We studied on improvement method of rutin content using mulberry leaf powder. Mulberry leaves were collected and then hot-air dried and powdered for experiment. As a result, we have developed a pre-treatment method that extracts mulberry leaf powder with water or fermented alcohol with reflux extractor and then increases the rutin content by improving the process. Citric acid (0.1 ~ 1%) and 1000 ml fermented alcohol (50 ~ 95%) or water (10 ~ 50 times) was extracted with 100 g of mulberry leaf powder using a reflux extraction device ($80{\sim}90^{\circ}C$, 1 hour, twice). The extracts were collected, filtered and concentrated. For the recrystallization, the concentrate was dissolved by adding distilled water and allowed to stand at a low temperature. Then, the supernatant was discarded by centrifugation, and only the residue was lyophilized to prepare a final powder. As a result, regardless of the concentration of citric acid added, the content of rutin was higher in 90% fermented alcohol extract. Whereas, in the case of extracting with water, citric acid 0.5% was added to water 25 times as much as the weight of mulberry leaf powder, and 2274.4 (mg / 100g) of rutin content was highest in the case of refluxing twice at $80^{\circ}C$ for 1 hour. The powder with increased rutin content is expected to be applicable to various foods as a food additive. In addition, it can contribute to the improvement of the farm income by promoting consumption of mulberry leaf while satisfying the consumers' desire for functional food intake.
In this study, whitening activity of Lithospermum erythrorhizon extracts were investigated according to several extraction processes: water extraction at $100^{\circ}C$ (WE100) and $60^{\circ}C$ (WE60), 70% ethyl alcohol extraction (EE) and ultra high pressure extraction (HPE) at 500 MPa for 30 minutes at $60^{\circ}C$. The extracts from ultra high pressure extraction showed the highest tyrosinase inhibition and melanogenesis inhibition activities as 52% and 79.5%, respectively, in adding $1mg/m{\ell}$ than others extraction processes. HPE extracts also showed the strong reducing power as 3.19 that absorbance at 450 mm. The contents of polyphenol in WE100, we measured as $10.1{\mu}g/m{\ell}$ in adding $1mg/m{\ell}$. Extracts have a high total flavonoid contents by HPE as $4.1{\mu}g/m{\ell}$ at $1mg/m{\ell}$. We can conclude that better whitening activity of extracts from high pressure extraction was due to high antioxidant activities which could be extracted by higher polyphenol and flavonoid contents in HPE than others.
Optimum extraction conditions were investigated for the manufacture of Kiwi liqueur. Eight-weeks of storage was found to be the optimal extraction time for unsliced fruit, but 4 week of storage was found to be optimal extinction time when the fruit was sliced into 4 pieces. There were no changes in the brix, pH and total acidity during extraction. However 24.92 ppm of methanol and acetaldehyde was detected after extracting the liqueur after 8 week extraction time when the fruit was sliced into 4 pieces. The sensory evaluation showed the highest over all acceptability. The Kiwi liqueur was contained 48.90 mg% of total phenolic compounds, 16.7 mg% of vitamin C, 1.77% of malic acid, 1.18% of fructose, 1.12% of glucose and 0.08% of maltose at the optimum extraction conditions. The sensory quality of the Kiwi liqueur was estimated to be best when 0.035% of stevioside, 0.0085% of citric acid and 0.005% of salts were added to the Kiwi liqueur.

This study examined the optimization of alcohol extraction conditions for maximizing the total polyphenols derived from apple pomace, by response surface methodology (RSM). The effects of four independent variables, including $X_1$ (ratio of solvent to sample content), $X_2$ (dipping time), $X_3$ (extraction time), and $X_4$(extraction temperature), were investigated at five levels using central composite design (CCD). $Y_1$ (yield) and $Y_2$ (total polyphenols) were chosen as dependent variables. The coefficients of determination, $R^2$, were greater than 0.900 (0.9042 and 0.9555). The results showed that the model fit was very significant (p<0.001). The optimum extraction conditions were as follows: 13.00 mL/g for the ratio of solvent to sample content, 89.02 min for dipping time, 180 min for extraction time, and $70^{\circ}C$ for extraction temperature. At these conditions, the predicted total polyphenol content was 29.68 mg catechin equiv./g.
This study was carried out to investigate the volatile constituents change according to extraction methods, parts and heating methods. The research results are as follows: 23 kinds of components were extracted by steam distillation method; 8 by head space method; 16 by ether extraction method; 9 by Tenax-GC. This fact indicated that volatile constituents differ depending upon the extracting method. When the volatile constituents were extracted by steam distillation method and analyzed by GC-MSD, the content of the volatile constituents was 41.71 mg% in the stem and 85.37 mg% in the leaf. A total of 23 components - 14 kinds of hydrocarbon, 5 of aldehyde, 4 of alcohol- were detected in the stem. A total of 31 components - 21 kinds of hydrocarbon, 5 of aldehyde, 4 of alcohol and 1 of ester were detected in the leaf. The leaf had comparatively more varied volatile constituents than the stem. In the stem, the octanal content was highest and the contents of limonene, sabinene, -terpinene and -myrcene were higher; in the leaf, the content of octanal was highest and the contents of isobicyclogermacrene, limonene, -farnesene, undecaethiol, -terpinene, sabinene, elemene, -myrcen were higher. These were the major volatile constituents of the water d.opwort. The volatile constituent formation changed as the water dropwort was heated. The volatile constituents of the water dropwort heated in 1000 $m\ell$ were the most similiar to those of the raw water dropwort, and volatile constituent content was highest.
