Glutamine synthetase (GS) of Kkbsiellu pmumonzae was purified and identified it's properties. It was determined to be composed of 12 identical subunits and it's molecular weight was about 600,000. It's optimum pH and temperature were identified as pH 7.0 and $37^{\circ}C$ respectively, and also there was no considerable variation of activity between pH 5 and 8. When GS was incubated at $57^{\circ}C$ for 10 min, it's activity was decreased to half of maximum activity. It was observed that K. pneumoniae has adenylylation-deadenylylation system which regulates activity of GS according to the quality and quantity of nitrogen source like GS of E. coli Also it's GS was very similar to that of E. coli. in structure deduced from the immunodiffuslon experiment using anti-E. coli GS antibody.

The purpose of this study is to find out the activity of gluteus medius muscle by the changes of load given to the upper limbs in single support phase. This study was twenty healthy men from A College. The left gluteus medius muscle was measured using SEMG(surface Electromyogram). Only the left foot was supported, and for the right foot, the right upper limbs were abducted while hip joint and the knee joint were bent at a $90^{\circ}$ angle. The study was made by giving weight using dumbbells, depending on the RM of the subject(0RM, 1RM, 3RM, 5RM, 7RM). Gluteus medius muscle showed a large activity for those given weight to the upper limbs(1RM, 3RM, 5RM, 7RM) than that without weight(0RM). There was a significant difference in the activity of gluteus medius muscle between each RM. Gluteus medius muscle is high active when weight is given to the upper limbs in single support phase.
Objectives : JiaoTeng-Yuan(교등원) is said to be a prescription for preservation of health in $\ulcorner$HuaTuo.ZhongZangJing (화타.중장경)$\lrcorner$. It is known to have the effect of Bu-Shen (보신: strengthening kidney) and Yi-Shou (익수: prolonging the span of one's life). This study investigates whether JTY is effective on inhibition of oxidation stress. Methods : Sprague-Dawley rats (12-week-old, weight $300{\pm}20g$) were divided into 3 groups. The normal group (n=8) was injected with PBS (1ml/body, s.c) subcutaneously behind the neck. The control group (n=8) was injected with D-galactose (50mg/kg, 1ml PBS/body, s.c) to induce pathological animals. The JTY group was injected with the same treatment as the control group, and fed containing JTY (10%). All groups were treated 1 time per day for 6 weeks. After the rats were sacrificed anti-oxidant enzyme (SOD, CAT, G-px) activity, GSH quantity of RBC and tissue (heart, liver and kidney), and plasma Vit-C quantity were examined. Also, the MDA levels of liver and kidney, lipofuscin of heart and endurance of erythrocyte membrane were measured. Results : In the JTY group, RBC's SOD activity decline was halted by 21 % of the normal level, compared to the control group, G-px activity (unit/g of Hb) increased significantly, compared to the normal group, and the level of Vit-C in plasma increased by 16%. Heart's SOD activity was kept at the same level as that of the normal group; and CAT activity decline was halted by 26%. Kidney's CAT and G-px activities were kept at the same level as that shown in the normal group, implying the existence of a halting effect. Liver also showed a slight halting effect against the decline of anti-oxidant ability, but the effect was not significant (${\alpha}=0.05$). A comparison between the levels of peroxide in SD rats showed that the level of TBARS in plasma increased significantly in the control group and that it was normal in the JTY group. The livers in the JTY group, compared to those in the control group, showed 36% halting effect of the normal level while their kidney's indicated the level significantly lower than the normal level. Heart's lipofuscin increased significantly in the control group, but was alike in both the JTY and the normal groups. Endurance of erythrocyte membrane (%) decreased significantly in the control group while it was kept at the similar level in both the JTY and the normal groups, indicating the halting effect. Conclusions : This study suggests that JTY is effective to defend oxidation stress caused by D-galactose in the animals. It showed that the anti-oxidant ability was maintained and strengthened. On the other hand, it reduced the level of peroxide in animals. In sum, JTY appeared to have the equilibrium normal physiological function in SD rat.

