Sildenafil Ameliorates Advanced Glycation End Products-Induced Mitochondrial Dysfunction in HT-22 Hippocampal Neuronal Cells

  • Objective : Accumulation of advanced glycation end-products (AGE) and mitochondrial glycation is importantly implicated in the pathological changes of the brain associated with diabetic complications, Alzheimer disease, and aging. The present study was undertaken to determine whether sildenafil, a type 5 phosphodiesterase type (PDE-5) inhibitor, has beneficial effect on neuronal cells challenged with AGE-induced oxidative stress to preserve their mitochondrial functional integrity. Methods : HT-22 hippocampal neuronal cells were exposed to AGE and changes in the mitochondrial functional parameters were determined. Pretreatment of cells with sildenafil effectively ameliorated these AGE-induced deterioration of mitochondrial functional integrity. Results : AGE-treated cells lost their mitochondrial functional integrity which was estimated by their MTT reduction ability and intracellular ATP concentration. These cells exhibited stimulated generation of reactive oxygen species (ROS), disruption of mitochondrial membrane potential, induction of mitochondrial permeability transition, and release of the cytochrome C, activation of the caspase-3 accompanied by apoptosis. Western blot analyses and qRT-PCR demonstrated that sildenafil increased the expression level of the heme oxygenase-1 (HO-1). CoPP and bilirubin, an inducer of HO-1 and a metabolic product of HO-1, respectively, provided a similar protective effects. On the contrary, the HO-1 inhibitor ZnPP IX blocked the effect of sildenafil. Transfection with HO-1 siRNA significantly reduced the protective effect of sildenafil on the loss of MTT reduction ability and MPT induction in AGE-treated cells. Conclusion : Taken together, our results suggested that sildenafil provides beneficial effect to protect the HT-22 hippocampal neuronal cells against AGE-induced deterioration of mitochondrial integrity, and upregulation of HO-1 is involved in the underlying mechanism.

중국약용식물의 최종당화산물 생성저해활성 검색 (VI) (Screening of Herbal Medicines from China with Inhibitory Activity on Advanced Glycation End Products (AGEs) Formation (VI))

  • Advanced glycation end products (AGEs) has been shown to play an important role in the development of the diabetic complications. The AGEs inhibitors or cross-link breakers attenuate various functional and structural manifestations of diabetic complications. In this study, 69 China herbal medicines have been investigated with an in vitro evaluation system using AGEs inhibitory activity. Of these, 28 herbal medicines $IC_{50}$=<50 ${\mu}g/ml$) were found to have stronger AGEs inhibitory activity compared with aminoguanidine ($IC_{50}$=59.77 ${\mu}g/ml$). Particularly, 5 herbal medicines, Camptotheca acuminata (stem, leaf), Eurya groffii (stem, leaf), Cornus Capitata (leaf), Mucuna birdwoodiana (root), Nelumbo nucifera (fruit, seed) showed more potent inhibitory activity (approximately 6-27 fold) than the positive control aminoguanidine.

Hot water extract of Loliolus beka attenuates methylglyoxal-induced advanced glycation end products formation in human umbilical vein endothelial cells

  • Over production of methylglyoxal (MGO) a highly reactive dicarbonyl compound, has been associated in progressive diabetes with vascular complication. Therefore, we investigated whether hot water extract of Loliolus beka meat (LBM-HWE) presents a preserve effect against MGO-induced cellular damage in human umbilical vein endothelial cells (HUVECs). The LBM-HWE extract showed to inhibit MGO-induced cytotoxicity. Additionally, the LBM-HWE reduced mRNA expression of pro-inflammatory cytokines, and reduced MGO-induced advanced glycation end product (AGEs) formation. Furthermore, LBM-HWE induced glyoxalase-1 mRNA expression and reduced MGO-induced carbonyl protein formation in HUVECs. The results implicate that LBM-HWE has protective ability against MGO-induced HUVECs toxicity by preventing AGEs formation. In conclusion, LBM-HWE could be used as a potential treatment material for the prevention of vascular complications of diabetes.

