  • Advanced glycation end products (AGEs) contribute to the progression of micro and macrovsacular complication of diabetes and therefore present a promising target for therapeutic agents. In this study, 40 Korean herbal medicines have been investigated with an in vitro evaluation system using AGEs inhibitory activity. Of these, 21 herbal medicines $(IC_{50}<50{\mu}g/ml)$ exhibited an inhibitory activity against AGEs formation compared with anminoguanidine $(IC_{50}=72.12{\mu}g/ml)$. Particularly, 7 herbal medicines, Actinidia arguta (root and stem), Crataegus pinnatifida (twig), Camellia japonica (whole), Kalopanax pictus (bark), Lagerstroemia indica (leaf-stem), Reynoutria sachalinensis (root) showed more potent inhibitory activity (approximately 3-10 fold) than the positive control aminoguanidine.

  • Two isoflavone C-glucosides, puerarin (1) and PG-3 (2), a but-2-enolide, $({\pm})-puerol$ B (3), two isoflavone O-glucosides, daidzin (4) and genistin (5), and three pterocarpans, (-)-medicarpin (6), (-)-glycinol (7) and (-)-tuberosin (8), were isolated from a MeOH extract of the roots of Pueraria Iobata, using an in vitro bioassay based on the inhibition of the formation of advanced glycation end products (AGEs) to monitor chromatographic fractionation. The structures of 1-8 were determined by spectroscopic data interpretation, particularly by 1D- and 2D-NMR studies, and by comparison of these data with values in the literature. All of the isolates (1-8) were evaluated for their inhibitory activity on AGEs formation in vitro. Of these, puerarin (1), PG-3 (2), and $({\pm})-puerol$ B (3) exhibited more potent inhibitory activity than the positive control aminoguanidine.

  • Hyperglycemia found in diabetes mellitus causes several physiological abnormalities including the formation of advanced glycation end products (AGEs) and oxidative stress. Accumulation of AGEs and elevation of oxidative stress plays major roles in the development of diabetic complications. Adiponectin secreted from adipocytes is known to improve insulin sensitivity and blood glucose level. Curcumin (CCM), a bioactive component of turmeric, has been reported as a potent antioxidant. Present work aimed to elucidate the roles of CCM in high glucose-induced protein glycation and intracellular events in mature adipocytes. The results demonstrated that CCM inhibited the formation of fluorescent AGEs by approximated 52% at 3 weeks of bovine serum albumin (BSA) glycation with glucose. Correspondingly, CCM decreased the levels of fructosamine and ${\alpha}-dicarbonyl$ compounds during BSA glycation with glucose. These data suggested that CCM might be a new promising anti-glycation agent. Also, CCM reduced high glucose-induced oxidative stress in a dose dependent manner, whereas CCM treatment time-dependently elevated the expression of adiponectin gene in 3T3-L1 adipocytes. The findings from this study suggested the possibility of therapeutic use of CCM for the prevention of diabetic complications and obesity-related diseases.

  • Advanced glycation end products (AGEs) has been shown to play an important role in the development of the diabetic complications. The AGEs inhibitors or cross-link breakers attenuate various functional and structural manifestations of diabetic complications. In this study, 69 China herbal medicines have been investigated with an in vitro evaluation system using AGEs inhibitory activity. Of these, 28 herbal medicines $IC_{50}$=<50 ${\mu}g/ml$) were found to have stronger AGEs inhibitory activity compared with aminoguanidine ($IC_{50}$=59.77 ${\mu}g/ml$). • Advanced glycation end products (AGEs) are final products formed by glycation reaction between reducing sugars and proteins, lipids or nucleic acids. These AGEs are related to progress of skin aging. In this study, we evaluate anti-aging activity of Defatted Torreya nucifera seed extract (DTSE) through antioxidant, anti-glycation, anti-elastase and inhibitory and breaking activity on the cross-linking of AGEs to collagen assay. Results showed that DTSE contained polyphenols and flavonoids. The $IC_{50}$ values of 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical and 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) radical scavenging activity were $16.4{\mu}g$ (Dried materials, DM)/mL and $16.7{\mu}g\;DM/mL$, respectively. DTSE also inhibited the formation of AGEs, elastase activity and cross-linking of AGEs to collagen as well as broke existing cross-linking of AGEs to collagen in a dose-dependent manner. Consequently, our findings suggest that DTSE could be useful as a cosmetic material with anti-aging activity.

  • Advanced glycation end products(AGEs) are largly involved in the pathogenesis of diabetic complications. It is obvious that inhibition of AGEs formation is important in preventing the occurrence and progression of diabetic complications. Therefore, to seek possible AGEs inhibitors in herbal medicines, unprocessed - und processed Puerariae Radix were tested. The inhibitory effect on AGEs formation was slightly increased through processing. Unprocessed-, processed Puerariae Radix and puerarin showed potential inhibitory action than that of positive control, amino guanidine HCI.
