Effects of photoperiod and light intensity on milk production and milk composition of dairy cows in automatic milking system

  • The purpose of this study was to determine the effects of photoperiod and light intensity on milk production, milk composition, hormones levels and blood metabolites indices of Korean Holstein dairy cows in automatic milking system (AMS). A total of 24 Holstein dairy cows were selected and used to four subsequent treatments for the experimental periods of 60 days. The light programs consisted of (1) Control: the natural photoperiod with 14.2 h of the light period and 9.4 h of the dark period (below 10 Lux); (2) T1: 16 h of the long day photoperiod (LDPP) with 50 Lux of light; (3) T2: 16 h of LDPP with 100 Lux of light; and (4) T3: 16 h of LDPP with 200 Lux of light, respectively. Importantly, there was a significant difference in the thurl activity of dairy cows between the different light intensity programs (p < 0.05). Milk yield was higher in T1 and T2 (40.80 ± 1.71 and 39.90 ± 2.02 kg/d, respectively) than those of Control and T3 (32.18 ± 1.51 and 35.76 ± 2.80 kg/d, respectively) (p < 0.05), but DMI was lower in T1, T2, and T3 compared to Control (p < 0.05). Also, milk fat percentage, the contents of milk fat and total solids were higher in T2 than those in the others (p < 0.05). The average daily melatonin level in milk was high to T3 (28.20 ± 0.43 pg/mL), T2 (24.62 ± 0.32 pg/mL), T1 (19.78 ± 0.35 pg/mL), and Control (19.36 ± 0.45 pg/mL) in order (p < 0.05). Also, the cortisol levels in milk and blood were lower in treatment groups than in Control (p < 0.05). The results of this study showed that it will be effective to improve the milk yield and milk composition, and to reduce the stress of dairy cows when the light conditions regulate to extend the photoperiod to 16 h at a light emitting diode (LED) intensity of 100 Lux under the AMS in dairy farm.

효소처리와 트레할로스를 첨가한 백설기의 품질 특성 변화 연구 (A Study on the Quality Characteristics of Backsulgi Prepared with Combined Treatment of Enzyme and Trehalose)

  • 본 연구는 백설기의 노화를 억제 및 저장 중 품질 향상을 위한 Novamyl의 최적 첨가량을 0.1%로 선정하고 트레할로스를 5%, 10%, 15% 첨가하여 백설기를 제조한 후 상 온(${25^{\circ}C}$)에서 0, 1, 2, 3일간 저장하면서 기계적 및 관능적 특성과 노화 속도 변화를 살펴보았으며 그 결과는 다음과 같다. 수분활성도는 트레할로스의 첨가량이 증가할수록 유의적으로 감소하였으며 저장 일수에 따른 차이는 없었다. 색도는 트레할로스를 첨가한 백설기의 명도(L값)가 유의적으로 낮게 나타났으며 적색도를 나타내는 a값은 첨가군이 대조구에 비해 낮게 나타났고 황색도(b값)는 첨가군이 다소 낮은 값을 보였으며 저장 일수가 증가함에 따라 황색도가 감소하는 경향을 보였다. 기계적 조직감에 대해서는 트레할로스의 첨가량이 증가할수록 경도가 감소하였고 저장 중 씹힘성의 감소폭이 감소하였다. Avrami 방정식으로 노화속도를 측정하였으며, Avrami exponent(n) 값과 시간상수(1/k) 값을 계산한 결과 트레할로스 첨가군이 대조구에 비해 노화속도가 감소하였음을 알 수 있었다. 관능검사 결과에서 15% 첨가구가 대부분의 항목에서 우수한 결과를 보였고 특히 전반적인 기호도 및 씹힘성 항목에서 높은 선호도 값을 보였으나 10% 첨가구와의 유의적인 차이는 보이지 않았다. 이와 같은 실험결과를 통해 백설기 제조 시 Novamyl 0.1%를 처리하고 트레할로스를 첨가할 경우 노화 억제와 품질 향상의 효과가 있었으며, 특히 기계적 측정 결과와 관능검사 결과를 종합했을 때, 트레할로스를 10% 첨가가 15%의 첨가에 비해 유의적인 차이가 적어 가장 적절한 것으로 나타났다. 효소의 첨가는 백설기의 노화를 억제하는 효과가 있지만 물성적인 측면에서 조직이 과도하게 물러지는 등의 문제를 보이는데 이에 트레할로스를 적당량 첨가함으로써 노화 억제를 향상시키고 물성적인 측면과 기호도를 향상시켜 저장성이 우수하고 소비자의 입맛에 적합한 백설기의 제조가 가능하다고 사료된다.

