Dynamics and Control Methods of Cyanotoxins in Aquatic Ecosystem

  • Cyanotoxins in aquatic ecosystems have been investigated by many researchers worldwide. Cyanotoxins can be classified according to toxicity as neurotoxins (anatoxin-a, anatoxin-a(s), saxitoxins) or hepatotoxins (microcystins, nodularin, cylindrospermopsin). Microcystins are generally present within cyanobacterial cells and are released by damage to the cell membrane. Cyanotoxins have been reported to cause adverse effects and to accumulate in aquatic organisms in lakes, rivers and oceans. Possible pathways of microcystins in Lake Suwa, Japan, have been investigated from five perspectives: production, adsorption, physiochemical decomposition, bioaccumulation and biodegradation. In this study, temporal variability in microcystins in Lake Suwa were investigated over 25 years (1991~2015). In nature, microcystins are removed by biodegradation of microorganisms and/or feeding of predators. However, during water treatment, the use of copper sulfate to remove algal cells causes extraction of a mess of microcystins. Cyanotoxins are removed by physical, chemical and biological methods, and the reduction of nutrients inflow is a basic method to prevent cyanobacterial bloom formation. However, this method is not effective for eutrophic lakes because nutrients are already present. The presence of a cyanotoxins can be a potential threat and therefore must be considered during water treatment. A complete understanding of the mechanism of cyanotoxins degradation in the ecosystem requires more intensive study, including a quantitative enumeration of cyanotoxin degrading microbes. This should be done in conjunction with an investigation of the microbial ecological mechanism of cyanobacteria degradation.

  • The objective of this research was to review the correlation between microalgae and agricultural water quality. Although microalgae has been considered as an essential factor for control1ing of water ecosystem, little attention has been paid for evaluating of microalgae as an important factor for water quality management. But it can be use to make us know the water pollution state at saprobic system, LTSI (Lake Tropic State Index), DAIpo (Diatom Assemblage Index to Organic Pollution), and AGP (Algal growth potential). In saprobic system, it is used microalgae such as Actinastrum hantzschii var. fluviatile, Asterionella gracillima, Coelastrum microporum, Synedra acus, Dictyosphaerium pulchellum, Micractinium pusillum, Cyclotella meneghiniana, Microcystis aeruginosa, Scenedesmus quadricauda, and Nitzschia palea for assessment water quality. In addition, they have ecologically significant characteristics such as dominant species, cosmopolitan species, redtide causative species etc. Also, microalgae such as Botryococcus braunii, B. sp., Chlorella vulgaris, C. sp., Phormidium sp., Scenedesmus quadricauda, Selenastrum capricornutum, Spirulina maxima, and S. platensis have an effect on improvement of water quality.

국내 담수역 남조류 원격탐사를 위한 피코시아닌 추출법 비교 연구 (A Study on Comparison of Phycocyanin Extraction Methods for Hyperspectral Remote Sensing of Cyanobacteria in Turbid Inland Waters)

  • Phycocyanin (PC) is one of the water-soluble accessory pigments of cyanobacteria species, and its concentration is used to estimate the presence and relative abundance of cyanobacteria. In laboratory experiments, PC content of field data were determined using Sarada's freeze-thaw method in algal bloom season. The effectiveness of three selected extraction methods (repeated freeze-thaw method, homogenization, power control) for PC were determined. The extraction efficiency of phycocyanin was the highest (of the methods compared) when a single freezing-thawing cycle was followed by pre-sonication. Applying this optimized method to surface water of Korean inland waters, the average concentration distribution was estimated at $2.9{\sim}51.9mg/m^3$. It has been shown that the optimized pre-sonication method is suitable to measure cyanobacteria PC content for the characterization of inland waters. The approach and results of this study indicates the potential of effective methods for remote monitoring and management of water quality in turbid inland waters using hyperspectral remote sensing.

Keeping house: evaluation of housekeeping genes for real-time PCR in the red alga, Bostrychia moritziana (Florideophyceae)

  • Biological response of cells to variable conditions should affect the expression level of certain genes. Quantification of these changes in target genes needs stable internal controls. Real-time quantitative polymerase chain reaction (PCR) has traditionally used reference or ‘housekeeping’ genes, that are considered to maintain equal expression in different conditions, to evaluate changes in target genes between samples and experimental conditions. Recent studies showed that some housekeeping genes may vary considerably in certain biological samples. This has not been evaluated in red algae. In order to identify the optimal internal controls for real-time PCR, we studied the expression of eleven commonly used housekeeping genes; elongation factor 1-alpha, glyceraldehyde-3-phosphate dehydrogenase, β-actin, polyubiquitin, 30S ribosomal gene, 60S ribosomal gene, beta-tubulin, alpha-tubulin, translation initiation factor, ubiquitin-conjugating enzyme, and isocitrate dehydrogenase in different life-history stages of Bostrychia moritziana. Our results suggest that glyceraldehyde-3-phosphate dehydrogenase (GAPDH) and 30S ribosomal gene, have the most stable gene expression levels between the different life history stages (male, female, carposporophyte, and tetrasporophyte), while the other genes are not satisfactory as internal controls. These results suggest that the combinations of GAPDH and 30S would be useful as internal controls to assess expression level changes in genes that may control different physiological processes in this organism or that may change in different life history stages. These results may also be useful in other red algal systems.

