Impairments of Learning and Memory Following Intracerebroventricular Administration of AF64A in Rats

  • Three types of learning and memory tests (Morris water maze, active and passive avoidance) were performed in rats following intracerebroventricular infusion of ethylcholine aziridium (AF64A). In Morris water maze, AF64A-treated rats showed the delayed latencies to find the platform iron 6th day after the infusion. In pretrained rats, AF64A caused the significant delay of latency at 7th days but not 8th day. In the active avoidance for the pretrained rats, the escape latency was significantly delayed in AF64A-treatment. The percentages of avoidance in AF64A-treated rats were less increased than those in the control. Especially, the percentage of no response in the AF64A-treated rats was markedly increased in the first half trials. In the passive avoidance, AF64A-treated rats shortened the latency 1.5 h after the electronic shock, but not 24 h. AF64A also caused the pretrained rats to shorten the latency 7th day after the infusion, but not 8th day. These results indicate that AF64A might impair the learning and memory. However, these results indicate that the disturbed memory by AF64A might rapidly recover after the first retrain. Furthermore, these results suggest that AF64A may be a useful agent for the animal model of learning for Spatial cognition .

Antipruritic Effect of DA-5018m A Capsaicin Derivative, in Mice

  • The antipruritic effect of DA-5018m a capsaicin derivative, was examined in mice. Male ICR mice were topically pretreated with Zostrix-HP (0.075% capsaicin cream), 0.1%, 0.3% DA-5018 cream or cream base (control) twice daily for 4 days. One hour after the last application, itch was induced either by compound 48/80 ($50{\mu}g$, s.c.) or leukotriene B4 (0.03 nmol, i.d.) injection into the rostral back of the animals, and the number of scratches made by the animals at the injection site was counted for 60 min post-injection. DA-5018 cream (both 0.1 and 0.3%) significantly inhibited compound 48/80-induced scratching when compared with the cream base control (p<0.01), which Zostrix-HP showed minimal inhibition of the scratching behavior. In leukotriene B4-induced itch model, Zostrix-HP and 0.3% DA-5018 cram significantly inhibited the scratching during the first 10-min period (p<0.01). The results suggest that DA-5018 cream can be used as an antipruritic agent and warrant clinical evaluation.

  • Investigations were carried out on a $\beta$-glucosidase produced by Aspergillus niger under solid-state fermentation conditions as a model of enzyme recovery from fermented wheat bran. The leaching efficiency of distilled water to recover the enzyme from the fermented bran was higher than acetate buffer, citrate buffer, citrate-phosphate buffer and 5% methanol; thus, the conditions were further optimized with distilled water as the extracting agent. After fermented bran was washed three times with distilled water for 1.5 hr each under shaking conditions at 1:5 solid to solvent ratio, a maximum recovery of 0.025 U/g of wheat bran was obtained.

  • The separation and concentration of L-phenylalanine (L-Phe) using a supported liquid membrane (SLM) is investigated. A cation complex agent, di-2-ethylhexyl phosphoric acid (D2EHPA), is used as a carrier in the SLM with n-Heptane as a solvent. The reaction order and equilibrium constant in the formation reaction of L-phe-carrier complex are obtained from the extraction experiment. A mathematical model for a carrier mediated counter transport process is proposed to estimate the diffusion coefficient of L-phe-carrier complexly in the liquid membrant. Permeation experiments of L-phe using a SLM are performed under various operating conditions and optimum conditions for the transport of L-phe are obtained. Concentration of L-phe in the strip phase against its concentration is observed. Transport rate of glucose through liquid membrane is less than that of L-phe in the competitive transport of L-phe and glucose. And the existence of glucose reduced the transport rate of L-phe. The performance of separation with continuous strip phase is increased due to the dilution effect in the strip phase.

  • In this study, Woongjin fermented red ginseng extract (WFRG) was evaluated for its potential ability to act as an adjuvant for the immune response of mice. For the in vitro study, macrophages were treated with serial concentrations ($1{\mu}g/mL$, $10{\mu}g/mL$, and $100{\mu}g/mL$) of WFRG. For in vivo studies, mice were administered different concentrations (10 mg/kg/day, 100 mg/kg/day, and 200 mg/kg/day) of WFRG orally for 21 days. In vitro, the production of nitric oxide and TNF-${\alpha}$ by RAW 264.7 cells increased in a dose-dependent manner. In vivo, WFRG enhanced the proliferation of splenocytes induced by two mitogens (i.e., concanavalin A and lipopolysaccharide [LPS]) and increased LPS-induced production of TNF-${\alpha}$ and IL-6, but not IL-$1{\beta}$. In conclusion, WFRG has the potential to modulate immune function and should be further investigated as an immunostimulatory agent.

