Development of Melanotropin Antagonists: Investigating Potent and Specific Ligands for New Receptors

  • ${\alpha}$-Melanotropin (Ac-Ser-Tyr- Ser-Met-Glu$\^$5/-His-Phe-Arg-Trp-Gly$\^$10/-Lys-Pro-Val-NH$_2$) is one of the first peptide hormones to be isolated and have its structure determined. It was early recognized to have essentially the same N-terminal tridecapeptide sequence as adrenocorticotropic hormone (ACTH) except that the N-terminal was acetylated in the case of ${\alpha}$-MSH but not in the case of ACTH, indicating that their biosyntheses were different (Figure 1). Subsequently it was discovered that ${\alpha}$-MSH and ACTH were derived from the same gene, currently referred to as proopiomelanocortin (POMC). Its original bioactivity was pigmentation, but it also was recognized that it may have activity in the central nervous system, though the precise nature of these central activities have been controversial. The recent cloning and expression of five melanocortin receptors, with the MC3 and MC4 receptors found primarily in the brain and the MC5 receptor (MC5-R) found throughout the body, has provided new impetus to understand the structure-activity relationships of ${\alpha}$-MSH at these receptors. The effects of ${\alpha}$-MSH on pigmentation are mediated by the MC1-R expressed specifically on the surface of melanocytes. Similarly the MC2-R is involved in the regulation of adrenal steroidogenesis by ACTH. However, given the complexity of expression of the MC3, MC4, and MC5 receptors, it has not been possible to identify any simple correlations between these receptors and the reported biological activities of the melanocortin peptides. Consequently, potent and receptor specific agonists and especially antagonists would be extremely valuable tools for the determination of the physiological roles of the MC3, MC4, and MC5 receptors. Though the extensive structure-activity relationships have provided much information on agonist activity related to pigmentary effects, only recently has it been possible to begin to systematically develop potent and selective antagonists.

Nanotubular Structure Formation on Ti-6Al-4V and Ti-Ta Alloy Surfaces by Electrochemical Methods

  • Nanotubular structure formation on the Ti-6Al-4V and Ti-Ta alloy surfaces by electrochemical methods has been studied using the anodizing method. A nanotube layer was formed on Ti alloys in 1.0 M $H_3PO_4$ electrolyte with small additions of $F^-$ ions. The nanotube nucleation and growth of the ${\alpha}$-phase and ${\beta}$-phase appeared differently, and showed different morphology for Cp-Ti, Ti-6Al-4V and Ti-Ta alloys. In the ${\alpha}$-phase of Cp-Ti and martensite ${\alpha}^{\prime}$ and in the ${\alpha}^{{\prime}{\prime}}$ and ${\beta}$-phase of the Ti-Ta alloy, the nanotube showed a clearly highly ordered $TiO_2$ layer. In the case of the Ti-Ta alloy, the pore size of the nanotube was smaller than that of the Cp-Ti due to the ${\beta}$-stabilizing Ta element. In the case of the Ti-6Al-4V alloy, the ${\alpha}$-phase showed a stable porous structure; the ${\beta}$-phase was dissolved entirely. The nanotube with two-size scale and high order showed itself on Ti-Ta alloys with increasing Ta content.

탁주 2단 담금시 ${\alpha}G-Hesperidine$의 주질 변화 (Change of Takju Qualities during the Second Brewing Process by Addition of ${\alpha}G-Hesperidine$)