Visual MODFLOW 지하수 유동 모델링을 위한 GIS 기반 전ㆍ후처리기 개발 (Development of the GIS Based Pre- and Post-Processing Tool for the Visual MODFLOW Groundwater Flow Modeling)

  • 본 연구에서는 지리정보시스템(GIS)을 사용하여 Visual MODFLOW 지하수 유동 모델링 툴에 필요한 입력 인자(자료)를 과학적으로 만들어줄 뿐만 아니라, 모델링을 위해 입력된 인자들과 모델링 후 생성된 결과물들을 DB화하고 이를 체계적으로 관리할 수 있도록 하는 Visual MODFLOW 지하수 유동 모델링을 위한 GIS 기반 전 후처리기를 개발하였다. 이 전 후처리기의 모듈로서 가장 특징적인 것은 모델구역 내에서 관정 주변에 상대적으로 조밀하게 즉, 셀의 크기가 다양한 그리드를 GIS ArcView에서 자동 또는 반자동으로 형성하는 툴을 개발하였다는 것이다. 모델링 구역 내에서 다양한 경계조건(boundary condition)을 반영한 DXF 생성을 위한 툴 개발도 들 수 있다. 아울러 2차원인 ArcView를 이용하여 3차원 수리지질구조를 생성하고, MODFLOW 프로그램의 입출력 자료형태와 동일한 위상구조를 유지한 채 전 후처리하도록 한 것은 이 연구의 가장 큰 성과이다.

CONTROL OF DIATOM BY PREOXIDATION AND COAGULATION IN WATER TREATMENT

  • Conventional coagulation is still the main treatment process for algae removal in water treatment. The coagulation efficiency can be significantly improved by the preoxidation of algae-containing water. Jar test was conducted to determine the optimal condition for the removal of diatoms, especially Cyclotella sp. by preoxidation and the subsequent coagulation. The effects of various concentration of PAC (Polyaluminum chloride) on coagulation with and without preoxidation using chlorine or potassium permanganate at different pHs (7.7 and 9.0) were evaluated. At pH 7.7, preoxidation with 2ppm $Cl_2$ followed by coagulation with 7.5 ppm PAC coagulant could reduce Cyclotella sp. concentration by 86%. At pH 9.0, preoxidation with 1 mg $KMnO_4/L$ followed by coagulation with 12.5 ppm PAC coagulant reduced Cyclotella sp. concentration by 85%. Non-linear regression was applied to determine the optimal condition. At pH 7.7 and 9.0, R was over 0.9, respectively. The pH of algal blooming water is over 9.0. Algae (diatom; Cyelotella sp.) can be controlled in the following ways: preoxidation with 1 mg $KMnO_4/L$ followed by coagulation with 12.5 ppm PAC coagulant can remove 80% algae from water. If water pH is adjusted to 7.7, it was expected that less amount of coagulant (7.5 or 10 mg PAC /L) after preoxidation ($Cl_2$ 2 ppm or $KMnO_4$ 0.33, 1 ppm) would be needed to achieve similar level of algae removal. The oxidation with 0.33ppm $KMnO_4$ followed by coagulation with 7.5 ppm PAC coagulant was preferable due to cost-effectiveness of treatment condition and color problem after treatment.

적조생물 Cochlodinium Polykrikoides에 대한 유류 및 유처리제의 영향 (Effects of Oils and Dispersant on the Red Tide Organism Cochlodinium Polykrikoides)

  • Oil spill caused severe effects on the marine fauna and flora due to direct contact of organisms with the oil and even in regions not directly affected by the spill. This study was conducted to understand the effects of the oil spill accidents and the use of dispersant on the red tide of Cochlodinium polykrikoides. Crude oil produced in Kuwait, bunker-C, kerosene and diesel oil, and a chemical dispersant produced in Korea, were added with a series of 10 ppb to 100 ppm in the f/2-Si medium at $20^{\circ}C$ under a photon flux from cool white fluorescent tubes of $100\;mol\;m^{-2}\;s^{-1}$ in a 14: 10 h L:D cycle for the culture of C. polykrikoides. In low concentrations of ${\leq}$ 1 ppm of examined oils no impact on the growth of C. polykrikoides was recorded, while in high concentration of ${\geq}$ 10 ppm, cell density was significantly decreased with the range of 10 to 80% in comparison with the control. The growth of C. polykrikoides after the addition of the dispersant and the mixtures combined with oils and a dispersant of ${\geq}$ 10 ppm appeared to decrease, whereas the growth of C. polykrikoides exposed to ${\leq}$ 100 ppb showed little serious impact. However, almost all the C. polykrikoides cells were died regardless of a dispersant and combined mixtures within a few days after the addition of high concentrations.

