  • 토양으로부터 분리한 Bacillus sp. YJ-63으로부터 항진균성 항생물질을 분리하여 그 구조를 분석한 결과는 다음과 같다. 항생물질은 7개의 $\alpha$-amino acid와 1개의 $\beta$-amino acid가 결합한 환상구조를 가지는 iturin E 계에 속하며 특히. $\beta$-amino acid의 methylene 수가 기존의 iturin E 보다 더 많은 것으로 나타났다.

Protein Hydrolysis with Formic Acid and Analysis of Amino Acid Using Butylthiocarbamyl - trimethylsilyl (BTC - TMS) Derivatives by Gas Chromatography

  • 단백질 구성 아미노산 분석을 위한 효과적인 가수분해 방법을 찾기 위하여 0.3% tryptamine을 함유한 6M formic acid와 6M HCI을 표준 아미노산과 단백질 표준품인 bovine serumn albumin 가수분해에 적용하여 표준아미노산의 회수율과 bovine serum albumin의 아미노산 조성을 분석하였고 GC에 의한 효과적인 아미노산 분석을 위하여 새로운 유도체인 butylthiocarbamyl-trimethylsilyl(BTC-TMS)유도체를 개발하여 분석한 결과는 다음과 같았다 표준아미노산의 회수율은 6M formic acid에 의한 가수분해방법이 6M HCI에 의한 가수분해 방법보다 상당히 정확하였고 특히 산 가수분해에서 tryptamine의 존재하에서도 잘 파괴되는 tryptophan의 경우 formic acid가수분해가 HCI가수분해보다 1.5배정도 높은 회수율을 보였다. Bovine serum albumin의 아미노산 조성을 583 아미노산 잔기로 환산하여 나타내었을 경우도 formic acid에 의한 가수분해가 HCI가수분해 경우보다 훨씬 정확하였고 이 때 tryptophan의 회수율도 훨씬 높게 나타났다. 다만 서열분석에서 분석되지 않는 cystine이 formic acid 가수분해시 분석되고 있어 이에 대한 정확한 검정이 필요하였다. BTC-TMS 유도체는 GC분석시 극성이 다소 낮은 DB-l7 column으로 분리가 잘되었고 재현성도 좋았으나 GC분석을 위한 대부분의 유도체에서와 마찬가지로 몇 가지 아미노산에서 두 개의 peak로 나타나는 결점이 있었다.

  • 구기자(枸杞子)의 총(總)아미노산조성(組成) 및 유리(遊離)당을 각각 amino acid autoanalyzer, HPLC로 분석(分析)한 결과(結果)는 다음과 같다. 1. 17종(種)의 총(總)아미노산이 검출(檢出)되었다. 즉 asparatic acid, threonine, serine, glutamic acid, proline, glycine, alanine, cystine, valine, methionine, isoleucine, leucine, tyrosine, phenr-lalanine, hisidtine, lysine, arginine인데 이중 threonine을 가장 많이 함유하였으며 aspartic acid는 가장 적었다. 2. 총(總) essential amino acid량은 총(總)아미노산량의53.93%로 양질(良質)의 아미노산을 함유하였다. 3. Glucose, fructose, saccharose 등 3종(種)의 유리(遊離)당이 검출(檢出)되었는데 이중 fructose를 가장 많이 함유하였다.

Amino Acid, Amino Acid Metabolite, and GABA Content of Three Domestic Tomato Varieties

  • To determine the nutritional value of domestic tomatoes, the levels of amino acids, amino acid metabolites, and the bioactive compound ${\gamma}-aminobutyric-acid$ (GABA) were analyzed in three domestic tomato varieties (Rafito, Momotaro, and Medison). Eighteen free amino acids were found, and total free amino acid content was 3,810.21~4,594.56 mg/100 g (dry weight). L-glutamic acid (L-Glu) was the most abundant amino acid, ranging from 1,866.60 mg/100 g for Momotaro to 2,417.45 mg/100 g for Medison. The next most abundant amino acids were L-glutamine (L-Gln) and L-aspartic acid (L-Asp). The three tomato varieties had a good balance of all the essential amino acids except tryptophan. Total essential amino acid content was 274.26~472.71 mg/100 g (dry weight). The following amino acid metabolites were found: L-carnitine (L-Car), hydroxylysine (Hyl), o-phosphoethanolamine (o-Pea), phosphoserine (p-Ser), ${\beta}-alanine$ (${\beta}-Ala$), N-methyl-histidine (Me-His), ethanolamine (EtNH2),and L-citrulline(L-Cit). Large quantities of GABA were found in all three varieties: 666.95-868.48 mg/100g (dry weight). These results support the use of these tomato varieties as nutritious food materials.

