HCl/에탄올로 유발된 급성 위염에서 Artemisia frigida Willd의 위장 보호 효과 (Gastroprotective Effect of Artemisia frigida Willd in HCl/Ethanol-induced Acute Gastritis)

  • Artemisia frigida Willd (AW, Fringed sagewort), which is widespread in Mongolia, is a well-known medicinal plant as a member of the Compositae family. This study aims to explore the gastroprotective effect of water extract of AW on 150 mM HCl/60% ethanol-induced acute gastritis in 5 week old male ICR mice. Total polyphenols, total flavonoid contents, and anti-oxidant activity in vitro in AW were evaluated. First, the gross area of gastric mucosal damage was measured. Then western blot analysis was conducted to determine the possible mechanisms of action underlying the effects of AW. AW administration decreased gastric mucosal damage. Moreover, the group with AW treatment effectively inhibited nicotinamide adenine dinucleotide phosphate (NADPH) oxidase expression associated with oxidative stress. AW treatment enhanced an anti-oxidant effect through the increase of anti-oxidant proteins. Besides, the increased expressions of inflammatory cytokines induced by nuclear factor-kappa B (NF-κB) activation are alleviated through AW treatment. Taken together, AW exerted a gastroprotective effect against gastric mucosal damage. These results indicate that AW could have the potential used as a natural therapeutic drug for the treatment of acute gastritis.

산약(山藥) 물 추출물의 급성 췌장염 보호 효과 (Protective effects of Dioscorea batas Decaisne water extract on acute pancreatitis)

  • Objectives : Dioscorea batas Decaisne (DB) has been known to be good for the digestive system on Eastern Asia. However, the protective effect of DB on acute pancreatitis (AP) has not been studied. In this study, we tried to investigate the protective effect of DB water extract on caerulein-induced AP. Methods : To measure the protective effect of DB on AP, Mice were injected with cholecystokinin analogue caerulein (50 ㎍/kg) hourly for 6 times. DB water extract (200 or 400 mg/kg) or saline (control group) was administered orally 1 h before the first injection of caerulein. The mice were sacrificed at 6 h after the last injection of caerulein. The pancreas tissues and serum samples were immediately taken for further analysis. Results : Administration of DB water extract showed the inhibitory effect on the increase of pancreas weight/body weight ratio, pancreatic histological damage. And the rise of serum lipase level was significantly reduced in DB water extract treatment group during AP in mice. However administration of DB water extract did not show significant reduction in serum amylase level. Also, mRNA levels of pro-inflammatory cytokines Interleukin (IL)-6 and Tumor necrosis factor (TNF)-𝛼 but not IL-1𝛽 were inhibited by administration of DB water extract. Conclusions : Taken together, we found that administration of DB water extract ameliorates the severity of caerulein-induced AP, which suggests the potential to be an effective treatment on AP.

Changing Gadolinium-Based Contrast Agents to Prevent Recurrent Acute Adverse Drug Reactions: 6-Year Cohort Study Using Propensity Score Matching

  • Objective: To determine the preventive effect of changing gadolinium-based contrast agents (GBCAs) to reduce the recurrence of GBCA-associated acute adverse drug reactions (ADRs). Materials and Methods: This retrospective, observational, single-center study-conducted between January 2016 and December 2021-included 238743 consecutive GBCA-enhanced MRI examinations. We focused on a subgroup of patients who experienced acute GBCA-associated ADRs during any of these examinations and subsequently underwent follow-up GBCA-enhanced MRI examinations up until July 2023. The follow-up examinations involved either the same (non-change group) or different (change group) GBCAs compared to the ones that initially caused the acute ADR. Baseline participant characteristics, generic profile of the GBCAs, administration of premedication, history of prior ADR to iodinated contrast media, and symptoms of GBCA-associated acute ADRs were retrospectively analyzed. Multivariable logistic regression with generalized estimating equations and propensity score matching were used. Results: A total of 1042 instances of acute ADRs (0.44%; 95% confidence interval [CI]: 0.41%-0.46%) were reported. Three-hundred and seventy-three patients underwent GBCA-enhanced MRI examinations after experiencing GBCA-associated acute ADRs within the study period; 31.9% (119/373) reexperienced acute ADRs at any of the follow-up examinations. The ADR recurrence was significantly lower in the GBCA change group than in the non-change group according to multivariable logistic regression (adjusted odds ratio [OR]: 0.35; 95% CI: 0.13-0.90; P = 0.03) and analysis with propensity score matching (14.3% [6/42] vs. 36.9% [31/84], respectively; OR: 0.32, 95% CI: 0.11-0.94; P = 0.04). A history of an ADR to iodinated contrast media (OR: 1.14, 95% CI: 0.68-1.90; P = 0.62) and premedication (adjusted OR: 2.09, 95% CI: 0.93-4.68; P = 0.07) were not significantly associated with GBCA-associated acute ADR recurrence. A separate analysis for recurrent allergic-like hypersensitivity reactions demonstrated similar results (adjusted OR: 0.20, 95% CI: 0.06-0.65; P < 0.01). Conclusion: Changing GBCAs may reduce the risk of GBCA-associated acute ADR recurrence.

