Biofunctional Activities of Citrus Flavonoids (감귤류 플라보노이드의 생리기능 활성)

  • This review showed a discussion on the biofunctional activities of citrus flavonoids. The major flavonoids of citrus species, hesperidin, hesperetin, naringin, and naringenin, were selected to evaluate their biological effects on the lipid metabolism in rats and hamsters, the proliferation of human hepatocyte HepG2 cells, and the antioxidative effect in lipid peroxidation models. These flavonoids showed hypotriglyceridemic effect in hamsters and hypochloesterolemic effect in rats. They also significantly inhibited the activities of phosphatidate phophohydrolase and acyl-CoA: cholesterol acyltransferase, which are key enzymes for biosynthesis of triglyceride and cholesterol, repectively, in vivo and in vitro experiments. These biofunctional activities by citrus flavonoids were shown more potent in the aglycone flavonoids, hespreretin and naringenin, than their corresponding glycoside flavonoids, hesperidin and naringin. These aglycone flavonoids also have inhibitory effects on proliferation of human hepatocyte cancer HepG2 cells. Hesperidin showed lowering activities of cellular triglyceride and cholesterol concentrations in HepG2 cells. Citrus flavonoids have significant importance in functional food industry as biofunctional active ingredients.

Changes in the Accumulation of Isoflavonoids in Soybeans by Stress (Stress에 의한 대두의 Isoflavonoid 화합물 축적변화)

  • The changes of the time-dependent accumulation of isoflavone aglycones(daidzein, genistein) and their glucosides(daidzin, genistin) by various stress-inducing treatment on cotyledon of soybeans(Dankyungkong and Paldalkong) were investigated. Levels of isoflavone aglycones and their glucosides in soybeans treated with UDP-glucose, $MgSO_4$ and $HgCl_2$ and infected with phytopathogen appeared to be higher than those of treatment with distilled water. When compared with data for control, the levels of isoflavone glucosides in citrate-treated soybeans were increased but those of their aglycones did not appear appreciable differences. In Paldalkong treated with UDP-glucose, $MgSO_4$ and $HgCl_2$ the levels of isoflavone aglycones and their glucosides were higher than in Dankyungkong. In particular, the accumulation of daidzein in Paldalkong was significantly higher than in Dankyungkong. By infection with Botrytis cinerea, the maximum amount of accumulation of daidzein in two cultivar did not to be a large different, but accumulation time appeared more rapidly in Paldalkong than Dankyungkong. The accumulation amount of daidzein and genistein in Dankyungkong treated with elicitors appeared to be similar at initial stage, but the level of daidzein after 48hours appeared to be higher than that of genistein. In Paldalkong, the level of daidzein was higher than that of genistein at all stage. The level of daidzein in soybeans infected with phytopathogen appeared to be higher than that of genestein.

Antioxidative and Antiaging Effects of Sorbus commixta Twig Extracts (마가목 가지 추출물의 항산화 및 항노화에 관한 연구)