일부 보건진료소에서 실시한 건강증진프로그램이 농촌여성노인의 노쇠에 미치는 효과 (The Effect of Health Promotion Program on the Frailty of Rural Elderly Women Implemented at Primary Health Care Posts)

  • 이 연구는 J시 일부 보건진료소에서 실시한 건강증진프로그램이 농촌여성노인 노쇠정도 확인을 위한 건강상태(지각된 건강상태, 노쇠 점수, 상 하체 유연성, 최대악력, 동적평형검사 Timed Up and Go)에 미치는 효과를 규명하기 위함으로 대한노인병학회에서 개발한 한국형 노쇠측정도구 8개 항목을 통해 보건진료소 관할지역 노인들의 노쇠수준을 파악하고, 농촌 여성노인들의 노쇠수준에 맞는 12주 건강증진프로그램을 적용하였다. 건강증진프로그램(프로그램명: ${\bigcirc}{\bigcirc}$보건진료소와 주민이 함께 만드는 하하호호 백세건강마을 만들기)은 J시 보건진료소 특성화 사업의 한 부분으로 연구자인 보건진료전담공무원이 전문가의 자문을 얻어 농촌여성 노인들에게 적합하도록 수정 보완하여 실시하였다. 연구 결과 노쇠측정도구 8개 항목을 통한 노쇠점수도 실험군이 대조군에 비해서 유의한 차이를 보였는데, 8개 항목 중 주관적인 현재 건강상태와 우울한 감정의 개선이 노쇠점수가 향상에 특히 도움이 되었다. 건강상태 변화로 왼쪽 상체유연성은 실험군이 약 2배(+17cm), 오른쪽 상체유연성은 +11cm, 하체유연성은 실험군이 +6cm 증가하였고, 대조군은 거의 변화가 없어 통계적으로 유의한 차이가 있었다. 노인의 근력평가로 최대악력은 실험군이 약+4kg 증가하였고, 동적 균형 검사(Timed Up and Go test)는 실험군이 3초 빨라지고 대조군은 거의 변화가 없어 통계적으로 유의한 차이가 있었다. 따라서 이 연구에서 실시한 건강증진프로그램이 상 하체 유연성 및 근력, 민첩성 등 운동능력을 향상하는 데 도움이 되었다고 볼 수 있다. 이상의 연구 결과를 바탕으로 의료취약지역 보건 진료소 관할 인구의 평균수명 연장과 고령화로 노쇠한 농촌 노인들의 특성을 반영한 근력강화 및 유연성 운동과 인지개선 활동 등 다양한 건강증진프로그램의 지속적인 개발과 프로그램의 표준화를 통한 확대 적용이 필요하다.

닭과 돼지의 항균펩타이드 NK-Lysin으로부터 조합된 펩타이드의 In Vitro 항균효과 (In Vitro Antibacterial Effects of the Chimeric Peptides from Chicken and Pig Antimicrobial Peptide NK-Lysin)