Quantitaties of flavonoids were determined by HPLC in three different ethanol extract (50, 70 and 95%, v/v) from Korean cherry. Antimicrobial activity of the ethanol extract and its solvent fraction (ethyl acetate and n-butanol fractions) against Staphylococcus aureus (ATCC 6538) and Staphylococcus epidermidis (ATCC 12228) was evaluated using MIC and disk test. Additionally, UV protective effect was also determined. The highest flavonoid quantity was found in 70% ethanol extract of Korean cherry. The quantities of quercitrin were 12.7 mg% and 2.1 mg% in ethyl acetate fraction and in buthanol fraction, respectively. In addition, the flavonoid in 70% (v/v) ethanol extract of Korean cherry included 14.9 mg% of quercitrin, 6.0 mg% of tannic acid and 5.2 mg% of catechin. Therefore, it was confirmed that main flavonoid of Korean cherry was quercitrin. For antimicrobial activity of ethanol extract of Korean cherry against Staphylococcus aureus (ATCC 6538) and Staphylococcus epidemidis (ATCC 12228), MIC and disk test using ethyl acetate fraction and butanol fraction were carried out. As a result, ethyl acetate fraction with higher quantity of flavonoid showed higher antimicrobial activity. Therefore, it was confirmed that the higher antimicrobial activity was brought about by flavonoid in Korean cherry. For UV protective effect of Korean cherry, ethanol extract showed the absorption zone of ultraviolet rays in 210∼380 nm including 290∼320 nm that is the wave length of UV-B harmful to the skin of human body. The pattern is the same as the absorption zone of 220∼300 nm in catechin and 210∼400 nm in quercitrin. Then, the functionalities of catechin and quercitrin were anticipated. It is expected that quercitrin in Korean cherry has the antimicrobial effect and UV protective effect related to the skin.

Recently our government operates earned value management system (EVMS) for improving work management system of the construction projects over specific scale. The EVMS is based on the integrated management between construction cost and schedule, the existing systems, however, are focused on the cost management by using wort quantity. This study suggests a work management mode1 to integrate construction cost with activity information. The model introduces a work task concept as a tool that can connect construction cost to activity information. The suggested model in this study is verified by using actual data for the applicability to practical construction projects.
Objectives : This study was carried out for the development of medicine for vitiligo treatment and focused on the effect of Psoraleae fructus extract on melanin synthesis of B16 melanoma cells. Methods : Activity of tyrosinase playing a vital role in the synthesis and quantity of melanin, which is the final product in cultured B16 melanoma cells, the effects of Psoraleae fructus extract were measured. Results : The results indicated that Psoraleae fructus extract increased beth the amount of melanin and the activity of tyrosinase according to concentration, also supported by western blot analysis. Conclusions : The results suggest that Psoraleae fructus extract has an advantageous effect on the promotion of melanin synthesis and will contribute to the development of vitiligo treatment through further related studies.

