Ethanol Production by a New Method of Alginate-Immobilization (새로운 Alginate 고정화 방법에 의한 에탄올 생산)

  • When the cells of yeast K35 were immobilized in Ca-alginate gel, cell concentration and viability decreased as alginate concentration increased. Considering the results, 2% (w/v) Ca-alginate concentration would be suitable. Among various concentrations of additives and cross-lin-king agent, the addition of 1.67% (w/v) of bentonite together with 0.33% (v/v) of glutaraldehyde (ABG bead) resulted in the highest ethanol production of 1.8%(w/v), using YPD medium containing 2% glucose. ABG bead seemed to be more resistant to phosphate ion than Ca-alginate bead. 0.33%(w/v) of phosphate was a proper concentration for the ethanol production by ABG bead. Scanning electron microscopic observation depicted that the immobilized cells on the bead surface were coated by alginate gel and that the cells in the internal bead were cross-linked with alginate matrix. When repeated-batch culture was performed with ABG bead for 40 days in a packed-bed reactor, ethanol concentration of about 90~110 g/l-gel was maintained. Cell viability was maintained around 70%, and outgrowing cell concentration was below 6.3% of total cell concentration. Consequently, the results showed that ABG head was a potential carrier for continuous production of ethanol compared to conventional Ca-alginate bead.

  • Two types of alginate gel beads capable of floating in the gastric cavity were prepared. The first, alginate gell bead containing olive oil(Al-Oil), is a hydrogel bead and its buoyancy is attributable to olive oil held in the alginate gel matrix. The model drug, metronidazole(MZ), contained in Al-Oil was released gradually into artificial gastric fluid. The profiles of MZ release from Al-Oil shown initial burst and after 90 min they were about 100%. The second, alginate gel bead containing curdlan microsphere(Al-C), is a gel bead with curdlan-MZ microsphere in the matrix. To sustained release rate of drug, alginate bead were prepared curdlan microsphere containing MZ. Results demonstrated that sustained delivery of MZ over 2h can be easily achieved while the bead remained float. The release properties of prepared alginate beads are applicable not only for sustained release of drugs but also for targeting the gastric mucosa.

Removal Characteristics of Lead by Immobilizing Agents and Immobilized Seaweed (고정화제와 고정화된 해조류에 의한 납의 제거 특성)

  • In this study, the characteristics of lead removal by PVA and alginate bead which used widely as immobilizing agents were investigated, and the difference of removal amounts between pure PVA/alginate bead and Sargassum thunbergii immobilized bead was studied. All PVA beads, pure and S. thunbergii immobilized, reached an equilibrium state in about 1 hour, and S. thunbergii immobilized bead adsorbed more lead than pure one. But in the case of alginate beads, they needed much time, about 5 hours, to reach an equilibrium state, and adsorbed lead four times higher than PVA beads. Therefore, it was considered that alginate beads had more mass transfer resistance and function groups which adsorb lead such as hydroxyl, carboxyl and etc. than PVA bead. To examine the continuous usage of alginate beads, the process of adsorption/desorption of lead was conducted repeatedly. As the process proceeded, the amounts of lead adsorption decrease, so it was indicated that the non-desorbed lead from alginate bead at first adsorption/desorption process remained constantly.

Development of Alginate-Celite Immobilization Technique for the Improvement of Ethanol Productivity (에탄올 생산성 향상을 위한 Alginate-Celite 고정화 방법의 개발)