  • The structure of tuguaconitine, MP $196{\sim}198^{\circ}C$, a new diterpene alkaloid isolated from the roots of Aconitum sibiricum P., was studied. Its empirical formula is $C_{23}H_{35}NO_7$ and spectroscopic evidence shows that the base contains three methoxyl, three hydroxyl and one N-ethyl groups. The acute toxicities of most of Aconitum species were strong and the acute toxicity of the alkaloid fraction of A. sibiricum was decreased by Glycyrrhizae, black bean or Zingiberis extract.

  • The acute toxicity of a recombinant human interferon $\alpha$A (code name: LBD-007) was evaluated in both sexes of ICR mice, 5 weeks old, by the oral, subcutaneous and intravenous routes of administration. Based on the results, LBD-007 was not considered to induce any toxic effect on the mice in mortalities, clinical findigs, body weights and gross findings. It is suggested that LD$_{50}$ values in mice would be $>48{\times}10^8$ IU/kg in the oral, subcutaneous or intravenous routes.s.

  • This is a case report about effect of syeom pharmacopuncture, bee venom and autohemo-seperated regeneration pharmacopuncture(ASRP) for acute traumatic subdeltoid bursitis with partial tear of subscapularis tendon, which was diagnosed by symptoms and MRI(Magnetic resonance imaging) and caused by bongchuna treatment. We evaluated the patient using Visual Analogue Scale(VAS) every two or four days and range of movement(ROM), physical examination of shoulder about one per ten days and observed improvement with reexamination by ultrasonography and MRI as well. Pharmacopuncture rapidly reduced pain and improved range of motion and function of shoulder in patients with acute sub-deltoid bursitis even though it was severe symptom. Our result suggest that autohemo-seperated regeneration pharmacopuncture might be effective in regenerating the tear of soft tissue such as subscapularis tendon.

  • The actue toxicity of a recombinant granulocyte macrophage colony-stimulating factor (code name: LBD-005) was evaluated in both sexes of Sprague-Dawley rats, 4 weeks old, by the oral, subcutaneous and intravenous routes of administration. LBD-005 in the acute toxicity study in the rats was not considerde to induce any toxicological effect on the rats in mortalities, clinical findings, body weights and gross findings. It is suggested that $LD_{50}$ values in rats would be >48 mg/kg in the oral route and >12 mg/kg in the subcutaneous or intravenous route.

An Acute Pathophysiology of Environmental Strains of Cryptococcus neoformans Isolated from a Park in Busan

  • The present study was carried out to elucidate whether an environmental strain of Cryptococcus neoformans (environmental C. neoformans) isolated from an environmental source in a park of Busan has an acute pathophysiological effect in rats. On the second day after peritoneal inoculation of environmental C. neoformans, adverse effects occurred from the viewpoint of hematology and biochemistry. Eosinophil damages and crystal formations were found in the blood. Disturbances in cytokines production were observed in the cerebral and pulmonary tissues. Fungal budding existed in the brain, lung, liver and kidney. Tissue injury findings such as inflammation, leukocyte infiltration, bleeding, or degeneration were found in the brain, lung, liver and kidney. The present data suggest that the environmental C. neoformans can cause systematically harmful effects even for short periods of infection (two days of cryptococcal infection) and the adverse effects are summarized as immune derangements and biochemical and/or histological dysfunction and injury on major organ such as the brain, lung, liver and kidney in the immunocompetent hosts. Further studies should be focused on comparing the differences between environmental and clinical strains of C. neoformans.

  • Gamiinjinoryung-San is composed of eight crude drugs including Artemisiae capillaris Herba which is widely used for the treatment of acute jaundice, acute and chronic hepatitis at the Oriental hospital of Kyung-Hee Medical Center. This study was conducted to investigate the effects of water extract of Gamiinjinoryung-San on the liver function. The results obtained were as follows; the extract markedly reduced LDH and ALP activities, but slightly decreased GOT and GPT activities elevated with D-galactosamine in rat serum. The liver protective activities were shown in $CCl_4-intoxicated\;rats$. The extract prevented the prolongation of sleeping time in $CCl_4-intoxicated\;rats$. The bile flow and the biliary bile acid secretion were significantly increased in normal rats.

  • This study was performed to investigate the acute toxicity of PEG-hemoglobin SB1, a blood substitute, in beagle dogs. The male and female dogs were administered intravenously at the doses of 0.4375, 0.875 and 1.75 g/kg body weight, respectively. After a single intravenous administration of SB1 to dogs, we observed them daily for 2 weeks. SB1 did not induce any toxic signs in the mortalities, clinical signs, body weight changes, and gross necropsy findings of dogs. Based on these results, acute toxicity, dogs SB1 may have no side effect and its $LD_{50}$ value may be over 1.75 g/kg (25 ml/kg) of body weight in dogs.
