톨페스큐짚과 볏짚에 비해 건초 형태의 조사료로부터의 $CO_2$ 생성량이 더 많았다(P<0.05~P<0.01). 메탄가스($CH_4$) 생성량 역시 모든 배양시간에서 추출 전에 비해 추출 후에 크게 감소되었으며(P<0.01~P<0.001), 12~24시간을 제외하고는 짚류에 비해 건초에서 현저히 높은(P<0.05) 것으로 나타났다. 본 시험의 결과를 종합하면, 조사료 자원에 대한 buffer 용해도와 단백질의 분획이 $In$$vitro$ VFA 농도와 분해율 및 gas ($CO_2$ 및 $CH_4$) 발생량 간 상호 밀접한 관계를 보이는 것으로 여겨진다. 이에 따라 조사료 이용 효율 개선을 위해 조사료자원에 대한 buffer 용해도와 단백질 분획을 반추동물 TMR 조제에 활용할 필요가 있는 것으로 여겨진다.
