Short-term Treatment of Daumone Improves Hepatic Inflammation in Aged Mice

  • Chronic inflammation has been proposed as one of the main molecular mechanisms of aging and age-related diseases. Although evidence in humans is limited, short-term calorie restriction (CR) has been shown to have anti-inflammatory effects in aged experimental animals. We reported on the long-term treatment of daumone, a synthetic pheromone secreted by Caenorhabditis elegans in an energy deficient environment, extends the life-span and attenuates liver injury in aged mice. The present study examined whether late onset short-term treatment of daumone exerts anti-inflammatory effects in the livers of aged mice. Daumone was administered orally at doses of 2 or 20 mg/kg/day for 5 weeks to 24-month-old male C57BL/6J mice. Increased liver macrophage infiltration and gene expression of proinflammatory cytokines in aged mice were significantly attenuated by daumone treatment, suggesting that short-term oral administration of daumone may have hepatoprotective effects. Daumone also dose-dependently suppressed tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$ )-induced nuclear factor-${\kappa}B$ (NF-${\kappa}B$) phosphorylation in HepG2 cells. The present data demonstrated that short-term treatment of daumone has anti-inflammatory effects in aged mouse livers possibly through suppression of NF-${\kappa}B$ signaling and suggest that daumone may become a lead compound targeting aging and age-associated diseases.

Process Development for Enhancement of High Temperature Thermoelectric Properties in a p-Type Skutterudite (P-형 Skutterudite 소재의 고온 열전물성 제어를 위한 공정 개발)

  • Power factor improvement at high temperatures has been a major research topic for the development of skutterudite thermoelectric materials. Here, we attempted to optimize the process parameters for manufacturing skutterudite materials, especially for p-type systems. We focused on the effect of aging time variation to maximize the high-temperature performance of the Ce-filled Fe3CoSb12 skutterudite system. The optimized aging time was concluded to be a key parameter for the formation of single-phase nanostructures in this p-type skutterudite system. The optimized condition was effective in reducing the bipolar effect at high temperature ranges by increasing the carrier concentration in the p-type system. To confirm the conclusions, the electrical conductivity, Seebeck coefficient, and power factor were measured. The results matched well with the microstructure and with those of an XRD analysis performed for the system.

Development of Scale and Empirical Study of Trust-Forming Factors for Anti-aging Products (항노화 상품의 신뢰형성요인 척도 개발과 실증연구)

  • In this study, the relationship between trust and purchase intention, which is an outcome variable, was empirically verified by discovering the trust-forming factors of anti-aging products and developing measurement variables. In addition to the existing trust-forming factors such as perceived quality, perceived reputation, brand recognition, and customization, a newly discovered factor is 'accredited certification'. Since the anti-aging industry in Korea is an industry that directly affects consumer health, it was determined that the existing trust-forming factors had limitations in building consumer trust, and a new measure was discovered. This study conducted a survey on anti-aging product consumers and analyzed the solidity and relationship of the questions. As a result of the analysis, it was found that perceived quality, brand recognition, customization, and accredited certification among the anti-aging product trust-forming factors had a positive (+) effect on trust, and perceived reputation had a negative (-) effect on trust. appeared to affect It was found that there was a positive (+) effect between trust and purchase intention, which are the trailing variables. This study proved the meaningful relationship between trust-forming factors and trust by strategically developing and empirically and practically examining meaningful and effective trust-forming factors and measurement items. At the same time, this study is expected to significantly contribute to the establishment of marketing strategies related to anti-aging products in the future by discovering key factors for forming trust that are important for marketing in the anti-aging industry.

Yeosin-san Increases Female Fertility through Inducing Uterine Receptivity and Ovarian Function

  • Despite the development of assisted reproduction technologies (ART) including in vitro fertilization (IVF), the poor ovarian response and endometrial receptivity remains clinically a major unmet need. Although these problems are difficulties to solve in infertility treatment, there are no good therapeutic option yet. Traditional herbal remedies and acupuncture, therefore are being proposed as alternative treatment. Our group found that traditional herbal medicines such as Paeonia lactiflora L.(PL, 芍藥), Cyperus rotundus L.(CR, 香附子), and Perilla frutescens (PF, 紫蘇葉) could improve endometrial receptivity. In this study, we found out Yeosin-san (如神散) as an optimal herbal formula via combination of the previously established herbal medicines. Yeosin-san is a traditional Korean medical formula which was established by Ziming Jin (陳自明) and recorded in Furendaiquanliangfang (婦人大全良方) at first. The formula traditionally used for treating abnormal uterine bleeding and leukorrhea. It showed a highest effect on leukemia inhibitory factor (LIF) expression and on the adhesion between trophoblastic cells and endometrial cells. In addition, it has been shown that the Yeosin-san not only increases the endometrial receptivity to improve the embryo implantation but also enhances the ovary function by expressing the angiogenesis-related genes. Here we suggest that Yeosin-san could be a novel and effective candidate for treating female infertility.

