  • 피부 각질층내의 아미노산 조성비율과 유사한 조성비로 amino acid complex을 제조하고, 제조된 amino acid complex가 피부에 미치는 영향을 알아보기 위해 세포 생존율, TIMP-1 발현 증가율, 보습 및 각질량 변화를 측정하였다. 연구 결과 amino acid complex는 독성이 없는 안전한 물질로 우수한 보습력 및 보습유지효과와 ascorbic acid와 유사한 TIMP-1 발현량 증가로 주름 예방 및 개선에 효과를 가지고 있으며, 각질량 감소 및 감소유지효과로 자극이 있는 AHA 대체 물질로의 가능성을 보였다. 따라서, amino acid complex는 다기능을 가진 노화방지 화장료 기능성분으로 유용하게 사용될 수 있음을 확인하였다.

  • Seed lipids and proteins of Crotalaria juncea L were analyzed for fatty acids and amino acids respectively. Gas chromatographic analysis of the oil gave palmitic acid (16.01%), stearic acid (7.29%), oleic acid (14.41%), linoleic acid (54.44%) and linolenic acid (7.86%). The defatted seed cake contained all the essential amino acids except methionine and six non-essential amino acids.

  • Amino acid cobalt(Ⅲ) complexes of 1,3-diminopropane-N,N'-di-α-(β-methyl)-pentanoic acid (H2dpdmp), uns-cis-[Co(dpdmp)(aa)] (aa=glycine, S-alanine, R-aspartic acid, sarcosine) have been prepared from the reaction between the uns-cis-[Co(dpdmp)Cl2]- complex and the corresponding amino acid. In the reaction with the uns-cis-[Co(dpdmp)Cl2]- complex, glycine and S-alanine have yielded both merridional and facial isomers, while R-aspartic acid and sarcosine, only merridional isomers. The stereospecific substitution reaction of R-aspartic acid to racemic uns-cis-[Co(dpdmp)Cl2]- complex has yielded two merridional diastereomers; ΛR-uns-cis- and ΛR-uns-cis-[Co(dpdmp)(R-asp)]. It is of interest to note that this is one of the few CoⅢ(ONNO)(aa) type complex preparations, which gives only one isomer with stereospecificity. On the other hand, two merridional products obtained from the reaction of sarcosine with racemic uns-cis-[Co(dpdmp)Cl2]- are turned out to be mixtures of optical isomers.

Modulation of Branched-Chain Amino Acid Metaolism by Exercise in Rats

  • A variety of important roles for branched-chain amino acids in metabolic regulation has been suggested. Branched-chain $\alpha$-keto acid dehydrogenase(BCKAD) complex is a rate limiting enzyme in branched-chain amino acid metabolism. The purpose of this study was to examine the effects of exercise on the activity and activity state of branched-chain $\alpha$-keto acid dehydrogenase in rat hert and liver thssues. Forty-eight Sprague-Dawley rats were assigned into three experimental groups : sedentary control, exercised, or exercised-rested. Submaximal exercise(running) for two hours significantly increased basal activity without a change in total activity in both tissues, with a concomitiant increase in activity state of the enzyme complex. At 10 min post-exercise, heart enzyme activity significantly decreased, though not to the control level, while liver enzyme activity remained unchanged. These data suggested that the exercise-induced increase in branched-chain $\alpha$-keto acid decarboxylation in rat tissues may not be the result of enzyme synthesis, but rather is due to increased activity of the BCKAD.

  • Solution equilibria of the thmary systems $UO_2(II)$, La(III), Ce(III)-N-(2-acetamido)-iminodiacetic acid and dicarboxylic amino acids are studied potentiometrically. The formation of 1:1:1 mixed ligand complexes are inferred from the potentiometric titration curves. The formation constants of the different binary and thmary complexes in such system are evaluated at t=$24\pm{0.1}^\circ{C}\;and\;\mu=0.1\;mol\;dm^{-3}(kNO_3)$. It is deduced that the temary complex is more stable than the corresponding binary amino acid anion complex. Moreover, the order og stability of the binary or temary complexes in terms of nature of amino acid and metal ion is investigated and discussed.

홍삼 추출물과 아미노산 복합제가 정상 Rat의 지구력 운동에 미치는 영향 (Effects of Red Ginseng and Amino Acid Complex in Exercise in the Rat)

  • Objective: Physical inactivity contributes to mortality rates and is now the fourth most frequent cause of death worldwide. Red ginseng is a medicinal herb that is often used as an ergogenic aid. In this study, red ginseng was administered to rats to test whether it affected their ability to exercise. Methods: Forty-five rats were randomly distributed and divided into five groups: normal (N, n=5), control (C, n=10), the group to which only red ginseng was administered (H, n=10), the group to which only amino acid complex was administered (A, n=10), and the group to which both red ginseng and amino acid complex were administered (HA, n=10). Once a day for three weeks, 333.3 mg/kg body weight per day (b.w./day) of red ginseng and 750 mg/kg b.w./day of amino acid preparation were administered to rats. After three weeks, body weight, swimming time, and the weight of the anterior tibialis muscle of rat were measured. Blood was taken for analysis using the cardiac puncture method. Results: The swimming time of group H (921.3±199.26 sec) showed significant improvement compared to that of group C (798.48±156.37 sec) (p<0.05). Conclusion: Red ginseng has improved swimming time in rat and can be used as an effective ergogenic aid.

