BACKGROUND: The aim of this study was to isolate and identify algicidal bacterium that tends to kill the toxic dinoflagellate Alexandrium catenella, and to determine the algicidal activity and algicidal range of algicide. METHODS AND RESULTS: Among of algicidal bacteria isolated in this study, NH-3 isolate was the strongest algicidal activity against A. catenella. NH-3 isolate was identified on the basis of biochemical characteristics and analysis of 16S rRNA gene sequences. The NH-3 isolate showed over 99% homology with Arthrobacter oxydans, and was designated as Arthrobacter sp. NH-3. The optimal culture conditions were $25^{\circ}C$, initial pH 7.0, and 2.0% (w/v) NaCl concentration. The algicidal activity of Arthrobacter sp. NH-3 was significantly increased to maximum value in the late of logarithmic phase. Arthrobacter sp. NH-3 showed algicidal activity through indirect attack, which excreted active substance into the culture filtrate. When 10% culture filtrate of NH-3 was applied to A. catenella, 100% of algal cells were destroyed within 30 h. In addition, the algicidal activities were increased in dose and time dependent manners. The pure algicide was isolated from the ethyl acetate extract of the culture filtrate of NH-3 by using silica gel column chromatography and high performance liquid chromatography (HPLC). We investigated the algicidal activity of this algicide on the growth of harmful algal bloom (HAB) species, including A. catenella. As a result, it showed algicidal activity against several HAB species at a concentration of $100{\mu}g/mL$ and had a relatively wide host range. CONCLUSION: Taken together, our results suggest that Arthrobacter sp. NH-3 and its algicide could be a candidate for controlling of toxic and harmful algal blooms.
In this study, a newly synthesized thiazolidinedione derivative, TD49 with a highly selective algicide to red tide, was examined in order to evaluate the bioconcentration on aquatic organisms of coast. BAF (accumulation of TD49 by aquatic food chain) and BCF (accumulation of TD49 by sea water) were examined employing the shrimp (Fenneropenaeus chinensis) as the feed organism, and the olive flounder Paralichthys olivaceus as a consumer in marine ecosystem. Bioconcentration degree in sea water showed that the order in P. olivaceus was viscera > gill > muscle. The average BCF values of TD49 were 67.70, 63.32 and 20.25 at viscera, gill and muscle, respectively. Bioaccumulation degree using feed showed that the order in the organs of P. olivaceus was viscera > gill > muscle. The average BAF values of TD49 were 175.89, 114.88 and 32.59 at viscera, gill and muscle, respectively. When compared with two results, the accumulation by the food and water was higher than that by water. After the elimination experiment in sea water, the TD49 concentration was 2.81 nmole/g in the viscera and were not found in the gill and the muscle. More than 50% of the accumulated TD49 were eliminated from viscera in 7 days and all the accumulated TD49 were eliminated from gill and muscle in 7 days. On the other hand, the octanol/water partition coefficient (log $K_{ow}$) was measured to be 3.66 and experimental BCF of this study was 67.7.
Cell-free culture broth of marine halophilic bacterium, Kordia algicida was shown to possess specific algicidal ability against red tide organism, Cochlodinium polykrikides. Physiochemical characteristics of algicidal material originated in the bacterial culture broth were analyzed that its molecular weight was estimated to a 3,000 dalton and it was stable in heat and pH treatment. The algicidal fraction against C. polykrikoides obtained from gel permeable chromatography contained high concentration of ammonium ion as analyzed by ICP/Mass spectrum. C. polykrikoides by the fraction was quickly lysed within 1 min. It was shown that the effective concentration for algicide against C. polykrikoides was over 1mM of ammonium chloride. On the other hand, other metal ions presented in the algicidal fraction showed no algicidal effect against C. polykrikoides. In additon, ammonium ion exhibited species-specific killing spectrum for two species of red tide organisms, C. polykrikoides and Gymnodinium sanguieum. Therefore, further researches on the killing mechanism against C. polykrikoides exerted by ammonium ion, and subsequent development of replaceable algicidal materials will perform to provide useful tools for the control of red tide.

