홍삼은 우리나라에서 오랜 역사동안 여러 질병을 치료하는데 사용되었다. 본 연구에서는 홍삼 에탄올 추출물에 감마선을 1~5kGy 범위에서 조사를 하는 새로운 기법을 개발하고자 한다. 감마선 조사된 홍삼 추출물(IHRG)의 진세노사이드의 조성변화를 관찰하기 위해서 HPLC 분석을 이용하였다. 그 결과, 홍삼 에탄올 추출물에 감마선 1 kGy와 5 kGy를 조사한 처리군에서 진세노사이드의 조성에 변화는 관찰되지 않았다. MTT 분석법을 이용하여 사람의 전립선암세포주인 PC-3세포에서의 IHRG의 세포독성을 살펴 본 결과, 감마선을 처리하지 않은 홍삼추출액(HRG) 보다 더 높은 세포독성을 보였다. $LD_{50}$ 농도가 IHRG-1(1 kGy)에서는 $30{\mu}g/mL$, IHRG-5 (5 kGy)에서는 $15{\mu}g/mL$로 나타났다. 이러한 세포독성이 Annexin V/PI 분석 및 핵의 염색법인 DAPI 염색을 통하여 IHRG를 처리한 군들에서 전형적인 apoptosis를 관찰할 수 있었다. 또한, 산화적 스트레스(ROS)의 유발이 IHRG 처리군에서 나타났다. BALB/c 마우스에 암세포를 이식시킨 모델에서 IHRG에 의한 암세포 증식억제 효과를 살펴 본 결과, 암세포 증식 억제율이 HRG에서 21.1%인 반면에, IHRG-1에서 56.9%, IHRG-5에서 76.1%로 나타났다. 이들 결과들 통해, HRG에 어떤 생리활성 물질이나 성분들이 감마선 조사에 의해 항암효과를 증대시킨 것으로 사료된다.
본 연구에서는 제주도에서 자생하는 37 종의 식물추출물에 대한 미백 및 항산화 효능을 조사하였다. 미백 효능은 in vitro tyrosinase 저해활성과 B16-F1 세포를 이용한 멜라닌 합성 저해효과를 조사하여 확인하였고, 항산화 효능은 DPPH (1,1-diphenyl-2-picrylhydrazyl)를 이용한 전자공여능 측정방법과 DCF-DA (dichlorofluorescin diacetate)를 이용한 V79-4 폐 섬유아세포에서 활성 산소종 소거작용 능력을 조사하여 측정하였다. 세포실험 조건에서 식물추출물들의 투여농도에 따른 세포독성 정도를 조사하기 위하여 MTT (3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl-2H-tetrazolium bromide) assay를 수행하였다. 미백 효능조사 실험에서 우수한 결과를 나타내었던 8종의 식물 추출물들은 토끼를 이용한 국소 독성 시험 및 인체 피부 자극 임상 시험에 적용하여 안전성을 조사하였다. 연구결과에서 높은 미백과 항산화 효능을 동시에 나타내었던 비수리(Lespedeza cuneata), 제주광나무(Ligustrum lucidum (stem)), 산뽕나무(Morus bombycis (stem)), 꿀풀(Prunella vulgaris var. lilacina)의 식물추출물들은 화장품 원료화에 매우 유리한 조건을 갖고 있음을 확인하였다.
Objectives : Polycyclic aromatic hydrocarbons (PAH) and toluene have been reported to induce reactive oxygen species and oxidative stress. This study was performed to investigate the effects of low level exposure to PAHs or toluene on the lipid peroxidation level in elementary school children and the elderly in a rural area. Methods : Forty seven elementary school children and 40 elderly people who were living in a rural area and not occupationally exposed to PAH or toluene were the subjects of this study. Information about active or passive smoking and diet was obtained using a self-administered questionnaire. The urinary 1-hydroxypyrene (1-OHP), 2-naphthol, hippuric acid and thiobarbituric acid reactive substance (TBARS) concentrations were measured, and these values were corrected with the urinary creatinine concentration. Results : In school children, the geometric means of the urinary 1-OHP, 2-naphthol, hippuric acid and TBARS levels were $0.02\;{\mu}mol/mol$ creatinine, $0.47\;{\mu}mol/mol$ creatinine, 0.14 g/g creatinine and $0.95\;{\mu}mol/g$ creatinine, respectively. Those values for the elderly were $0.07{\mu}mol/mol$ creatinine, $1.87{\mu}mol/mol$ creatinine, 0.11 g/g creatinine and $1.18\;{\mu}mol/g$ creatinine, respectively. The mean levels of urinary 1-OHP, 2-naphthol and TBARS were significantly higher in the elderly subjects than in the children. The urinary TBARS level was not correlated with the urinary 1-OHP, 2-naphthol and hippuric acid, but they were correlated with the age of the subjects. Conclusions : These results suggest that low level inhalation exposure to PAH or toluene does not markedly increase lipid peroxidation, and age is a significant determinant of lipid peroxidation.

