Effects of Protein and Fiber on Antioxidant Enzyme Activites of Brain in Ethanol-Treated Rats (에탄올을 투여한 흰쥐 노조직의 항산화효소계 활성에 미치는 단백질과 섬유소의 영향)

  • This study was to investigate the effect of dietary protein and fiber on the antioxidant enzyme activities of brain in acute or chronic ethanol-treated rats. Male Sprague-Dawley rats were fed on diets containing two levels of protein(7%, 20%) with two levels of fiber (5%, 10%) Rats were administered 40%(v/v) ethanol(5g/kg body weight)orally 90min before decaptiation in acute ethanol-treated groups and 25%(v/v) ethanol(5g/kg body weight) once a day for 5 weeks in chronic ethanol treated-groups. The rats were sacrificed after 5 weeks of feeding periods. Superoxide dismutase and gluthathione S-transferase activities were lower in chronic ethanol-treated groups than acute ethanol-treated groups whereas catalase and glutathuone peroxidase activities were significantly increased by chronic ethanol treatment. Low protein supplement accelerated to change of their activities however dietary fiber levels did not affect antioxidant enzyme activities. Chronic ethanol treatment and/or low protein supplement results in increasing the brain lipid peroxide content but in lowering glutathione level. (Korean J Nutrition 33(6) ; 613~618, 2000)

  • Image scrambling is an important technology in information hiding, where the Arnold transformation is widely used. Several researchers have proposed the application of $H{\acute{e}}non$ map in square image scrambling, and certain improved technologies require scrambling many times to achieve a good effect without resisting chosen-plaintext attack although it can be directly applied to non-square images. This paper presents a non-square image scrambling algorithm, which can resist chosen-plaintext attack based on a chaotic two-dimensional Sine Logistic modulation map and $H{\acute{e}}non$ map (2D-SLHM). Theoretical analysis and experimental results show that the proposed algorithm has advantages in terms of key space, efficiency, scrambling degree, ability of anti-attack and robustness to noise interference.

Study on Acute Subcutaneous Toxicity of Hydroxyapatite Sinter Produced from Tuna Bone in Sprague-Dawley Rats (참치뼈로부터 제조한 Hydroxyapatite 소결체의 랫드에 대한 급성피하독성시험)

  • This study was performed to evaluate the actue sbucutaneous toxicity of hydroxyapatite sinter produced from tuna bone in Sprague-Dawley(SD) rats. Hydroxyapatite sinter was administrated at dose levels of 5000, 2500, 1250, 625, 312.5 and 0 mg/kg. After single subcutaneous adiminstration to both sexes to both sexes SD rats, we observed rats for 14 days. Hydroxyapatite sinter did not induce any toxic signs inmortalities, clinical findings, body weights and gross findings of the rats. In view of result, it was impossible to estimate LD/ sub 50/ values in SD rats. In conclusion, these results suggest that hydroxyapatite sinter produced from tuna bone has no effect on acute subcutaneous toxicity in SD rats.

  • Acute oral toxicity of Bifidobacterium breve K-110, Bifidobacterium breve K-111, Bifidobacterium infantis K-525 were studied in Sprague-Dawley rats of both sexes. In this study, we examined number of deaths, clinical signs, bod weight and gross findings for 14 day after single oral administration of B. breve K-110,B. breve K-111 or B. infantis K-525 with different levels. They did not show any toxic effect in rats and oral LD$_{50}$ value was over 5 g/kg in rats.s.

  • SKI306X is a herbal extract prepared from three herbs Clematis mandshurica, Trichosanthes kirilowii and Prunella vulgaris. It showed strong antiinflammatory actions on carrageenan-induced edema, acetic acid-induced pain, adjuvant-induced arthritis, and oxygen radical-generated reactions. In this study, the acute toxicity of SKI306X was evaluated in rats by a single oral administration. Thirty male and thirty female rats were divided into 6 groups according to the dose levels, respectively. After oral administration of SKI306X with several doses (5.0 g/kg, 3.3 g/kg, 2.2 g/kg, 1.5 g/kg, 1.0 g/kg), mortality, clinical signs, body weight, and gross findings in organs were examined. No toxic effect was shown in terms of mortality, clinical signs, body weight changes and gross findings. It is suggested the LD$_{50}$ of SKI306X would be more than 5.0 g/kg in rats.s.

