  • Discoloration by heating and gamma-radiation of aqueous solutions of two coal-tar food dyes used in meat products was observed. Tartrazin with sodium tri-polyphosphate or cinnamon oil was slightly discolorized by heating at $75^{\circ}C$, but Amaranth was comparatively resistant. Effect of gamma rays of coal-tar food dyes was generally increased with irradiation doses, and especially by the addition of sodium tri-polyphosphate and cinnamon oil. This tendency was higher in Tartrazin than Amaranth. The addition of sodium nitrate and ascorbic acid, and storage at low temperature, however, seems to be effective in decreasing the discoloration of irradiated solutions.

Optimization of Extraction Conditions of Polyphenolic Compounds from Amaranth Leaf using Statistically-based Optimization (통계학적 최적화를 이용한 아마란스 잎으로부터 폴리페놀 열수추출조건 최적화)

  • This study examined the optimization of hot-water extraction conditions for maximizing the total polyphenol compounds (TPC) extracted from amaranth leaf. The effects of three independent variables, including extraction temperature, extraction time and ethanol concentration on TPC were investigated using central composite design (CCD). The concentration of TPC increased with increased levels of extraction temperature and time. The extraction temperature and the ethanol concentration showed the significant effect on TPC production (p<0.05). The predicted values at the optimized condition were acceptable when compared to the experimental values ($R^2=0.9566$). The optimum extraction conditions were as follows: temperature of $90.1^{\circ}C$, time of 50 min and ethanol concentration of 61.6% (v/v) for the maximum TPC of 12.6 mg GAE/g DM.

Comparisons of Characteristics of Amaranth Starches Isolated from five Cultivars Grown in Korea (국내산 아마란스로부터 분리한 전분의 특성 비교)

  • The physicochemical and pasting properties of amaranth starches isolated from five cultivars, Andy, Suvarna, Nu World, K266-1, K432, grown in Korea, were investigated. The shapes of starch granules were all polygonal and size was in the range $1.14{\sim}1.48\;{\mu}m$ for all five cultivars. X-ray diffractograms were shown typical A type diffraction patterns for all amaranth starches. The protein and crude lipid contents of starches were $0.13{\sim}0.23%$ and $0.01{\sim}0.05%$, respectively. The apparent amylose contents ranged from $2.79{\sim}4.35%$ and the water binding capacities were $128.05{\sim}135.80%$. The transmittances of 0.1% amaranth starch suspensions except K266-1 increased rapidly above $65^{\circ}C$, thereafter increased slowely. The initial pasting and peak temperature ranges of five cultivars by RVA were $71.3{\sim}73.7^{\circ}C$ and $81.5{\sim}84.0^{\circ}C$, respectively. The peak and cooling viscosities followed the order : Nu World>Andy>Suvarna>K432>>K266-1. Nu World was shown the highest peak (166), final (103) and cooling viscosities (30 RVU). K266-I exhibited the lowest setback (-38) of all five cultivars. Gelatinization (To) and peak temperature (Tp) of amaranth starches in DSC thermograms were $65.7{\sim}68.0^{\circ}C$ and $70.6{\sim}75.8^{\circ}C$, respectively. Functional Cosmetic Effect of Porcine Placeta (Porcine Placenta의 기능성 화장품소재 특성)

  • Porcine placenta was treated with alkali, acid and enzyme treatment to obtain extracts. Heavy metal contents such as Pb, As, and Hg were low enough to satisfy cosmetic agent standard. As a result of safety test(MTT assay), porcine placenta extracts showed over 80% of cell viability at $50{\mu}g/ml$, and cell toxicity was relatively lower. From antioxidation test using DPPH free radical scavenging assay, antioxidation effect was highest as 63% at $50{\mu}g/ml$ when porcine placenta was treated with alkali in pH 9. From whitening effect test using tyrosinase inhibition assay, tyrosinase inhibition effect was 30% at $50{\mu}g/ml$ concentration in alkali treated procine placenta, however, the efficiency was lower compared with arbutin or vitamin C. In anti-wrinkle effect test from elastase inhibition assay, elastase inhibition effects were 20~30% at $50{\mu}g/ml$ for 5 kinds of porcine placenta treatments, which was superior to standard, and especially, protease treated extracts showed best results. Skin formulation including 1% porcine placenta was made and the formulation was very stable for temperature and storage period. From this research, porcine placenta extract showed high potential for anti-wrinkle functional cosmetic agent.

Effect of Growing Period on the Dry Matter Productivity and Grain Yield of Amaranth (Amarnathus caudatus) (아마란스의 생육기간에 따른 건물생산성 및 종실 수량)

  • Amaranth (Amaranth caudatus) is attracting attention as a preference crop in Gyeongsangbuk-do province. To determine its growth potential and cultivation requirements, we investigated its growth characteristics, dry matter productivity, and grain yield according to the growing period. Growth and dry matter productivity were significantly higher for plants that were sown on May $10^{th}$ when the temperature was the highest, whereas the yield was significantly higher for plants that were sown on April $10^{th}$. Amaranth grain yield ranged from 96 to 243 kg according to the sowing date and cultivation year. The optimum harvest time for plants that were sown on April $10^{th}$, May $10^{th}$, and June $10^{th}$ were 120, 110, and 110 days after seeding, respectively. The mean temperature and growing period had a significant quadratic function with yield. Based on these equations, the optimum growing temperature was estimated as $20.6^{\circ}C$ and the optimum growing period as 104-119 days after seeding.