Parkinson's disease (PD) is a neurodegenerative disability caused by a decrease of dopaminergic neurons in the substantia nigra (SN). Although the etiology of PD is not clear, oxidative stress is believed to lead to PD. Catalase is antioxidant enzyme which plays an active role in cells as a reactive oxygen species (ROS) scavenger. Thus, we investigated whether PEP-1-Catalase protects against 1-methyl-4-phenylpyridinium (MPP+) induced SH-SY5Y neuronal cell death and in a 1-methyl-4-phenyl-1,2,3,6-trtrahydropyridine (MPTP) induced PD animal model. PEP-1-Catalase transduced into SH-SY5Y cells significantly protecting them against MPP+-induced death by decreasing ROS and regulating cellular survival signals including Akt, Bax, Bcl-2, and p38. Immunohistochemical analysis showed that transduced PEP-1-Catalase markedly protected against neuronal cell death in the SN in the PD animal model. Our results indicate that PEP-1-Catalase may have potential as a therapeutic agent for PD and other oxidative stress related diseases. [BMB Reports 2015; 48(7): 395-400]
Coptis japonica (C. japonica) is a perennial medicinal plant that has anti-inflammatory activity. C. japonica contains numerous biologically active alkaloids including berberine, palmatine, epi-berberine, and coptisine. The most well-known anti-inflammatory principal in C. japonica is berberine. For example, berberine has been implicated in the inhibition of iNOS induction by cytokines in microglial cells. However, the efficacies of other alkaloids components on microglial activation were not investigated yet. In this study, we investigated the effects of three alkaloids (palmatine, epi-berberine and coptisine) from C. japonica on lipopolysaccharide (LPS)-induced microglial activation. BV2 microglial cells were immunostimulated with LPS and then the production of several inflammatory mediators such as nitric oxide (NO), reactive oxygen species (ROS) and matrix metalloproteinase-9 (MMP-9) were examined as well as the phosphorylation status of Erk1/2 mitogen activated protein kinase (MAPK). Palmatine and to a lesser extent epi-berberine and coptisine, significantly reduced the release of NO, which was mediated by the inhibition of LPS-stimulated mRNA and protein induction of inducible nitric oxide synthase (iNOS) from BV2 microglia. In addition to NO, palmatine inhibited MMP-9 enzymatic activity and mRNA induction by LPS. Palmatine also inhibited the increase in the LPS-induced MMP-9 promoter activity determined by MMP-9 promoter luciferase reporter assay. LPS stimulation increased Erk1/2 phosphorylation in BV2 cells and these alkaloids inhibited the LPS-induced phosphorylation of Erk1/2. The anti-inflammatory effect of palmatine in LPS-stimulated microglia may suggest the potential use of the alkaloids in the modulation of neuroinflammatory responses, which might be important in the pathophysiological events of several neurological diseases including Alzheimer's disease (AD), multiple sclerosis (MS), Parkinson's disease (PD) and stroke.