The plasma membrane and microsomes, isolated from the cells treated with hematoporphyrm derivative (HpD) for 1 and 24 h, accumulated the aggregated porphyrin. The quantity of aggregated porphyrin was same in the plasma membrane and microsomes after isolating them from cells treated with HpD for 1 h whereas the microsomes accumulated higher quantity of aggregated porphyrin when cells were treated with HpD for 24 h. Photodynamic action of aggregated porphyrin on plasma membrane and microsomes was investigated using lipid specific fluorescent probes: 1,6-diphenyl-1,3,5-hexatrine (DPH) and 1-(4-trimethylammonium), 6-diphenyl-1,3,5-hexatrine(TMA-DPH). The time dependent anisotropy of these probes in the membranes was measured and the decay of anisotropy was analyzed using wobbling in cone model. Upon irradiation both the plasma membrane and the microsomes showed an increase in the limiting anis~)tropy and order parameter and a decrease in the cone angle of the lipid probes. The increase in the limiting anisotropy was pronounced in membranes isolated from the cells treated with HpD for 24 h. Photoinduced change in the limiting anisotropy was dependent on the duration of incubation of cells with HpD before isolating the membranes. In both the membranes. the membrane core was affected more as compared to the outer leaflet. In addition to the structural changes, a decrease in Na$^+$-K$^+$-ATPase and NADPH cyt c reductase activity was also observed upon irradiation of HpD treated cells. Inhibition in NADPH cyt c reductase was more when cells were treated with HpD for 24 h, however, Na$^+$-K$^+$-ATPase activity did not depend on the duration of the treatment of cells with HpD before irradiation. Our results suggest that the extent of photoinduced perturbations in the membranes varies as a function of duration of the treatment of cells with HpD and the membrane core is more susceptible to the photodynamic action of aggregated porphyrin.
This study is to find out effects of hot water soluble extract from green tea, one of the Korean favorites, on the calcium metabolism and bone strength in body. To do so, calcium, phosphate, creatinine concentration and ALP activity in blood and the content of calcium and ash in the organ, the length, weight, strength in bone were measured. In addition, to find the calcium metabolism, the level of calcium intake, excretion, retention were measured. Twenty male Sprague-Dawley rats were divided into two groups and isoloated soy protein was provided as the source of protein and CaCO₃ was provided as the source of calcium. 0.5% hot water soluble extract from green tea was provided to the green tea groups and for the control group deionized water was provided. The results are as follows ; 1. There is no difference between the experimental groups in diet intake, weight gain, and the feed intake. 2. Feed efficiency ratio was low in the group which hot water soluble extract from green tea was provided. 3. There is no difference between groups the level of calcium, phosphorus, creatinine and ALP activity in serum. 4. There is no difference between groups weight, contents of ash and calcium in kidney and liver. 5. There is no difference between groups in calcium intake, absorption, excretion, and retention. 6. There is no difference between groups weight, length and strength in bone. In summary, when hot water soluble extract from green tea was provided with the amount of 150-200mg, which is taken when people generally drink as favorite tea, weight gain was reduced due to the decrease of feed efficiency ratio. However, it did not affect the availability of calcium in body at all. Thus, even if a big quantity of green tea powder or solid of hot green tea extract is not provided, the quantity obtained when people drink green tea lowers the feed efficiency ratio without reducing availability of calcium in body.
This study is to find out the antioxidative effect and serum lipid composition of wild grape seed powder in vivo. 20 white Sprague Dawley rats of six weeks old were divided into 2 groups and AIN-93 basic diet, high fat and cholesterol were provided. And they were examined to know how wild grape seed powder worked for antioxidative effect and serum lipid composition. For the comparing group, wild grape seed powder consisting 5% of the diet weight was provided and the quantity of protein, fat, carbohydrate, and cellulose was controlled following the analysis of the ingredients. The rats were fed for four weeks with experimental diet. Serum lipid and the antioxidant enzyme activity in blood and liver microsome were measured after 4 weeks of experiment. The results are as follows; There was no difference between the experimental groups in the initial body weight, final body weight, weight gain and FER. Food intake was higher in the group wild grape seed powder was provided than in the control group(p<0.05). Serum total cholesterol in the control group was significantly higher than that in the group wild grape seed powder was provided.(p<0.05). There was no difference serum HDL cholesterol and LDL cholesterol between the groups. Serum triglyceride showed no significant difference between the groups. In blood, glutanthione peroxidase activity was higher in the group supplemented with wild grape seed powder than in the control group. The glutathione reductase activity of blood showed no difference between the groups. In liver, the glutanthione peroxidase activity was higher in the group supplemented with wild grape seed powder than in the control group(p<0.05). Glutathione reductase activity in liver showed no difference in accordance with the supplementation of wild grape seed powder.

Purpose : The aim of this study was to compare the effects of the abdominal draw-in exercise (ADIE) and the dead bug exercise (DBE) on the pelvic anterior tilt angle and the activities of the gluteus maximus (GM), erector spinae (ES), and semi tendinosus (ST) during prone hip extension. Methods : A total of 22 female adults with weak abdominal muscles were divided into two groups: ADIE group (n=11) and DBE group (n=11). The muscle activities of the GM, ES, and ST along with the pelvic anterior tilt angle during prone hip extension were measured using a wireless surface electromyograph and gyro sensor before performing the prescribed exercise. Two groups conducted the assigned exercise for 10 minutes. After the exercise, their muscle activities and the pelvic anterior tilt angle were equally re measured. Results : In the DBE group, the muscle activity of GM was significantly increased after the intervention (p<0.05). However, there was no significant difference between the two groups in the amount of increase in the activity of GM (p>0.05). Moreover, in both groups, the activity of ES and the pelvic anterior tilt angle decreased significantly after the intervention (p<0.05) The decreased quantity in the pelvic anterior tilt angle and in the activity of ES showed no difference between the two groups (p>0.05). In the activity of ST, there was no significant difference within and between the two groups (p>0.05). Conclusion : Therefore, we suggest that ADIE and DBE are effective for women with weak abdominal muscles since the ES activity and pelvic anterior tilt angle are reduced during prone hip extension.
