Enzymatic Characteristics and Applications of Microbial Chitin Deacetylases (미생물 Chitin Deacetylase의 특성과 응용)

  • Chitin deacetylase (CDA; EC 3.5.1.41) catalyzes the hydrolysis of N-acetamide bonds of chitin, converting it to chitosan. Chitosan has several applications in areas such as biomedicine, food ingredients, cosmetics, pharmaceuticals, and agriculture. In this paper, occurrence, assay and purification protocols, enzymatic characteristics, substrate specificity, and mode of action of microbial CDAs have been described. Several lines of evidence have substantiated the biological roles involved in cell wall formation and plant-pathogen interactions for fungal CDAs. The gene structure of CDAs has been compared with other family 4 carbohydrate esterases which deacetylate a wide variety of acetylated poly/oligo-saccharides. The use of CDAs for the conversion of chitin to chitosan, in contrast to the presently used chemical procedure, offers the possibility of a controlled, non-degradable process, resulting in the production of well-defined chitosan oligomers and polymers. Insect pathogen that can secrete high levels of chitin-metab­olizing enzymes including CDA can be a possible alternative for new pest management tools.

Antibiotic Production of Pseudomonas otitidis PS and Mode of Action (Pseudomonas otitidis PS 균주의 항생물질 생산과 작용 기작)

  • An isolate capable of inhibiting the growth of gram-positive bacteria was obtained from the soil of Mushim stream, Cheongju. The isolate was identified as Pseudomonas otitidis PS by 16S rRNA gene sequence analysis. P. otitidis PS produced antibiotics as a secondary metabolite when cultured in 1% soybean meal with 0.5% glucose. The maximum yield was about 0.1%. The antibiotic substance of P. otitidis PS extracted using ethyl acetate displayed a minimum inhibitory concentration of $2{\mu}g/ml$ for Staphylococcus aureus KCTC 1261. The antibiotic substance produced an orange halo on chrome azurol S agar due to siderophore activity. Growth inhibition was decreased when the iron was depleted. Since the antibiotic activity was lost upon the addition of the reducing agent ascorbic acid or during anaerobic culture, it was considered that antibiotic of P. otitidis PS strain exerts its bactericidal effect by the generation of reactive oxygen species.

Inhibitory Effect of Nisin upon Kimchi Fermentation (김치발효에 대한 Nisin의 저해효과)

  • To examine the inhibitory effect of nisin on Kimchi, comparison of the fermentation pattern was studied between the Kimchi added nisin (100 WIg) and the control Kimchi at 15cC. The Kimchi added nisin was showed pH 4.03 at 7 days fermentation, on the other hand the control Kimchi showing pH 4.04 at 5 days. And total acidity (lactic acid%) showed 0.47,0.69,0.88 at 5,9, 14 days fermentation compared with the control Kimchi showing 0.57,0.93, 1.13 respectively. Maximum growth of lactic acid bacteria on the control Kimchi indicated 1.8, 1.6 x 108 CFU/ml at 7 days but on presence of nisin indicated 9.5,6.4 x 107 CFU/ml at 5 days. In conclusion, nisin showed inhibitory action to Kimchi fermentation from pH, total activity and lactic acid bacteria count results.

  • Streptomyces griseorubiginosus var. soyoensis previously identified, produced two kinds of antifungal antibiotics, trans-cinnamamide and another new substance. The latter was identified to be a new substance of tetraene family by establishment of UV, IR, NMR, mass spectra and chemical reactions and rotatively named as Tetraene KM-A. Through an antimicrobial activity test using serial agar dilution method, Tetraene KM-A showed strong growth inhibitory activity against fungi and yeasts, but not against procaryotes tested. The inhibitory action of Tetraene KM-A on fungi was remarkably ineffective when some of sterols were added to the cultural media. $LD_{50}$ of the Tetrene KM-A to mice and rats by intravenous injection were 84.3 and 90.4 mg/kg respectively. $LD_{50}$ to mice by oral feeding was 1503mg/kg.

The Production of Cyclodextrin Glucanotransferase by Bacillus sp. and Its Utilization (Bacillus sp. 의 Cyclodextrin Glucanotransferase 생산 및 이용에 관한 연구)

  • A highly cyclodextrin glucanotransferase producing strain of Bacillus sp. was isolated from soil, and basic studies on the characteristics of the strain and its enzyme, conditions for the enzyme production, and the enzyme utilization were carried out. The isolated strain was aerobic, motile, endospore-forming and rod-shaped bacterium. Optimum pH and temperature for the enzyme action were 6.0 and 45$^{\circ}C$, and the enzyme was stable within 5$0^{\circ}C$, and between pH 6.0 and 10.0. The highest yield of the enzyme was obtained using the medium containing 2% corn starch as a carbon source, and 5% corn steep liquor, 0.1% urea and 0.25% ammonium sulfate as nitrogen sources. The fermentation conditions for the enzyme production in a jar fermentor were cetermined to be 3$0^{\circ}C$, 200rpm, 0.6vvm and 60hr cultural period. Stevioside transglycosylation catalyzed by this enzyme was identified by high performance liquid chromatography.

