Possible Negative Effect of Pigmentation on Biosynthesis of Polyketide Mycotoxin Zearalenone in Gibberella zeae

  • We investigated a possible coordination between the biosyntheses of two polyketides in the cereal head blight fungus Gibberella zeae, zearalenone (ZEA) and aurofusarin (AUR), which are catalyzed by the polyketide synthases (PKS) PKS4/PKS13 and PKS12, respectively. To determine if the production of one polyketide influences that of the other, we used four different transgenic strains of G zeae; three were deficient for either ZEA or AUR or both, and one was an AUR-overproducing strain. The mycelia of both the wild-type and ${\Delta}PKS4$ strain deficient for ZEA produced AUR normally, whereas the mycelia of both the ${\Delta}PKS12$ and ${\Delta}PKS4::{\Delta}PKS12$ strain showed no AUR accumulation. All the examined deletion strains caused necrotic spots on the surface of com kernels and were found to produce the nonpolyketide mycotoxins trichothecenes to the same amount as the wild-type strain. In contrast, the AUR-deficient ${\Delta}PKS12$ strains produced greater quantities of ZEA and its derivatives than the wild-type progenitor on both a rice substrate and a liquid medium; the AUR-overproducing strain did not produce ZEA on either medium. Furthermore, the expression of both PKS4 and PKS13 was induced earlier in the ${\Delta}PKS12$ strains than in the wild-type strain, and there was no difference in the transcription of PKS12 between the two strains. Therefore, these results indicate that the ZEA biosynthetic pathway is negatively regulated by the accumulation of another polyketide (AUR) in G zeae.

  • The effect of an artificial diet on developmental rate, a life history parameter, was examined for the swallowtail butterfly Papilio xuthus. Artificial insect diets are an essential component of many insect rearing systems that produce insects for research purposes. Complex agar-gelled diets are generally prepared in large batches and used shortly after preparation because the degradation of perishable diet ingredients, such as vitamins and fatty acids, can adversely affect insect quality (Brewer 1984). However, the timing of diet preparation may be inconvenient, and large batches wasteful, if the unused excess is discarded. The percentage of pupation varied considerably, with no significant differences among diets, on which a maximum pupation percentage of 83% was observed. Pellet-type diets were investigated with the aim of developing a more easily prepared diet. The extrusion of the artificial diet under high temperature and pressure may induce desirable chemical and physical changes in the extruded product. The purpose of the present study was to develop an artificial diet for rearing P. xuthus.

  • The functional packaging film, that keeps the freshness of the fresh produce, has composed of the ferro fluid particles, chitin and chitosan. The film exhibited selectivity in permeability for carbon dioxide and ethylene gases. Antagonistic effect of the film for Fusarium, Botrysphania, Altanaria and E. coli was excellent. The film kept the favorable taste of Kimchi and Korean strawberry well.

Screening of Deoxynivalenol Producing Strains from Agricultural Products by Immunoanalytical Method (면역분석기법에 의한 농산물에서의 Deoxynivalenol생성균 검색)

  • In order to evaluate the safety of agricultural products in Korea, we carried out work by screening of Fusarium species. which can produce deoxinivalenol(DON) from agricultural products in Western Gyeongnam, Korea. From 215 samples of soil agricultural products, 129 strains of Fusarium species were obtained. The production of DON was verified by thin layer chromatography(TLC) As the results of TLC, 25 strains were identified as DON producing strain. But only 10 strains were identified as DON producing strains by enzyme-linked immunosorbent assay (ELISA). The maximum DON producing strain No.41 was isolated from corn. In conclusion. the above results indicate that DON producing fungi contaminated agricultural products in Korea. Therefore further studies are required to accumulate more detailed data about the contamination of DON in various agricultural products.

  • To address complex solutions to tack1e the cost rise with the increasing of the prices of crude oil, and to lower warming gas discharge in agricultural sector in line with the framework Convention on Climate Change, we need to net only develop the methods of simply reducing energy use ratio, but also effectively energize resources discharged in the course of agricultural production aimed at enabling repealed farming, and develop systems to reinvest such energy into agricultural production and apply them to actual farming. To that end, specifically, we need to reduce the energy cost in agricultural production, and produce and harness bio gas that makes it possible to stabilize the continued supply of energy. Thus, in this research, we seek to discuss energy use volume and warming gas discharge, and the characteristics of bio gas and its use methods alike, in agricultural sector, and thus present methods for environment-friendly and continual development in agriculture.

  • The principle goal of environmentally-friendly agriculture is to maintain and conserve water and agriculture environment including drinking water resources by properly using agricultural materials such as agricultural chemicals, chemical fertilizers, and other agricultural materials according to act 19 of foster law of environmentally-friendly agriculture. To achieve these goals, we have to establish Integrated Nutrient Management(INM) and Integrated Pesticide Management(IPM) which are most important core technologies for environmentally-friendly rice cultivation. However, there are lack of criteria and technology for evaluation category according to soil management and its soil classes to practice an environmentally-friendly agriculture. Therefore, we should eatablish the standards to produce the safe agricultural products based on the soil physical and chemical characteristics which are basic properties of soil to accomplish the principle aims of environmentally-friendly agriculture.

Detection of Extracellular Enzyme Activities in Ganoderma neo-japonicum

  • The ability of Ganoderma to produce extracellular enzymes, including ${\beta}$-glucosidase, cellulase, avicelase, pectinase, xylanase, protease, amylase, and ligninase was tested in chromogenic media. ${\beta}$-glucosidase showed the highest activity, among the eight tested enzymes. In particular, Ganoderma neo-japonicum showed significantly stronger activity for ${\beta}$-glucosidase than that of the other enzymes. Two Ganoderma lucidum isolates showed moderate activity for avicelase; however, Ganoderma neojaponicum showed the strongest activity. Moderate ligninase activity was only observed in Ganoderma neo-japonicum. In contrast, pectinase, amylase, protease, and cellulase were not present in Ganoderma. The results show that the degree of activity of the tested enzymes varied depending on the Ganoderma species tested.

Efficient Propagation by Bioreactor System of Korean Native Seosanjong in Ginger

  • For the purpose of establishing an efficient propagation through airlift bioreactor system of Zingiber of officinale Rosc. Korean native Seosanjong, the effect of different factors and bioreactor on cultured plantlets were investigated. The highest number of plantlets, fresh weight per plant was obtained from explants when cultured in MS liquid medium including 0.3 mg/L NAA and 2.0 mg/L kinetin for 40 days. A 10 L bottle type bubble bioreactor, compared with 250 mL Erlenmeyer flask, was more efficient, producing 4.7 plantlets or from 1.5 to 1.6 times more than did the Erlenmeyer flask. The results demonstrate the rapid mass propagation of airlift bioreactor to produce normal ginger.
