Safety Evaluation of Chrysanthemum indicum L. Flower Oil by Assessing Acute Oral Toxicity, Micronucleus Abnormalities, and Mutagenicity

  • Chrysanthemum indicum is widely used to treat immune-related and infectious disorders in East Asia. C. indicum flower oil contains 1,8-cineole, germacrene D, camphor, ${\alpha}$-cadinol, camphene, pinocarvone, ${\beta}$-caryophyllene, 3-cyclohexen- 1-ol, and ${\gamma}$-curcumene. We evaluated the safety of C. indicum flower oil by conducting acute oral toxicity, bone marrow micronucleus, and bacterial reverse mutation tests. Mortality, clinical signs and gross findings of mice were measured for 15 days after the oral single gavage administration of C. indicum flower oil. There were no mortality and clinical signs of toxicity at 2,000 mg/kg body weight/day of C. indicum flower oil throughout the 15 day period. Micronucleated erythrocyte cell counts for all treated groups were not significantly different between test and control groups. Levels of 15.63~500 ${\mu}g$ C. indicum flower oil/plate did not induce mutagenicity in S. Typhimurium and E. coli, with or without the introduction of a metabolic activation system. These results indicate that ingesting C. indicum flower oil produces no acute oral toxicity, bone marrow micronucleus, and bacterial reverse mutation.

ICR 마우스를 이용한 평위산과 발효평위산 급성독성 연구 (A Study on Acute Oral Toxicity of Pyungwi-san and Fermented Pyungwi-san in ICR Mice)

  • Objectives: This study was conducted to investigate the acute toxicity of Pyungwi-san(Pingwei-san in Chinese) in ICR mice, according to KFDA and OECD guideline. Methods: In the present study, 15 male and female ICR mice administrated singly by gavage at dose levels of 0 and 2000 mg/kg of Pyungwi-san. During the experimental period, no treatment-related death was observed. There were no adverse effects on clinical signs, body weight, and gross findings at all treatment groups. Results: These results showed that the single oral adminstration of Pyungwi-san (Pingwei-san) did not cause any toxic effect at the dose levels of 2000 mg/kg in rats. Conclusions: Taken together, the median lethal dose($LD_{50}$) of Pyungwi-san (Pingwei-san) was considered to be over 2000 mg/kg body for both sexes.

마우스에 대한 Surfactin C의 급성경구독성시험 (Acute Oral Toxicity of Surfactin C in Mice)

  • This study was carried out to investigate the acute toxicity of surfactin C in mice. Surfactin C was administered orally at does of 0, 381, 610, 977, 1562 and 2500 mg/kg. Number of deaths, clinical signs, body weights, feed and water consumptions, and biochemical examinations were investigated for 14 days after single oral administration of surfactin C. $LD_{50}$ value was over 2500mg/kg in mice. In addition, no differences were found between control and treated groups in clinical signs, body weight gains, hematology, serum chemistry, feed and water consumptions. The results indicate that surfactin C did not show any toxic effects at 2500 mg/kg in mice.

ICR 마우스를 이용한 인삼패독산(人蔘敗毒散)의 급성독성 연구 (Acute Toxicity Study on Insampaedok-san Extracts in Mice)

  • Objective : This study was carried out to investigate the acute toxicity and safety of Insampaedok-san extract in ICR Mice. Methods : SPF ICR male and female mice were administered orally with Insampaedok-san extract of 0 (control group), 1250, 2500 and 5000 mg/kg. After single administration, we daily examined number of deaths, clinical signs, gross findings and changes of body weight for 14 days. Hematological parameters and isolated organ weights were determined after 14 days of administration. Results : No dead animal and no significant changes of body weights were found during experimental period. In addition, no differences were found between control and all of treated groups in clinical signs, organ weights and hematology, and other findings. Conclusions : Insampaedok-san extract did not show any toxic effects and oral LD50 values of the extracts was over 5000 mg/kg in ICR mice.

오약순기산(烏藥順氣散)의 소염 진통작용에 관한 연구 (Studies on the Anti-inflammatory and Analgesic Activities of Ohyaksungisan)

  • Ohyaksungisan, combined preparation of crude drugs, has been used for hemiplegia, arthralgia and paralysis in traditional Korean medicine. The anti-inflammatory activity of the aqueous extract from Ohyaksungisan(OSSE) was investigated on acetic acid-induced edema and adjuvant arthritis in rats. Acute toxicity and analgesic action in mice were also examined. Its anti-inflammatory activity on 5% acetic acid-induced edema and adjuvant arthritis was observed with oral administration. The acute toxicity showed 10% mortality at 2400 mg/kg (p.o), but was not showed at 1200 mg/kg (i.p). OSSE was showed to have significant analgesic action (P<0.05) at 150 mg/kg and this action was strengthened at 300, 600 mg/kg. The anti-inflammatory effect was showed significant preventive effect on the hind paw edema from 90 min. and the adjuvant arthritis, when orally administered for 19 days. showed significant inhibitory effect on the hind paw edema from the 5th day.