  • 2단 담금시 ${\alpha}G$-헤스페리딘에 대한 막걸리의 주질 개선 효과 가능성 여부를 검토하였다. 알코올 생성량이 대조구보다 증가하였으며 경시적으로 생성량에는 큰 차이를 나타내었다. 환원당은 ${\alpha}G$-헤스페리딘을 첨가한 경우2담 담금2일째는 많은 양의 환원당이 생성되었으나 4일-6일 사이에는 감소하였다. ${\alpha}G$-헤스페리딘 첨가의 경우산도는 담금2일째 이후에는 거의 변화하지 않았다. 또 ${\alpha}G$-헤스페리딘을 첨가한 경우 fusel oil의 생성이 적게 일어났다. 효모는 ${\alpha}G$-헤스페리딘의 경우에 발효 전기간 동안 효모증식이 잘 이루어졌다. 저장 중 막걸리의 침전속도는 ${\alpha}G$-헤스페리딘을 첨가한 경우는 대조구에 비해서 거의 1.5배 낮은 속도로 침전되었으며 침전량은 경시적으로 거의 변화하지 않았다. ${\alpha}G$-헤스페리딘을 첨가한 경우 막걸리의 떫은맛과 쓴맛은 다소 감소하고 탁도, 신선함은 대조구보다 높은 것으로 나타났다. 전체적인 기호도는 대조구보다 ${\alpha}G$-헤스페리딘을 첨가한 것이 좋은 것으로 나타났다. 막걸리는 저장 중 발생하는 이취는 ${\alpha}G$-헤스페리딘을 첨가한 경우 저장 6일째부터 감지되었다.

이산화탄소와 α-Tetralol과의 2성분계 고압상평형 측정 (High Pressure Binary Phase Equilibria Measurements of α-Tetralol with Carbon Dioxide)

  • 313.2K, 343.2K 그리고 373.2K 온도와 압력 6.0MPa에서 35.0MPa 범위까지 이산화탄소와 방향족 탄화수소인 1,2,3,4-tetrahydro -1-naphthol($={\alpha}$-tetralol)과의 상평형 실험을 각 온도에서 압력을 변화시키면서 수행하였다. 상평형 장치는 기상과 액상을 동시에 순환시키는 순환형이 사용되었으며, 실험분석은 미량시료채취법으로 시료를 채취하여 분석하였다. 이산화탄소와 ${\alpha}$-tetralol계의 실험은 기상과 액상에서의 상평형데이터 및 혼합물 밀도의 측정이 실험조건별로 이루어졌다. 실험결과 일정압력에서 이산화탄소의 용해도는 액상에서 온도가 증가함에 따라 감소하였고, 기상에서 ${\alpha}$-tetralol의 용해도는 온도가 증가할수록 증가하였다. 혼합물 밀도는 압력이 증가할수록 기상, 액상 모두 혼합물 임계밀도쪽으로 접근해 감을 알 수 있었다. 또한 열역학적 해석을 위하여 3차 상태방정식인 Peng-Robinson식을 이용하여 이산화탄소 -${\alpha}$-tetralol계의 실험치를 상호 연관시켜 이론치를 계산하여 실험치와 비교한 결과 AAD가 K(1)의 경우 1.08%~8.98%이고 K(2)의 경우는 45.71%~72.34% 이었다.

Fe-29%Ni-17%Co 저열팽창 합금의 피로 특성에 미치는 알파상의 영향 (Effects of Alpha Phase on the Fatigue Properties of Fe-29%Ni-17%Co Low Thermal Expansion Alloy)

  • The effect of alpha phase on the fatigue properties of Fe-29%Ni-17%Co low thermal expansion alloy was investigated. Two kinds of alloys (Base alloy and Alpha alloy) were prepared by controlling the minimal alloy composition. Microstructure observation, tensile, high-cycle fatigue, and low-cycle fatigue results were measured in this study. The Base alloy microstructure showed typical austenite ${\gamma}$ phase. Alpha alloy represented the dispersed phase in the austenite ${\gamma}$ matrix. As a result of tensile testing, Alpha alloy was found to have higher strengths (Y.S. & T.S.) and lower elongation compared to those of the Base alloy. High cycle fatigue results showed that Alpha alloy had a higher fatigue limit (360MPa) than that (330MPa) of the Base alloy. The Alpha alloy exhibited the superior high cycle fatigue property in all of the fatigue stress conditions. SEM fractography results showed that the alpha phase could act to effectively retard both fatigue crack initiation and crack propagation. In the case of low-cycle fatigue, the Base alloy had longer fatigue life in the high plastic strain amplitude region and the Alpha alloy showed better fatigue property only in the low plastic strain amplitude region. The fatigue deformation behavior of the Fe-29%Ni-17%Co alloy was also discussed as related with its microstructure.

