IDCC 9203은 모유 영양아의 분변으로부터 분리하였으며 분자생물학적 동정 결과 Lactobacillus johnsonii로 밝혀졌다. L. johnsonii IDCC 9203은 pH 2.3으로 조정된 MRS 액체배지에서 2시간 반응시 81%의 생존율을 보였으며, 0.3% bile이 함유된 MRS 액체배지에서는 5시간 반응시 57%의 생존율을 보여 산과 담즙에 대한 내성이 있는 것으로 나타났다. L. johnsonii IDCC 9203은 in vitro에서 Caco-2 cells에 대한 부착능이 우수하여 S. typhimurium KCTC 2054의 부착을 경쟁적으로 50%이상 감소시켰다 또한 in vivo에서 L. johnsonii IDCC 9203은 항생제로 교란된 mice의 장내에 정착하여 증식하였으며 이상 증식된 salmonella ($10^9CFU/g$ feces)를 초기 수준 ($(10^5CFU/g$ feces)로 감소시켰다. 이상의 결과에서 L. johnsonii IDCC 9203은 사람 및 동물에 유용한 프로바이오틱 특성을 가지고 있어 정장용 유산균으로 개발될 수 있을 것으로 기대된다.
묵은지 스타터로 개발된 Lb. curvatus ML17의 생균제적 특성 및 기능성 조사를 통하여 기능성 소재로서의 개발 가능성을 검토하였다. Lb. curvatus ML17은 용혈현상을 일으키지 않았으며 ${\beta}$-glucosidase와 ${\beta}$-glucuronidase의 발암효소 활성이 없는 것으로 나타나 균주에 대한 안전성을 확인하였다. 장내 생존성 확인 실험에서는 인공위액에서 2시간 처리한 다음 인공담즙에서 24시간 처리한 결과 초기 균수 대비 12.30%의 생존율을 나타내었다. Caco-2 cell에 대한 부착율은 초기 접종균수가 많을수록 높게 나타났으며, $5.66{\times}10^9$ CFU/mL로 처리 시 17.31%의 부착율을 나타냈는데 20.78%의 부착율을 나타낸 양성 대조군 Lb. rhamnosus GG에 근접하게 높은 부착율을 나타내어 장내환경에서 바람직한 작용을 할 수 있음을 보여주었다. 유해균주에 대한 생육저해 활성 측정에서는 Micrococcus luteus, Bacillus cereus, Salmonella enterica subsp. enterica, Pseudomonas aeruginosa에 대하여 우수한 항균 활성을 나타내었다. DPPH radical 소거능을 이용한 항산화 활성 측정 결과 72.88%로 양성 대조군인 0.1 mg/mL ascorbic acid보다 높은 free radical 소거능을 나타내었다. AGS 위암세포 및 HT-29 결장암세포에 대한 성장 억제 효과에서는 Lb. curvatus ML17의 배양 상등액 $80{\mu}L/mL$ 처리 시 각각 82.91% 및 79.35%의 암세포 성장 억제 효과를 나타내었다. 이와 같은 결과로 Lb. curvatus ML17이 묵은지를 포함한 발효스타터로 사용되었을 경우 식품의 보존성 향상과 품질 유지 및 균주에 의한 건강 기능성을 부여할 수 있으며 식품, 사료, 의약 분야의 다양한 산업적 활용이 기대된다.
Mucin2 (MUC2), an important regulatory factor in the immune system, plays an important role in the host defense system against bacterial translocation. Probiotics known to regulate MUC2 gene expression have been widely studied, but the interactions among probiotic, pathogens, and mucin gene are still not fully understood. The aim of this study was to investigate the role of MUC2 in blocking effects of probiotics on meningitic E. coli-induced pathogenicities. In this study, live combined probiotic tablets containing living Bifidobacterium, Lactobacillus bulgaricus, and Streptococcus thermophilus were used. MUC2 expression was knocked down in Caco-2 cells by RNA interference. 5-Aza-2'-deoxycytidine (5-Aza-CdR), which enhances mucin-promoted probiotic effects through inducing production of Sadenosyl-L-methionine (SAMe), was used to up-regulate MUC2 expression in Caco-2 cells. The adhesion to and invasion of meningitic E. coli were detected by competition assays. Our studies showed that probiotic agents could block E. coli-caused intestinal colonization, bacteremia, and meningitis in a neonatal sepsis and meningitis rat model. MUC2 gene expression in the neonatal rats given probiotic agents was obviously higher than that of the infected and uninfected control groups without probiotic treatment. The prohibitive effects of probiotic agents on MUC2-knockdown Caco-2 cells infected with E44 were significantly reduced compared with nontransfected Caco-2 cells. Moreover, the results also showed that 5-Aza-CdR, a drug enhancing the production of SAMe that is a protective agent of probiotics, was able to significantly suppress adhesion and invasion of E44 to Caco-2 cells by upregulation of MUC2 expression. Taken together, our data suggest that probiotic agents can efficiently block meningitic E. coli-induced pathogenicities in a manner dependent on MUC2.

