Decomposition of Hexahydro-1,3,5-trinitro-1,3,5-triazine by Gamma Ray Irradiation (감마선 조사에 의한 hexahydro-1,3,5-trinitro-1,3,5-triazine(RDX)의 분해)

  • The purpose of this study was to evaluate the potential of a gamma ray irradiation to decompose hexahydro-1,3,5-trinitro-1,3,5-triazine (RDX) in an aqueous solution. The decomposition reaction of RDX by gamma ray irradiation was a first-order kinetic over the applied initial concentrations (10-40mg/L). The dose constant was strongly dependent on the initial concentration of the RDX. The removal of RDX was more efficient at pH below 3 and at pH above 11 than at neutral pH (pH 5-9). The required irradiation dose to remove 99% of the RDX (40mg/L) was 4, 8 and 1 kGy, at pH 2, 7 and 13, respectively. The dose constant was increased by two folds and over twelve folds at pH 2 and 13, respectively, when compared with that at pH 7. When an irradiation dose of 20 kGy was applied, the removal efficiencies of TOC were 80, 57 and 91% at pH 2, 7 and 13, respectively. Ammonia and nitrate were detected as the main nitrogen byproducts of RDX and formic acid was detected as an organic byproduct. The results showed that a gamma ray irradiation was an attractive method for the decomposition of RDX in an aqueous solution and it was found that a strong alkaline pH over 12 should be applied to the decomposition reaction of RDX.

Effect of H2SO4 and Reaction Time on Synthesis of 5Mg(OH)2·MgSO4·3H2O Whiskers using Hydrothermal Reaction (수열반응을 이용한 침상형 5Mg(OH)2·MgSO4·3H2O 합성에 대한 H2SO4와 반응 시간의 영향)

  • Magnesium hydroxide sulfate hydrate (MHSH) whiskers were synthesized via a hydrothermal reaction by using MgO as the reactant as well as the acid solution. The effects of the H2SO4 amount and reaction time at the same temperature were studied. In general, MHSH whiskers were prepared using MgSO4 in aqueous ammonia. In this work, to reduce the formation of impurities and increase the purity of MHSH, we employed a synthesis technique that did not require the addition of a basic solution. Furthermore, the pH value, which was controlled by the H2SO4 amount, acted as an important factor for the formation of high-purity MHSH. MgO was used as the raw material because it easily reacts in water and forms Mg+ and MgOH+ ions that bind with SO42- ions to produce MHSH. Their morphologies and structures were determined using X-ray diffraction (XRD) and scanning electron microscopy (SEM).

Effect of Defaunation on In Vitro Fermentation Characteristics and Methane Emission When Incubated with Forages

  • An in vitro study was conducted to determine the effects of defaunation (removal of protozoa) and forage sources (rice straw, ryegrass and tall fescue) on ruminal fermentation characteristics, methane ($CH_4$) production and degradation by rumen microbes. Sodium lauryl sulfate, as a defaunation reagent, was added into the mixed culture solution to remove ruminal protozoa at a concentration of 0.375 mg/ml. Pure cellulose (0.64 g, Sigma, C8002) and three forage sources were incubated in the bottle of culture solution of mixed rumen microbes (faunation) or defaunation for up to 24 h. The concentration of ammonia-N was high under condition of defaunation compared to that from faunation in all incubations (p<0.001). Total VFA concentration was increased at 3, 6 and 12 h (p<0.05~p<0.01) but was decreased at 24 h incubation (p<0.001) under condition of defaunation. Defaunation decreased acetate (p<0.001) and butyrate (p<0.001) proportions at 6, 12 and 24 h incubation times, but increased propionate (p<0.001) proportion at all incubation times for forages. Effective degradability of dry matter was decreased by defaunation (p<0.001). Defaunation not only decreased total gas (p<0.001) and $CO_2$ (p<0.01~0.001) production at 12 and 24 h incubations, but reduced $CH_4$ production (p<0.001) at all incubation times for all forages. The $CH_4$ production, regardless of defaunation, in order of forage sources were rice straw > tall fescue > ryegrass > cellulose (p<0.001) up to 24 h incubation.

