Verification of Enhanced Phosphate Removal Capability in Pure Cultures of Acinetobacter calcoaceticus under Anaerobic/Aerobic Conditions in an SBR

  • Laboratory experiments were conducted using pure cultures of Acinetobacter under an-aerobic/aerobic cyclic conditions to explain the release and uptake of soluble phosphate in an activated sludge process showing enhanced biological phosphate removal (EBPR). Under anaerobic/aerobic cyclic conditions in a Sequencing Batch Reactor (SBR), COD uptake concurrent with soluble phosphate release by Acinetobacter was not significant during the anaerobic periods, indicating that EBPR would not be established in pure cultures. However Acinetobacter cells accumulated higher phosphate content (5.2%) in SBR than that obtained (4.3%) from batch experiments. These results suggest that Acinetobacter sp. may not follow the proposed pattern of behavior of poly-P bacteria in EBPR activated sludge Plants.

Microbiological, PH and Sensory Evaluations of Refrigerated Pork Loins Treated with Lactococcus lactis ATCC l1454 (Lactococcus lactis ATCC 11454로 처리한 냉장 돼지고기 등심의 미생물, pH 및 관능평가)

  • Microbiological, pH and sensory evaluations of pork loins treated with lactic acid cultures (Lactococcus lactis subsp. lactis ATCC 11454 grown in 10% milk solids) during storage at 4$^{\circ}C$ were assessed. Pork loins were immersed in solutions containing 3.96 log units, 4.10 log units, or 4.23 log units of individual lactic acid cultures for 2 min. Pork loins treated with lactic acid cultures had a inhibitory effect (P<0.05) on the growth of aerobic palate counts during storage of 3 days at 4$^{\circ}C$. Pork loins treated with lactic acid cultures during storage of 3 days at 4$^{\circ}C$ significantly (P<0.05) decreased pH values compared to those of controls. Sensory evaluations for odor and appearance of pork loins treated with lactic acid cultures during storage of 9 days at 4$^{\circ}C$ were a "liked less" to "typical" category compared to those of controls.

Global Regulation of Gene Expression in the Human Gastric Pathogen Helicobacter pylori in Response to Aerobic Oxygen Tension Under a High Carbon Dioxide Level

  • The human gastric pathogen Helicobacter pylori (Hp) has been considered a microaerophile. However, we recently reported that, when supplied with 10% $CO_2$, Hp growth is stimulated by an atmospheric level of $O_2$, suggesting that Hp is a capnophilic aerobe. In this study, we investigated the effects of aerobic $O_2$ tension on Hp cells by comparing gene expression profiles of cultures grown under microaerobic and aerobic conditions in the presence of 10% $CO_2$. The results showed that overall differences in gene expression in Hp cells grown under the two $O_2$ conditions were predominantly growth-phase-dependent. At 6 h, numerous genes were down-regulated under the aerobic condition, accounting for our previous observation that Hp growth was retarded under this condition. At 36 h, however, diverse groups of genes involved in energy metabolism, cellular processes, transport, and cell envelope synthesis were highly up- or down-regulated under the aerobic condition, indicating a progression of the cultures from the log phase to the stationary phase. The expression of several oxidative stress-associated genes including tagD, katA, and rocF was induced in response to aerobic $O_2$ level, whereas trxA, trxB, and ahpC remained unchanged. Altogether, these data demonstrate that aerobic $O_2$ tension is not detrimental to Hp cells but stimulates Hp growth, supporting our previous finding that Hp may be an aerobic bacterium that requires a high $CO_2$ level for its growth.

Physiological Response of Escherichia coli W3110 and BL21 to the Aerobic Expression of Vitreoscilla Hemoglobin

  • The aerobic growth and metabolic performance of Escherichia coli strains BL21 and W3110 were studied when the Vitreoscilla hemoglobin (VHb) was constitutively expressed in the chromosome. When VHb was expressed, acetate production decreased in both strains and was nearly eliminated in BL21. Transcriptional levels of the glyoxylate shunt genes decreased in both strains when VHb was expressed. However, higher transcription of the α-ketoglutarate dehydrogenase genes were observed for W3110, while for BL21 transcription levels decreased. VHb expression reduced the transcription of the cytochrome bo3 genes only in BL21. These results are useful for better selecting a production host.