Depigmenting Effects of Mistletoe (Viscum album var. coloratum) Extracts (겨우살이 추출물의 미백 효과)

  • Melanin pigments are the main cause of skin color. They are produced in melanocytes and then transferred to keratinocytes, which eventually gives the skin surface a variety of colors. Although many skin-lightening or depigmenting agents have been developed, the demand for materials to reduce pig- mentation is still increasing. Here, we tried to find materials for skin-lightening or depigmentation using natural compounds and found that mistletoe (Viscum album var. coloratum) extracts (ME) had an inhibitory effect on tyrosinase activity. As a result, ME significantly reduced pigmentation in human primary melanocytes. In addition, a promoter reporter assay revealed that ME inhibited the transcription of microphthalmia-associated transcription factor (MITF), melanophilin (MLPH), tyrosinase-related protein-2 (TRP-2), and tyrosinase (TYR) genes in HM3KO melanoma cells. In addition, ME decreased the protein level for pigmentation-related molecules, such as TYR and TRP-1. Furthermore, it markedly inhibited the melanogenesis of zebrafish embryos, an in vivo evaluation model for pigmentation. To elucidate the action mechanism of ME, we investigated its effects on intracellular signaling. Eventually, the ME dramatically decreased the phosphorylation of the cAMP responsive element binding protein (CREB), AKT, and ERK. The data suggest that ME may inhibit the melanogenesis pathway by regulating the signaling pathway related to pigmentation. Taken together, these data propose that ME can be developed as a depigmenting or skin-lightening agent.

The Study on the Development of the Korean Elderly's Successful Aging Scale (한국 노인의 성공적 노화 척도 개발을 위한 연구)

  • In phase 1 of the study, data for the construct and items of KESAS(The Korean Elderly's Successful Aging Scale) were collected by indepth interviews with a purposive sampling group of 25 elderly people and focus group interviews with four groups. Content analysis was applied to the data and from this analysis were emerged six key categories of successful aging. In phase 2 of the study, 78 items were collected from the data of 2 sets of interviews mentioned above. Through pilot test, preliminary 66 items are selected. In phase 3 of the study, 597 cases collected from the survey were divided randomly into 345 developmental samples and 252 validity samples. The items were examined exploratorily with the developmental samples and confirmatorily factor analysis with validity samples. Confirmatory factor analysis supported the hypothesized six-factor structure of KESAS, which finally resulted in consisting of 31 items. Six factors are as follows: 'autonomous life', 'self-completion orientation', 'positive life participation', 'satisfaction with one's offsprings', 'self-acceptance', 'other-acceptance'. Cronbach's alpha estimate and split-half estimates of the scale were .903 and .845, which confirms that the scale has reliability. This six-factor model exhibited a good fit on the basis of the overall fit measure criteria(TLI=.983, RMSEA=.057) aquired by using the confirmatory factor analysis. Moreover, KESAS was significantly related highly(r=.72) to the 'Quality of Life for the Korean Elderly'. This proved concurrent validity of the scale.

Development of Scale on Selection, Optimization, Compensation(SOC) Model as Successful Aging Strategies of Korean Elderly (한국노인의 성공적 노화 전략으로서의 선택·최적화·보상(SOC) 척도 개발에 관한 연구)

  • The purpose of this study is to develop the scale on Selection, Optimization, Compensation(SOC) model as successful aging strategies of Korean Elderly. In first phase of the study, 64 pilot items were collected from researcher's indepth interviews with a purposive sample group of 24 elderly people(16 items) and original SOC scale(48 items). To analyze the factor structure and to verify the validity of the scale, 592 questionnaires collected from survey were divided randomly into 300 developmental samples and 292 validity samples. The items were examined exploratory with developmental samples and confirmatory factor analysis with developmental samples. Two factor analysis supported four factor structure of the SOC consisted of 20 items. Four factors are as follows: 'Elective Selection', 'Loss-Based Selection', 'Opimization', 'Compensation'. The cronbach's alpha estimate of the scale was .930. This scale of four factor model exhibited good fit, assessed by overall fit measure criteria(TLI=.939, CFI=.947, RMSEA=.058). The result of analysis by item response theory for SOC scale is satisfatory. Also, SOC scale was significantly related to the two successful aging scales for Korean elderly and life satisfation scale(SWLS). These results proved the validity of the scale.