Preparation of Visible-light Active TiO2 Nanotubes by Solution Method (액상법에 의한 가시광감응성 티타니아 나노튜브의 제조)

  • N-doped $TiO_2$nanotubes have been newly prepared by two stage sol-gel and strong-alkali hydrothermal process using $TiCl_4$ and hydrazine/ammonia aqueous solution as raw materials. These nanotubes revealed a well developed anatase crystalline phase and perfect nanotube morphology with the diameter around 10nm and the wall thickness below 3 nm. Also, they showed a superior visible light activity and yellowish color due to the light absorption redshifted by ~35 nm and ~25 nm compared to undoped $TiO_2$ nanotubes and anatase nanoparticles, respectively.

Continuos-Flow culture of Hepatocytes in Sugar-derivatized poly (lactide-co-glycolide) Scaffolds Prepared by Gas-foaming/salt-leaching Method

  • Highly open porous polymer matrices are required for high density cell seeding, efficient nutrient, and oxygen supply to the cells cultured in the three dimensional matrices. However, there are severe problems of mass transfer limitations within the cell/scaffolds culture system. Thus we hypothesize that continuos-flow culture conditioning of cells with the scaffolds may improve the cell viability and the differentiated function. In this study, we fabricated porous PLGA scaffolds by using gas-foaming/salt-leaching method as previous described. Viscous PLGA gel paste contains ammonium bicarbonate particulates, acting as a gas-foaming agent as well as a salt-leaching porogen, were cast into Teflon mold and dried. Ammonium bicarbonate salt upon contact to an acidic aqueous solution evloves gaseous ammonia and carbon dioxide by itself. And we conjugated galactose moiety [AGA; $N-(aminobuty1)-O-{\beta}-D-galactopyranosyl-(1{\rightarrow}4)-D-glucoamide]$ to the terminal end group of a PLGA to increase the cell adhesion and matain the differentiated function of hepatocytes. Cell-seeded scaffolds were secured in a flow bioreactor chamber and exposed to continuous flow at 5 ml/min. As a result of our study, the high yield of hepatocytes attachment was accomplished by increasing the concentration of PLGA-AGA conjugate in polymer scaffolds and cells in the scaffolds under continuos flow condition maintained a high level of viability and albumin secretion rate of cultured hepatocytes showed a higher level that of control groups.

회분식과 유가식 배양에 의한 Motierella alpina로부터의 Arachidonic acid의 생산

  • In the batch culture experiments, the addition of $MnSO_4$, was examined in flask culture, and then the optimal amounts of $MnSO_4$ was investigated in 2.5- L jar-fermenter. As a results, 0.005% $MnSO_4$ was found to enhance the ARA yield of 1.14- fold. Also the addition of $KH_2PO_4_4$ was investigated in 2.5-L jar-fermenter and the ARA yield was enhanced 1.20-fold. Fed batch culture study shown a relatively high productivity of cell mass (62.1 g/L) and ARA content (12.0 g/L) when 14% ammonia solution were alternatively used to control the pH and nitrogen source in the cultivation period.

Changes in Chemical Components of Soybean Cheese Making from Cow한s Milk Added Soybean Curd (우유 첨가두부를 이용한 대두 치이즈 제조 중 화학성분의 변화)

  • In order to making the good quality soybean cheese, it is prepared byover growing cow's milk added soybean curd with Actinomucor elegans and the nold-overgrown curd soked in salt-brine/ethanol mixture. The physicochemical changes and sensory evaluation were investigated during the aging period. Crude protein, carbohydrate, crude fat ash contents of the pehtze were increased by elapsing the fermentation time, whereas moisture's decreased . Amino-N and ammonia -N contents of cow's milk added pehtze and soybean phetze were increased 17.25%, 7.23% and 16.16%, 8.42% respectively. Total nitrogen content of the pehtze was decreased by elaping the aging time but soaking solution's increased. Free amino acid content of soybean cheese was increased as a result of the proteolytic action of molds. As a result, sulfur containing amino acid such as methionine and cysteine of the cow's milk added soybean cheese were enriched 1.3 times more than the soybean cheese. Flavor, taste and texture of the cow's milk added soybean cheese were higher than soybean cheese.

