To select submerged macrophytes to suppress growth of Microcystis aeruginosa through releasing allelochemicals, we conducted growth experiments with water from patches of submerged macrophytes and with aqueous extracts of those submerged macrophytes. In the first experiment, growth rates of M. aeruginosa decreased as biomass of Myriophyllum spicatum and Hydrilla verticillata increased. In the second experiment, M. aeruginosa showed approximately 50% growth reduction with extracts from M. spicatum and 24% reduction with extracts from Ottelia alismoides. Both M. aeruginosa growth experiments with water and plant extracts suggest that M. spicatum would be the best candidate to reduce M. aeruginosa growth.
This study is the first attempt to understand the feeding physiology of a sea-urchin larva on a red-tide dinoflagellate. Fifteen day old larvae of S. intermedius capture C. polykrikoides cells by localized reversal of ciliary beats. No failure to transporte the algal cells from theciliated band to mouth and no rejection at the mouth suggest that C. polykrikoides has no feeding deterrence to S. intermedius larvae. The trend obtained for the clearance rate of S. intermedius larvae is similar to that of other sea urchin larvae. Thus, the clearance rate decreased as the algal concentration increased. Maximum clearance rate of S. intermedius on C. polykrikoides was 17.7 $\mu l$/larva/hr. Ingestion rate rapidly increased at lower algal concentrations and saturated at higher concentrations. There was no inhibition in ingestion rate at the highest prey concentration of ca. 3000 cells/ml. Maximum ingestion rate of S. intermedius on C. polykrikoides was 131 ngC/larva/d, which is higher than that reported for the larvae of the mussel Mytilus gal-lotrovincialis, but lower than that of the ciliate Strombidinopsis sp. The grazing rate, calculated by combining the field data on algal abundances with experimental data on ingestion rate, suggests that due to its low abundance, sea urchin Iarva has no significant grazing impact on C. polykrikoides population.
Interactive effects of UV-B and Cd on growth, pigment, photosynthesis, and lipid peroxidation have been studied in Anabaena, Microcystis, and Nostoc; all the tested cyanobacteria showed differential sensitivity to different dosage of UV-B and Cd alone as well as in combination. Phycocyanin was severely affected by UV-B and Cd by all the strains; the degree of pigment bleaching was most pronounced in Anabaena followed by Microcystis and Nostoc. $UV-B_2+Cd_2$ produced nearly 83, 78, and 65% inhibition of phycocyanin in Anabaena, Microcystis, and Nostoc, respectively. The above treatment also significantly decreased the contents of Chl ${\alpha}$ and carotenoid. Presence of capsule in Microcystis protected the phycocyanin bleaching as compared to decapsulated cells. Laboratory-grown Microcystis revealed about 75 and 80% inhibition, following $UV-B_2+Cd_2$ treatment, respectively. in capsulated and decapsulated cells. Damage caused by Cd was more pronounced than UV-B. Inhibition of photosynthesis did occur in all the test strains, being maximum in Anabaena. PS II was the most sensitive component of the electron transport chain, showing 84, 80, and 70% inhibition in Anabaena, Microcystis, and Nostoc, respectively. As compared to control, significant lipid peroxidation (6.5-fold higher) was observed in Anabaena with $UV-B_2+Cd_2$, $^{14}C-uptake$ was more susceptible to Cd and Uv-B than oxygen-evolution. Approximately 84, 80, and 76% inhibition of $^{14}C-uptake$ was observed in Anabaena, Microcystis, and Nostoc, respectively. Similarly, $UV-B_2+Cd_2$ inhibited APT content of Anabaena by 87%. This ,study suggests that inhibition of carbon fixation was due to decreased ATP content of the test cyanobacteria by UV-B+Cd, where Anabaena was the most sensitive and Nostoc the most tolerant.
