자하거(紫何車) 약침액(藥針液)이 B16 melanoma 세포주의 멜라닌 합성에 미치는 영향 (The effects of Hominis Placenta extract on Melanin synthesis of B16 melanoma cells)

  • Objective : This research was carried out for the development of medicine for vitiligo treatment and focused on the effect of Hominis Placenta extract on Melanin synthesis of B16 melanoma cells. Methods : Acitivity of tyrosinase playing a vital role in synthesis of Melanin and the quantity of Melanin, which is the final product in cultured B16 melanoma cells, effects of Hominis Placenta extract were measured. Results: The results indicated that Hominis Placenta extract increased both the amount of Melanin and the activity of tyrosinase according to the concentration, and they also supported by western blot analysis. Conclusion : The results suggests that Hominis Placenta extract has an advantageous effect on the promotion of Melanin synthesis and will contribute to the development of vitiligo treatment through further related studies.

화산회토(火山灰土)의 특성(特性)에 관(關)하여 (Characteristics of Volcanic Ash Soils)

  • Volcanic Ash Soils are widely distributed in Jeju island, and constitute the important upland soils which are either presently being cultivated or are suitable for reclaiming. The characteristics of Volcanic Ash Soils according to data made available by previous studies in Jeju and the outside of the country are as following: The most conspicuous mineralogical property is the presence of amorphous mineral colloids. The colloids have large and highly reactive surface to which the common physical and chemical properties are related. Soils are low in bulk density and higher both in porosity and permeability. Accumulation of humus in the upper part of soil is found in great quantity. Cation exchange capacity is high mainly due to high humus content, but the absorbing intensity of ammonium and potassium is weaker than that of crystalline clays. The phosphate absorption coefficient is extremely high and deficiency of minor element may occur both crops and animals. Soils are densely populated with actinomycetes and anaerobic bacteria. Nitrification and activity of urease are distinctly stronger than that of non-Volcanic Ash Soils.

  • 일상생활 및 제반 활동을 유지시키는 중요한 사회 기반 시설은 상수도이다. 본 논문에서는 상수도 관리의 기본이 되는 유량의 계측과 분배, 관리를 위한 상수도 유수율 관리 시스템의 설계 및 구현 방법을 제안한다. 본 논문에서 제안하는 상수도 유수율 관리 시스템의 개발은 상수도 시설의 유지관리와 상수도 시스템의 효과적인 운영을 위한 데이터를 체계적으로 관리하여 양질의 수량 공급과 합리적인 용수 수요량의 예측 및 누수 방지를 위한 상수도 기반 시설의 설계 방향을 제시할 것으로 판단된다.

미생물을 이용한 미역폐기물의 저장 및 알긴산염 저분자화 (Storage of Waste-Brown Seaweed and Degradation of Alginate Using Microorganism)

  • We studied a storage of waste-brown seaweed at room temperature and degradation of alginate in seaweed by microorganism DS-02. The seaweeds, mixed with 5.0 wt% DS-02 and sealed in vinyl package without any other treatment, could be stored longer than 1 year without spoilage at room temperature. During the storage process, the alginate of seaweed was decomposed by enzyme of DS-02 and the molecular weight of alginate decreased to about 1/10 of initial quantity. DS-02 growed as fast as it had maximum weight after 24 hour culture and it's enzyme had a maximum activity of alginate degradation at $40^{\circ}C.$ The seaweed sample became particles in DS-02 culture solution and the M. W of alginate decreased to about 1/10 of initial value after 24 hour decomposition. The effect of alginate degradation with DS-02 was similar to that of degradation with 3.0 M HCI solution for 24 hour.

4.16kV 전동기 고정자 권선의 절연열화 특성 (Characteristics of Insulation Aging in 4.16kV Motor Stator Windings)

  • Nondestructive tests were performed on two 4.16kV motors. Epoxy-mica coupler(80pF) was connected to 4.16 kV motor stator windings. The voltage applied to the stator windings was 2.4 kV, 3.0kV, 3.5kV and 4.16kV, respectively. Digital partial discharge detector(PDD) and turbine generator analyzer(TGA) were used to measure partial discharge(PD) activity. TGA summarizes each plot with two quantities such as the normalized quantity number(NQN) and the peak PD magnitude(Qm). The PD levels in pC were measured with PDD. PD patterns of stator windings were indicated the internal discharge. PD patterns are consistent with the result of measurement using PDD and TGA.