The Clinical Efficacy of Chungpyesagan-tang on Acute Stroke (급성기 중풍 환자에 대한 청폐사간탕의 임상적 효능)

  • To assess the clinical efficacy of Chungpyesagan-tang on acute stroke, we prescribed this medicine to 88 acute stroke patients without thrombolytic treatment. The rate of progressive stroke type was 2.3%, remarkably lower than in previous reports. 1.1 % felt an itching sensation, and 17.0% complaxined of loose stool and diarrhea. Chungpyesagan-tang decreased Stroke-Pattern Identification and National Institute of Health Stroke Scale (NIHSS), and increased Modified Barthel Index (MBI). Thus, we could suggest that this medicine has a desirable effect to reduce the severity of stroke and improve functional recovery. As to the laboratory findings, ALT had anupward tendency and increased over normal value in 16 cases (18.2%). However, the mean serum level 2 weeks later was within the normal value and the other hepatic enzymes did not increase.

Acute Toxicity Study on Scopoliae Rhizoma in Mice (낭탕근의 급성독성 연구)

  • Scopoliae rhizoma is a perennial herb which has a similar effect with atropine on the cardiovascular system. It is also known to have a seditive and anticonvulsant activity on the central nerve system. In order to evaluate an acute toxicity of Scopoliae rhizoma, the present study was performed after administration the Scopoliae rhizoma prepared by both decoctional and frozen dried extract through three different routes (oral; 5,000 mg/kg, intraperitoneal; 2,000 mg/kg, subcutaneous; 5,000 mg/kg) to the female ICR mice. In the group treated intraperitoneally with a frozen dried extract, abnormal clinical signs such as decreased activity, crouch, potosis and abnormal walking were observed for 40 rain after administration. With regard to WBC, decreased number of lymphocyte and increased number of monocyte and granulocyte were also observed in the animals received intraperitoneally with Scopoliae rhizoma extract. Taken together, what toxicity of Scopoliae rhizoma was shown differently depending on its type for administration may be resulted in the differency of administered dose. The results provided here support a pharmacological and toxicological consideration for its clinical use in the regard of oriental medicine.

Quantitative Analysis of Orcinol and Acute Toxicity of Gyrophora esculenta (석이중 오르시놀 정량 및 급성독성)

  • In previous study, Gyrophora esculenta showed significant inhibitory effect on $\alpha$-glucosidases in vitro and blood glucose elevation in vivo. In the isolating process of active substance, orcinol was separated from Gyrophora esculenta. Orcinol is known to be toxic, therefore, in this study, it was analysed by the TLC densitometry method for quantitative determination from Gyrophora esculenta. The average amount of orcinol of Gyrophora esculenta was 0.2%. For the purpose of removing orcinol, the water extract of Gyrophora esculenta was sequentially fractionated by organic solvents, and the acute toxicity of each fraction was assessed in mice. Among them, the LD50 of butanol fraction was 1.19 g/kg(p.o.) and the weight increase of mice in that group was somewhat retarded.

Acute Oral Toxicity of Chitooligosaccharides in Mice (Chitooligosaccharides의 마우스에 대한 급성 경구독성)

  • Chitooligosaccharides were prepared by enzymatic hydrolyzing of crab shell chitosan. Low molecular weight chitooligosaccharides(LMW-chitooligosaccharides), 64.3% of which was composed of trimer, tetramer, and pentamer, was obtained by hydrolyzing chitosan with the chitosanase originated Bacillus pumilus BN-262. High molecular weight chitooligosaccharides(HMW-chitooligosaccharides), 49.3% of which was composed of chitooligosaccharides over heptamer, was obtained by hydrolyzing chitosan with the cellulase originated Trichoderma viride. Acute oral toxicity of chitooligosaccharides were tested in mice. Chitooligosaccharides did not have any toxic effect in mice and oral LD$\_$50/ value of chitooligosaccharides was over 5.0g/kg in mice.

Reduction of Hepatic Glutathione by Acute Taurine Treatment in Male Mice (숫컷 생쥐에서 타우린 투여에 의한 간내 글루타치온의 감소)

  • Effect of taurine treatment on metabolism of glutathione (GSH) was studied in adult male ICR mice. An acute injection of taurine (250 mg/kg, ip) resulted in a significant decline of hepatic GSH level at t = 6 hr, but plasma GSH level was not altered. The activity of GSH-related enzyme in liver, such as GSH peroxidase, GSSG reductase, GSH S-transferases, ${\gamma}$-glutamylcysteine synthetase or ${\gamma}$-glutamyltranspeptidase, was not affected by taurine at t = 2.5 or 6 hr. Plasma cysteine and cystine levels were elevated rapidly following taurine treatment. Hepatic cysteine level was decreased by taurine, reaching a level approximately 70% of control at t = 4 and 6 hr. In conclusion, the results indicate that an acute dose of taurine decreases hepatic GSH level by reducing the availability of cysteine, an essential substrate for synthesis of this tripeptide in liver. It is also suggested that taurine may decrease the cysteine uptake by competing with this S-amino acid for a non-specific amino acid transporter.