암소저장(暗所貯藏) 및 일사광선조사(日射光線照射) 조건하(條件下)에서의 일부(一部) 항산화제(抗酸化劑)의 항산화효과에 대하여 (Relative Effectiveness of Some Antioxidants in A Dark and A Sunlight-irradiated Condition)

  • 일부(一部) 식품항산화제(食品抗酸化劑)를 주로하는 항산화제(抗酸化劑)들, 즉 BHA, BHT, PG, ${\alpha}-tocopherol$, ascorbic acid (As.A), pyrogallol, 및 antage 3C를 식용대두유(食用大豆油)에 각각(各各) 0.05%의 농도(濃度) 첨가(添加)하여, 일사광선조사(日射光線照射) 조건하(條件下) 및 암소저장(暗所貯藏) 조건하(條件下) $(45.0{\pm}0.5^{\circ}C)$에서의 이들 항산화제(抗酸化劑)의 식용대두유(食用大豆油)에 대한 과산화물형성(過酸化物形成) 억제효과와 유리지방산형성(遊離脂肪酸形成) 억제효과를 비교(比較) 검토(檢討)하였다. 암소저장(暗所貯藏) 조건하(條件下)에서 저장(貯藏)할 때는 ${\alpha}-tocopherol$ 을 제외(除外)한 다른 항산화제(抗酸化劑)들은 비교적 강(强)한 과산화물형성(過酸化物形成) 억제효과를 보였으며, 특히 PG, BHT, antage 3C, As.A 등의 억제효과는 매우 강(强)하였다. 일사광선조사(日射光線照射) 조건하(條件下)에서도 BHT, PG 등은 강(强)하고 지속적(持續的)인 효과를 나타냈었다. 한편 동일조건하(同一條件下)에서의 pyrogallol, antage 3C의 억제효과는 BHA, BHT, PG 등의 식품항산화제(食品抗酸化劑)의 효과를 훨씬 능가하였었다. ${\alpha}-Tocopherol$의 억제효과는 상기의 두 조건하(條件下)에서 다 같이 저장일수(貯藏日數)가 경과(經過)함에 따라 급속(急速)히 상실(喪失)되었다. 일반적(一般的)으로 암소저장조건하(暗所貯藏條件下)에서의 각(各) 항산화제(抗酸化劑)의 과산화물형성(過酸化物形成) 억제효과는 일사광선조사(日射光線照射) 조건하(條件下)에서보다 훨씬 더 효과적인 듯하였다. 각(各) 항산화제(抗酸化劑)의 유리지방산형성(遊離脂肪酸形成) 억제효과는 암소저장(暗所貯藏) 조건하(條件下)에서 더 강력(强力)하고 효과적인 듯하였다. 특히 pyrogallol 은 상기의 두 조건하(條件下)에서 다 같이 강력(强力)한 억제효과를 보여주었다. 또한 모든 항산화제(抗酸化劑)의 일사광선조사(日射光線照射) 조건하(條件下)에서의 유리지방산형성(遊離脂肪酸形成) 억제효과는 동일조건하(同一條件下)에서의 과산화물형성(過酸化物形成) 억제효과와 비교할 때 매우 현저하였다.

