Expression of $\beta$-amylase Gene and Degradation of Starch Granules of Germinating Rice Seed under Low Temperature and Submerged Soil Condition (저온.담수토양에서 벼종자 $\beta$-아밀라제 유전자 발현과 호분층 인접 배유의 전분분해 양상)

  • This study was conducted to determine $\beta$-amylase gene expression and degradation of starch granules in the endosperm near scutellar epithelium of rice cultivars under the submerged soil at hypoxia 18$^{\circ}C$, which is practically important condition for farmers in temperate regions. In case of cv. Janghyangdo, accumulation of $\beta$-amylase mRNA was detected in the aleurone layer on the ninth day after seeding. However that of cv. Suwon 287 and Norm 6 were not detected in the aleurone layer in submerged soil(hypoxia) at 18$^{\circ}C$. $\beta$-amylase of cv. Janghyangdo was synthesized de novo in aleurone cells not in the scutellar epithelium. Degradation of starch granules in the endosperm near scutellar epithelium of c.v. Janghyangdo and Ginbozu, which have a strong $\beta$-amylase activity, was greater than that of cv. Suwon 287 and Norm 6 with no $\beta$-amylase activity in submerged soil(hypoxia) at 18$^{\circ}C$. This result may indicate that $\beta$-amylase gene expression and degradation of starch granules of germinating rice seed are related to the emergence of rice under the submerged soil condition at low temperature.

Electron Microscopic Study for the Influence of Soaking in Hot Water and Prochloraz Solution on Spore and Mycelium of Fusarium fujikuroi Infected in Rice Seed (온탕소독과 prochloraz 침지소독이 벼 종자에 감염된 Fusarium fujikuroi의 포자와 균사의 형태에 미치는 영향에 대한 전자현미경적 연구)

  • This experiment was conducted to find the causes of ineffective seed disinfection methods such as rice seeds soaking in hot water and prochloraz EC solution when the rice seeds were severely infected by Bakanae disease. In case of rice seeds collected from severely diseased field by Bakanae disease, the pathogen as the forms of spores and mycelium were infected in plumule and inner and outer integument of embryo, aleurone layer, and pericarp layer. When the rice seeds were soaked in hot water, the appearances of spores and hypha on the outer pericarp layer were severely disordered, however those of inner region of outer integument and aleurone layer were shown normal. The membrane of hypha on the outer pericarp layer was destroyed within 24 hours, while some spores were healthy and germinated 7 days after soaking, when the rice seeds soaked 24 hours in 125 ppm prochloraz solution at $30^{\circ}C$. These results indicated that the seed disinfection methods were ineffective on the Bakanae disease severely infected rice seed because the hot water did not transmit the pericarp layer of rice seed and also prochloraz solution did not effectively destroy the spore of pathogen.

  • This study was performed to develop experimental model for milling degree and whiteness from analysis the CBB (colored bran balance) index using NMG dying and image processing. The endosperm layer increased and the pericarp, aleurone layer and CBB index decreased as the milling degree and whiteness increased. So, CBB index was closely linked to milling degree and whiteness. A empirical CBB index model was developed according to the milling degree and whiteness. The model were found to fit adequately to all test data with a coefficient of determination of 0.99, and therefore the CBB model proved to be quite applicable.

  • We have successfully used the low-pressure BioWare gene gun, developed for gene transfer in animal cells, for plant tissues. The BioWare device is easy to manipulate. Just 50 psi helium pressure was sufficient to transfer foreign genes into the aleurone layer and embryo of maize without causing tissue damage in the impact area. As shown by expression signals from invasive histochemical ${\beta}-glucuronidase$ (GUS) activity, the foreign reporter gene expressed well in bombarded tissues. This successful GUS-transient expression extends the application of this low-pressure gene gun from animal cells to plant tissues.

  • Barley is considered as one of the most important cereals for Korean in terms of production and utilization. In this review we concentrate mainly on the chemical structure of husk, aleurone layer, endosperm and embryo (especially endosperm). The relationships among the structure, characteristics and role of chemical components in barley kernel are also discussed. The nature of the fine-structure and minor components is not fully identified, however, a lot of investigation and progress in this area probably contributes the development of new technique for better utilization of barley in the future.

Physicochemical Properties of Sugary-Endosperm Mutants in Rice (벼 고당미 변이계통 배유의 이화학적 특성)

  • Some physicochemical properties of sugary rices were comparatively analyzed with the original cultivars, using Hwachung su-1, su-2, and Nampung su lines bred in Seoul Nat'l Univ., and EM 5 bred in Kyushu Univ. Sugary rice grains were smaller in width and thickness, and the relative dry weight of 1000 grains of sugary rices ranged from 49% to 72%, compared to normal ones. No distinct starch granules were found in endosperm of sugary rices exposured by SEM, with structural variations between Hwachung su-1 and su-2 lines. Sugary rices showed the significant increases in sucrose, glucose and fructose content, especially much higher in sucrose content showing 5.84-8.79%, in comparison with the normal ones, 1.11∼2.65%. However, sucrose and total sugar content varied within sugary lines carrying the same sugary gene, and were significantly lower in Hwachung su-2. Sugary rices, except Hwachung su-1, showed the same or lower ADV and harder gel characteristics, compared to normal ones. Protein and lipid content were much higher in sugary rices, due to the increased relative proportion of aleurone layer in brown rice caused by thicker aleurone layer than in normal rices and the reduction of endosperm part. However, there were no remarkable changes in amino acid and fatty acid compositions.

