A tide-simulating apparatus was developed for culturing marine macroalgae. The objective of this study was to introduce a novel tide-simulating apparatus that can simulate a diurnal or semi-diurnal tidal cycle in the laboratory. In this apparatus, the seaweeds are move up and down and the water level remains the same during the simulated tidal cycle. The apparatus consists of 18 cylindrical culture tanks (3 blocks $\times$ 6 culture tanks) with 12 cm diameter and 24.5 cm long containing up to 2.5 L of seawater. There is a horizontal plate which covered all 18 culture tanks, and it is raised and lowered by a programmable motor that can regulate exposure time. In one application, seaweeds are attached to braided twine hung on Plexiglas air-tubing. The air-tubing is attached to a lid that is set on a horizontal plate. This apparatus is made of colorless Plexiglas to maximize light transmittance. This apparatus is easily disassembled and transportable to any indoor laboratory, wet laboratory, greenhouse, etc. This apparatus also offers considerable flexibility in terms of design. The size of culture tank can be redesigned by either increasing the height of cylinder or/and using a different diameter of cylindrical Plexiglas, therefore, larger/taller thalli can be cultivated. Growth rates of three eulittoral Porphyra species from different tidal elevations have been compared using this device.

The objective of this study was to assess the variation in the concentration of assimilable organic carbon (AOC) in a drinking water resource, and investigate the characteristics of AOC derived from algae. The seasonal change in AOC at the Kamafusa dam corresponded to changes in the algal cell number. In order to understand the relationship between AOC and algae in a water resource and water purification plant, two kinds of laboratory experiment were performed. The algal culture experiment showed that extracellular organic matter (EOM) that was released during the growth of Phormidium tenue with M-11 medium led to significant increases in the AOC concentration, but no significant variation in the AOC concentration was observed with CT medium containing a high dissolved organic carbon concentration. The chlorination experiment showed that the AOC included in EOM was not easily removed by chlorination, although the AOC included in intercellular organic matter released from the algal cells by chlorination was removed under conditions where residual chlorine was detected.
The relative focus about phosphorus (P) which causes eutrophication characterized by increased growth of undesirable algae has increased in recent years. Phosphorus forms in soil and water include both organic and inorganic forms. There are also a large number of soil P determination methods that have been designed to account for various types of P and mechanisms controlling the chemistry of P in soil, water, and residual materials for environmentally relevant forms of P. However, phosphorus forms in soil, water, and residual materials are also difficult to standardize with any reasonable consensus, due to the number of different disciplines involved. Hence, it is essential to accurately define how P can be measured in soil, water, or residual material samples to avoid potential misinterpretations or inappropriate recommendations in determining amount and types of P. Therefore, we reviewed the testing methods which have appeared in the scientific literature to provide an overview of the soil test P most commonly used.

A morphologically distinct lineage within the Bostrychia moritziana-B. radicans species complex is described as a new species. Bostrychia anomala has thalli with branched monosiphonous filaments with apical cell divisions. The species has terminal tetrasporangial stichidia, each subtending cell bearing tetrasporangia with 2 cover cells. Discharged spores divide transversely, the lower cell first forming a narrow rhizoid and the upper cell forming a monosiphonous shoot. Females have subterminal procarps and males have terminal spermatangial stichidia. Carposporophytes are spherical. Isolates in culture show a pattern of cell death not associated with injury, reminiscent of programmed cell death. Bostrychia anomola shows cell death at intervals along the filaments resulting in division of adjacent cells on either side of the dead cell re-joining the filament; cell division of only one adjacent cell resulting in branching at that site; or filaments fragmenting at the cell death point with adjacent cells forming new apical cells, a means of thallus propagation. The cell death pattern could be a method of filament propagation in the mangrove environment where sexual reproduction is rare.