비자 유박 추출물이 최종당화산물과 콜라겐 교차결합에 미치는 효과 (Effect of Defatted Torreya nucifera Seed Extract on the Cross-linking of Advanced Glycation End Products to Collagen)

  • 최종당화산물은 환원당과 단백질, 지질 또는 핵산 사이의 당화반응에 의해서 생성되는 물질로 이물질은 피부 노화 과정에 밀접하게 연관되어 있다고 보고되어 있다. 본 연구에서는 항산화 활성, 최종당화산물 생성 저해, 콜라겐과 최종당화산물의 교차결합 저해 및 억제, 엘라스타제 활성 저해 효능 시험을 통해서 비자나무 유박 추출물의 항노화 효과를 평가하였다. 시험 결과, 비자나무 유박 추출물은 폴리페놀과 플라보노이드를 함유하고 있으며, DPPH와 ABTS 자유라디칼을 50% 소거할 수 있는 농도는 각각 $16.4{\mu}g$ (Dried materials, DM)/mL과 $16.7{\mu}g\;DM/mL$ 로 관찰되었다. 또한, 비자나무 유박 추출물은 농도 의존적으로 최종당화산물 생성과 엘라스타제 활성을 저해하였고, 콜라겐과 최종당화산물의 교차결합을 저해 및 억제하였다. 따라서 본 연구 결과를 바탕으로 비자나무 유박 추출물은 항노화 효능이 있는 화장품 소재로서 활용가치가 있음을 확인하였다.

호두 열매 추출물의 메틸글라이옥살 유도 신장 세포손상 억제 효과 및 당화억제 효능 (Antiglycation and Protective Effect of Juglans regia L. in MGO-induced Renal cell Death)

  • Methylglyoxal is a highly reactive precursor which forms advanced glycation end products (AGEs). AGEs and methylglyoxal are known to induce various diseases such as diabetes, vascular disorders, Diabetes Mellitus (DM), and neuronal disorders. Juglans regia L is an important food commonly used worldwide, having nutritious components, including phenolic compounds. Since ancient times, Juglans regia L have been differently applied by various countries for health and in diverse diseases, including arthritis, asthma, skin disorders, cancer, and diabetes mellitus. However, the effect of diabetes-induced renal damage against AGEs remains unclear. This study evaluates the anti-glycation and renal protective effects of ethanol extract of Juglans regia L against methylglyoxal-induced renal tubular epithelial cell death. Exposure to methylglyoxal resulted in reduced cell viability in NRK-52E cells, but co-treatment with Juglans regia L extracts significantly increased the cell viability. In addition, we examined the anti-glycation effect of Juglans regia L extracts. Compared to the positive control aminoguanidine and Alagebrium, treatment with Juglans regia L extracts significantly inhibited the formation of AGEs, collagen cross-linking, and breaking collagen cross-linking. Taken together, our results indicate that Juglans regia L is a potential therapeutic agent for regulating diabetic complications by exerting anti-glycation and renal protective activities.

Effect of Alpinia officinarum on the Formation of Advanced Glycation Endproducts and Aldose Reductase Activity

  • The inhibition effects of an Alpinia officinarum (AO, Zingiberaceae) on the formation of advanced glycation end products, aldose reductase, and scavenging effect on 1,1-diphenyl-2-picryl-hydrazil (DPPH) radical for the prevention and/or treatment of diabetic complications were investigated. The ethyl acetate fraction of AO was the most effective among all fractions. Through the tests with electron impact-mass spectrometry and nuclear magnetic resonance, two compounds (compound 1 and 2) finally obtained from the ethyl acetate fraction of AO were identified as galangin (1) and kaempferide (2), respectively. In addition, the compound 1 and 2 and the ethyl acetate fraction were compared for the prevention effect on advanced glycation end products, aldose reductase, and the scavenging effect on DPPH radical. The ethyl acetate fraction was significantly more effective than the 2 compounds for those preventive activities.

  • Hwanggeum-tang (HGT) was recorded in Dongeuibogam as being able to treat Sogal whose concept had been applied to Diabetes Mellitus (DM). Advanced glycation end products (AGEs) play important roles in the development of diabetic complications such as atherosclerosis by eliciting inflammatory responses. In this study, we examined the suppressive effects of HGT against inflammation elicited by AGEs. AGEs treatment increased the expression of pro-inflammatory cytokine gene TNF-${\alpha}$; chemokines MCP-1, IP-10; pro-inflammatory cyclooxygenase COX-2 on the THP-1 cells. HGT had suppressed the expression of pro-inflammatory genes and protein levels in AGE-treated THP-1 cells. HGT had also decreased intracellular ROS production stimulated by AGEs. These results suggest that HGT has beneficial effects for the improvement diabetic vascular complication through suppressing inflammatory responses elicited by AGEs.