팔당호 주변에서 자생하는 식물체를 이용하여 실험실 내에서 배양된 남조류에 투여하여 조류증식 억제 효율을 조사하였다. 조류 성장 억제 효과에 있어 육상식물을 이용하는 경우 조류 밀도가 10$^{4}$ cells/ml 범위에서의 0.5g/l 투여량은 전혀 효과를 나타내지 못했던 반면, 2.5 g/l 투여량에서는 95% 이상의 효과를 보였다. 조류 밀도가 10$^{6}$ cells/ml 범위에서의 육상식물 투여량은 2.5 g/l 이상이 필요했다. 조류성장 억제에 육상식물은 잣잎, 수생식물은 애기마름이 큰 효과를 보였다. 애기마름 투여조는 조류 밀도가 10$^{6}$ cells/ml 범위에서도 2.5 g/l 투여량으로 50%의 억제 효율이 나타났다. 식물을 이용한 조류 증식 억제 효과는 식물 투여 후 7일째 후 가장 효과가 좋은 것으로 나타났다. 습체, 건체, 추출물 비교시 추출물이 가장 효과가 좋은 것으로 육상식물 투여조의 경우 1.25 g/l 투여량에 비해 2.5 g/l 투여량에서 현저한 효과를 보였고, 애기마름은 1.25 g/l 투여량에서도 99%의 효과를 보였다. 건체식물은 습체식물에 비해 초기에도 높은 효과를 보였다. 조류의 비증식속도로 보았을 때도 식물체 추출물을 투여했을 때 가장 효과가 좋았고 다음으로 건체, 습체식물 투여순으로 조류 증식억제 효과를 나타내었다.
A new direct colorimetric assay of microcystin in water and algal samples is proposed consisting of two procedures as follows: 1) the elimination of phosphorus in the sample and concentration of microcystin using a C(sub)18 cartridge, 2) the detection of the released phosphorus by the ascorbic acid method and determination of protein phosphatase (PP) inhibition by microcystin. The optimum amounts of phosphorylase ${\alpha}$ and PP-1 in 50 ${\mu}$L concentrated sample were 50$\mu\textrm{g}$/50${\mu}$L buffer and 1.0unit/50${\mu}$L buffer, respectively, for the best assay. The pH for the maximum activity of PP-1 was 8. The minimum detectable concentration for this method was about 0.02$\mu\textrm{g}$/L, which is sufficient to meet the proposed guideline level of 1$\mu\textrm{g}$ microcystin/L in drinking water. Consequently, it would seem that the proposed direct colorimetric assay using PP is a rapid, easy, and convenient method for the detection of microcystin in water and algal samples.
For several decades, lactic acid bacterium (Lactobacillus graminis: LAB) has been generally recognized as safe. To develop the pan-environmental bio-control agent, algicidal activity of the live LAB cell and its culture filtrate (CF) was examined against Microcystis aeruginosa. LAB cells perfectly lysed M. aeruginosa within 3 days, while the CF had a less effect than the live cells, approximately 78% inhibition of algal growth during a same culture period. The concentration of microcystin in alone culture of M. aeruginosa was $7.1{\mu}gL^{-1}$, but gradually increased and leach $158.5{\mu}gL^{-1}$ on 10 days. However, LAB cells clearly decreased the microcystin by $10.3{\mu}gL^{-1}$ in the same period, approximately 93.5%. CF of LAB showed a strong algicidal activity over 75% between pH 2-7, 91.3% by the treatment of proteinase K, 87.8% by below 3 kDa in particle size, and 75.3% by heat treatment, respectively. Of five solvents, fractions of CF passed through solvents diethyl ether and ethyl acetate showed an obvious algicidal activity in the algal-lawn test. Among 5 fractions purified by silica-gel TLC plate, two spots showed a most strong removal activity on M. aeruginosa. Another analysis of GC indicate that CF contained six representative fatty acids. Even though most of these substance have been known as an anti-algal substance against M. aeruginosa, oleic acid is the most effective. These results suggested that the culture filtrate or specific substances, like a fatty acids, in comparison with live L. graminis can be a successful and eco-friendly agent to control Microcystis bloom.
