Immune-Enhancing Alkali-Soluble Glucans Produced by Wild-Type and Mutant Saccharomyces cerevisiae

  • The alkali-soluble glucan of the yeast cell wall contains $\beta-(1,3)-$ and (1,6)-D-linkages and is known to systemically enhance the immune system. In the previous study [6], in order to isolate cell wall mutants, a wild-type strain was mutagenized by exposure to ultraviolet light, and the mutants were then selected via treatment with laminarinase $(endo-\beta-(1,3)-D-glucanase)$. The mass of alkali- and water-soluble glucans produced by the mutant was measured to be 33.8 mg/g of the dry mass of the yeast cell. Our results showed that the mutants generated the amount of alkali-soluble glucan 10-fold higher than that generated by the wild-type. Structural analysis showed that the alkali-soluble glucan from the mutants was associated with a higher degree of $\beta-(1,6)-D-linkage$ than was observed in conjunction with the wild-type. Yeast cell wall $\beta-glucan$ was shown to interact with macrophages via receptors, thereby inducing the release of tumor necrosis factor alpha $(TNF-\alpha)$ and nitric oxide. Alkali-soluble $\beta-glucans$, both from water-soluble and water-insoluble glucan, exhibited a higher degree of macrophage activity with regard to both the secretion of tumor necrosis factor alpha $(TNF-\alpha)$ and nitric oxide and direct phagocytosis, than did the positive control ($1{\mu}g$ of lipopolysaccharide).

클링커 제조 조건에 따른 수용성 6가 크롬 용출 특성 (Leaching Properties of Water-Soluble Hexavalent Chromium by Manufacturing Condition of Cement Clinker)

  • One of the trace constituents included in cement clinker, chromium, has become prominent and highly noticed lately as a social issue both inside and outside of this country because it affects the human body negatively. The purpose of the present study was to investigate leaching properties of water-soluble hexavalent chromium by different manufacturing conditions of cement clinker. Raw materials were prepared to add different $SiO_2$, $Al_2O_3$ and $Fe_2O_3$ sources. After the raw materials, such as limestone, sand and clay, iron ore was pulverized and mixed, and the raw meal was burnt at $1450^{\circ}C$ in a furnace with an oxidizing atmosphere. Leaching of soluble hexavalent chromium showed a tendency to decrease with an increasing LSF and IM. However, leaching of soluble hexavalent chromium increased with an increasing S.M. Alkali contents of iron source minerals is closely related to the leaching properties of soluble hexavalent chromium. Green sludge has the highest content of alkali added; leaching of water-soluble hexavalent chromium was mostly high. In order to reduce the water-soluble hexavalent chromium in cement, reducing the alkali content in raw materials is important.

  • 최근에 날로 강화되는 환경 규제, 그리고 수요자의 필요에 의해 수용성 그리고 알칼리 가용성 점착제에 대한 많은 제품 개발이 이루어지고 있다. 본 연구실에서는 그러한 특성을 정량적으로 정확하게 측정할 수 있는 시험법을 개발하여 KS 및 ISO 시험법으로 제정하였다. 본 연구에서는 이 시험법을 이용하여 수용성과 알칼리 가용성 점착제의 시간에 따른 수용화와 알칼리 가용화 정도를 측정하였고 측정결과 분석을 통해 본 시험법이 안정된 수용성 및 알칼리 가용화 데이터를 생성하고 있음을 보여주었다.

  • Recently, the attraction for water-based ink is increasing, Because Solvent ink, based on organic solvent, cause bad effect on environment and rise the cost. Instead of organic solvent that cause environment pollution as well as cost-rise by rise of petroleum price, Binder based on alkali-soluble resin is popular and studied by many other country. However, in domestic, research and development for alkali-soluble resin is not like, because of lack of understanding environmental pollution and safety, therefor alkali-soluble resin development and research for the properties of the water-based ink used by alkali-soluble resin is not study very well. In this paper, we studied dispersion of carbon black in acrylic resin solution and the properties of water-based ink used by carbon black and acrylic binder.

타닌접착제를 위한 침엽수 수피의 알칼리성 추출물의 양적 변이 (Quantitative Variation of Alkali-soluble Extracts from Coniferous Barks for Tannin-based Adhesives)

  • Alkali-soluble extracts were prepared from barks of Japanese larch(Larix leptolepis). Siberian larch(Larix gmelineii) and Radiata pine(Pinus radiata) The effect of various factors, such as particle size, liquor ratio, extraction temperature, and reaction time, on the extracts yield was investigated. Particle sizes affected the alkali-soluble extracts; the finer the particle size, the higher extracts and extract efficiency. High temperature and high liquor ratio were more effective. In the range of 0.25% to 1% NaOH concentration, alkali extracts were increased with the increasing alkali concentration. However, extract yields were leveled off above 0.5% alkali concentration. Extractions with NaHCO3 were almost equivalent to those of NaOH extraction. 1% NaOH and 1% NaHCO3 resulted in the highest yields of alkaline extracts. Extracts from Japanese larch were lower than those from Siberian larch and Radiata pine barks. Siberian larch bark was the best raw material for tannin adhesives, because its extractive yield was higher than those of the other barks. Concerning pH of alkaline media during extraction, small increases of the extract yields were resulted at range of pH 7 to pH 9, while a large increases from pH 9.0 to pH 12. This phenomenon is attributed to higher alkalinity.