  • In this study, the antioxidative effects and inhibitory effects on tyrosinase and elastase of Sorbus commixta (S. commixta) twig extracts were investigated. The aglycone fraction of S. commixta twig extract showed the prominent free radical (1,1-diphenyl-2-picrylhydrazyl, DPPH) scavenging activity($FSC_{50}$, $13{\mu}g/mL$). Reactive oxygen species (ROS) scavenging activities of S. commixta twig extracts on ROS generated in $Fe^{3+}-EDTA/H_2O_2$ system were investigated by the luminol-dependent chemiluminescence assay. The 50 % ethanol extract among extracts showed the most prominent ROS scavenging activity ($OSC_{50}$, $0.189{\mu}g/mL$). The cellular protective effects of extract/fractions of S. commixta twig on the rose-bengal sensitized photohemolysis of human erythrocytes were investigated. The 50 % ethanol extract and ethyl acetate fraction showed the cellular protective effects against ROS in a concentration dependent manner ($5{\sim}50{\mu}g/mL$). The inhibitory effect of S. commixta twig extract on tyrosinase was investigated to assess the whitening efficacy. The ethyl acetate ($IC_{50}$, $113.2{\mu}g/mL$) and aglycone fraction($IC_{50}$, $105.3{\mu}g/mL$) on tyrosinase showed more remarkable inhibitory effect than arbutin($IC_{50}$, $226.88{\mu}g/mL$), known as the whitening agent. The inhibitory effect of aglycone fraction ($IC_{50}$, $6.9{\mu}g/mL$) on elastase was simillar to quercetin($IC_{50}$, $6.1{\mu}g/mL$), flavonoid known as reference compound. These results indicate that extract/fractions of S. commixta twig can function as antioxidants in biological systems, particularly skin exposed to UV radiation by scavenging $^1O_2$ and other ROS, and protect cellular membranes against ROS. S. commixta twig extracts can be applicable to new functional cosmetics for anti-aging products.

Antioxidative and Inhibitory Activities on Tyrosinase of Hippophae rhamnoides Leaf Extracts (비타민나무 잎 추출물의 항산화 및 타이로시네이즈 저해활성)

  • In this study, the antioxidative and inhibitory effects on tyrosinase and elastase of Hippophae rhamnoides (H. rhamnoides) leaf extracts were investigated. The ethyl acetate fraction of H. rhamnoides extracts showed more effective free radical (1,1-diphenyl-2-picrylhydrazyl, DPPH) scavenging activity ($FSC_{50}$ = 4.68 ${\mu}g$/mL). Reactive oxygen species (ROS) scavenging activity ($OSC_{50}$) of the aglycone fraction in the luminol-dependent $Fe^{3+}$-EDTA/$H_2O_2$ system was 0.19 ${\mu}g$/mL. The aglycone fraction exhibited more prominent cellular protective effects (${\tau}_{50}$, 133.3 min at 10 ${\mu}g$/mL) in the $^1O_2$-induced photohemolysis of human erythrocytes. The inhibitory effect ($IC_{50}$) of the aglycone fraction on tyrosinase was 54.86 ${\mu}g$/mL, and more effective than arbutin known as whitening agent. These results indicate that fractions of Hippophae rhamnoides extract can be used as antioxidants in biological system, particulaly skin exposed to UV radiation by quenching and/or scavenging $^1O_2$ and other ROS, and protecting cellular membranes against ROS.

Changes of physiochemical properties and biological activity during the fermentation of Doenjnag with bitter melon (Momordica charantia L.) (여주 함유 된장의 발효 중 이화학적 특성 및 생리활성의 변화)

  • This study evaluated the changes of physiochemical properties, phytochemical compounds (isoflavones and phenolic acids), and biological activity during the fermentation of Doenjang without and with bitter melon powder (BMP). The pH decreased from 6.41-5.83 to 5.81-5.24, during the fermentation of Doenjang, while the acidity increased from 0.42-0.65% to 1.28-1.48%. The viable cell numbers of Bacillus and Yeast, salinity, and total amino acid contents increased at the end fermentation (60 day). Also, the fermented Doenjang (FD) with 10% BMP showed the highest ${\gamma}$-aminobutyric acid (GABA, 129.87 mg/100 g) contents, among all the Doenjang samples. The FD exhibited significantly higher inhibitory activities than unfermented Doenjang (UFD) on radicals and ${\alpha}$-glucosidase. The phytochemical compounds including isoflavone-aglycones and phenolic acids increased, whereas isoflavoneglycosides decreased in the BM following fermentative processing. Moreover, the total phenolic, isoflavone-aglycone, and phenolic acid contents were markedly increased, leading to a general increase in antioxidant and ${\alpha}$-glucosidase inhibition activities after fermentation. These results suggest that BMP may be used to prepare a new type of fermented Doenjang with improved antioxidant and antidiabetic activities.