In an effort to identify a new environment-friendly algicide, we examined the ability of extracts from the leaves and stems of nine Korean oak tree species to inhibit growth of the bloom-forming cyanobacterium, Microcystis aeruginosa. At a concentration of 100 mg $L^{-1}$, five of the oak tree extracts (QAT-L, QAT-5, QAS- L, QGI-5, and QSA- L) decreased the cell density of M. aeruginosa by over 90% for 7 days. At a concentration of 20 mg $L^{-1}$, the same five extracts inhibited the growth of M. aeruginosa by approximately 50%. The minimum concentration of oak tree extracts required for effective inhibition of M. aeruginosa (20 mg $L^{-1}$) is comparable to that of the known algicide, tannic acid (17 mg $L^{-1}$), which is thought to be one of the main active ingredients in the oak tree extract. These findings suggest that oak extracts may be useful as an environment-friendly algicide to control the bloomforming cyanobacterium, M. aeruginosa, in eutrophic waters.
In order to perform an acute toxicity assessment of a new algicide, thiazolidinedione derivative (TD53) with enhanced solubility and lower toxicity to marine ecosystem, representative 3 organisms: plant plankton (Skeletonema costatum), animal plankton (Daphnia magna), fish (Paralichthys olivaceus) related in the food chain of marine ecosystem according to OECD standard methods were employed in the exposure experiment. The exposure assessment showed that $EC_50$ of S. costatum in 96-hour, $EC_50$ of D. magna in 48-hour and $LC_50$ of P. olivaceus in 72-hour for TD53 were $1.53\;{\mu}M$, $0.61\;{\mu}M$ and $2.14\;{\mu}M$ respectively. NOEC (No Observed Effect Concentration) and PNEC (Predicted No Effect Concentration) were calculated to be $0.25\;{\mu}M$ and 6.10 nM, respectively from $EC_50$ of most sensitive strain, D. magna. Comparing with the results of toxicity assessment previously performed by using Ulva pertusa Kjellman accepted as an ISO standard method, the values of PNEC showed 3.7 times lower toxicity in case of this study employing 3 organisms, indicating that if the organisms which are more representative and sensitive in marine ecosystem are further investigated, more accurately and validly predicted toxicity of TD53 could be applied in field.
The potential of calcium peroxide to act as an agent for waterblooming control was In-vestigated by examining the growth inhibition of Microcystis aerusinosa. Due to the chemical nature of calcium peroxide, it can remove dissolved phosphate by forming an Insoluble precipitate, generating radicals, coagulant, and oxygen as byproducts as it dissolves in water. The growth of M. aerusinosa was severely inhibited and the chlorophyll-n concentration was drastically decreased in the presence of calcium peroxide. With 200 ppm of calcium peroxide dosage, a chlorophyll-a concentration of 1,700 mg/m$^3$ was lowered to below 10% of its initial concentration after 4 days. One possible explanation for this growth Inhibition is the removal of the available phosphate by calcium peroxide.
We evaluated the field application feasibility that biologically derived substances (Naphthoquinone derivate: NQ 2-0) can be used for the eco-friendly mitigation of natural harmful cyanobacterial blooms in freshwater. We conducted a 30 ton scale mesocosm experiment to investigate the effects of NQ 2-0 on biotic and abiotic factors in water collected from Gi-heung reservoir. In the mesocosm experiments, the abundance of Microcystis sp. was continuously increased in the control. However, the Microcystis sp. cell density was sharply decreased on the $10^{th}$ day. In the treatment, NQ 2-0 showed the strong and selective algicidal activity toward the target cyanobacteria (Microcystis sp.). Accordingly, the algicidal activity of NQ 2-0 compound increased gradually until $10^{th}$, $15^{th}$ days and algal biomass was decreased to 99.4 and 100 %, respectively. NQ 2-0 compound was not only selective algicidal activity but also the growth of other phytoplankton and increased the Shannon-Wiener diversity index of phytoplankton. In the mesocosm experiments, the dynamics of biotic (bacteria, heterotrophic nanoflagellate, ciliates, zooplankton) and abiotic (water temperature, dissolved oxygen, pH, conductivity, nutrients) factors remained unaffected. These results suggest that NQ 2-0 could be a selective and ecologically safe algicide to mitigate harmful cyanobacterial blooms. In addition, it is believed that NQ 2-0 will play a major role in forming a healthy aquatic ecosystem by facilitating habitat and food supply of aquatic organisms.