  • 가축에서 항생제의 남용은 항생제 내성균의 발생을 유도하므로 가축과 사람의 건강을 위협할 수 있다. 이에 따라 항생제를 대체할 수 있는 물질 중 하나로 항균펩타이드(antimicrobial peptides)가 연구되고 있다. 항균펩타이드는 12-50개의 아미노산으로 구성된 펩타이드로 숙주에 감염된 다양한 박테리아를 사멸시키고 면역작용을 조절하는 등 선천성 면역에 중요한 물질이다. 본 연구에서는 닭과 돼지의 NK-2를 조합하여 cpNK라는 chimeric 펩타이드를 합성하였으며 이것의 항균효과를 검증하였다. 그 결과, cpNK는 그람음성균인 E. coli와 Salmonella spp. 그리고 그람 양성균인 L. monocytogenes과 S. aureus를 사멸시켰으며, cpNK는 그람음성균에서 더 강력한 살균효과를 보였다. 특히, cpNK는 메티실린 내성 황색포도상구균 (methicillin-resistant Staphylococcus aureus, MRSA)에 대해서도 항균효과를 보여주었다. 또한, 27aa인 cpNK의 크기를 줄인 21aa의 cpNK-a1과 13aa의 cpNK-a2를 합성하여 마찬가지로 항균효과를 측정한 결과, cpNK보다는 약한 항균효과를 보였지만 여러 병원균의 생존율을 감소시켰다. 따라서 본 연구를 통해 우수한 항균효과를 갖는 cpNK와 cpNK로부터 유도된 펩타이드들을 항생제 대체제로 사용할 수 있는 가능성을 확인할 수 있었다.

Occurrence of an invertase producing strain of Aspergillus niger LP5 isolated from longan pollen and its application in longan syrup production to feed honey bees (Apis mellifera L.)

  • Background: In northern Thailand, the longan flower is the principal nectar source for honey production. Microorganisms play a critical function in the agricultural ecology. The morphological characteristics of fungal species found in longan pollen were studied. Aspergillus spp. were found to be invertase-producing strains and were employed in the longan syrup production process. The purpose of this study was to evaluate the effects of invertase-added longan syrup on the adult honey bee population numbers that were fed by this syrup for 16 weeks. Results: Different fungal species were found in longan pollen samples. Aspergillus was the main genus, with three predominant sections: Nigri, Flavi, and Terrei. Other isolated species were Trichoderma spp., Rhizopus spp., Neurospora spp., Chaetomium spp., Fusarium spp. and Penicillium spp. However, Aspergillus spp. is the only fungal species that produces the enzyme invertase. The invertase-producing strains belonging to the Aspergillus section Nigri were found to be A. niger LP5 with an optimum activity at pH 6.0 and 60℃. When A. niger LP5 invertase was used for longan syrup processing, the highest levels of glucose (3.45%) and fructose (2.08%) were found in invertase added longan syrup (C), while fresh (A) and boiled longan syrup (B) had lower contents of both sugars. The sucrose content was detected in (A) at 4.25%, while (B) and (C) were at 4.02% and 3.08%, respectively. An appropriate amount of sugar to feed and maintain the honey bee population was considered. The data showed no statistically significant differences between the two selected forms of longan syrup compared to the sugar syrup examined by the adult honey bee population. Conclusions: The main species of isolated fungi from longan pollen were Aspergillus spp. The discovery of an invertase-producing strain of A. niger LP5 has enabled its application for enzyme utilization in the invert sugar preparation process. The adult worker bee populations fed by longan syrup from both boiled and invertase-added sources showed an increasing trend. Artificial syrup made from longan fruit to feed honey bees when natural food sources are limited can be applied.

EQ-5D 지수를 활용한 만성질환별 삶의 질의 성별 및 연령에 따른 변화: 2017~2019년 국민건강영양조사 자료를 이용하여 (The Age and Sex-specific Quality of Life by Chronic Disease Using the EQ-5D Index : Based on the 2017-2019 Korea National Health and Nutrition Examination Survey)

  • Objectives: This study analyzed the decline in quality of life according to age in the chronic disease patient group, quantified it as a quantitative index, and compared it by sex and chronic disease. Methods: In the 2017-2019 Korea National Health and Nutrition Examination Survey database, 11,473 adults aged 19 years or older, excluding cancer patients, were analyzed for age-specific changes in the EQ-5D Index by chronic disease. The decline in quality of life according to age in patients with chronic diseases was analyzed by linear regression analysis while controlling for general characteristics. Then, linear regression analysis was performed according to sex. Results: In the case of the control group, the quality of life decreased by 0.0004 for every 1-year increase in age(P<0.001). By chronic disease, asthma(β=0.0019, P<0.001), arthritis(β=0.0017, P=0.002), thyroid dis- ease(β=0.0016, P=0.015), dyslipidemia(β=0.0011, P=0.020), and hypertension(β=0.0009, P=0.027) mostly showed a greater decrease in quality of life than the control group. In addition, when divided into two groups by sex, hypertension(β=0.0012, P=0.029), thyroid disease(β=0.0041, P=0.038), and arthritis(β=0.0022, P<0.001) showed a significant decrease in quality of life only in male. Diabetes(β=0.0056, P=0.038), dyslipi- demia(β=0.0022, P=0.001) significantly decreased quality of life only in female. Conclusions: Chronic disease had a negative impact on patients perception of quality of life, and the more severe the pain and activity limitation due to the chronic disease, the more severe it was. It also showed different patterns according to sex. Therefore, it is necessary to allocate more medical resources and provide policy support to prevent chronic diseases, which are serious social problems.

