Characterization of a Restriction Endonuclease AspJI from Alcaligenes sp. J-482 (Alcaligenes sp. J-482 로부터 분리한 제한효소 AspJI의 특성)

  • About 500 bacterial and fungal strains from a wide variety of natural habitats were screened for a new type II restriction endonuclease. Among the 500 species, we selected one species that produced a new restriction endonuclease. This strain has an optimum temperature of $30^{circ}C$ for growth. Morphological, cultural, and physiological characteristics were examined for identification of the isolated strain J-482. This strain was found to belong to the genus Alcaligenes. The restriction endonuclease was named as AspJI and partially purified from Alcaligenes sp. J-482 by DEAE-Sephadex A-50 column chromatography and gel filtration. Most of other nucleases were removed by the purification steps. The AspJI has a substrate specificity to ${lambda}$ DNA, pBR322 and Adenovirus-2 DNA. For its maximal activity, the isolated enzyme requires $MgCl_2$, which should be at least 12.5 mM and it does not need any other cofactors. It is maximally active in the absence of NaCl and is completely inactivated at 100 mM NaCl. The pH and temperature optima for activity were pH 7.5 and $37^{circ}C$, respectively. The DNA fragments generated by digesting ${lambda}$ DNA, pBR322, and Adenovirus-2 DNA with AspJI were the same as that produced by AatII. This suggests that AspJI is an isoschizomer of AatII.

Effect of Dietary Level of $\beta$-Carotene on Antioxidants Contents and Hepatic Morphology in Rats ($\beta$-Carotene 의 수준별 공급에 따른 흰주의 항산화 물질과 간조직의 형태적 변화)

  • This study was designed to investigate the effect of dietary level of $\beta$-carotene on level of antioxidant nutrients of rat tissues. Male Sprague Dawley rats at the age of 30days were fed on diets containing different levels of $\beta$-carotene(0, 10, 120, 1200, 12000mg per kg diet). Body weight gain of rats fed with 12000mg $\beta$-carotene diet was significantly decreased, but liver and heart weights were not significantly different among groups, The content of total glutathione tended to decrease significantly in 12000mg $\beta$-carotene diet group when compared to $\beta$-carotene restriction group(BC O). However, total vitamin C content of liver showed the tendency to increase by $\beta$ -carotene supply up to 1200mg. But this tendency was not found in plasma, The content of zinc in liver and plasma was significantly decreased by $\beta$-carotene restriction. Alkaline phosphatase activity was significantly higher in 12000mg diet group. In case of $\beta$-carotene restriction group, fibroblasts were proliferated in portal endothelium, and the vacuolar size was enlarged more than the nuclear, In 12000mg diet group, hepatic vacuoles were extended, but their size was regular and the lysis of hepatocytes was observed. Also, fibroblasts were proliferated in portal endothelium and the regular vacuolar size was extended.

Effects of Different Root Restriction Media on Root Activity and Seedling Quality and Early Growth Parameters of Runner Plantlets of Strawberry After Transplanting

  • The present study aimed to determine the influence of various root restriction media on seedling quality and early growth of strawberry after transplanting. The root activity of the seedlings, measured 20 days after fixation, was considerably higher (0.096, 0.090, and $0.063mg{\cdot}g^{-1}{\cdot}h^{-1}$ at 420, 450, and 480 nm, respectively) in expanded rice hull (ERH) treatment than in the sandy loam and loamy sand treatments. The volumetric water content (VWC) of the root media tested across 3 irrigation regimes (15 d, 30 d, 45 d) in the nursery field was highest in sandy loam (65.0-66.8%), followed by 59.4-61.3% in loamy sand and 38.6-45.3% in ERH. When growth parameters of runner plantlets were compared, ERH treatment was found to result in the highest crown thickness and fresh weights of root and above-ground parts. This had a favorable influence on above-ground tissue growth after transplanting to plastic house soil. As mentioned above, ERH treatment resulted in the highest seedling quality and early growth after transplanting. The results of this study would serve as useful on-site data for the production of high-quality strawberry seedlings.