  • Fed-batch fermentation was used to produce the high concentrations of poly-$\beta $-hydroxybutyrate (PHB) and poly-$\beta $-(hydroxybutyrate-co-hydroxyvalerate) (PHB/V). Specific growth rate ($\mu $), yield of cell from glucose (Y$_{x/s}$) were calculated from the two samples in 3 to 5 hours of interval and they were reflected on the determination of glucose feeding rate to maintain the glucose concentration at around 10 g/l in the culture broth. PHB was accumulated after the nitrogen became limited at 60 g/l of dry cell weight by changing ammonia water to 4N-NaOH solution. As results, the final dry cell weight (DCW) of 170 g/l, PHB of 115 g/l were obtained in 50 hours and the overall productivity was 2.4 g/l$\cdot $h. After PHB accumulation, cosubstrate of glucose and propionic acid (PA) was fed to accumulate PHB/V. But, PA feeding rate was decreased from 3 g/l$\cdot $h to 1 g/l$\cdot $h to prevent PA from accumulating to high level in the broth, which is very inhibitory to the cells. As results, DCW, PHB and PHV were 147.5 g/l, 90 g/l and 8 mole % of hydroxyvalerate, respectively.

Emulsion liquid membranes for cadmium removal: Studies of extraction efficiency

  • Emulsion liquid membrane (ELM) process suffers from emulsion instability problem. So far, emulsion produced by mechanical methods such as stirrer and homogenizer has big size and high emulsion breakage. This paper discussed the application of emulsion produced by sonicator to extract cadmium in a batch ELM system. The emulsions consist of N,N-Dioctyl-1-octanamine (trioctylamine/TOA), nitrogen trihydride (ammonia/NH4OH), sorbitan monooleate (Span 80), and kerosene as carrier, stripping solution, emulsifying agent, and organic diluent, respectively. Effects of comprehensive parameters on extraction efficiency of Cd(II) such as emulsification time, extraction time, stirring speed, surfactant concentration, initial feed phase concentration, carrier concentration, volume ratio of the emulsion to feed phase, and pH of initial feed phase were evaluated. The results showed that extraction efficiencies of Cd(II) greater than 98% could be obtained under the following conditions: 15 minutes of emulsification time, 4 wt.% of Span 80 concentration, 4 wt.% of TOA concentration, 15 minutes of extraction time, 250 rpm of stirring speed, 100 ppm of initial feed concentration, volume ratio of emulsion to feed phase of 1:5, and initial feed pH of 1.53.

Acidic Properties and Catalytic Activity of Titanium Sulfate Supported on TiO2

  • Titanium sulfate supported on $TiO_2$was prepared by impregnation of powdered $TiO_2$with an aqueous solution of titanium sulfate followed by calcining in air at high temperature. For Ti$(SO_4)_2/TiO_2$ samples calcined at 300 $^{\circ}C$, no diffraction lines of titanium sulfate are observed at $Ti(SO_4)_2$loading up to 30 wt%, indicating good dispersion of $Ti(SO_4)_2$ on the surface of $TiO_2$. The acidity of the catalysts increased in proportion to the titanium sulfate content up to 20 wt% of $Ti(SO_4)_2$. 20 wt% $Ti(SO_4)_2/TiO_2$ calcined at 300 $^{\circ}C$ exhibited maximum catalytic activities for 2-propanol dehydration and cumene dealkylation. The catalytic activities for these reactions, were correlated with the acidity of catalysts measured by ammonia chemisorption method.