Anti-aging Activity of Aralia Cordata Thunb. by Inhibiting Oxidized Low-dencity Lipoprotein Production in Rats

  • Aralia cordata Thunb. (Araliaceae, ACT) is an remarkable herbal plant that has been widely used in traditional oriental medicine for the treatment of inflammatory diseases and cardiovascular disorders. In this study, we have established a vascular aging model in rats by orally administrating excessive vitamin $D_2$ (500,000 IU/kg/day) for 4 days followed by feeding high cholesterol diet for 16 weeks and then rats were randomly divided into control group, high cholesterol diet (HCD) group, HCD+ACT (30 mg/kg) and HCD+ACT (60 mg/kg) group. ACT (30, 60) significantly reduced total cholesterol (TC) content compared with HCD, but no significant differences in the serum lipids. Secondly, we measured the serum levels of Oxidized Low-dencity Lipoprotein (OxLDL) and malondialdehyde (MDA) in order to further investigate the anti-vascular aging mechanism of ACT. The results, ACT (30, 60) treatments decreased OxLDL, MDA content and increased Cu/Zn superoxide dismutase activity compared with HCD treatments. The results suggested that ACT inhibited OxLDL production rather than serum lipids lowering and that ACT could be used as potential anti-atherosclerotic agent in aged cells.

Neuroprotective Effect of Heyneanol A from the roots of vitis amurensis Rup (왕머루포도 뿌리에서 분리한 heyneanol A의 신경보호효과)

  • According to the report of Korea National Statistical Office in 2007, cerebral vascular disease is second cause of deaths in Korean. Cerebral ischemia is one of the main reason of cerebral vascular diseases. To evaluate the neuroprotective effect of heyneanol A against cerebral ischemia, we used the middle cerebral artery occlusion model (MCAO). Heyneanol A from root of vitis amurensis Rup is a tetramer of resveratrol as known anti-oxidant and anti-cancer agent. Although the effects of resveratrol in the various fields have been well established, little is known of the effects of heyneanol A. In this study, heyneanol A reduced infarction and edema volume by 33.5% and 57% compared with control groups (vehicle), respectively. Also, neurological score was decreased by heyneanol A. It's effects were more potent than resveratol. Taken together, these results exerted that heyneanol A has a neuroprotective effect against cerebral ischemia.

  • A common control model used to implement computer integrated manufacturing(CIM) is based on the hierarchical decomposition of the shop floor activities, in which supervisory controllers are responsible for all the interactions among subordinates. Although the hierarchical control philosophy provides for easy understanding of complex systems, an emerging manufacturing paradigm, agile manufacturing, requires a new control structure necessary to accommodate the rapid development of a shop floor controller. This is what is called autonomous agent-based heterarchical control. As computing resources and communication network on the shop floor become increasingly intelligent and powerful, the new control architecture is about to come true in a modern CIM system. In this paper, heterarchical control is adopted and investigated, in which a controller for a unit of device performs three main functions - planning, scheduling and execution. Attention is paid to the planning function and all the detailed planning activities for heterarchical shop floor control are identified. Interactions with other functions are also addressed. In general, planning determines tasks to be scheduled in the future. In other words, planning analyzes process plans and transforms process plans into detailed plans adequate for shop floor control. Planning is also responsible for updating a process plan and identifying/resolving replanning activities whether they come from scheduling or execution.

  • This paper implements the graphic service template authoring tool based on the service template object model of describing the meta information of the services in the semantic web service environment. Our proposed system provides the robot services by constructing web services automatically and making the appropriate service plans. Moreover, it can create, append, delete, and update the service templates of URC based on STDL, and provide the graphic function on service template resources. In order to provide the user friendly environment in the service template phase, we implement the various editing environment : flow view style, grid view style, and text view style. We also provide the easy editing function by realizing abstract service block based on the robot API. Finally we can offer the intelligent and autonomous service of service agent based on semantic information.

  • This research designed and implemented a supply chain risk management platform and applied it to a case study of reduced-fat Lanna pork sausage as a new product development project. The proposed framework has three stages: risk identification, risk assessment, and risk mitigation. Seventeen risk agents with 17 risk events were identified based on SWOT analysis and the Porter Five Forces concept through the process of planning, sourcing, making and delivering, partially captured from the supply chain operations reference model in the first stage. In the second stage, an house of risk (HOR) framework was applied to present the impacts of each risk agent. In the third stage, eight risk agents with high impact were selected to design 21 preventive actions. Finally, three preventive actions with the highest effectiveness to difficulty ratio scores-'sales evaluation of familiar products', 'increasing distribution channels and promotions to improve sales', and 'work flow improvement for work safety'-were then recommended for this new product development.