A new cold-adapted Arctic strain of Haematococcus pluvialis from Blomstrandhalv${\o}$ya Island (Svalbard) is described. This strain is predominantly always in non-motile palmelloid stage. Transmission electron microscopy showed the presence of very thick cell wall and abundant lipid vesicles in the palmelloids, including red and green cells. The external morphology of the non-motile palmelloid and motile bi-flagellated cells of our strain is similar to H. pluvialis; however it differs from H. pluvialis in physiology. Our strain is adapted to live and produce astaxanthin in the low temperature ($4-10^{\circ}C$), whilst the usual growth temperature for H. pluvialis is between $20-27^{\circ}C$. Phylogenetic analysis based on 18S rRNA gene data showed that our strain nested within the Haematococcus group, forming a sister relationship to H. lacustris and H. pluvialis, which are considered synonymous. Therefore, we identified our Arctic strain as H. pluvialis.
Organic substances are released from phytoplankton cells during all phases of growth. The type and amounts of organic substance excreted and the effects of nutrient limitation are often highly species-specific. Dinoflagellate, Cochlodinium polykrikoides grown in batch culture produced an exopolysaccharide. Exopolysaccharide and intracellular polysaccharide concentrations increased as C. polykrikoides cultures progressed from exponential phase, through stationary phase, to declining phase. In the exponential phase, the concentration of exopolysaccharide was relatively low, but in the stationary phase, it showed a rapid increase which seemed to coincide with the depletion of nitrate from the medium. Of the 20 amino acids analyzed, proline dominated in the organic matter of all cultures ranging from 48.2 to 79.9 nmol L–1, and constituting the 20-90% of total amino acids, and followed by histamine varying from 0.7 to 47.5 nmol L–1. Leucine and cysteine were also abundant in the stationary phase. The release rates of exopolysaccharide and intracellualr polysaccharide were higher the end of stationary phase than in the exponential phase. Exopolysaccharide concentration per cell was more than two times higher during the end of stationary phase than that in exponential phase. C. polykrikoides produced extracellular polysaccharide at a rate of 47.04 pg cell–1 day–1.
부식산(腐植酸)이 식물생장(植物生長)에 있어 고염 피해(被害)에 미치는 영향을 구명(究明)하기 위하여 옥수수 및 조류(藻類)를 사용(使用)한 영양액 재배시험(栽培試驗) 결과(結果)는 다음과 같다. 1) 옥수수에 있어서는 소량이 부식산(腐植酸)(탄소(炭素)로 5ppm)처리(處理)가 고염에 의(依)한 생육피해(生育被害)를 현저히 저하 시켰으며 2) 조류(藻類)에 있어서도 소량의 부식산(腐植酸)(20ppm) 처리가 고염(高鹽)에 의(依)한 피해(被害)를 현저히 저하 시켰으나 부식산처리의 농도간(濃度間)에는 큰 차이가 없었다.
Although chemical oxygen demand (COD) is an important issue for wastewater treatment, COD reduction with microalgae has been less studied compared to nitrogen or phosphorus removal. COD removal is not efficient in conventional wastewater treatment using microalgae, because the algae release organic compounds, thereby finally increasing the COD level. This study focused on enhancing COD removal and meeting the effluent standard for discharge by optimizing sludge inoculation timing, which was an important factor in forming a desirable algae/bacteria consortium for more efficient COD removal and higher biomass productivity. Activated sludge has been added to reduce COD in many studies, but its inoculation was done at the start of cultivation. However, when the sludge was added after 3 days of cultivation, at which point the COD concentration started to increase again, the algal growth and biomass productivity were higher than those of the initial sludge inoculation and control (without sludge). Algal and bacterial cell numbers measured by qPCR were also higher with sludge inoculation at 3 days later. In a semi-continuous cultivation system, a hydraulic retention time of 5 days with sludge inoculation resulted in the highest biomass productivity and N/P removal. This study achieved a further improved COD removal than the conventional microalgal wastewater treatment, by introducing bacteria in activated sludge at optimized timing.
Three economic marine algae that have been used as food and carrageenan sources were collected from Korea during a survey of marine algal flora. They share the generic features of Hypnea, and three major clades supported by the sectional features were confirmed in a phylogenetic tree based on rbcL sequences. The first species, which belongs to a species group corresponding to the sect. Spinuligerae, nests in the same clade with Hypnea yamadae in a genetic distance of 0%. It is morphologically characterized by an entangled base, subcompressed or subterete to terete axes, somewhat percurrent main axis, irregularly alternately branching with wide angle, and rarely hooked spinous branchlets. The second one is also referred to the sect. Spinuligerae and formed the same clade as Hypnea cenomyce. The genetic distance between both sequences was calculated as 0.0-0.1%, which is considered to be intraspecific. This species is distinct by somewhat entangled thallus at the basal part, percurrent axis, short spine-like branchlets densely covering the axis, and medullary lenticular thickenings. The third alga, which forms a species group corresponding to the sect. Pulvinatae, nests in the same clade as Hypnea nidulans (no intraspecific divergence). It shows occasionally epiphytic habitat rather than epilithic habitat of low mat-forming growth and percurrent erect main axes with dense lateral branchlets. Based on these morphological and molecular data, the three Korean species are identified as H. yamadae, H. cenomyce, and H. nidulans. This is the first record of the Hypnea species in Korea.
Two new species of Pseudostaurosiropsis are described based on material collected from rivers in southern United States of America, P. californicus sp. nov. and P. elaboratus sp. nov. Both species have specific characters that set them apart from taxa currently ascribed to the genus. Pseudostaurosiropsis californicus sp. nov. has lanceolate valves with subrostrate, broadly rounded apices, an axial area at the same level as the virgae in internal and external views and both are at the same level as striae in external view, the spine tips are dentate and their growth is produced by filamentous extensions, and it has equal-sized apical pore fields on both valve extremes. On the other hand, P. elaboratus sp. nov. has heavily silicified valves, the virgae are slender than the striae in outer view and wider in inner view, it only has a single, externally occluded apical pore field on one valve extreme, and the spines have a solid core. All species within Pseudostaurosiropsis are contrasted with one another and unique features are described for each based on literature and newly collected image data from type material. The genus is reconsidered and two distinguishing features are recognized: rotae externally occluding the areolae and areolae that are funnel-shaped. These features are contrasted with those in other genera and additional published species that should be included in Pseudostaurosiropsis are discussed.