In-silico annotation of the chemical composition of Tibetan tea and its mechanism on antioxidant and lipid-lowering in mice

  • BACKGROUND/OBJECTIVES: Tibetan tea is a kind of dark tea, due to the inherent complexity of natural products, the chemical composition and beneficial effects of Tibetan tea are not fully understood. The objective of this study was to unravel the composition of Tibetan tea using knowledge-guided multilayer network (KGMN) techniques and explore its potential antioxidant and hypolipidemic mechanisms in mice. MATERIALS/METHODS: The C57BL/6J mice were continuously gavaged with Tibetan tea extract (T group), green tea extract (G group) and ddH2O (H group) for 15 days. The activity of total antioxidant capacity (T-AOC) and superoxide dismutase (SOD) in mice was detected. Transcriptome sequencing technology was used to investigate the molecular mechanisms underlying the antioxidant and lipid-lowering effects of Tibetan tea in mice. Furthermore, the expression levels of liver antioxidant and lipid metabolism related genes in various groups were detected by the real-time quantitative polymerase chain reaction (qPCR) method. RESULTS: The results showed that a total of 42 flavonoids are provisionally annotated in Tibetan tea using KGMN strategies. Tibetan tea significantly reduced body weight gain and increased T-AOC and SOD activities in mice compared with the H group. Based on the results of transcriptome and qPCR, it was confirmed that Tibetan tea could play a key role in antioxidant and lipid lowering by regulating oxidative stress and lipid metabolism related pathways such as insulin resistance, P53 signaling pathway, insulin signaling pathway, fatty acid elongation and fatty acid metabolism. CONCLUSIONS: This study was the first to use computational tools to deeply explore the composition of Tibetan tea and revealed its potential antioxidant and hypolipidemic mechanisms, and it provides new insights into the composition and bioactivity of Tibetan tea.

농촌거주 노인정 급식도입을 위한 식생활 비교 평가 연구 (Comparative Analysis of Dietary Intake for Introduction of Meal Service in Pavilion of the Elderly living in Rural Area)

  • Aging rate of rural area in our country is relatively high compared to that of the urban area. Thus, the introduction of meal service for the elderly residing in the rural area is necessary for their better living quality. Food habit, health and the nutritive intake conditions during the busy farming season were surveyed and comparative analysis of dietary intake for the introduction of meal service in pavilion of the elderly living in Chungnam, Kangwon, Jeonnam and Kyungbuk was performed for basic reference data of meal service introduction to the pavilion of the elderly in rural area. In general subject, the male elderly had a significant difference in marital state and showed that 79.4% was married and 20% was separated by death(P<0.05). In allowance, there were no significant difference but most of them lived with less then three hundred thousand won and especially, female lived with less then one hundred thousand won. In health state, the female elderly showed significant difference on difficulty with every day activity but with small trouble although they had to prepare their own meal(P<0.05). The condition dental health conditions of the female elderly had a significant difference showing bad conditions in following order; Kangwon(48%)>Chungnam(38.1%)>Kyungbuk(22.9%)>Jeonnam(22.5%)(p<0.05). The female elderly showed a significant difference in usage of denture and number of the female elderly without using the denture were very high(p<0.05). In nutrition intake condition, amount of sodium was very high but intakes of fiber and calcium were relatively 1ow(P<0.05). Meal service introduction in the pavilion of the elderly is suggested for the improvement of the life quality of the elderly in rural area. When developing the menu for them, conditions such as shortage of the fiber and calcium in diets, the dental conditions should be considered.