시멘트 클린커 생성과정에 미치는 Potassium의 영향 III. 불안정 알칼리의 상태 및 생성기구 (Influence of Potassium on the Cement Clinker Formation : III. THe State and Formation Mechanism of Unstable Alkali)

  • The formation mechanism of unstable alkali and its existing states in the clinker were studied. The relation of unstable alkali content vs. other water-soluble components porosity and the distributionof potassium were investigated. The results are as follows :1) Two states of unstable alkali seem to exist in clinker ie compounds of $K_2O-Al_2O_3$ system and free $K_2O$ 2) The content of water-soluble $Al_2O_3$ tends to increase with increase of unstable alkali content, 3) Most of alkalies in clinker are concentrated in liquid phase at high temperature. Therefore it is possible to make various $K_2O-Al_2O_3$ system compounds according to the content of $K_2O$ in the liquid phase of clinker. In this experiment we found out a $K_2O-Al_2O_3$ compound of high $Al_2O_3$ content (34%) and high $K_2O$ content (33%) in clinker with 1.09% unstable alkali. 4) The porosity of clinker tends to increase with increase of unstable alkali content. 5) The amount of trapped alkali vapor may increase in closed pore in the clinker with high alkali and low $SO_3$ condecent. Therefore free $K_2O$ is the condensed alkali on the wall of closed pore in the clinker.

감과실의 성숙과 추숙중의 세포벽 구성성분의 변화 (Changes in the Cell Wall Components of Persimmon Fruits during Maturation and Postharvest)

  • 감과실의 성숙과 추숙중 세포벽의 함량은 감소하는 경향이었고, 성숙중에는 pectin질과 알칼리 가용성 hemicellulose의 함량은 증가하는 반면 산 가용성 hemicellulose와 lignin은 감소하였다. 그러나 연시에서는 pectin질과 알칼리 가용성 hemicelloluse의 감소와 산가용성 hemicellulose의 증가가 뚜렷하였다. 그리고 성숙과 추숙중에 cellulose의 함량은 증가하였으며, 생체당 세포벽 다당류의 함량은 감소하였다. 총 pectin질과 불용성 pectin질의 함량은 성숙중에는 증가하였으나 연시에서는 현저히 감소하였으며, 반면에 수용성 pectin질의 함량은 성숙과 추숙중에 증가하였다.

알칼리 용해성 아크릴계 수분산 중합체를 사용한 소각로 비산재의 처리에 관한 실험적 연구 (An Experimental Study on the Treatment of Waste Ash from the Incinerator by Alkali Soluble Acrylic Copolymer Emulsion)

  • 각종 중금속을 흡착하는 알칼리 용해성 아크릴계 수분산 중합체의 특성을 활용하여 광산 폐기물 및 도시폐기물의 소각재 등에서 용출되는 중금속의 처리 방법에 대하여 실험 하였으며 그 결과를 고찰하였다. 실험결과, 수은, 납, 카드뮴, 구리 이온 등에 대한 킬레이트 반응에 의한 알칼리 용해성 아크릴계 수분산 중합체의 효과적인 흡착 효과가 확인되었다. 수중에서 불분리성이고 각종 중금속의 용출을 방지하는 친환경성 가소성 그라우트의 특성을 활용하여 광산 폐기물 및 도시폐기물의 소각재 등에서 용출되는 중금속의 처리를 위한 기초적 용출실험을 실시하였으며 그 결과를 고찰하였다. 실험결과, 알칼리 용해성 수분산 중합체를 첨가한 친환경성 가소성 그라우트의 $Cr^{6+}$ 이온에 대한 효과적인 중금속 흡착 효과가 확인되었다.

  • Effect of alkali $SO_3$ and $C_2S$ content and cooling condition of clinker on the formation of unstable alkali and cooling condition of clinker on the formation of unstable alkali was investigated. As a result alkali and $SO_3$ content of clinker were the main factors of unstable alkali formation and equation (7) in which the two factors were the variables showed a good agreement with the observed results. Water soluble alkali content of sulphate was decreased with increase of $C_2S$ content of clinker resulted by the increase of solid solution rate of alkali to clinker minerals. But change of unsatble alkali content accordint to the increase of $C_2S$ content was slight. A increasing tendency of ignition loss of clinker with increase of unstable alkali content was found.