Influence of Abiotic Treatments on Isoflavone Accumulation in Soybean Seeds during Germination (물리적 요인이 콩 발아 중 isoflavone 함량에 미치는 영향)

  • Isoflavones, a group of secondary metabolites present in plants and especially abundant in soybeans, are garnering increasing interest because of their medicinal properties. Numerous environmental factors influence isoflavone accumulation in plants. Successful attempts to increase isoflavone contents in soybean seeds have not been widely reported. In this report, we examined an effective method for producing a high amount of isoflavones during soybean seed germination, by treating the seed with 4 abiotic factors. Soybean seeds were exposed to ultrasound (50 Hz) and heat ($40^{\circ}C$) for 30minutes, then germinated for 5 days in darkness. Another sample was subjected to water deficit during germination in darkness, and the other sample was subjected to light conditions during the germination period. The results showed that the ultrasound treatment increased isoflavone content during soybean germination. Three days after germination, the ultrasound-treated seeds presented the highest isoflavone content (6,042.1 ug/g dry weight) compared to the other treatments, which was also 1.3-fold increases over the content of the raw seed's before germination. With respect to each type of isoflavone group, aglycone (partially genistein) content was markedly higher, whereas glucoside was lower than that of the untreated samples. In other experiment, total isoflavone content after heat, water deficit, and light treatments was 5,600.0, 4,740.1, and 5,631.4 ug/g, respectively. These present work suggests that ultrasound treatment is a novel approach for improving the production of isoflavones in soybean seeds during germination.

항암제로서 새로운 Anthracycline 유도체의 합성

  • 1960년대 이후 광범위한 항암제로서 그 임상효과가 탁월한 물질로 알려진 Anthracycline계 항생물질이 화학구조상 B환의 Hydroquinone형에 의한 생체내 산화환원 반응으로 이해 심근 독성에 영향을 준다고 알려져 있다. 이에 본 연구실에서는 Anthracycline의 Aqlycone 부분 12번 탄소의 전자친화력을 증가시키는 11번 탄소의 Hydroxy group이 제거되고 9번 탄소의 Hydroxy group을 Amine으로 대체함과 동시에 4번 탄소의 Methoxy group이 제거된 보다 독성이 적을 것이라 생각되는 Anthracycline의 새로운 Aglycone 유도체를 Design 하여 전합성 하였다.

  • Saponins are glycosidic compounds present in many edible and inedible plants. Structurally, they are composed of a lipid-soluble aglycone consisting of either a sterol or, more commonly, a triterpenoid and water-soluble sugar residues differing in type and amount of sugars [1]. Because of their amphiphilic nature, they are highly surface-active. Their biological activity is closely related to the chemical structures that determine the polarity, hydrophobicith and acidity of compounds [1]. (omitted)

  • Three crude saponins having the same aglycone were isolated from the root of Caltha minor $N_{AKAI}$ which has been known to be of effect as emetics in the folk medicines of Korea. And they were confirmed to their saponoidal properties through the methods of hemolytic test and $L_{IEBERMANN}-B_{UCHARD}$ reaction.

Anthracycline의 합성에 관한 연구

  • Anthracycline의 합성에 있어서 aglycone 뿐만 아니고 sugar도 대단히 중요한 역할을 한다. Anthracycline에 이용되는 sugar인 daunosamine의 전합성의 확립을 통하여 daunosamine의 anlogue을 합성할 목적으로 daunosamine의 전합성을 검토하고 있다. Methyl D-mannopyranoside을 원료로 하여 0-methylation, PhCH(OMe)$_2$ 에 의한 OH의 보호, n-BuLi을 이용한 위치선택적 deblocking, oxime생성, 이어서 Vitride를 이용한 환원으로 daunosamine 합성에 있어서 중요한 중간체인 methyl-3-amino-4,6-benzylidene-2,3-dideoxy-$\alpha$-D-hexopyranoside을 합성하였다.