Background: Invasive infections due to foodborne pathogens, including Salmonella enterica serovar Typhimurium, are prevalent and life-threatening. This study aimed to evaluate the effects of ginsenoside Rg3 (Rg3) on the adhesion, invasion, and intracellular survival of S. Typhimurium. Methods: The impacts of Rg3 on bacterial growth and host cell viability were determined using the time kill and the 3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl-2H-tetrazolium bromide assays, respectively. Gentamicin assay and confocal microscopic examination were undertaken to determine the effects of Rg3 on the adhesive and invasive abilities of S. Typhimurium to Caco-2 and RAW 264.7 cells. Quantitative reverse transcription polymerase chain reaction was performed to assess the expression of genes correlated with the adhesion, invasion, and virulence of S. Typhimurium. Results: Subinhibitory concentrations of Rg3 significantly reduced (p < 0.05) the adhesion, invasion, and intracellular survival of S. Typhimurium. Rg3 considerably reduced (p < 0.05) the bacterial motility as well as the release of nitrite from infected macrophages in a concentration-dependent manner. The expression of genes related to the adhesion, invasion, quorum sensing, and virulence of S. Typhimurium including cheY, hilA, OmpD, PrgK, rsgE, SdiA, and SipB was significantly reduced after Rg3 treatment. Besides, the compound downregulated rac-1 and Cdc-42 that are essential for actin remodeling and membrane ruffling, thereby facilitating Salmonella entry into host cells. This report is the first to describe the effects of Rg3 on "trigger" entry mechanism and intracellular survival S. Typhimurium. Conclusion: Rg3 could be considered as a supplement agent to prevent S. Typhimurium infection.
Probiotics are generally excreted within a few days if their ingestion in feces at the same rate as or even more quickly than a transit marker (meaning not clear). Ability of probiotics to adhere to intestine prolongs their persistence in gastrointestinal tract, allowing them to exert healthful effects longer. Hydrophobicities, zeta potentials, Alcian blue-binding capacities, and sedimentation profiles of Pichia farinosa SKM-1, P. anomala SKM-T, and Galactomyces geotrichum SJM-59 were determined to evaluate characteristic properties of cell surfaces responsible for adhesion. Results of intestinal Caco-2 cell line in vitro and murine intestine in vivo studies revealed these strains exhibit adhesive properties regardless of their cell surface hydrophobicity.
갓김치로부터 분리한 probiotic L. acidophilus GK20 및 L. paracasei GK74를 단독 혹은 혼합 배양하여 제조한 요구르트를 저장하는 동안 이화학적 및 미생물학적 특성과 스타터의 위산이나 담즙산에 대한 저항성, Caco-2 cell에 대한 부착능, 항균, 항산화 및 효소적 활성에 대한 prebiotic FOS(fructooligosaccharide)의 영향을 살펴보았다. FOS를 첨가했을 때 요구르트 내의 스타터 균수, 총산도 및 점도는 유의하게 높아짐과 동시에 pH는 감소되었다(p<0.05). 또한 E. coli ATCC 11229와 S. enteritidis ATCC 13076은 probiotics 스타터가 생산한 항균물질에 의해 저해되었으며, FOS를 첨가한 synbiotic 요구르트의 항균활성은 더욱 증가되었다. 게다가 FOS (1.0%)를 첨가하여 혼합 스타터로 발효시킨 요구르트에서 가장 높은 단백질 분해능 ($1.06{\pm}0.06$ unit) 및 ${\beta}$-galactosidase 활성 ($20.14{\pm}0.31$ unit)을 나타내었다. 하지만 비록 L. acidophilus GK20과 L. paracasei GK74 모두 장관액에 대한 저항성, 장관상피세포에 대한 부착능 및 DPPH radical 소거능이나 환원력과 같은 항산화 활성을 나타내었지만, 이들 활성이 FOS에 의해 증가되진 않았다. 결과적으로 L. acidophilus GK20과 L. paracasei GK74 혼합 배양에 FOS를 첨가한 synbiotic 요구르트는 장내환경 개선과 건강기능 향상에 유용한 것으로 여겨지며, 생리활성은 $4^{\circ}C$에서 7일간 저장 하에서도 일정하게 유지되었음을 확인하였다.
