INFLUENCE OF DIRECT-FED MICROBIALS ON RUMINAL MICROBIAL FERMENTATION AND PERFORMANCE OF RUMINANTS: A REVIEW

  • Direct-fed microbials (DFM) have been used to enhance milk production in lactating cattle and to increase feed efficiency and body weight gain in growing ruminants. Primary microorganisms that have been used as DFM for ruminants are fungal cultures including Aspergillus oryzae and Saccharomyces cerevisiae and lactic acid bacteria such as Lactobacillus or Streptococcus. Attempts have been made to determine the basic mechanisms describing beneficial effects of DFM supplements. Various modes of action for DFM have been suggested including : stimulation of ruminal microbial growth, stabilization of ruminal pH, changes in ruminal microbial fermentation pattern, increases in digestibility of nutrients ingested, greater nutrient flow to the small intestine, greater nutrient retention and alleviation of stress, however, these responses have not been observed consistently. Variations in microbial supplements, dosage level, production level and age of the animal, diet and environmental condition or various combinations of the above may partially explain the inconsistencies in response. This review summarizes production responses that have been observed under various conditions with supplemental DFM and also corresponding modification of ruminal fermentation and other changes in the gastrointestinal tract of ruminant animals.

Composite action of hollow concrete-filled circular steel tubular stub columns

  • To better understand the influence of hollow ratio on the hollow concrete-filled circular steel tubular (H-CFT) stub columns under axial compression and to propose the design formula of ultimate bearing capacity for H-CFT stub columns, 3D finite element analysis and laboratory experiments were completed to obtain the load-deformation curves and the failure modes of H-CFT stub columns. The changes of the confinement effect between core concrete and steel tube with different hollow ratios were discussed based on the finite element results. The result shows that the axial stress of concrete and hoop stress of steel tube in H-CFT stub columns are decreased with the increase of hollow ratio. AfteGr the yield of steel, the reduction rate of longitudinal stress and the increase rate of circumferential stress for the steel tube slowed down. The confinement effect from steel tube on concrete also weakened slowly with the increase of hollow ratio. Based on the limit equilibrium method, a simplified formula of ultimate bearing capacity for the axially loaded H-CFT stub columns was proposed. The predicted results showed satisfactory agreement with the experimental and numerical results.

Effect of Low Doses of Genistein and Equol on Protein Expression Profile in MCF-7 Cells

  • Although action modes of equol and genistein have been extensively studied, their precise roles in tumor cells remain elusive. To address possible effects of these compounds on protein expression in mammary tumor cells, proteins modulated in MCF-7 mammary tumor cells when incubated in absence and presence of 10 uM equol or genistein were identified through 2-dimensional gel electrophoresis, MALDI-TOF MS/MS, and NCBInr database search using Mascot software. Most proteins differentially expressed in MCF-7 cells after treatment with 10 uM genistein or equol were identified as being the same. Exposure to both compounds caused decreased cellular expression of RNA-binding protein regulatory subunit and oncogene DJ1 tubulin beta-1 chain, and increased expression of heterogeneous ribonucleoproteins F and L, KH-type splicing regulatory protein, and translation elongation factor EF-Tu precursor. Genistein and equol at dose used in this study showed common action mechanism.

U-플랜지 트러스 복합보의 휨 내력에 대한 실험 연구 (Experimental Study on the Flexural Capacity of the U-Flanged Truss Hybrid Beam)

  • U-flanged truss beam is composed of u-shaped upper steel flange, lower steel plate of 8mm or more thickness, and connecting lattice bars welded on the upper and lower sides. The hybrid beam with U-flanged steel truss is made in the construction site through pouring the concrete, and designated as U-flanged truss hybrid beam. In this study the structural experiments on the 4 hybrid beams with the proposed basic shapes were performed, and the flexural capacities from the tests were compared with those from the theoretical approach. The failure modes of each specimen were quite similar. The peak load was reached with the ductile behavior after yielding, and the failure occurred through the concrete crushing. The considerable increasement of deformation was observed up to the concrete crushing. The composite action of concrete and steel member was considered to be reliable from the behavior of specimens. The flexural strength of hybrid beam has been evaluated exactly using the calculation method applied in the boubly reinforced concrete beam. The placement of additional rebars in the bottom instead of upper side is proposed for the efficient design of U-flanged truss hybrid beam.

Theoretical and experimental study on shear strength of precast steel reinforced concrete beam

  • With the aim to put forward the analytical model for calculating the shear capacity of precast steel reinforced concrete (PSRC) beams, a static test on two full-scale PSRC specimens was conducted under four-point loading, and the failure modes and strain developments of the specimens were critically investigated. Based on the test results, a modified truss-arch model was proposed to analyze the shear mechanisms of PSRC and cast-in-place SRC beams. In the proposed model, the overall shear capacity of PSRC and cast-in-place SRC beams can be obtained by combining the shear capacity of encased steel shape with web concrete determined by modified Nakamura and Narita model and the shear capacity of reinforced concrete part determined by compatible truss-arch model which can consider both the contributions of concrete and stirrups to shear capacity in the truss action as well as the contribution of arch action through compatibility of deformation. Finally, the proposed model is compared with other models from JGJ 138 and AISC 360 using the available SRC beam test data consisting of 75 shear-critical PSRC and SRC beams. The results indicate that the proposed model can improve the accuracy of shear capacity predictions for shear-critical PSRC and cast-in-place SRC beams, and relatively conservative results can be obtained by the models from JGJ 138 and AISC 360.