Association of Genetic Polymorphism of IL-2 Receptor Subunit and Tuberculosis Case

  • Tuberculosis (TB) is infectious disease caused by Mycobacterium tuberculosis (MTB) infection. It is known that not only the property of microorganism but also the genetic susceptibility of infected patients is controlled. Interleukin 2 (IL-2) is a cytokine belonging to type 1 T helper (Th1) activity. In addition, IL-2, when infected with MTB, binds IL-2 receptor and promotes T cell replication and is involved in granuloma formation. The aim of this study was to investigate the genetic polymorphisms of the IL-2 receptor gene in tuberculosis patients and normal individuals. We analyzed 22 SNPs in three genes using the genotype data of 443 tuberculosis cases and 3,228 healthy controls from the Korea Association Resource for their correlation with tuberculosis case. IL2RA, IL2RB, and IL2RG genes were genotyped of 16, 4, and 2 SNPs, respectively. Among three genes, only IL2RA gene polymorphisms showed statistically significant association with tuberculosis case. 6 SNPs with high significance were identified in the IL2RA gene. In addition, the linkage disequilibrium (LD) structure of IL2RA gene was confirmed. SNP imputation of IL2RA gene was performed, it was confirmed that more SNPs were significant between case and control. If we look at the results of IL2RA gene analysis above, we can see that genetic polymorphism in the gene expressing $IL-2R{\alpha}$ will regulate the expression level of $IL-2R{\alpha}$, and the change in the immune system involved in $IL-2R{\alpha}$. In this study, genetic polymorphism that may affect host immunity suggests that susceptibility to tuberculosis may be controlled.

판상형 충전제의 함량과 배향에 따른 PP복합체의 열팽창계수 영향 연구 (Empirical Study on Effects of Disk Shape Filler Content and Orientation on Thermal Expansion Coefficient of PP Composites)

  • 두 가지의 종횡비(${\rho}_{\alpha}=a_1/a_3$ and ${\rho}_{\beta}=a_1/a_2$)에 의해 특징지어진 3차원 타원체($a_1$ > $a_2$ > $a_3$)를 사용하여 polypropylene 복합체의 판상형 충전제 함량과 방향이 열팽창률에 미치는 영향에 대해 실험적인 연구를 수행하였다. 측정된 실험적인 값은 Lee와 그의 연구자들이 제안한 이론적인 모델에 의한 계산 값과 비교분석되었다. 판상형 충전제로는 운모와 탈크가 사용되었다. 실험 결과로 운모의 경우 종횡비는 ${\rho}_{\alpha}=13.5$, ${\rho}_{\beta}=1.8$이 사용되어, 20 wt% 함량일 때 ${\alpha}_{11}/{\alpha}_m$는 약 0.56으로 감소하였으나, ${\alpha}_{33}/{\alpha}_m$는 오히려 1.018로 증가함을 보였다. 탈크의 경우의 종횡비는 ${\rho}_{\alpha}=3.7$, ${\rho}_{\beta}=1.4$이었고 ${\alpha}_{11}/{\alpha}_m$는 0.63으로 감소하였다. 결국 운모와 탈크 모두 그 함량증가에 따라 종단방향과 횡단방향에서 열팽창률은 감소하였으나 수직방향에서는 초기 낮은 충전제 함량에서는 오히려 증가하는 경향을 보였다.

  • 이 논문은 판상형 충전제의 함량과 배향에 따라 폴리프로필렌 복합체의 영률 변화에 미치는 영향을 연구하였다. 이론적인 예측은 Lee와 그의 연구자들에 의해 제안된 두 개의 종횡비, ${\rho}_{\alpha}=a_1/a_3$${\rho}_{\beta}=a_1/a_2$를 이용하여 3차원 타원체의 형상을 계산하였고 실험값과 비교 분석하였다. 판상형 충전제로 운모와 탈크를 사용하였다. 충전제의 형태는 SEM을 이용해 관찰하였고, 종횡비는 고분자의 흐름을 기준으로 좌표축을 설정하여 통계적 방법으로 계산하였다. 충전제가 운모인 경우에 ${\rho}_{\alpha}=13.5$${\rho}_{\beta}=1.8$이었고, 탈크의 경우는 ${\rho}_{\alpha}=3.8$${\rho}_{\beta}=1.4$이었다. 또한 충전제의 2개의 종횡비와 함량이 기계적 물성에 미치는 영향을 실험적으로 연구하였다. 운모의 경우 30 wt% 함량일 때 $E_{11}$이 약 2.7배 향상되었다. 탈크의 경우는 $E_{11}$이 약 2.3배 향상하였다.