Efficacy of Amaranth(Amaranthus spp. L.) Plant as a Natural Dye Resource: Focused on Wool Dyeing (아마란스 식물의 천연염재로서의 유효성 연구: 모직물 염색을 중심으로)

  • In this study, the efficacy of Amaranth(Amaranthus spp. L.) as a natural dye resource was investigated for wool fabrics. It is known that a large amount of flavonoid and anthocyanin colorant are contained in leaves and stems, as well as red flowers. The optimum condition of dyeing was 1.3% of dye concentration(o.w.b.) at 100℃ for 60 minutes, resulting the K/S value, 23.43 and R Munsell color on the wool fabrics. Al, Fe, Zinc and Titanium were used as a mordant. The mordant improved the dye uptake, regardless of the mordant type and mordant method. The pre-mordanting method was more effective than the post-mordanting method. Al pre-mordanted fabric showed the highest K/S, 30.02. Light fastness and washing fastness were high in grades 4-5 and 5, and rubbing fastness was good in grades 4 and 4-5 in dry condition, but low in grades 2-3 and 3 in wet condition. The dry cleaning fastness was excellent in all 5 grades. However, the alkaline perspiration fastness ratings were low in grades 2-3 and 3. The results show Amaranthus spp. L. colorant can be used as a functional natural dye for wool fabrics.

Energy Generation Coupled to Azoreduction by Membranous Vesicles from Shewanella decolorationis S12

  • Previous studies have demonstrated that Shewanella decolorationis S12 can grow on the azo compound amaranth as the sole electron acceptor. Thus, to explore the mechanism of energy generation in this metabolism, membranous vesicles (MVs) were prepared and the mechanism of energy generation was investigated. The membrane, which was fragmentized during preparation, automatically formed vesicles ranging from 37.5-112.5 nm in diameter under electron micrograph observation. Energy was conserved when coupling the azoreduction by the MVs of an azo compound or Fe(III) as the sole electron acceptor with $H_2$, formate, or lactate as the electron donor. The amaranth reduction by the vesicles was found to be inhibited by specific respiratory inhibitors, including $Cu^{2+}$ ions, dicumarol, stigmatellin, and metyrapone, indicating that the azoreduction was indeed a respiration reaction. This finding was further confirmed by the fact that the ATP synthesis was repressed by the ATPase inhibitor N,N'-dicyclohexylcarbodiimide (DCCD). Therefore, this study offers solid evidence of a mechanism of microbial dissimilatory azoreduction on a subcell level.

Evaluation of Genetic Structure of Amaranth Accessions from the United States

  • Amaranths (Amaranthus sp.), an endemic American crop, are now grown widely across the world. This study used 14 simple sequence repeat (SSR) markers to analyze the genetic diversity of 74 amaranth accessions from the United States, with eight accessions from Australia as controls. One hundred twenty-two alleles, averaging eight alleles per locus, were observed. The average major allele frequency, expected heterozygosity, and polymorphism information content (PIC) were 0.44, 0.69, and 0.65, respectively. The structure analysis based on genetic distance classified 77 accessions (94%) into three clusters, while five accessions (6%) were admixtures. Among the three clusters, Cluster 3 had the highest allele number and PIC values, while Cluster 2 had the lowest. The lowest FST was between Clusters 1 and 3, indicating that these two clusters have higher gene flow between them compared to the others. This finding was reasonable because Cluster 2 included most of the Australian accessions. These results indicated satisfactory genetic diversity among U.S. amaranths. These findings can be used to design effective breeding programs involving different plant characteristics.

Analysis of Oxalic Acid of Various Vegetables Consumed in Korea

  • Many vegetables contain oxalate at various levels depending on their type and family. Oxalate is known to reduce mineral bioavailability from foods. The following study was conducted to determine soluble and total oxalate contents in 32 plant samples commonly consumed in Korea using high-performance liquid chromatography (HPLC). Chard, amaranth, and spinach contained total oxalate of >1,000 mg/100 g. Approximately 45% of the oxalate in amaranth is insoluble, whereas 74.46 and 92.45% of the oxalates in chard and spinach, respectively, was soluble oxalates, which may be removed by blanching. Eggplant, carrot, leak, ginger root, spinach, burdock, and sweet pepper contained more than 90% soluble oxalate in total oxalate content. However, all oxalates detected in lettuce and celery were insoluble. Oxalate was not detected in shepherd's purse, bellflower root, garlic, radish root, broccoli, cabbage, onion, lotus root, adlay, cucumber, kale, and pumpkin. These observations provide useful information needed for selection of vegetables.

Effect of Sodium Taurodeoxycholate on Biliary Excretion of Amaranth as an Anionic Model Drug in Rats (음이온 모델 화합물 아마란스의 담즙배설에 미치는 타우로데옥시콜레이트의 영향)

  • Plasma disappearance of amaranth (AM), a model compound of organic anionic drugs, was retarded by intravenous infusion of taurodeoxycholate (TDC), a representative bile acid, in the rat. Biliary excretion accounted for 30-60% of the systemic excretion of AM. AM seemed to be metabolised in the hepatocyte to form a compound that is excreted more rapidly into the bile than AM itself, considering apparent biliary clearance, $CL_{bil}$, is much larger than systemic clearance, $CL_s$. Decrease in $CL_{bil}$ by TDC infusion might be due to elevated plasma level rather than decreased biliary excretion of AM. Decreased distribution or urinary excretion of AM by TDC was supposed to be one of the probable reasons of elevated plasma level. Competitive inhibition between AM and TDC on tissue distribution and urinary excretion might explain the mechanism. The effect of TDC on the $CL_{bil}$ of methylene blue, a cationic dye, was quite different from that of AM, as reported previously by us. More intensive study would be necessary to elucidate the difference of biliary excretion between organic anions and cations.