The solvent extracting of the tobacco has been carried out in order to find out any effects for the removal and/or reduction of nicotine from tobacco without affecting the tobacco flavor. We have now completed the physico-chemical characterization of the solvent untreated/treated tobaccos and 2) the contents of the tar and nicotine in the smoke. The results obtained ate like followings; 1. 3% alcohol extraction reduced the total nitrogen content for the Bright by 36.4%. However, it could not reduce the sugar content. 2. Nicotine content in tobacco leaves was inversely proportional to the dipping time Essentially same results were obtained in anacidic solution and even in a solution containing both alkali and acid. 3. Derivative thermogravimetric(DTG) curves indicated test extraction was effective by removing tobacco leaf components which could decompose or volatilize between the temperature of $150^{\circ}C\;and\;350^{\circ}C$. 4. 3% alcohol extracted leaf have the substantial improvements as following; (1) bulk filling power of 10% and 24% for Burley and Bright tobacco, respectively, (2) combustibility shortening of about 3 minute and 2 minute for Burley and Bright tobaccos, respectively, (3) reduction ratio(%) of nicotine was found to be shout 61.5% and 54.5% for Burley and Bright tobacco, respectively, (4) reduction ratio (%) of tar was obtained to he about 25% and 15% for Burley and Bright tobacco, respectively.

Recently, biohydrocarbons are gathering an interest as a new bioenergy due to the versatile applicability. In the present work, a process is proposed for the recovery of lipids from Recombinant E. coli MG1655 which provides longer chain fatty acids. After the growth of the recombinant E. coli, the cells were disrupted by high pressure homogenizer for obtaining intracellular lipids and the resulting solutions were centrifuged and extracted. For the efficient cell disruption with high pressure homogenizer, the pressure higher than 5,000 psi was required. In addition, under the conditions of applied pressure 5,000 to 20,000 psi, 1~3 pass homogenizing was enough for the more than 90% cell disruption. As organic solvents for extraction of lipid, hexane/isopropyl alcohol and ethyl acetate/ethanol systems showed excellent extracting power. With these solvent systems, the 60% lipid could be recovered. Moreover it was found that the extracted lipids contained long-chain fatty acids such as $C_{12}$, $C_{14}$, $C_{16}$ and $C_{18}$.
Background: The presence of antioxidants was not confirmed in the medicinal plant Equisetum hyamale grass. Purposes: This study was to determine the antioxidant and elastase inhibition effects of extracts of E. hyemale. Methods: Antioxidant functions of E. hyemale stems and roots were measured and extracted with hot water (HW) and ethyl alcohol (70EOH, 100EOH). Results: The extraction yield of stems was higher in HW extraction than in ethyl alcohol extraction. The polyphenol content was significantly higher in the root extract than in the stem. Total flavonoid content of 70EOH extract was significantly higher in root extract than in stem. Hemicelluloses pre-extracted from Korean mixed wood chip were investigated as a wet-end additive. Hemicelluloses dissolved in hot water and alkali solution were isolated by ethyl alcohol precipitation from pre-extractives. They showed molecular weight of 9,000 ~ 27,000 g/mol as revealed by size exclusion chromatography. The reduction of molecular weight through hot water extraction was caused by autohydrolysis. Chemical composition of the hemicelluloses were analyzed with high-performance liquid chromatography and UV-Vis spectroscopy. As the surface charge of isolated hemicelluloses were negative, the adsorption of hemicelluloses onto softwood unbleached kraft pulp fiber was promoted by poly-DADMAC. The physical properties of handsheets increased as the molecular weight of hemicellulose increased. On the other hands, the optical property decreased with hemicellulose adsorption.
To demonstrate the effect of formulation conditions and evaluations of structural integrity from ovalbumin containing poly lactide glycolide copolymer (PLGA) microspheres for Vaccine delivery, OVA microspheres were prepared by a W/O/W multiple emulsion solvent extraction technique. Dichloromethan (DCM) and Ethyl acetate (EA) were applied as an organic phase and poly vinyl alcohol (PVA) as a secondary emulsion stabilizer. Microspheres were characterized for particle size, morphology (optical microscopy and Scanning Electron Microscope (SEM)). Protein denaturation was evaluated by size exclusion chromatography (SEC), SDS-PAGE and isoelectric focusing (IEF). Residual organic solvent was estimated by gas chromatography (GC) and differential scanning calorimetry (DSC). Optical photomicrograph and SEM revealed that micro spheres were typically spherical but various morphologies were observed. Mean particle size $(d_{vs})$ of microspheres were in the range of $3{\sim}50{\mu}m$. Also, The protein stability was not affected by the fonnulation process and residual organic solvent was beyond the detection below 0.1ppm. These results demonstrated that micro spheres might be a good candidate for the parenteral vaccine delivery system.