  • The optimal initial pH for the ethanol production by Saccharomyces K35 was found to be 5.0, and about 80% of yield was obtained when 200g/$\ell$ of glucose was used as a substrate, which showed sugar tolerant. As the additives and cross-linking agent, the addition of 1.67%(w/v) Celite R-634 together with 0.33%(v/v) of glutaraldehyde(ACG bead) resulted in better stability, ethanol productivity and cell viability than Ca-alginate bead. Also, ACG bead seemed to be more resistant to phosphate ion than Ca-alginate bead, considering outgrowing cell concentration in the media. Scanning electron microscopic observation depicted that the surface of ACG bead was almost similar to the original state but not for Ca-alginate bead. When repealpd-batch culture was performed with Ca-alginate bead for 60 days in a 500m1 Erlenmeyer flask, ethanol and cell concentration were maintained about 138g/$\ell$-gel and 29~30g/$\ell$-gel, respectively, up to 40 days(7th run number), and then both were rapidly decreased. In the case of ACG bead, ethanol and cell concentration were maintained about 130~150g/$\ell$-gel and 32~35g/$\ell$-gel, respectively, up to 60days(10th run number). Cell viability was maintained about 70%, and outgrowing cell concentration was below 5.8% of total cell concentration.

  • Alginic acid is a hydrophilic , colloidal polysaccharide obtained from cell wall of seaweed or brown algae and has a broad range of applications. Alginlc acid becomes alginate gel bead due to its cation-induced gelation. Dried alginate beads can be reswollen according to environmental pH. The purpose of this paper is to explore the possible applicability of alginate beads as an oral controlled release system of ibuprofen. In this experiment ibuprofen was incorporated in alginate beads and alginate beads were treated with various methods. Ibuprofen release from alginate beads in phosphate buffer (pH 7.4) was laster than in distilled water and dilute HCl. The release of ibuprofen was more sustained in bead than simple mixture and coprecipitate of ibuprofen and sodium alginate. The dissolution rate of ibuprofen was decreased in using of bead that hardened with formaldehyde. The dissolution rate of the drug from the bead was the fastest in 12 hour dried beads, 1.5%-sodium alginate concentration and 1%-calcium chloride concentration. Sodium alginate bead can be used as a sustaind release drug delivery system of water-insoluble drugs.

Characteristics of Salt Adsorption by Calcium Alginate Beads (칼슘알긴산비드에 의한 염분의 흡착특성)

  • The adsorption characteristics of sodium chloride into Ca-alginate beads have been investigated and the result were as follows: Sodium chloride uptake by Ca-alginate beads increased with time. The highest uptake volume of sodium chloride was 4.2g after 10 minutes. The uptake volume by Fe, Ca, Ba, and Sr-alginate beads was 5.6g, 4.2g, 4.2g and 4.0g, respectively but in case of Fe-alginate beads, the induced hydrogel beads were very fragile and the strength of Fe-alginate beads were weaker than Ca- and Ba-alginate beads. Mg-alginate bead was not formed and Ca-, Ba- and Sr-alginate beads had a similar uptake volume about 4.2g, respectively. The uptake volume of sodium chloride by CaCl$_2$concentration(0.1M. 0.2M and 1M), curing solution, was 4.8, 4.2g and 4.1g, respectively. The uptake volume by sodium alginate concentration(0.6%, 1% and 2%) was 2.8g, 4.0g, and 4.4g, respectively and Ca-alginate bead size was not effected in uptake sodium chloride. The uptake rate on initial sodium chloride concentration(4%, 8%, 12% and 16%) was 30%, 28%, 27% and 25%, respectively. The uptake rate on basic pH(10.0) was higher than when compared to other neutral pH(6.8) and acidic pH(4.0). The initial uptake velocity of sodium chloride from immobilization beads with salt resistant bacteria was lower than that of non-immobilization beads. The uptake rate of sodium chloride was decreased according to elongation of curing time. Reusability of Ca-alginate beads was possible but according to reutilization, the salt uptake volume of beads was also decreased. The uptake volume of sodium chloride from Doengjang by Ca-alginate beads on time course(3, 6, 12, and 24 hour) was revealed 5g, 6g, 7g and 7g, respectively.

