The uncertainty of extreme wind speeds is one key contributor to the uncertainty of wind loads and their effects on structures. The probability distribution of annual extreme wind speeds may be characterized using a classical Gumbel Type distribution. The expression that establishes the relationship between the extreme wind speeds at different recurrence periods and the corresponding coefficients of variation is formulated, and its efficacy is validated. The coefficients of variation are calibrated to be about 0.125 and 0.184 according to defined Chinese and US design specifications, respectively. Based on the wind data of 54 cities in China, 49 meteorological stations in the US, 3 stations in Singapore, the coefficients span intervals of (0.1, 0.35), (0.08, 0.20) and (0.06, 0.14), respectively. For hurricanes in the US, the coefficients range approximately from 0.3 to 0.4. This convenient technique is recommended as one alternative tool for coefficient of variation analyses in the future revisions of related codes. The sensitivities of coefficients of variation for 49 meteorological stations in the US are quantified and demonstrated. Some contradictions and incompatibilities can be clearly detected and illustrated by comparing the coefficients of variation obtained with different combinations of recurrence period wind data.

Bacterial cell-based biosensors, or whole-cell bioreporters (WCBs), are an alternative tool for the quantification of hazardous materials. Most WCBs share similar working mechanisms. In brief, the recognition of a target by sensing domains induces a biological event, such as changes in protein conformation or gene expression, providing a basis for quantification. WCBs targeting heavy metal(loid)s employ metalloregulators as sensing domains and control the expression of genes in the presence of target metal(loid) ions, but the diversity of targets, specificity, and sensitivity of these WCBs are limited. In this study, we genetically engineered the metal-binding loop (MBL) of ZntR, which controls the znt-operon in Escherichia coli. In the MBL of ZntR, three Cys sites interact with metal ions. Based on the crystal structure of ZntR, MBL sequences were modified by site-directed mutagenesis. As a result, the metal-sensing properties of WCBs differed depending on amino acid sequences and the new selectivity to Cr or Pb was observed. Although there is room for improvement, our results support the use of currently available WCBs as a platform to generate new WCBs to target other environmental pollutants including metal(loid)s.
Purpose: Two-stage ${\bar{X}}$ chart is a useful tool for process control when a surrogate variable may be used together with a performance variable. This paper extends the two-stage ${\bar{X}}$ chart to a three stage version by decomposing the first stage into the preliminary stage and the main stage. Methods: The expected cost function is derived using Markov-chain approach. The optimal designs are found for numerical examples using a genetic algorithm combined with a pattern search algorithm and compared to those of the two-stage ${\bar{X}}$ chart. Sensitivity analysis is performed to see the parameter effects. Results: The proposed design outperforms the optimal design of the two-stage ${\bar{X}}$ chart in terms of the expected cost per unit time unless the correlation between the performance and surrogate variables is modest and the shift in process mean is smallish. Conclusion: Three-stage ${\bar{X}}$ chart may be a useful alternative to the two-stage ${\bar{X}}$ chart especially when the correlation between the performance and surrogate variables is relatively high and the shift in process mean is on the small side.
Postharvest diseases cause losses in a wide variety of crops around the world. Irradiation, a useful nonchemical approach, has been used as an alternative treatment for fungicide to control plant fungal pathogens. For a preliminary study, ionizing radiations (gamma, X-ray, or e-beam irradiation) were evaluated for their antifungal activity against Botrytis cinerea, Penicillium expansum, and Rhizopus stolonifer through mycelial growth, spore germination, and morphological analysis under various conditions. Different fungi exhibited different radiosensitivity. The inhibition of fungal growth showed in a dose-dependent manner. Three fungal pathogens have greater sensitivity to the e-beam treatment compared to gamma or X-ray irradiations. The inactivation of individual fungal-viability to different irradiations can be considered between 3-4 kGy for B. cinerea and 1-2 kGy for P. expansum and R. stolonifer based on the radiosensitive and radio-resistant species, respectively. These preliminary data will provide critical information to control postharvest diseases through radiation.

Raman spectroscopy provides many advantages compared to other common analytical techniques due to its ability of rapid and accurate identification of unknown specimens as well as simple sample preparation. Here, we described potential of Raman spectroscopic technique as an efficient and high throughput method to detect plants infected by economically important viruses. To enhance the detection sensitivity of Raman measurement, surface enhanced Raman scattering (SERS) was employed. Spectra of extracts from healthy and Turnip yellow mosaic virus (TYMV) infected Chinese cabbage leaves were collected by mixing with gold (Au) nanoparticles. Our result showed that TYMV infected plants could be discriminated from non-infected healthy plants, suggesting the current method described here would be an alternative potential tool to screen virus-infection of plants in fields although it needs more studies to generalize the technique.