위치 종속 유사도 스펙트럼을 이용한 단백질 서열의 아미노산 조성 추정 (Estimating Amino Acid Composition of Protein Sequences Using Position-Dependent Similarity Spectrum)

  • 단백질의 아미노산 조성은 생물정보학의 여러 문제를 해결하기 위한 기초적인 정보로 자주 활용된다. 본 논문에서는 아미노산간의 진화적인 연관성을 정의한 BLOSUM 행렬에서 유도한 유사도 함수를 사용하여 아미노산 조성을 결정한다. 이러한 방법은 생물학적인 연관성이 있는 단백질 서열일수록 비슷한 아미노산 조성을 갖도록 한다. 또한 단백질의 구조와 기능에 중요한 역할을 하는 위치-특이적인 아미노산의 분포를 추정하기 위해서 레이더나 음성 신호의 스펙트럼 분석에 사용되는 개념인 시간-종속 분석, 시간 해상도와 주파수 해상도의 개념을 적용하였다. 제안한 방법을 단백질의 세포내 위치예측에 적용하여 기존의 아미노산 조성 추정 방법을 사용하는 것보다 크게 향상된 성능을 보임을 확인하였다.

The Analysis of Proximate Composition, Minerals and Amino Acid Content of Red Alga Pyropia dentata by Cultivation Sites

  • In this study, the proximate composition of Pyropia dentata was compared by the analysis of minerals and amino acids that were produced at Seocheon, Chungcheongnam-do and at Wando and Jangheung, Jeollanam-do. Moisture, ash and crude proteins were analyzed using the AOAC method, and crude lipid was analyzed using the Soxhlet method, and inorganic analysis was performed using ICP-OES. Amino acid was used for the amino acid analyzer. The proximate composition analysis, moisture content, maximum values were displayed in Wando while the maximum content of ash appeared at Jangheung's Pyropia. Crude proteins was appeared at Seocheon's Pyropia and the maximum amount of crude lipid, carbohydrate was appeared in Wando's Pyropia. Inorganic analysis, calcium, potassium, magnesium, sodium, phosphorus, cobalt, chromium, copper, iron, manganese, nickel, zinc were analyzed from the Seocheon's Pyropia, however, cobalt, chromium, and nickel was not detected in Wando's Pyropia. For amino acid analysis, a total of 17 amino acids were detected: leucine, valine, aspartic acid, glutamic acid, and the amount of alanine accounted for about 57 % of total amino acids(26.1-28.7 %). The proximate composition, minerals, and amino acid of Pyropia dentata were different depending on the coastal ecological habitats.

Comparison of the Amino-Acid Content in Pharmacopuncture Extracts Taken from a Scorpion's Body and from Its Tail

  • Objective: This study was conducted to investigate the amino-acid compositions of pharmacopuncture extracts taken from the body and from the tail of Buthus martensii Karsch, which are frequently prescribed in Oriental medicine. Methods: Amino acids in hot water and 70% ethanol extracts taken from the scorpion's whole body and from its tail were screened by using high performance liquid chromatography (HPLC). The experiments were performed with linearity, precision and accuracy. Results: The results of the amino-acid-composition analysis showed that the Buthus martensii Karsch extracts contained various amino acids such as aspartic acid, histidine, alanine, tyrosine, and cystine. The amino-acid analysis showed that the hot water extract was more beneficial than the ethanol extract, except for histidine. The amino acids from the tail and the body of the scorpion were compared, and the concentration of aspartic acid in the extract from the scorpion's tail was two times that found in the extract from its body. The results of validation experiments were all satisfactory. Conclusion: Studies on the ingredients in extracts from a scorpion other than buthotoxin may demonstrate that the antiepileptic efficacy, anticancer activity, anti-thrombotic action and analgesic effect are enhanced. Using only the tail of the scorpion when pharmacopuncture is dispensed may be beneficial because the extracts from the tail of the scorpion have higher potency than those from the whole body.

  • Seed lipids and proteins of Crotalaria juncea L were analyzed for fatty acids and amino acids respectively. Gas chromatographic analysis of the oil gave palmitic acid (16.01%), stearic acid (7.29%), oleic acid (14.41%), linoleic acid (54.44%) and linolenic acid (7.86%). The defatted seed cake contained all the essential amino acids except methionine and six non-essential amino acids.

Phylogenetic study of penicillium chrysogenum based on the amino acid sequence analysis of chitin synthase

  • The phylogenetic study of Penicilium chrysogenum was performed based on amino acid sequence comparison of chitin synthase. Phylogenetic trees were constructed with the deduced amino acid sequences of the highly conserved region of chitin synthease gene fragments amplified by PCR. The BlasP similarity searcch and the bootstrap analysis of the deduced amino acid sequences of chitin synthase from P. chrysogenum with those form other fungi showed a close evolutionary relationship of Penicillium to ascomycetous fungi, especially to genus Aspergilus. The result from bootstrap analysis of the deduced amino acid sequences of the Class II chitin synthase from ascomyceteous fungi supported the usefulness of the Class II chitin synthease for phylogenetic study of filamentous fungi.