동력경운기의 경사지견인 및 주행특성에 관한 연구 (II)-동력경운기-트레일러계의 욍골동 및 동횡전도한계 (Study on the Travel and Tractive Characteristics of The Two-Wheel Tractor on the General Slope Ground (II)-Dynamic Side-overturn of the Tiller-trailer System-)

  • Power tiller is a major unit of agricultural machinery being used on farms in Korea. About 180.000 units are introduced by 1977 and the demand for power tiller is continuously increasing as the farm mechanization progress. Major farming operations done by power tiller are the tillage, pumping, spraying, threshing, and hauling by exchanging the corresponding implements. In addition to their use on a relatively mild slope ground at present, it is also expected that many of power tillers could be operated on much inclined land to be developed by upland enlargement programmed. Therefore, research should be undertaken to solve many problems related to an effective untilization of power tillers on slope ground. The major objective of this study was to find out the travelling and tractive characteristics of power tillers being operated on general slope ground.In order to find out the critical travelling velocity and stability limit of slope ground for the side sliding and the dynamic side overturn of the tiller and tiller-trailer system, the mathematical model was developed based on a simplified physical model. The results analyzed through the model may be summarized as follows; (1) In case of no collision with an obstacle on ground, the equation of the dynamic side overturn developed was: $$\sum_n^{i=1}W_ia_s(cos\alpha cos\phi-{\frac {C_1V^2sin\phi}{gRcos\beta})-I_{AB}\frac {v^2}{Rr}}=0$$ In case of collision with an obstacle on ground, the equation was: $$\sum_n^{i=1}W_ia_s\{cos\alpha(1-sin\phi_1)-{\frac {C_1V^2sin\phi}{gRcos\beta}\}-\frac {1}{2}I_{TP} \( {\frac {2kV_2} {d_1+d_2}\)-I_{AB}{\frac{V^2}{Rr}} \( \frac {\pi}{2}-\frac {\pi}{180}\phi_2 \} = 0 $$ (2) As the angle of steering direction was increased, the critical travelling veloc\ulcornerities of side sliding and dynamic side overturn were decreased. (3) The critical travelling velocity was influenced by both the side slope angle .and the direct angle. In case of no collision with an obstacle, the critical velocity $V_c$ was 2.76-4.83m/sec at $\alpha=0^\circ$, $\beta=20^\circ$ ; and in case of collision with an obstacle, the critical velocity $V_{cc}$ was 1.39-1.5m/sec at $\alpha=0^\circ$, $\beta=20^\circ$ (4) In case of no collision with an obstacle, the dynamic side overturn was stimu\ulcornerlated by the carrying load but in case of collision with an obstacle, the danger of the dynamic side overturn was decreased by the carrying load. (5) When the system travels downward with the first set of high speed the limit {)f slope angle of side sliding was $\beta=5^\circ-10^\circ$ and when travels upward with the first set of high speed, the limit of angle of side sliding was $\beta=10^\circ-17.4^\circ$ (6) In case of running downward with the first set of high speed and collision with an obstacle, the limit of slope angle of the dynamic side overturn was = $12^\circ-17^\circ$ and in case of running upward with the first set of high speed and collision <>f upper wheels with an obstacle, the limit of slope angle of dynamic side overturn collision of upper wheels against an obstacle was $\beta=22^\circ-33^\circ$ at $\alpha=0^\circ -17.4^\circ$, respectively. (7) In case of running up and downward with the first set of high speed and no collision with an obstacle, the limit of slope angle of dynamic side overturn was $\beta=30^\circ-35^\circ$ (8) When the power tiller without implement attached travels up and down on the general slope ground with first set of high speed, the limit of slope angle of dynamic side overturn was $\beta=32^\circ-39^\circ$ in case of no collision with an obstacle, and $\beta=11^\circ-22^\circ$ in case of collision with an obstacle, respectively.