The integrity of steam generator tubes in nuclear power plant should be maintained sufficiently during operation. For sake of this, complicated assessment procedures are required such as fracture mechanics analysis, etc. The integrity assessment of tubes has been performed by using conventional deterministic approaches while there are many uncertainties to carry out a rational evaluation. In this respect, probabilistic integrity assessment is considered as an alternative method for integrity assessment. The objectives of this study are to develop an integrity assessment system based on probabilistic fracture mechanics and to predict the failure probability of steam generator tubes containing an axial through-wall crack. The developed integrity assessment system consists of three evaluation modules, which apply first order reliability method, second order reliability method and Monte Carlo simulation method, respectively. The system has been applied to predict failure probability of steam generator tubes and the estimation results showed a promising applicability of the probabilistic integrity assessment system.
In this study, we explored a new approach for generating ions of organics and biomolecules using contactless atmospheric pressure ionization (C-API). That is, a tapered capillary (~20 cm) was connected to a syringe, which was coupled to a syringe pump for providing a given flow rate to introduce sample solution to the proximity of a mass spectrometer. The gas phase ions derived from analytes were readily formed in the capillary outlet, which was very close to the mass spectrometer (~1 mm). No external electric connection was applied on the capillary emitter. This setup is very simple, but it can function as an ion source. This approach can be readily used for the analysis of small molecules such as amino acids and large molecules such as peptides and proteins. The limit of the detection of this approach was estimated to be ~10 pM when using bradykinin as the sample. Thus, we believe that this approach should be very useful for being used as an alternative ion source because of its low cost, high sensitivity, simplicity, and ease of operation.

Fucoidan (fucan sulfate) is a fucose-containing sulfated polysaccharide from brown algae such as Fucus vesiculosus, Ecklonia kurome, and Cladosiphon okamuranus. The aim of this study was to investigate the effect of fucoidan on the expression of diabetes-related genes in mouse cell line 3T3-L1. 3T3-L1 adipocytes were cultured for 48 hr with or without fucoidan (10, 100, and 500 ppm) on a 60 mm dish. Reverse transcription polymerase chain reaction (RT-PCR) was used for measurement of peroxisome proliferators activated receptor ${\gamma}\;(PPAR{\gamma})$, CCAAT/enhancer binding protein ${\alpha}\;(C/EBP{\gamma})$, and glucose transporter 4 (GLUT4) RT-PCR analysis revealed that expression level of GLUT4, $PPAR{\gamma}$, and $C/EBP{\alpha}$ mRNAs increased with fucoidan treatment from 10 to 500 ppm in a dose-dependent manner. Fucoidan appears to enhance insulin sensitivity by increasing the expression level of diabetes-related genes in 3T3-L1 adipocytes. Therefore, fucoidan is potentially useful as a natural therapeutic material for hyperglycemia in type II diabetes patients.
As an alternative to radioimmunoassay a simple and highly sensitive enzyme immunoassay (EIA) was developed and validated for androstenedione quantification in plasma of Karan Fries bulls using second antibody coating technique. The wells of the microtitreplate were coated with affinity-purified goat immunoglobulin (antirabbit IgG) that binds the hormone specific antibody. The EIA was performed to analyze androstenedione directly in $40{\mu}l$ of bull plasma. The androstenedione standards ranged from 0.20 to 200 pg/$40{\mu}l$/well and the sensitivity of the assay was 5 pg/ml plasma. Serially diluted bull plasma containing high endogenous androstenedione showed good parallelism with bovine androstenedione standard curve. Intra- and inter-assay coefficients of variation (CV) were found to be 8 and 9%, respectively. Peripheral plasma androstenedione concentrations determined in young and adult bull samples ranged between 104-990 pg/ml and 184-2040 pg/ml, respectively.
A rapid, accurate and precise method for the determination of 22 amino acids in Isatidis Radix by Hydrophilic Interaction Ultra-High-Performance Liquid Chromatography Coupled with Triple-Quadrupole Mass Spectrometry (HILIC-UPLC-MS/MS) was established. Chromatographic separation was carried out on a Acquity UPLC BEH Amide column ($2.1mm{\times}100mm$, $1.7{\mu}m$) with gradient elution of acetonitrile (containing 0.05% formic acid and 2 mM ammonium formate) and water (containing 0.15% formic acid and 10 mM ammonium formate) at a flow rate of 0.4 mL/min; Waters Xevo$^{TM}$ TQ worked in multiple reaction monitoring mode. All components were separated in 17 min. All calibration curves were linear ($R^2$ > 0.991) over the tested ranges. The limits of detection (LOD) and limits of quantitation (LOQ) for these compounds were 0.21-79.55 and 0.72-294.23 ng/mL, respectively. The average recoveries were in the range of 93.75-104.16% with RSD value less than 6.56%. Therefore, this method could be an alternative assay for the determination of 22 amino acids in Isatidis Radix due to its rapidness, sensitivity, less sample and solvent consumption.