Eletron Microscopic Observation of Calcium-Acetylated Seaweed Alginate Gel

  • Seaweed alginate was acetylated by activated carbon immobilized Pseudomonas syringae in a fluidized bed, up-flow reactor. The acetylation degree of seaweed alginate was about 30%. Calcium-acetylated seaweed alginate gel bead was made and compared to calcium-seaweed alginate gel bead by the scanning electron microscopy (SEM). Structural difference of two gel beads may results from increased viscosity and decreased affinity of acetylated seaweed alginate for calcium ion. On the basis of interior and exterior structure of calcium-acetylated seaweed alginate gels and property of acetylated seaweed alginate, it seems that acetylated seaweed alginate is used for the supporter for electrophoresis and packing materials for liquid chromatography and gel filtration.

Survival of Bifidobacterium breve in Acidic Solutions and Yogurt, Following Immobilization in Calcium Alginate Beads

  • Sodium alginate was used to immobilize Bifidobacterium breve ATCC 15700 cells. The ability of the Ca-alginate beads to protect the B. breve ATCC 15700 was evaluated under different conditions including alginate concentration, bead size, pH, hydrogen peroxide, and storage period. The survival of the B. Breve ATCC 15700 was estimated in pasteurized yogurt, containing either the immobilized or free cells, throughout the storage period. The survival cells in bead after exposure to acidic solution (pH 3.0) increased with increase of both the alginate gel concentration and bead size. Also, immobilized cells in alginate bead were more resistant than the free cells to hydrogen peroxide, storage period, and the environment inside yogur. When retreated beads with skim milk and nonretreated beads were tested in acidified pH 3.0 TPY media including acetic and lactic acid, the number of viable cells in the retreated bead was approximately 10-fold higher than that of nonretreated beads. This suggests that the skim milk operated as a material decreasing the diffusion of acid and hydrogen perosicde into alginate gels. From this research, it was found that yogurt itself supported immobilized cells with an improved protection from the extreme acidity in yogurt.

Assessment of Applicability of a Calcium Carbonate-Alginate Beads as Neutralizer for the High Cell Density Cultivation of Isolated Sourdough Lactic Acid Bacteria (Sourdough에서 분리된 유산균의 고농도 배양을 위한 중화제로서 Calcium Carbonate-Alginate Bead의 이용가능성 평가)

  • Lab scale experiments were conducted in order to assess the applicability of $CaCO_{3}$-alginate beads as neutralizer for the high cell density cultivation and prepare the direct vat inoculation cultures of isolated sourdough lactic acid bacteria. With increasing the amount of bead and decreasing the diameter of bead in acidic solution, the neutralizing effect of $CaCO_{3}$-alginate bead became higher. In batch process with $CaCO_{3}$-alginate beads, Lactobacillus amylovorus DU-21 isolated from sourdough showed the highest viable cell counts and optical density in MRS broth. The values of viable cell counts and optical density were 9.996 log CFU/mL and 3.97, respectively. Experiments on the conditions which increase viability during lyophilization were carried out and the following results were obtained; 15% glycerol revealed the high cryoprotective effect on the concentrated cultures during lyophilization among the two cryoprotective agents. Consequently, $CaCO_{3}$-alginate beads and 15% glycerol were found to be useful not only to cultivate Lactobacillus amylovorus DU-21 but also to preserve strain.

Adsorption Characteristics of Nickel, Zinc and Cadmium Ions using Alginate Bead (Alginate Bead를 이용한 니켈, 아연, 카드뮴의 흡착특성에 관한 연구)

  • This study investigated the adsorption characteristics of nickel, zinc and cadmium ions from the aqueous solution onto the alginate bead. Adsorption equilibrium capacities of the heavy metal ions increased with increasing initial pH of the solution. The adsorption equilibrium isotherm of the heavy metal ions was well represented by Langmuir equation. The magnitude of adsorption capacity of the heavy metal ions onto alginate bead was the order of cadmium > zinc > nickel. Kinetic parameters were measured in a batch adsorber to analyze the adsorption rates of the heavy metal ions. The internal diffusion coefficient of the heavy metal ions in the intraparticle were determined by comparing the experimental concentration curves with those predicted from the surface diffusion model (SDM) and pore diffusion model (PDM). The internal diffusion of the heavy metal ions in the intraparticles was explained by PDM.