Worldwide development of harmful algal blooms causes serious problem for public health and fisheries industries. To evaluate the algicidal impact on the harmful algae bloom species in aquatic ecosystems of coast, a new algicide thiazolidinedione derivative (TD49) were tentatively examined in the growth stages (i.e., lag, logarithmic and stationary phase) of rapidophyceae $Heterosigma$$akashiwo$, $Chattonella$$marina$ and $Chattonella$ sp..Three strains could easily destroy in the lag phase due to relatively weak cell walls than those of the logarithmic and stationary phase. It is thought that inoculation of TD49 substances into initial or developmental natural blooms with a threshold concentration ($2{\mu}M$) can maximize the algicidal activity. Also, bio-chemical assays revealed that the algicidal substances from all culture strains were likely to be extracellular substances because those cells have easily destroyed in cell walls. On the other hand, natural zooplankton communities were influenced within the exposure experiments of $2{\mu}M$, which is showed the maximum algcidal activity of tested organisms. These results indicate that although the TD49 substance is potential agents for the control of $H.$$akashiwo$, $C.$$marina$ and $Chattonella$ sp. in the enclosed eutrophic bay and coastal water, more detailed research of acute toxicity effect on high trophic organism in marine ecosystems need to be conducted.

This study aimed to understand the changes in microbial community after algicide dosing to control the fish-killing dinoflagellate Cochlodinium polykrikoides in 10L microcosm. Based on our microcosm experiments, the algicidal activity for C. polykrikoides of yellow clay at the concentrations of 4g and 10g per 10 L was < 20%. At $0.8{\mu}M$ concentration of thiazolidinedione(TD49), the population of C. polykrikoides was controlled to be > 85%. In microbial community, a significant increase in heterotrophic bacterial (HB) abundance was observed at day 1 in the TD49 and yellow clay treatments including control. The HB remained high for 2 days and then gradually decreased. In contrast, the abundance of heterotrophic nanoflagellates (HNFs) increased significantly on days 3 and 5 in the TD49 treatments, indicating that the decline in HB was probably a result of predation by the high density of HNFs. In addition, fluctuations in the aloricate ciliate Uronema sp., which feed on bacteria, was clearly correlated with fluctuations in HB abundance, with a lag period of 1-3 days. Therefore, the short-term responses of the HNF and Uronema sp. may have been a result of the rapidly increasing of HB abundance, which is related to degradation of the dense C. polykrikoides bloom, particularly in the TD49 treatment. In addition, large aloricate ciliate Euplotes sp. was significantly increased after reproduction of HNFs and Uronema sp. Consequently, the algicide TD49 had positive effect on the microbial communities, which indicates that the microbial loop was temporarily enhanced in the microcosm by energy flow from HB through HNFs to ciliate.
A thiazolidinedione derivative, TD49 with the highly selective algicide to red tide was newly synthesized and its acute toxicity was examined in order to evaluate the effect on aquatic ecosystems of coast. Major three species having important role in the food chain of marine ecosystem, such as Skeletonema costatum of microalgae, Daphnia magna of crustacea, Paralichthys olivaceus of flatfish fingerling were employed for the acute toxicity assessment. $EC_50$ or $LC_50$ as the assessment criterion was investigated to each specie, and NOEC (No Observed Effect Concentration) and PNEC (Predicted No Effect Concentration) from most sensitive specie to toxicity of TD49 were further calculated. $EC_50$ of S. costatum in 96-hour, $EC_50$ of D. magna in 48-hour, and $LC_50$ of P. olivaceus in 72-hour for TD49 were $0.34\;{\mu}M$, $0.68\;{\mu}M$, and $0.58\;{\mu}M$, respectively. NOEC from the results of S. costatum was estimated to be $0.20\;{\mu}M$ and PNEC was estimated as 3.40 nM by applying factor value of 100 to $EC_50$$0.34\;{\mu}M$ of S. costatum. In addition, it was revealed that Solutol used as the dispersing agent of TD49 had very little toxic influence under the concentration range of $0{\sim}0.4\;{\mu}M$ used in TD49 toxicity experiment. Although the estimated concentration of TD49 that will be sprayed onto the coastal field for the algicide is higher than NOEC value, it is considered that the spraying concentration would not be a considerable problem due to a dilution effect by tide at the opened coast.