Malus melliana (Hand.-Mazz.) Rehder (M. melliana)는 장미과에 속하는 중국 자생 식물 중 하나로 현재까지 보고된 생리활성은 전무하다. 본 연구에서는 M. melliana 에탄올 추출물(MMEE)의 항산화 및 항염증 생리활성을 DPPH 라디칼 소거능, ROS 소거능, NO 생성 저해능 및 Western blot hybridization을 통한 연관 단백질 발현분석을 통해 평가하였다. MMEE의 항산화능을 DPPH 라디칼 소거능을 통해 분석한 결과 양성 대조군으로 사용한 대표적인 항산화제인 아스코르빈산과 유사한 정도의 높은 소거활성을 보여 MMEE가 매우 강한 항산화능을 보유함을 확인하였다. 또한 RAW 264.7 세포주에서 $H_2O_2$에 의해 유도된 ROS에 대한 MMEE의 소거능을 분석한 결과, 농도의존적인 강한 ROS 소거능을 보였다. 뿐만 아니라 대표적인 항산화 효소인 HO-1 및 그 전사 인자인 Nrf2의 단백질 발현에 미치는 영향을 분석한 결과 MMEE에 의해 HO-1 및 Nrf2의 발현이 증가됨을 보였다. 한편 MMEE가 LPS에 의해 유도된 NO 생성에 미치는 영향을 분석한 결과 농도의존적인 NO 생성 저해능을 보였으며 이는 NO 생성 단백질인 iNOS의 발현 저해에서 기인함을 확인하였다. 이러한 결과를 통해 MMEE의 높은 항산화능과 항염증 활성을 확인하였으며 향후 잠재적인 기능성 소재로서 유용하게 활용될 수 있을 것으로 판단된다. 추후 계속적인 연구를 통해 활성 물질의 규명이 필요할 것으로 판단된다.
본 연구는 꼭두서니의 뿌리인 천초근의 물 추출물과 에탄올 추출물을 이용하여 대장암 세포에 대한 암세포 성장 억제 및 사멸효과가 있는지 알아보고자 수행하였다. ERA(천초근 에탄올 추출물)은 폴리페놀($45.77{\pm}2.03mg/g$)과 플라보노이드($22.82{\pm}1.33mg/g$)를 함유하고 있었으며, $H_2O_2$에 의해 증가된 ROS(reactive oxygen species)를 억제하는 효과를 나타냈지만 WRA(천초근 물 추출물)은 효과가 없었다. 또한 ERA는 $500{\mu}g/m{\ell}$의 농도로 대장암 세포주(HCT-116)에 처리했을 때 세포사멸을 유도할 뿐만 아니라 caspase-3 단백질 활성화, DNA fragmentation 및 apoptotic cell death를 일으키는 것으로 확인되었다. 이는 ERA가 HCT-116 세포주에 대해 apoptosis(세포자멸사)를 통해 항암효과를 나타내는 것으로 생각되지만 다른 연구결과들과 비교하였을 때 농도 대비 효능이 미미하다. 따라서 천초근 에탄올 추출물에 대장암 세포의 성장을 억제하는 유효성분을 분석하여 그 효능을 탐색하는 추가실험이 필요할 것으로 생각된다.
In this paper, AgCl/Ag3PO4/diatomite photocatalyst is successfully synthesized by microemulsion method and anion in situ substitution method. X-ray diffraction (XRD), photoelectron spectroscopy (XPS), scanning electron microscopy (SEM), energy dispersive spectroscopy (EDS), transmission electron microscopy (TEM), and ultraviolet-visible spectroscopy (UV-Vis) are used to study the structural and physicochemical characteristics of the AgCl/Ag3PO4/diatomite composite. Using rhodamine B (RhB) as a simulated pollutant, the photocatalytic activity and stability of the AgCl/Ag3PO4/diatomite composite under visible light are evaluated. In the AgCl/Ag3PO4/diatomite visible light system, RhB is nearly 100 % degraded within 15 minutes. And, after five cycles of operation, the photocatalytic activity of AgCl/Ag3PO4/diatomite remains at 95 % of the original level, much higher than that of pure Ag3PO4 (40 %). In addition, the mechanism of enhanced catalytic performance is discussed. The high photocatalytic performance of AgCl/Ag3PO4/diatomite composites can be attributed to the synergistic effect of Ag3PO4, diatomite and AgCl nanoparticles. Free radical trapping experiments are used to show that holes and oxygen are the main active species. This material can quickly react with dye molecules adsorbed on the surface of diatomite to degrade RhB dye to CO2 and H2O. Even more remarkably, AgCl/Ag3PO4/diatomite can maintain above 95 % photo-degradation activity after five cycles.