  • Airlift bioreactors are extensively used in the fields of aerobic fermentation, animal and plant cell cultures. This review article evaluates the present research activities in the field of airlift bioreactors. The published research works on the design parameters such as types, location and properties of gas sparser, hydrodynamic properties such as phase holdups mixing, liquid circulation rate, mass and heat transfer rates are summarized. Also, recommendations are presented for designing airlift bioreactors.

  • Open heart surgery patients have additional risks arising from the special nature of the operative procedure. And postoperative infections in patients with extracorporeal circulation are associated with high incidence of serious sequelae. To investigate the incidence and organisms of contamination, and the relation between the duration of extracorporeal circulation and contamination, following study was done. Eighty-four of the open heart surgery patients were examined with cultures from the blood and priming solution before and after bypass. 1. Cultures before bypass were sterile, but 2 cases of cultures from the blood and priming solution after bypass were positive, and culture positive group was 5% of all patients. 2. The organisms were Gram-negative, aerobic coliform bacilli; 3. The culture positive group had significantly longer bypass time.

Growth of Issatchenkia orientalis in Aerobic Batch and Fed-batch Cultures

  • The aerobic batch growth of Issatchenkia orientalis DY252 with glucose and fructose medium was investigated at 32$\^{C}$ and pH 5.0. Aerobic ethanol production was evident with yeast I, orientalis. A diauxic lag of about 1 h between growth on glucose and growth on ethanol during batch culture was observed. However, no diauxic growth occurred with fructose. As the incubation temperature was increased from 32 to 39$\^{C}$, viability at the end of each batch culture declined significantly, from 93 to 43%, Unlike the effect of temperature, viability was not greatly affected by incubation pH, and cell yield values in a range of 0.45-0.48 were obtained. In order to overcome overflow metabolism, a fedbatch culture under glucose limitation was carried out. Compared with aerobic batch culture, about 10% improvement in cell yield was achieved with a fed-batch culture in optimal conditions.

Isolation of Hydrogen-producing Bacteria from Granular Sludge of an Upflow Anaerobic-Sludge Blanket Reactor

  • H$_2$-producing bacteria were isolated from anaerobic granular sludge. Out of 72 colonies (36 grown under aerobic conditions and 36 under anaerobic conditions) arbitrarily chosen from the agar plate cultures of a Suspended sludge, 34 colonies (15 under aerobic conditions and 19 under anaerobic conditions) produced H2 under anaerobic conditions. Based on various biochemical tests and microscopic observations, they were classified into 13 groups and tentatively identified as follows: From aerobic isolates, Aeromonar spp. (7 strains), Pseudomonas spp. (3 strains), and Vibrio spp. (5 strains); from anaerobic isolates, Actinomyces spp. (11 Strains), Clostridium 5pp. (7 strains). and Porphyromonas sp. When glucose was used as the carbon substrate, all isolates showed a similar cell density and a H$_2$ production yield in the batch cultivations after 12 h (2.24-2.74 OD at 600 nm and 1.02-1.22 mol H$_2$/mol glucose, respectively). The major fermentation by-products were ethanol and acetate for the aerobic isolates, and ethanol, acetate and propionate for the anaerobic isolates. This study demonstrated that several H$_2$ producers in an anaerobic granular sludge exist En large proportions and their performance in terms of H$_2$ production is quite similar.

Effects of Starter Cultures on the Quality Traits of Electron Beam Irradiated Fermented Meat during Aging (전자선 조사된 원료육과 Stater Culture의 사용이 발효육의 숙성 중 품질에 미치는 영향)

  • The microbiological and physicochemical properties of irradiated (2 kGy) or non-irradiated fermented meats processed with or without a commercial starter culture were evaluated during fermentation and aging. The pH of irradiated (2 kGy) fermented meats with starter cultures dramatically decreased during fermentation and aging (p<0.05), and the final pH was 4.25. The total aerobic counts and lactic acid bacteria counts reflected the addition of the starter culture. As the fermentation progressed, the total aerobic counts closely paralleled the lactic acid bacteria counts. The TBARS values of irradiated fermented meats increased regardless of the treatment during fermentation and aging. These results show that the irradiated(electron-beam) meat/fat resulted in the reduction of the total microbes and survives lactic acid bacteria. The use of starter cultures in meat batters post-irradiation may be useful for the production of fermented meats.