p38 MAPK Inhibitor NJK14047 Suppresses CDNB-Induced Atopic Dermatitis-Like Symptoms in BALB/c Mice

  • Atopic dermatitis (AD) is a chronic inflammatory skin disorder. Suppression of MAPKs and NF-κB is implicated as a vital mechanism of action of several traditional Chinese medicines for AD therapy. Although overexpression of MAPK mRNA in the skin tissue has been shown in the AD model, the roles of each MAPK in AD pathogenesis have rarely been studied. This study examined the effect of NJK14047, an inhibitor of p38 MAPKs, on AD-like skin lesions induced in BALB/c mice by sensitization and challenges with 1-chloro-2,4-dinitrobenzene (CDNB) on dorsal skin and ears, respectively. After induction of AD, NJK14047 (2.5 mg/kg) or dexamethasone (10 mg/kg) was administrated for 3 weeks via intraperitoneal injection. Following its administration, NJK14047 suppressed CDNB-induced AD-like symptoms such as skin hypertrophy and suppressed mast cell infiltration into the skin lesions. It also reduced CDNB-induced increase in TH2 cytokine (IL-13) and TH1 cytokines (interferon-γ and IL-12A) levels but did not decrease serum IgE level. Furthermore, NJK14047 blocked CDNB-induced lymph node enlargement. These results suggest that NJK14047, a p38 MAPK inhibitor, might be an optimal therapeutic option with unique modes of action for AD treatment.

Systems pharmacology approaches in herbal medicine research: a brief review

  • Herbal medicine, a multi-component treatment, has been extensively practiced for treating various symptoms and diseases. However, its molecular mechanism of action on the human body is unknown, which impedes the development and application of herbal medicine. To address this, recent studies are increasingly adopting systems pharmacology, which interprets pharmacological effects of drugs from consequences of the interaction networks that drugs might have. Most conventional network-based approaches collect associations of herb-compound, compound-target, and target-disease from individual databases, respectively, and construct an integrated network of herb-compound-target-disease to study the complex mechanisms underlying herbal treatment. More recently, rapid advances in high-throughput omics technology have led numerous studies to exploring gene expression profiles induced by herbal treatments to elicit information on direct associations between herbs and genes at the genome-wide scale. In this review, we summarize key databases and computational methods utilized in systems pharmacology for studying herbal medicine. We also highlight recent studies that identify modes of action or novel indications of herbal medicine by harnessing drug-induced transcriptome data.

Finite element modelling of the shear behaviour of profiled composite walls incorporating steel-concrete interaction

  • The novel form of composite walling system consists of two skins of profiled steel sheeting with an in-fill of concrete. The behaviour of such walling under in-plane shear is important in order to utilise this system as shear elements in a steel framed building. Steel sheet-concrete interface governs composite action, overall behaviour and failure modes of such walls. This paper describes the finite element (FE) modelling of the shear behaviour of walls with particular emphasis on the simulation of steel-concrete interface. The modelling of complex non-linear steel-concrete interaction in composite walls is conducted by using different FE models. Four FE models are developed and characterized by their approaches to simulate steel-concrete interface behaviour allowing either full or partial composite action. Non-linear interface or joint elements are introduced between steel and concrete to simulate partial composite action that allows steel-concrete in-plane slip or out of plane separation. The properties of such interface/joint elements are optimised through extensive parametric FE analysis using experimental results to achieve reliable and accurate simulation of actual steel-concrete interaction in a wall. The performance of developed FE models is validated through small-scale model tests. FE models are found to simulate strength, stiffness and strain characteristics reasonably well. The performance of a model with joint elements connecting steel and concrete layers is found better than full composite (without interface or joint elements) and other models with interface elements. The proposed FE model can be used to simulate the shear behaviour of composite walls in practical situation.

  • The purpose of this study is to arrange the design elements and methods for sustainable community. After made the concept of sustainable community clear in the contemporary architecture and urban design, this study examines rural ecovillage, urban demonstration project, co-housing, new urbanism and urban village, and ecological township as the application modes of sustainable community. Through the cases of the application mode, the design elements for sustainable community were summarized as the followings: the symbiosis system with the nature, permaculture and edible landscape, identity, the technology for saving of resources and energy, the traffic system for the reduction of private motor use, mixed use development, and the housing with diversity. And the design methods for sustainable community were summarized as the followings: strategic planning, visioning, community action planning, workshops and charrette, environmental evaluation, and participation techniques.

The Role of Bronchodilators in Preventing Exacerbations of Chronic Obstructive Pulmonary Disease

  • Bronchodilators are the cornerstone of symptomatic chronic obstructive pulmonary disease (COPD) treatment. They are routinely recommended for symptom reduction, with a preference of long-acting over short-acting drugs. Bronchodilators are classified into two classes based on distinct modes of action, i.e., long-acting antimuscarinics (LAMA, once-daily and twice-daily), and long-acting ${\beta}2$-agonists (LABA, once-daily and twice-daily). In contrast to asthma management, evidence supports the efficacy of both classes of long-acting bronchodilators as monotherapy in preventing COPD exacerbations, with greater efficacy of LAMA drugs versus LABAs. Several novel LAMA/LABA fixed dose combination inhalers are currently approved for COPD maintenance treatment. These agents show superior symptom control to monotherapies, and some of these combinations have also demonstrated superior efficacy in exacerbation prevention versus monotherapies, or combinations of inhaled corticosteroids plus LABA. This review summarizes the current data on clinical effectiveness of bronchodilators alone or in combination to prevent exacerbations of COPD.