Replicative Senescence in Cellular Aging and Oxidative Stress (세포 노화에 있어서 복제 세네센스 현상과 산화적 스트레스의 영향)

  • Explanted mammalian cells perform a limited number of cell division in vitro and than are arrested in a state known as replicative senescence. Such cells are irreversibly blocked, mostly in the G1 phase of cell cycle, and are no longer sensitive to growth factor stimulation. Thus replicative senescence is defined as a permanent and irreversible loss of replicative potential of cells. For this characteristic, replicative senescence seems to evolve to protect mammalian organism from cancer. However, senescence also contributes to aging. It seems to decrease with age of the cell donor and, as a form of cell senescence, is thought to underlie the aging process. Extensive evidence supports the idea that progressive telomere loss contributes to the phenomenon of cell senescence. Telomeres are repetitive structures of the sequence (TTAGGG)n at the ends of linear chromosomes. It has been shown that the average length of telomere repeats in human somatic cells decreases by 30∼200 bp with each cell division. It is generally believed that when telomeres reach a critical length, a signal is activated to initiate the senescent program. This has given rise to the hypothesis that telomeres act as mitotic clocks to regulate lifespan. One proposes that cumulative oxidative stress, mainly reactive oxygen species generated from mitochondria, may mainly cause telomere shortening, accelerating aging. Here, the biological importance and mechanism of replicative senescence were briefly reviewed. Also it was summarized that how oxidative stress affects replicative senescence and telomere shortening.

Deficiencies of Homer2 and Homer3 accelerate aging-dependent bone loss in mice

  • Homer proteins are scaffold proteins that regulate calcium (Ca2+) signaling by modulating the activity of multiple Ca2+ signaling proteins. In our previous report, Homer2 and Homer3 regulated NFATc1 function through its interaction with calcineurin, which then acted to regulate receptor activator of nuclear factor-kappa B ligand (RANKL)-induced osteoclastogenesis and bone metabolism. However, to date, the role of Homers in osteoclastogenesis remains unknown. In this study, we investigated the roles of Homer2 and Homer3 in aging-dependent bone remodeling. Deletion of Homer2/Homer3 (Homer2/3 DKO) markedly decreased the bone density of the femur. The decrease in bone density was not seen in mice with Homer2 (Homer2-/-) and Homer3 (Homer3-/-) deletion. Moreover, RANKL treatment of bone marrow-derived monocytes/macrophages in Homer2/3 DKO mice significantly increased the formation of multinucleated cells and resorption areas. Finally, Homer2/3 DKO mice decreased bone density in an aging-dependent manner. These findings suggest a novel potent mode of bone homeostasis regulation through osteoclasts differentiation during aging by Homer proteins, specifically Homer2 and Homer3.

A Study on the Sulfate Attack Resistance of Concrete Using EAF Slag as Fine Aggregate (전기로슬래그 잔골재를 사용한 콘크리트의 황산염침식 저항성에 관한 연구)

  • The purpose of this study is to investigate the sulfate attack resistance of concrete using the EAF(electric arc furnace) Slag as fine aggregate. In order to figure out the effects of magnesium sulfate solution on the durability of concrete using the EAF Slag as fine aggregate, the experiments for the immerging test in the 10% magnesium sulfate solution was executed by selecting factors such as aging processes, replacement ratio(0, 10, 20, 30, 50%), and duration of immerging. The specimens were made with various EAF slag replacements for fine aggregates and with W/C ratio fixed 0.45. compressive strength and S.D.F(Sulfate Deterioration Factor), weight change, and SEM(Scanning Electron Microscope) were tested. From the test results, EAF slag aggregate treated with accelerated aging is better than treated with air aging. The compressive strength and resistance to the sulfate attack is slightly improved with an increase in the EAF slag aggregate treated with accelerated aging replacement for aggregate.