마주송이풀의 생리활성(生理活性) (Biological Activities of Pedicularis resupinata var. oppositifolia)

  • The plant of Pedicularis resupinata var. oppositifolia(Scrophulariaceae) is described to be used for treatment of rheumatoid arthritis, malignant abscess (tumor), urolith and diuretics in oriental medicinal books. This study is concerned with some general pharmacological activities. It was observed that the methanol extract of the aerial part of this plant showed some effects on acute toxicity, carrageenin induced edema in rat paw, bile juice secretion and antibacterial effect. From this result it was obtained that the methanol extract revealed the antiinflammatory activity and bile juice secretion increasing effect. Thus, it was fractionated and buthanol fraction has obvious inhibition effect on edema and bile juice secretion increasing effect. The total extract showed low acute toxicity i.e., the minimum lethal dose was more than 8000mg/kg in oral administration in mice.

Lactobacillus plantarum AF1이 생성한 조항진균 물질의 마우스에 대한 급성독성 (Acute Toxicity of Crude Anti-fungal Compounds Produced by Lactobacillus plantarum AF1)

  • 본 연구는 김치로부터 항진균 활성을 보이는 유산균인 Lactobacillus(Lb.) plantarum AF1이 생산하는 조항진균 물질의 부분 정제물이 천연 식품보존제 및 사료보존제로서 사용 가능성을 알아보기 위하여 안전성을 평가하고자 단회 급성독성시험을 실시하였다. 급성독성시험을 위하여 경구로 1회 시험물질을 최고 용량(2,000 mg/kg)으로 하여 ICR계통 암수 마우스에게 용량별 각 군당 10 마리씩 투여한 후 14일간의 일반증상, 사망률, 체중, 임상증상 및 육안적 소견을 관찰하였다. 단회 경구투여한 후 모든 시험군에서 사망례가 관찰되지 않았으며, 시험동물은 시험 종료 시까지 계속 생존하여 평균치사량을 산출할 수 없었다. 경구투여한 후 마우스의 체중변화에 있어서도 암수 모두 대조군과 시료물질 투여군 사이에 유의성 있는 차이는 보이지 않았으며, 용량 의존적인 차이도 볼 수 없었다. 경구투여한 후 시험 종료한 다음 생존동물 모두를 부검하여 주요 장기의 육안적 소견을 관찰한 결과 대조군과 시료투여군 모두에서 외관상이나 내부 장기에 어떠한 이상소견이나 병변이 관찰되지 않았다. 이상의 결과로부터 시험물질인 Lb. plantarum AF1이 생산한 조항진균 물질을 경구투여 시 ICR계통 암수 마우스에서 독성학적인 변화가 관찰되지 않았으며, $LD_{50}$은 경구투여 시 2,000 mg/kg 이상인 저독성의 안전한 물질로 사료된다.

사상자 중 Torilin의 분리 및 진통소염작용 (Isolation of Torilin from Torilis japonica Fruit and Its Analgesic and Anti-inflammatory Activities)

  • Torilin was isolated from haxane fraction of Torilis Fructus extract. Torilin produced inhibition of the acetic acid-induced and phenylquinone-induced writhing syndrome at the oral doses of 30 and 90 mg/kg in mice. It also increased the pain threshold at the oral doses of 30, 90 and 270 mg/kg in the tail pressure method and the Randall-Selitto method. However, it did not show a hypothermic action at the oral doses of 30 and 90 mg/kg in mice. The compound exhibited strong anticarrageenan activity at the oral doses of 90 and 270 mg/kg in rats, and had inhibitory effect on the vascular permeability at the oral doses of 30 and 90 mg/kg in mice. It also showed potent inhibition of leucocyte emigration in CMC-pouch at the doses of 3 and 9 mg/rat, sc. The acute toxicity of torilin was very weak: the $LD_{50}$ values were more than 5000 mg/kg, po and 2000 mg/kg, ip in mice. From the above mentioned results, it was suggested that torilin had potent analgesic and anti-inflammatory activities.

Acute oral and subcutaneous toxicity of Aloewhite in Mice

  • -Acute oral and subcutaneous toxicity of Aloewhite(30% aloesine) were carried out in ICR mice. In this study, we daily examined number of deaths, clinical signs, body weights, and pathological examinations for 14 days after single oral and subcutaneous administration of Aloewhite with different dose levels. Aloewhite did not show any remarkable toxic effect in mice. These results suggest that oral and subcutaneous LD$\sub$50/ values in mice were over 6.8g/kg and 10g/kg, respectively.

합성세제 및 계면활성 성분의 독성학적 연구 (Toxicological Studies on Surfactants and Synthetic Detergents)

  • Synthetic detergents and surfactants are in widespread usage as household and industrial detergents. Potential human toxic hazard arises following percutaneous absorption or oral ingestion of solution residues from kitchen and feeding utensils, fruits, and vegetables and contaminated water supplies. A toxicological investigations was performed with the synthetic detergents and surfactants [linear alkyl benzene sulfonate (LAS), ${\alpha}$-olefin sulfonate (AOS), sodium lauryl sulfonate (SLS), sodium lauryl ester sulfonate (SLES)], In acute toxicity, agents were administered subcutaneously into ICR mice. In acute study, after lowering of spontaneous motility, respiratory failure, death appeared, vomitting was often associated with salivation and or retching. No sex difference was observed in LD$\sub$50/ of mice. In subacute toxicity, agents were administered orally into SD rats. Body weight increase was suppressed and there was no adverse effect on food and water consumption. The weight of organs were not changed by agents as compared with control group. No specific change was observed in biochemical and hematologicalor data.