부영양호수의 남조 Microcystis aeruginosa 제어를 위해 호수 바닥층에서 분리한 살조세균과 섬모충을 현장여과수를 이용하여 조류배양조건과 동일한 조건에서 단일 또는 혼합적용하여 그 결과를 서로 비교하였다. 두 생물제재는 저자들의 선행연구와 같이 단일 적용시 매우효과적인 반면, 혼합적용시 오히려 조류의 성장을 촉진하였다. 결국 남조 Microcystis제어를 위한 섬모충이나 박테리아의 적용은 다른 한 생물군의 낮은 밀도를 요구하였으며, 이처럼 동일 조류에 대한 제어능을 갖는 두 생물제재의 배타적인 관계는 앞으로 부영양호수의 남조대발생 제어에 귀중한 자료로서 제공될 것이다.
Fourier Transform Infrared (FTIR) spectroscopy was used to study carbon allocation patterns in response to N-starvation in the nearly ubiquitous diatom Chaetoceros muellerii. The role of gene expression, protein synthesis and transamination on the organic composition of cells was tested by using specific inhibitors. The results show that inhibition of key processes in algal metabolism influence the macromolecular composition of cells and and prior cell nutritional state can influence a cell's response to changing nutrient availability. The allocation of C can thus lead to different organic composition depending on the nutritional context, with obvious repercussions for the trophic web. This also shows that C allocation in algal cells is highly flexible and that C (and the energy associated with its allocation) can be variably and rapidly partitioned in algal cells in response to relatively short term perturbations. Furthermore, the data confirm and extend the utility of infrared spectroscopy as a probe of the metabolic state of autotrophic cells.
Blue-Min was initially developed as an adsorbent for harmful gas removal and recently improved to apply to livestock, agriculture and aquaculture as an assistant feed. In the Blue-Min treatment, growth of harmful algae (Cochlodinium polykrikoides and the others causing the red-tide in the ocean) were inhibited below 10% in comparison with control and coagulation removal of harmful alge with Blue-Min treatment was more efficient than that of yellow loess treatment. It would be expected that the Ble-Min can be useful for the extirpator against the red-tide organisms and restrain the toxic algal growth around the fish aquaculture using the assistant feed. Recently, its utility has become to be diverse as it was revealed that aquaculture productivity increase by its application and, in addition, that it improve the water quality or sediment conditions in the aquaculture of Chinese White Shrimp. When Blue-Min was treated with the proper dose, the growth inhibition of Microcystis aeruginosa and lsochrysis galbana, which are typical red-tide organisms in freshwaters and food organisms in aquaculture, respectively, were less than that of marine red-tide organisms, while their growth slightly increased with low concentration treatiment. In addition, polyunsaturated fatty acids (PUFA) content of I. galbana slightly increase with the Blue-Min treatment. Through our research, the Blue-Min has diverse and comples function against various biological organisms and is proved as a biological activator or depressor.
국내에서 널리 분포하여 쉽게 구할 수 있는 천연재료인 상수리나무, 밤나무, 녹차, 황토, 맥반석, 제올라이트와 혼합물 등 7가지 물질의 남조와 녹조류 성장 억제능을 파악하기 위하여, 단순 추출법을 이용하여 얻은 각 물질을 농도별(1, 5, 10 mg $L^{-1}$)로 배양조류에 처리하고 각 조류의 성장 특성을 조사하였다. 분리형 남조 Microcystis aeruginosa, 군체형 M. aeruginosa 등은 저농도의 상수리나무, 밤나무, 제오라이트에 의해서 60% 이상의 높은 조류억제능을 보였으며, 녹조 Scenedesmus quadricauda는 녹차, 상수리나무에서 50% 이하의 억제능을 보였으며, 제오라이트에서는 65% 이상 성장이 억제되었다. 녹차를 포함한 대부분의 물질에서 낮은 농도인 1 mg $L^{-1}$에서 녹조 Chlorella vulgaris의 성장을 촉진하였으며, 황토는 S. quadricauda 성장을 촉진하였다. 따라서 부영양호수에서 흔히 발생되는 남조발생 억제에는 상수리나무, 밤나무, 제올라이트가 적합할 것으로 판단되었으며, 녹차, 황토나 맥반석 등은 녹조류 대량배양이나 배지로서 이용 가능성이 높다고 사료되었다.