  • These studies were conducted to find out the polishing methods that improve yield and quality of the polished barley. Four varieties of hulled barley, Dongbori 1. Bunong, Kangbori and Suwon 182 which were produced in Suwon, Korea in 1979, were subjected to this experiment. The polishing machine, manufactured by Satake Co, was used as test mill. Increasing the roller speed of polishing machine causes more polished barley in a unit period. The speed influenced more in length than thickness or width of grain. Therefore the shape of grain became bold type as the speed increased. The optimum roller speed was 1,300rpm in ideal shape of polished barley. The lowest hardness was observed in the husk layer and the hardness was found in the decreasing order of the aleurone, testa, peri carp and the endosperm layer. The thickness of bran layer, weight of 1,000 kernel and hardness of polished barley were greatly different according to barley varieties. Also the length, thickness, width and the ratio of length to width of barley grain were significantly different in barley varieties. The ratio of length to width of the polished barley was 1.59 in Suwon 182, 1.53 in Bunong, 1.51 in Kangbori and 1.26 in Dongbori 1.

Effects of the Fractions of Oryza sativa cv. Heugjinmi on Plasma Glucose and Lipid Levels in Streptozotocin-Induced Diabetic Rats (흑진미 과피 용매분획물 투여시 당뇨 유발 흰쥐의 혈당 및 혈중 지질함량에 미치는 영향)

  • We have investigated the effects of the solvent fractions from the aleurone layer of Oryza sativa cv. Heugjinmi on plasma glucose and lipid levels in streptozotocin-induced diabetic rats. Sprague-Dawley rats were divided into 6 groups, normal, diabetic control, and four experimental groups. Diabetes mellitus was induced by injection of streptozotocin(45 mg/kg body weight) into tail vein, and fraction of Oryza sativa cv. Heugjinmi was administered orally in experimental rats for 14 days. The body weight, glucose, cholesterol, HDL-cholesterol, triglyceride, free fatty acid level, and aminotransferase activities were analyzed. It was identified that acetone and anthocyanin fractions significantly reduced the levels of glucose, triglyceride, and free fatty acid in plasma of diabetic rats. And HDL-cholesterol levels of acetone and anthocyanin groups were higher than those of diabetic control group. From these data, it can be postulated that acetone and anthocyanin fractions might possess rather potent hypoglycemic and hypotriglyceridemic properties in streptozotocin-induced diabetic rats.

Timing of Fusarium Head Blight Infection in Rice by Heading Stage

  • Fusarium graminearum causes the devastating plant disease Fusarium head blight and produces mycotoxins on small cultivated grains. To investigate the timeframe of F. graminearum infection during rice cultivation, a spore suspension of F. graminearum was applied to the rice cultivars Dongjin 1 and Nampyeongbyeo before and after the heading stage. The disease incidence rate was the highest (50%) directly after heading, when the greatest number of flowers were present, while only 10% of the rice infected 30 days after heading showed symptoms. To understand the mechanism of infection, an F. graminearum strain expressing green fluorescent protein (GFP) was inoculated, and the resulting infections were visually examined. Spores were found in all areas between the glume and inner seed, with the largest amount of GFP detected in the aleurone layer. When the inner part of the rice seed was infected, the pathogen was mainly observed in the embryo. These results suggest that F. graminearum migrates from the anthers to the ovaries and into the seeds during the flowering stage of rice. This study will contribute to uncovering the infection process of this pathogen in rice.

Ultrastructure of Compound Starch Granules and Protein Bodies of Starchy Endosperm Cell in Rice (쌀 배유세포 전분복합체와 단백질체의 미세구조)

  • The ultrastructure of the compound starch granules and the protein bodies of Odaebyeo rice of early matured variety were examined by light microscope and electron microscope. The endosperm cell appealed rectangular or octangular shape on the cross section. The thickness of cell wall containing of membraneous materials was about $0.5\;{\mu}m$ in diameter. The starch cell was filled compactly with globular or oval shaped compound starch granules with the size of $20{\sim}25\;{\mu}m$ in diameter. The compound starch granules were consisted of central core starch granule and concentrical $2{\sim}3$ layers of starch granules. The average thickness of the starch granules were about $5\;{\mu}m$. Most protein bodies were found in the aleurone layer The globular protin bodies were scattered near the compound starch granules and $2.5{\sim}3\;{\mu}m$ in diameter. The protein bodies composed of central electron dense materials and peripheral electron loose materials in limiting membrane.