Effects of Phytase and Enzyme Complex Supplementation to Diets with Different Nutrient Levels on Growth Performance and Ileal Nutrient Digestibility of Weaned Pigs

  • An experiment was conducted to investigate the effect of microbial phytase ($Natuphos^{R}$) supplementation in combination with enzyme complex (composed of enzymes targeted to SBM dietary components such as $\alpha$-galactosides and galactomannans; $Endo-Power^{R}$) to diet with low nutrient levels on growth performance and ileal nutrient digestibility of weaned pigs. A total of 210 crossbred weaned pigs (Landrace$\times$Yorkshire$\times$Duroc), 6.68$\pm$0.98 kg of initial body weight, were randomly allotted to five dietary treatments, based on weight and age, according to a randomized complete block design. There were three pens per treatment and 14 pigs per pen. The dietary treatments were 1) CON (Control diet with no phytase and enzyme complex (EC)), 2) LP+EC 100 (Control diet with 0.15% unit lower available phosphorus (aP) level+0.1% phytase (500 FTU/kg diet) and 0.1% enzyme complex), 3) LP+EC 80 (Control diet with 0.15% unit lower aP level+0.08% phytase (400 FTU/kg diet) and 0.08% enzyme complex, 4) LPEA+EC 100 (Control diet with 0.15% unit lower aP and 3% lower ME and amino acid levels (lysine, methionine, threonine and typtophan)+0.1% phytase (500 FTU/kg diet) and 0.1% enzyme complex), 5) LPEA+EC 80 (Control diet with 0.15% unit lower aP and 3% lower ME and amino acid levels+0.08% phytase (400 FTU/ kg diet) and 0.08% enzyme complex). For the determination of ileal nutrients digestibility, a total of 15 T-cannulated pigs (initial body weight; 7.52$\pm$1.24 kg; 3 replicates per treatment) were used in the present study. Piglets were weighted and allotted into same dietary treatments as one in growth trial and phase I experimental diets were provided for ileal digestibility study. There was no significant difference (p>0.05) in average daily gain (ADG) and average daily feed intake (ADFI) among dietary treatments during the whole experimental period (0 to 5 weeks). However, piglets in LP+EC 100 group had a significantly higher gain/feed ratio (G:F) than piglets had in control (p<0.05). Crude protein, energy and phosphorus digestibilities were significantly improved when both of phytase and enzyme complex were supplemented at the revel of 0.1%, respectively to diets with low nutrient level (aP or (and) ME and amino acids) (p<0.05). Piglets in LP+EC 100 and LPEA+EC 100 groups showed significantly higher phosphorus content (%) in bone than that of piglets in control group (p<0.05). Supplementation of both of phytase and enzyme complex at 0.1%, respectively, to diet with low nutrient levels (aP or (and) ME and amino acids) significantly improved total ileal essential amino acid and nonessential amino acid digestibilities compared to control group (p<0.05). In conclusion, the results from the present study suggest that the simultaneous inclusion of phytase and enzyme complex to diets at recommended level is advantageous with respect to improving growth performance and nutrient digestibility of weaned pigs and may contribute to increased economic return when added to corn-soy based weaned pig diets.

Mechanisms of amino acid sensing in mTOR signaling pathway

  • Amino acids are fundamental nutrients for protein synthesis and cell growth (increase in cell size). Recently, many compelling evidences have shown that the level of amino acids is sensed by extra- or intra-cellular amino acids sensor(s) and regulates protein synthesis/degradation. Mammalian target of rapamycin complex 1 (mTORC1) is placed in a central position in cell growth regulation and dysregulation of mTOR signaling pathway has been implicated in many serious human diseases including cancer, diabetes, and tissue hypertrophy. Although amino acids are the most potent activator of mTORC1, how amino acids activate mTOR signaling pathway is still largely unknown. This is partly because of the diversity of amino acids themselves including structure and metabolism. In this review, current proposed amino acid sensing mechanisms to regulate mTORC1 and the evidences pro/against the proposed models are discussed.

Mislocalization of TORC1 to Lysosomes Caused by KIF11 Inhibition Leads to Aberrant TORC1 Activity

  • While the growth factors like insulin initiate a signaling cascade to induce conformational changes in the mechanistic target of rapamycin complex 1 (mTORC1), amino acids cause the complex to localize to the site of activation, the lysosome. The precise mechanism of how mTORC1 moves in and out of the lysosome is yet to be elucidated in detail. Here we report that microtubules and the motor protein KIF11 are required for the proper dissociation of mTORC1 from the lysosome upon amino acid scarcity. When microtubules are disrupted or KIF11 is knocked down, we observe that mTORC1 localizes to the lysosome even in the amino acid-starved situation where it should be dispersed in the cytosol, causing an elevated mTORC1 activity. Moreover, in the mechanistic perspective, we discover that mTORC1 interacts with KIF11 on the motor domain of KIF11, enabling the complex to move out of the lysosome along microtubules. Our results suggest not only a novel way of the regulation regarding amino acid availability for mTORC1, but also a new role of KIF11 and microtubules in mTOR signaling.