우르솔산(Ursolic acid)은 다양한 약재, 과일 그리고 야채 등으로 부터 분리되는 식물성 생리활성물질로서, 천연 항산화제로 널리 알려져 있지만, 배아줄기세포에서 우르솔산의 기능에 대한 연구는 아직까지 잘 이해되지 않고 있다. 본 연구에서는 저산소 상태에서 우르솔산이 배아줄기세포의 성장에 미치는 효과에 대해 조사하였다. 48시간 동안의 저산소 환경은 배아줄기세포의 미분화상태 및 전능성을 유지에 있어서 영향을 미치지 않았지만, 세포 내 활성산소종의 지속적인 생산과 이로 인한 lactate dehydrogenase 활성을 촉진 시켰다. 저산소에 의해 유도된 배아줄기세포의 산화적 스트레스는 $30{\mu}M$의 우르솔산의 처리로 인해 유의적으로 감소되었으며, 이러한 우르솔산의 활성산소 소거능력은 항산화제인 N-acetyl-cysteine (NAC)의 효과와 유사하였다. 저산소 환경은 또한 유의적으로 배아줄기세포의 생존과 증식을 감소 시킬 뿐만 아니라, 세포의 자살 및 노화를 유도함이 관찰되었지만, 강한 항산화 능력을 지닌 우르솔산은 세포를 저산소로부터 보호하여 정상수준으로 세포의 생존과 증식을 유지시키고, 세포 자살 관련 단백질(cleaved caspase-3, Bcl-2, 그리고 cIAP) 및 세포 노화 관련 단백질(beta-galactosidase)을 조절하는 탁월한 약리학적 효과를 가지고 있었다. 따라서 본 연구결과를 통해, 우르솔산은 강력한 천연 항산화제이며, 저산소에 의해 유도된 유해 기작을 제어함으로서, 배아줄기세포의 전능성을 유지시킬 수 있는 기능성 물질이라는 것을 알 수 있었다.
Background: T-cell acute lymphoblastic leukemia (T-ALL) is a kind of aggressive hematological cancer, and the PI3K/Akt/mTOR signaling pathway is activated in most patients with T-ALL and responsible for poor prognosis. 20(S)-Ginsenoside Rh2 (20(S)-GRh2) is a major active compound extracted from ginseng, which exhibits anti-cancer effects. However, the underlying anticancer mechanisms of 20(S)-GRh2 targeting the PI3K/Akt/mTOR pathway in T-ALL have not been explored. Methods: Cell growth and cell cycle were determined to investigate the effect of 20(S)-GRh2 on ALL cells. PI3K/Akt/mTOR pathway-related proteins were detected in 20(S)-GRh2-treated Jurkat cells by immunoblotting. Antitumor effect of 20(S)-GRh2 against T-ALL was investigated in xenograft mice. The mechanisms of 20(S)-GRh2 against T-ALL were examined by cell proliferation, apoptosis, and autophagy. Results: In the present study, the results showed that 20(S)-GRh2 decreased cell growth and arrested cell cycle at the G1 phase in ALL cells. 20(S)-GRh2 induced apoptosis through enhancing reactive oxygen species generation and upregulating apoptosis-related proteins. 20(S)-GRh2 significantly elevated the levels of pEGFP-LC3 and autophagy-related proteins in Jurkat cells. Furthermore, the PI3K/Akt/mTOR signaling pathway was effectively blocked by 20(S)-GRh2. 20(S)-GRh2 suppressed cell proliferation and promoted apoptosis and autophagy by suppressing the PI3K/Akt/mTOR pathway in Jurkat cells. Finally, 20(S)-GRh2 alleviated symptoms of leukemia and reduced the number of white blood cells and CD3 staining in the spleen of xenograft mice, indicating antitumor effects against T-ALL in vivo. Conclusion: These findings indicate that 20(S)-GRh2 exhibits beneficial effects against T-ALL through the PI3K/Akt/mTOR pathway and could be a natural product of novel target for T-ALL therapy.

② 상기 이용제한 규정에 따라 서비스를 이용하는 회원에게 서비스 이용에 대하여 별도 공지 없이 서비스 이용의
일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.
[부 칙]
1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