Comparison of Factors Influencing Health-Related Quality of Life between Young-Aged and Old-Aged Patients with Cancer: Analysis of the 2015 Korea Health (노인 암환자의 건강관련 삶의 질 영향요인: 2015년 한국의료패널 자료 분석)

  • Purpose: This study aimed to examine factors influencing health-related quality of life (HRQOL) and compare them between young-aged and old-aged patients with cancer. Methods: Data of 291 patients (young-aged: 168, old-aged: 123) were obtained from the 10th wave of the 2015 Korea Health Panel Survey. The HRQOL was measured using the Korean version of Euro-QoL-5D. Independent t-test, analysis of variance, and multiple regression analysis were performed to identify factors influencing HRQOL. Results: The average HRQOL score was 0.87±0.10 and 0.82±0.15 among young-aged and old-aged, respectively. The factors differed partially between the two groups. For young-aged, the influencing factors were activity restriction, subjectively perceived health status, and smoking. For old-aged, the influencing factors were activity restriction, subjective health status, and unmet healthcare needs. Conclusion: Strategies to improve the HRQOL of elderly adults need to be developed considering the age group. Additionally, studies that include clinical factors such as symptoms are required to prepare need-based practical approaches for better quality of life of such patients.

The Family Characteristics Affecting the Socio-Cultural Adjustment of North Korean Defectors (가족특성에 따른 새터민의 사회문화적 적응)

  • As the number of North Koreans defectors entering South Korea with their family members increases, it becomes more important to explore the family characteristics that facilitate their adjustment. The data used in the present study was collected from a survey of 195 North Korean defectors. The aim of this study was to investigates how the structural and relational aspects of family influence the socio-cultural adjustment of North Korean defectors. The structural characteristics of family life included whether or not the defectors exited with their family members, whether they were living with family members, and whether they were living with family members under the age of 18. The relational characteristics included family cohesion and adaptability. The study results showed that living with young family members was negatively associated with the social activity restriction problems commonly perceived by North Korean defectors. Family cohesion was also negatively associated with the social activity restriction problems and positively associated with the social isolation problems. The results of this study also showed that family adaptability was not significantly associated with any socio-cultural adjustment problems.

Effects of general and oral health on quality of life in the elderly living alone and with family (독거노인과 가족동거노인의 건강 및 구강건강이 건강 관련 삶의 질에 미치는 영향)

  • Objectives: The purpose of this study was to investigate the effects of general and oral health on quality of life in the elderly living alone and with family. Methods: We analyzed data from the $6^{th}$ Korea National Health and Nutrition Examination Survey. Distribution of the elderly living alone and with family based on the general characteristics and general and oral health was analyzed using complex-sample chi-square tests. Multiple logistic regression was used to analyze the factors affecting quality of life by calculating the 95% confidence intervals. Results: In the elderly living alone, the quality of life significantly correlated with restriction of activity, perceived general and oral health status, perceived stress, and speech difficulties. Further, in the elderly living with family, lower quality of life significantly correlated with restriction of activity, perceived health status, walking days per week, life time smoking history, Community Periodontal Index, and chewing and speech difficulties. Conclusions: The elderly are concerned with self-maintenance of general and oral health. Therefore, systematic policies related to health services need to be developed and operated at the national level. It is especially necessary to take social interest in the elderly living alone and a more continuous and professional approach in their health care.

Effects of chronic caloric restriction on kidney and heart redox status and antioxidant enzyme activities in Wistar rats

  • Caloric restriction (CR) has been associated with health benefits and these effects have been attributed, in part, to modulation of oxidative status by CR; however, data are still controversial. Here, we investigate the effects of seventeen weeks of chronic CR on parameters of oxidative damage/modification of proteins and on antioxidant enzyme activities in cardiac and kidney tissues. Our results demonstrate that CR induced an increase in protein carbonylation in the heart without changing the content of sulfhydryl groups or the activities of superoxide dismutase and catalase (CAT). Moreover, CR caused an increase in CAT activity in kidney, without changing other parameters. Protein carbonylation has been associated with oxidative damage and functional impairment; however, we cannot exclude the possibility that, under our conditions, this alteration indicates a different functional meaning in the heart tissue. In addition, we reinforce the idea that CR can increase CAT activity in the kidney. Moreover, CR caused an increase in CAT activity in kidney, without changing other parameters. Regional Difference of ROS Generation, lipid Peroxidation, and Antioxidant Enzyme Activity In Rat Brain and Their Dietary Modulation