유치원 교육과정 기반 영양·식생활 교육 내용 체계화: 질적 기초 연구 (Systematization of food and nutrition education content based on national kindergarten curriculum: a qualitative formative study)

  • Objectives: This study is intended to develop a curriculum for kindergarten food and nutrition education aimed at preschool children, reflecting government policy and meeting the demands of preschool settings. Methods: Existing educational materials were analyzed, and key elements of the 2019 Revised Nuri Curriculum ("Nuri Curriculum") and Guidelines for Nutrition and Food Education in Kindergartens, Elementary, Middle, and High Schools ("Guidelines") were examined as foundational information for developing the curriculum for food and nutrition education. Results: Basing ourselves on the five domains of the Nuri Curriculum, "Physical Activity and Health," "Communication," "Social Relationships," "Art Experience," and "Natural Science Inquiry," we integrated three areas from the Guidelines, namely "Dietary Habits and Health," "Dietary Habits and Safety," and "Dietary Habits and Culture," to structure the curriculum for kindergarten food and nutrition education. Three specific domains, "Nutrition and Health," "Food and Culture," and "Safe Dietary Practices," were tailored for preschool children, each comprising core concepts, content elements, and educational materials. In the "Nutrition and Health" domain, core concepts such as "nutrition" were addressed through content elements such as "balanced eating" and "vegetables and fruit," while "health" included elements such as "eating regularly" and "nutrients for disease prevention," each with two educational content components. The "Food and Culture" domain focused on "food" with content on "local foods (vegetable-garden experience)" and "food culture" with content on "our dining table (rice and side dishes)," "our agricultural products," "global cuisine (multiculture)," and "considerate dietary practices," each with four educational content components. The "Safe Dietary Practices" domain included core concepts such as "hygiene" with content on "hand-washing habits" and "food poisoning management," and "safety" with content on "food labeling." Conclusions: The systematized curriculum for kindergarten food and nutrition education aligns with the Nuri Curriculum and is interconnected with the Guidelines. This curriculum can be used as foundational material for developing educational resources tailored to the characteristics of preschoolers, contributing to effective implementation in early childhood education.

여름철 활엽수림에서의 휴식이 온열환경 쾌적성 및 인체의 생리ᐧ심리적 반응에 미치는 영향 (Thermal Comfort and the Physiological and Psychological Effects of Spending Time in Broad-Leaved Forests in Summers)

  • 활엽수림은 여름철에 착엽을 통해 일사를 차단하여 열 스트레스를 완화하는 역할을 하는 것으로 알려져 있지만, 이와 관련된 데이터의 축적은 부족한 실정이다. 따라서 본 연구는 여름철 활엽수림에서의 휴식이 온열환경 쾌적성 및 인체의 생리・심리적 반응에 미치는 영향을 분석하는 것을 목적으로 하였다. 연구대상자는 남녀 대학생 32명(21.5±2.1세)으로 하였다. 연구대상자는 두 명이 한 조를 이루어 각각 실험지(수관울폐도: 76.6%)와 대조실험지(25.9%)로 이동하였고, 일련의 실험과정을 이후 장소를 교대하여 같은 실험을 진행하였다. 실험과정은 다음과 같다. 연구대상자는 의자에 앉아 휴식한 후, 눈을 감고 5분 동안 안정을 취하였다. 이때의 온열환경 쾌적성을 평가하고 심박변이도, 심박수, 이마체온을 연속적으로 측정하였다. 이후 혈압 및 맥박수를 측정하였으며, 주관평가를 진행하였다. 그 결과, 실험지는 통계적으로 유의미한 예상평균한서감지수 및 예상불만족률의 감소, 부교감신경활동의 증가, 심박수・이마체온・이완기혈압・맥박수의 감소가 나타났으며, 주관적 온열감 및 쾌적감이 개선되었다. 결론적으로, 수관울폐도가 높은 활엽수림은 여름철 온열 스트레스를 감소시키며, 인체를 생리・심리적으로 안정시킨다는 것을 확인하였다.