Resolution of L-Carnitine from DL-Carnitine by Resting Cells of the Enterobacter sp. NH-104

  • For the resolution of L-carnitine from DL-carnitine, resting cells of Enterobacter sp. NH-104, which had a higher capacity of D-carnitine decomposition, were harvested at maximal specific activity of D-carnitine decomposition of 47.05 unit/mg cell. The cells were frozen at $-80^{\circ}C$ to assess functions as enzyme sources. Optimal concentration of cells and DL-carnitine were 17 g/$\ell \; and \; 20 g/\ell$, respectively, and reaction buffer was best at 75 mM of Tris. HCl. Optimal temperature and pH were $36^{\circ}C$ and 8.2, respectively. When the reaction at optimal conditions was carried out for 14 h, the optical purity was 98.21 %, and the quantity and yield of remaining L-carnitine were 4.432 g/$\ell$ and 44.32%, respectively.

  • The lambda Integrase (Int) carries out site-specific recombination between the two partner DNA sequences, attachment P (attP) and attachment B (attB). In order to study the recombination mechanism, a large quantity of pure integrase is required. Then, we constructed an int gene inserted recombinant plasmid (pNYL3) by using the pQE31 HIS-Tag vector, and produced the fusion protein, 6xHIS-Int from the E. coli TG1 strain carrying the pNYL3 plasmid. The recombinant protein produced was purified by phosphocellulose and Ni$^{++}$-NTA affinity column chromatographies. The result of the in vitro recombination assay using the standard reaction mixture containing 6xHIS-Int and partially purified integration host factor (IHF) showed that the 6xHIS-Int tagged recombination Integrase had the full recombination activity.

Synthesis and Biological Activity of Fungal Metabolite, 4-Hydroxy-3-(3'-Methyl-2'-Butenyl)-Benzoic Acid

  • 4-Hydroxy-3-(3'-methyl-2'-butenyl)-benzoic acid (HMBA) was previously isolated from Curvularia sp. KF119 as a cell-cycle inhibitor. However, the present study used a novel and practical synthetic method to prepare a large quantity of HMBA. The synthetic HMBA was found to inhibit the cell-cycle progression of HeLa cells with a comparable potency to the natural fungal metabolite. The inhibition of the cell-cycle progression by the synthetic HMBA involved both the activation of $p21^{WAF1}$ and the inhibition of cyclin D1 expression in the cells. Consequently, this new synthetic procedure provides an easy and convenient way to produce or manipulate the original fungal metabolite.

  • Media company is not a business which sells news and information but a company sells audience's attention. Advertisers estimate audience's quantity and quality which pay attention to media and pay the cost. Thus drawing audience's attention is a natural and instinctive behavior for survival of media company. News doesn't deliver the fact just as it. That's impossible. News is a commodity made by processing and selection of the media company and journalist. On the process, judge of values is involved and limitation of time and place of media is considered. If scientists understand media's character truly, their misunderstanding about media company and journalist may be cleared up. In this society, media is not a being to ignore, particularly for big science like space science which spends huge public capital. Nowadays, space science meets the time to take the leap in Korea. However that can be crisis cause of uncertainty of science activity. When the crisis which no one desires happens, preparation needs for new opportunity. We can take the crisis as a chance. Understanding about media and public will be the first step for this preparation.

양수발전소 발전-전동기 운전중 감시 시스템의 개발 (Development of On-Line Monitoring System for Pumped Storage Generator/Motor)

  • On-line monitoring system(OMS) has been developed for the pumped storage generator/motor The OMS is applied to diagnosis of the PD(partial discharge) activity of stator insulation, the shorted-turn of rotor winding and the variation of the air-gap between stator and rotor. The OMS consists of DAS(data acquisition system), main server system, gateway and display PC. The DAS measures the PD, the shorted-turn and air-gap from three sensors installed on the generator/motor. The gateway controls the data which sent by DAS. The main server system saves the data, analyzes the data and conducts the diagnostic algorithm. The display PC shows the diagnostic results of partial discharge, shorted-turn and air-gap. Field tests were conducted using PDA(partial discharge analyzer). The results of the OMS and PDA measurements can be directly correlated with normalized quantity number(NQN), PD magnitude(Qm) and PD pattern.