  • One of the potential causes of age-related neuronal damage can be reactive oxygen species (ROS), as the brain is particularly sensitive to oxidative damage. In the present study, we investigated the effects of aging and dietary restriction (DR) on ROS generation, lipid peroxidation, and antioxidant enzymes in cerebrum, hippocampus, and cerebellum of 6-, 12-, 18-, and 24-month-old rats. ROS generation significantly increased with age in cerebrum of ad libitum (AL) rats. However, no significant age-difference was observed in hippocampus and cerebellum. DR significantly decreased ROS generation in cerebrum and cerebellum at 24-months. On the other hand, the increased lipid peroxidation of AL rats during aging was significantly reduced by DR in all regions. Our results further showed that catalase activity decreased with age in cerebellum of AL rats, which was reversed by DR, although SOD activity had little change by aging and DR in all regions. In a similar way, glutathione (GSH) peroxidase activity increased with age in cerebrum of AL rats, while DR suppressed it at 24-months. These data further support the evidence that the vulnerability to oxidative stress in the brain is region-specific.

Influence of Energy Restriction and Pre-incubation Holding Period of Eggs on Fertility and Hatchability in Aged Broiler Breeders

  • The effects of controlled energy restriction and duration of pre-incubation egg holding on fertility, hatchability and hatch losses were evaluated in aged broiler breeders (64 wk). The energy (ME) required for maintenance, activity, growth and anticipated egg production was calculated and offered to a control group (283-471 kcal/kg) from 21-64 weeks of age. In three other groups, ME was quantitatively reduced either by 20% (SER; severe energy restriction) or 10% (MER; moderate energy restriction) and increased by10% (EEF; excess energy feeding) over the control group (CER; controlled energy restriction). Each diet was offered to 130 pullets in individual cages, and the quantity of ME increased with age. At the end of 64 weeks, fertile eggs were collected from each dietary group for 11 consecutive days and grouped under 4 holding periods based on the length of storage (2, 5, 8 or 11 d). The influence of energy regimes, egg holding intervals and their interaction was evaluated on fertility, hatch losses and hatchability. Broiler breeders maintained on SER regime (231-419 kcal/d) produced maximum number of eggs (993) followed by MER (819), CER (624) and EEF (438) during the 11-day period. The percent fertility and hatchability was significantly (p$\leq$0.05) higher in SER and MER groups compared to CER and EEF. However, energy regimes did not influence the loss in egg weight during pre-incubation storage, shell weight, shell thickness or hatch losses as dead germs and dead in shell. The improvement in hatchability in SER and MER groups appeared to be closely related to higher fertility and lower embryonic mortality. Holding of eggs for 11 days showed a linear loss in egg weight with the length of storage, but did not influence the fertility and hatch losses. The percent hatchability on eggs set was maximum when storage period was restricted to 5 days. The interaction between energy regimes and egg holding periods exhibited better hatchability results with SER regime when eggs were held for 5 days. Response to MER was not different from SER. It was obvious that energy restriction during production period had a positive influence on egg number, fertility and hatchability in aged breeders. At 64 weeks of age, holding of fertile eggs for 5 days prior to incubation was adequate for optimum hatchability in breeders.

Recombinant Expression and Purification of Functional XorII, a Restriction Endonuclease from Xanthomonas oryzae pv. oryzae

  • An endonuclease from Xanthomonas oryzae pathovar oryzae KACC 10331, XorII, was recombinantly produced in Escherichia coli using a T7 system. XorII was purified using a combination of ion exchange and immobilized metal affinity chromatography (IMAC). An optimized washing protocol was carried out on an IMAC in order to obtain a high purity product. The final amount of purified XorII was approximately 2.5 mg/L of LB medium. The purified recombinant XorII was functional and showed the same cleavage pattern as PvuI. The enzyme activity tested the highest at $25^{\circ}C$ in 50 mM NaCl, 10 mM Tris-HCl, 10 mM $MgCl_{2}$, and 1 mM dithiothreitol at a pH of 7.9.