본 연구에서는 전통발효식품에서 분리한 유산균 200여종에 대하여 프로바이오틱 특성을 확인하였다. 내담즙성 및 내산성이 높게 측정된 유산균 4종을 선발하였다. 선발된 유산균은 Lb. plantarum SRCM102224, Lb. plantarum SRCM102227, Lb. paracasei SRCM102329, Lb. paracasei SRCM102343이다. 항균활성, 내산성, 내담즙성, 용혈성, 세포표면 소수성, 장내 세포 부착성, 항생제 내성을 조사하였다. 선발된 유산균 4종 중 SRCM102343은 세포표면 소수성이 95.9%로 대조구 Lb. rhamnosus GG는 13.4%보다 높게 측정되었다. 4종의 유산균의 장내상피세포 caco-2에 대한 부착능을 조사하였다. 그 결과, SRCM102343와 SRCM102329의 Lb. rhamnosus GG보다 부착율이 높았다. 선발된 유산균 4종에 대하여 항균활성 조사 결과, 그 중 3종이 Escherichia coli ATCC 10798, Staphylococcus aureus KCCM 11593, Listeria invanovii KCTC3444, Bacillus cereus ATCC11778 및 S. enterica serovar. Typhi KCTC1926에 대하여 항균활성이 뛰어났다. 이들 결과를 바탕으로 선발된 균주는 프로바이오틱의 가능성으로 기능성식품에 활용이 기대된다.
The aim of this study was to enhance the antioxidant activity of salted squid by inoculation of two Weissella spp. strains (W. cibaria FB-069 and W. viridescens FB-077) isolated from traditional Korea salted squid. The safety and probiotic potential characteristics of these two strains were evaluated. The safety of these strains was analyzed based on hemolytic activity, mucin degradation, biogenic amino production, and resistance to antibiotics. These lactic acid bacteria showed probiotic potential, including resistance to gastrointestinal tract conditions, adhesion to Caco-2 cells, and aggregation. The low-salted squid fermented with Weissella strains had consistently higher antioxidant activity through changing their amino acid profiles. Therefore, W. cibaria FB-069 and W. viridescens FB-077 might be good candidates for fermentation of salted squid to develop functional food with enhanced health benefits.
다양한 건강기능식품 중 프로바이오틱스는 전 세계적으로 가장 방대한 시장을 이루고 있으며, 장 건강이 우리 몸의 전반적인 면역작용을 조절하며 질병을 예방하고, 나아가 치료에 도움을 줄 수 있다는 연구결과에 따라 지속적으로 프로바이오틱스에 대한 관심이 증가하고 있다. 본 연구에서는 무화과식초가 가진 항균능을 이용하여 유제품과 드레싱 소스를 개발하여 그 선호도를 조사하고 대장세포에 미치는 영향을 분석하고자 하였다. 일반우유에 5% 무화과식초를 첨가하였을 때 마시는 요거트와 리코타치즈에 대한 가장 만족도가 높았다. 제조된 유제품에서 분리한 균은 Leuconostoc lactis이며 1.0×107~1.0×108 CFU/mL 이상의 세균수가 측정되어 식품의 기준 및 규격의 기준치를 만족하였다. 인체의 소장 상피세포와 흡사하게 증식되는 Caco-2 세포주에서 세포생존율을 관찰한 결과, 유청을 10% 농도로 처리하였을 때 세포생존율이 19% 가량 유의적으로 증가하였다. 농도별로 유청을 처리하였을 때 최대 58%의 항산화능을 보였다. 또한 유청에서 분리된 Leuconostoc lactis는 우수한 부착능이 관찰됨에 따라 장건강에 도움을 주는 유제품임을 확인하였다.
In this study, the survival capacity (acid and bile salt tolerance, and adhesion to gut epithelial cells) and probiotic properties (enzyme activity-inhibition and anti-inflammatory activities, inhibition of adipogenesis, and stress hormone level reduction) of Lactiplantibacillus plantarum LRCC5314, isolated from kimchi (Korean traditional fermented cabbage), were investigated. LRCC5314 exhibited very stable survival at ph 2.0 and in 0.2% bile acid with 89.9% adhesion to Caco-2 intestinal epithelial cells after treatment for 2 h. LRCC5314 also inhibited the activities of α-amylase and α-glucosidase, which are involved in elevating postprandial blood glucose levels, by approximately 72.9% and 51.2%, respectively. Treatment of lipopolysaccharide (LPS)-stimulated RAW 264.7 cells with the LRCC5314 lysate decreased the levels of the inflammatory factors nitric oxide, tumor necrosis factor (TNF-α), interleukin (IL)-1β, and interferon-γ by 88.5%, 49.3%, 97.2%, and 99.8%, respectively, relative to those of the cells treated with LPS alone. LRCC5314 also inhibited adipogenesis in differentiating preadipocytes (3T3-L1 cells), showing a 14.7% decrease in lipid droplet levels and a 74.0% decrease in triglyceride levels, as well as distinct reductions in the mRNA expression levels of adiponectin, FAS, PPAR/γ, C/EBPα, TNF-α, and IL-6. Moreover, LRCC5314 reduced the level of cortisol, a hormone with important effect on stress, by approximately 35.6% in H295R cells. L. plantarum LRCC5314 is identified as a new probiotic with excellent in vitro multifunctional properties. Subsequent in vivo studies may further demonstrate its potential as a functional food or pharmabiotic.