Properties of Lipases and Palm Oil Assimilating Patterns in Palm Oil Fermentation (팜유발효에 있어서 리파제의 특성과 팜유자화와의 관계)

  • In order to elucidate the patterns of natural oils and fats assimilation by microorganisms, lipases properties of yeast and bacterium strain, Torulopsis candia Y-128 and Acinetobacter calcoaceticus KB-2, which could assimilate palm oil efficiently, were investigated. T candida Y-128 attached palm oil droplets directly, and assimilated unsaturated fatty acid more easily than saturated acids liberated by the action of its lipase. Lipase of A. calcoaceticus KB-2 was extracellular and appeared quickly from the beginning of log phase of growth, whereas lipase of f candida Y-128 appealed intracellular. The lipases of two strains seem to be only enough to utilize the lipid materials for their own growth, without accumulation of lipases in the culture broth. Lipases of the strains have 1 (3-)-positional specificities on triglycerides. The patterns of palm oil assimilation showed that two strains attached droplets of lipid materials directly and split off fatty acids at 1 (3-)-position of triglycerides first, and assimilated the reaction products via fatty acids metabolic pathway.

Isolation and Characterization of a Feather Degrading Alkalophilic Streptomyces sp. TBG-S13A5 and its Keratinolytic Properties

  • Keratinases are of particular interest because of their action on insoluble keratins and generally on a broad range of protein substrates. Alkalophilic and neutrophilic actinomycete strains isolated from different soil samples, rich in keratinaceous substances were screened for keratinolytic activity. An alkalophilic isolate, TBG-S13A5, was found to possess good keratinolytic activity and was able to utilize feather as the sole carbon and nitrogen source. TBG-S13A5 exhibited an off-white aerial mass color with a rectus-flexibilis type of spore chain. The morphological, microscopical and biochemical characters were comparable with that of Streptomyces albidoflavus. Fatty acid methyl ester profiling (FAME) and 16S rDNA sequence analysis confirmed its identity as a strain of S. albidoflavus. Under submerged fermentation conditions, maximum protease production was recorded on the $5^{th}$ day of incubation at $30^{\circ}C$, using basal broth of pH 9.0 with 0.25% (w/v) white chicken feather. This strain could affect feather degradation when the initial pH was 8 and above and maximum protease production was recorded when the initial pH was around 10.5. The effectiveness of the crude enzyme in destaining and leather dehairing were also demonstrated.

  • This study tried to suggest the development of a writing education model for Christian Universities based on both Hobbs' media education model and Vanhoozer's media text analysis model. This model consists of 6 steps- Christian worldview establishment, access, analysis and evaluation, creation, reflection and social action. This was developed in order to be applied for the class "reading and writing" of liberal arts. Also, this is an appropriate model for media text writing education aiming at an alternative creation activity through a critical comprehension of the complex texts consisting of sounds, images, letters and so on. Furthermore, this is designed to train the capable persons having intelligence, character, and spirituality balanced, whom the education of Chrisitian universities aims at. Finally, this model pursues the student-friendly and amalgamative text writing appropriate for a new era and has an advantage to raise the power of various forms of letter writing which all the universities should stress as well.

Alcohol Fermentration of Naked Barley without Cooking (쌀보리의 무증자 Alcoho 효소에 관한 연구)

  • Alcohol fermentation of uncooked naked barley was carried out by the combined action of the maceration enzyme from black Aspergillus niger and the glucoamylase from Rhizopus sp. The combined enzyme preparation was found to be effective in maceration and saccharification of the raw naked barley starch. The Hydrolysis rate measured by the amount of glucose liberated reached more than 70% at pH 4.5 and 3$0^{\circ}C$ after 76 hrs. For alcohol fermentation without cooking, the naked barley mash of 18% initial total sugar was pretreated with concentrated sulfuric acid (0.15 weight % of the mash volume) at 55$^{\circ}C$ for 2hr, and used for alcohol fermentation. A simultaneous saccharification and fermentation was carried out at pH 4.8 and 3$0^{\circ}C$ for 96 hrs. Under this fermentation condition, 3.5% increase in alcohol yield together with 2.0% increase in alcohol concentration were obtained when compared with the conventional cooking fermentation.

Enhancing mechanism of the saccharification of uncooked starch in an agitated bead reaction system (무증자전분의 분쇄마찰매체에 의한 효소당화촉진 Mechanism의 규명)

  • In an agitated bead reaction system, the enzymatic saccharification of uncooked starch was substantially enhanced. The enhancement mechanism was investigated front the view of the structural aspect of starch. The mechanical impact caused by the movement of the attrition-milling media resulted neither the destruction of microcrystalline structure nor the fragmentation of starch granule. instead, the most distinct phenomenon was the swelling of starch granule up to about 2.5 times, and the swelling mechanism was not similar with that caused by cooking. However, in the case of the enzyme addition in the attrition coupled reaction system, the swollen starch was easily fragmented into the large number of small particles by the synergistic action of the enzyme and milling-media. The exposed surface area of the fragmented particles plays the major role in enhancing the saccharification. The saccharification rate was quite different depending on the source of starch, the reason was discussed in terms of the granular structure of uncooked starches.