Omega-7 producing alkaliphilic diatom Fistulifera sp. (Bacillariophyceae) from Lake Okeechobee, Florida

  • Incorporating renewable fuel into practice, especially from algae, is a promising approach in reducing fossil fuel dependency. Algae are an exceptional feedstock since they produce abundant biomass and oils in short timeframes. Algae also produce high-valued lipid products suitable for human nutrition and supplement. Achieving goals of producing algae fuels and high-valued lipids at competitive prices involves further improvement of technology, especially better control over cultivation. Manipulating microalgae cultivation conditions to prevent contamination is essential in addition to promoting optimal growth and lipid yields. Contamination of algal cultures is a major impediment to algae cultivation that can however be mitigated by choosing extremophile microalgae. This work describes the isolation of alkali-tolerant / alkaliphilic microalgae native to South Florida with ideal characteristics for cultivation. For that purpose, water samples from Lake Okeechobee were inoculated into Zarrouk's medium (pH 9-12) and incubated for 35 days. Selection resulted in isolation of three strains that were screened for biomass and lipid accumulation. Two alkali-tolerant algae Chloroidium sp. 154-1 and Chlorella sp. 154-2 were poor lipid accumulators. One of the isolates, the diatom Fistulifera sp. 154-3, was identified as a lipid accumulating, alkaliphilic organism capable of producing 0.233 g L-1 d-1 dry biomass and a lipid content of 20-30% dry weight. Lipid analysis indicated the most abundant fatty acid within Fistulifera sp. was palmitoleic acid (52%), or omega-7, followed by palmitic acid (17%), and then eicosapentanoic acid (15%). 18S rRNA phylogenetic analysis formed a well-supported clade with Fistulifera species.

산업단지 종말처리장 방류수 유입에 따른 소류지 부영양화 제어 가능성 연구 (Feasibility study on the control of the eutrophication by flowing the industrial wastewater effluents to the lake)

  • 본 논문은 A 시의 산업단지 종말처리장의 방류수 유입에 따른 K 소류지의 녹조 제어 가능성을 평가하고자 하였다. 완전혼합연속반응기로 모형화한 호소를 총인과 엽록소-a의 농도를 예측하였다. 정상상태일 때 총인과 엽록소-a 농도는 각각 $0.29\;/m^3$$4.0\;g/m^3$를 나타났고 실측치와 유사한 결과를 보였다. 비정상상태에서 종말폐수방류수가 최소유량 또는 최대유량으로 소류지로 유입될 때 시간경과에 따른 총인과 엽록소-a 농도 변화를 예측하였다. 또한, 현재 심각한 부영양화 단계에 있는 소류지를 중영양 또는 빈영양 단계로 전환하기 위해 소류지로 유입되는 총인부하량 ($L_p$)과 호소깊이/체류시간 ($H/{\tau}w$)값의 변화를 통해 현실적으로 적용 가능한 제어방법을 제안하였다. 이와 같은 분석기법이 소류지내 총인농도의 제어전략을 수립하기에 적합한 도구로 사용될 수 있음을 확인하였다.

Changes in Phosphorus and Sediment Oxygen Demand in Coastal Sediments Promoted by Functionalized Oyster Shell Powder as an Oxygen Release Compound

  • In this study, we performed a sediment elution experiment to evaluate water quality in terms of phosphorus, as influenced by the dissolved oxygen consumed by sediments. Three separate model column treatments, namely, raw, calcined, and sonicated oyster shell powders, were used in this experiment. Essential phosphorus fractions were examined to verify their roles in nutrient release from sediment based on correlation analyses. When treated with calcined or sonicated oyster shell powder, the sediment-water interface became "less anaerobic," thereby producing conditions conducive to partial oxidation and activities of aerobic bacteria. Sediment Oxygen Demand (SOD) was found to be closely correlated with the growth of algae, which confirmed an intermittent input of organic biomass at the sediment surface. SOD was positively correlated with exchangeable and loosely adsorbed phosphorus and organic phosphorus, owing to the accumulation of unbound algal biomass-derived phosphates in sediment, whereas it was negatively correlated with ferric iron-bound phosphorus or calcium fluorapatite-bound phosphorus, which were present in the form of "insoluble" complexes, thereby facilitating the free migration of sulfate-reducing bacteria or limiting the release from complexes, depending on applied local conditions. PCR-denaturing gradient gel electrophoresis revealed that iron-reducing bacteria were the dominant species in control and non-calcined oyster shell columns, whereas certain sulfur-oxidizing bacteria were identified in the column treated with calcined oyster powder.
