Effect of Temperature and Salt Concentration on Kimchi Fermentation (김치발효에 미치는 온도 및 식염농도의 영향)

  • Chemical and microbial changes during Kimchi (a group of Korean seasoned pickles) fermentation were carried out at various temperatures and salt concentrations. The time reaching optimum ripening of Kimchi varied depending upon fermentation temperature and salt concentration. At high temperature and low salt content Kimchi fermentation was faster than at low temperature and high salt content. The ratio of volatile to non-volatile acids reached its maximum at the optimum ripening time of Kimchi and decreased thereafter. Leu. mesenteroids, Lac. brevis, Lac. plantarum, Ped. cerevisiae, Str. faecalis and low acid producing Lactobacilli were isolated from Kimchi samples. However, the main microorganism responsible for Kimchi fermentation was Leu. mesenteroides and Lac. plantarum was the main acidifying organism. Total viable count increased rapidly in the beginning of fermentation and reached its maximum number at optimum ripening time and then decreased slowly as the acidity of Kimchi increased. While the total aerobic bacteria and fungi decreased during Kimchi fermentation, the yeast increased significantly at lower temperature.

  • Cadmium pentachlorophenolxanthate has been synthesized with pentachlorophenol, carbondisulfide sodium hydroxide and cadmium chloride and its fungicidal activity was tested to Pyricularia oryzae and Cochliobolus miyabeanus. The results are summarized as followes; 1) The sample V synthesized by acidifying the crude PCP-xanthate solution at pH 8, then treated with $BaCl_2\;and\;CaC_2$ showed the highest purity, and the yield was $85.1\%$. 2) The same sample of $50\%$ wettable dust formulation inhibited conidial germination of P. oryzae and C. miyabeanus completely at 10 and 20 ppm, respectively. (Fig. 1) 3) The sample of $1.5\%$ dust formulation inhibited mycelial growth of P. oryzae and C. miyabeanus with 96 and $65\%$, respectively. The results were similar to those obtained by Ceresan calcium (Fig. 2) 4) Both dust and wettable powder formulations had no phytotoxcity on rice seedlings.

Effects of Korean Red Ginseng marc with aluminum sulfate against pathogen populations in poultry litters

  • Background: The aim of this study was to evaluate the effects of Korean Red Ginseng marc with aluminum sulfate as litter amendments on ammonia, soluble reactive phosphorus, and pathogen populations in poultry litters. Methods: Increasing levels of Korean Red Ginseng marc with aluminum sulfate were applied onto the surface of rice hull as a top-dress application; untreated rice hulls served as controls. Results: Treatment with Korean Red Ginseng marc with aluminum sulfate or aluminum sulfate alone resulted in lower litter pH (p < 0.05), as compared with that of the controls. There were some differences (p < 0.05) between treatments with Korean Red Ginseng marc with aluminum sulfate or aluminum sulfate alone and controls at 2-4 wk (not at 1 wk). Ammonia levels reduced on an average by 29%, 30%, and 32% for 10 g, 20 g Korean Red Ginseng marc with aluminum sulfate, and aluminum sulfate alone, respectively, as compared with controls at 4 wk. During the experiment, Korean Red Ginseng marc with aluminum sulfate or aluminum sulfate treatment had an effect (p < 0.05) on soluble reactive phosphorus content, as compared with the controls (not at 4 wk). A decrease in Salmonella enterica and Escherichia coli was observed (p < 0.05) in litter amended with both Korean Red Ginseng marc with aluminum sulfate and aluminum sulfate alone, as compared with the control, except at 1-3 wk for Salmonella enterica and 1 wk and 4 wk for Escherichia coli, respectively. Conclusion: The results showed that using Korean Red Ginseng marc with aluminum sulfate (blends), which act as acidifying agents by reducing the pH of the litter, was equally effective as aluminum sulfate in reducing the environmental impact.

A Study of Vegetation and Soil Characteristic of Urban Forest in Daejeon City (대전시 도시숲의 식생 및 토양특성에 관한 연구)

  • This study was conducted to suggest appropriate methods for management of urban forest after investigating the present condition and problems of urban forests by analyzing vegetation and soil properties in urban forests in the Daejeon. On the basis of our research, Pinus rigida dominate Gyejoksan and Bomunsan. Pinus densiflora dominate Wolpyeong park and Quercus acutissima dominate Namsun park. On the basis of our result of analysis of soil chemical properties, all investigated areas have low pHs, available phosphates and exchangeable cations. They indicate that the soil of those areas have been acidifying progressively. Soil hardness measurements were conducted to know the conditions of trampled soils and the results of them show that soil hardness in Namsun park was higher than the others. This indicates that human interference affect the health of the urban forest.

Relationship between RNA- and Protein-Synthesis and Cell Wall Acidification in Auxin-Mediated Elongation of Sunflower Hypocotyls (해바라기 하배축의 오옥신 유도 신장에서 RNA 및 단백질의 합성과 세포벽 산성화의 관계)

  • The roles of RNA- and protein-synthesis and $H^{+}$ excretion in 1AA ($10\;\mu\textrm{M}$)-induced elongation were investigated using abraded hypocotyl segments of sunflower (Helianthus annuus L.). The response of elongation initiated about 13 min after IAA treatment. Removal of cuticle, acting as diffusion barrier for inhibitors, by mechanical abrasion of hypocotyl segments enhanced the effect of inhibitors markedly, but the degree of abrasion for the saturated effect of inhibition was different among inhibitors. The elongation induced by 1M was completely inhibited when cycloheximide ($10\;\mu\textrm{M}$) was applied to abraded hypocotyl segments as shortly as 4 min before the onset of the growth response (= 10 min after administration of IAA). Cordycepin ($200\;\mu\textrm{M}$) prevented completely 1AA-induced elongation when applied as shortly as 19 min before the onset of the growth response (=5 min before administration of 1AA). Vanadate (1 mM) inhibited both lAA-induced elongation and medium acidification via lAA-induced $H^{+}$ excretion to apoplast. Cycloheximide and cordycepin also prevented lAA-induced $H^{+}$ excretion strongly. However, inhibition by cycloheximide of lAA-induced elongation was not alleviated by acidifying the cell wall to pH 4.5. The results indicate that, a few minutes before the initiation of growih, protein synthesis is demanded for the initiation of 1AA-induced elongation and the $H^{+}$ excretion to cell wall, and that the H+ excretion, even though it may be necessary for elongation, does not seem to bring about acid growth simply through acidifying cell wall.l wall.

Separatipon of Oryzanol from the Refining By-Product of Rice Bran Oil (미강유 정제 부산물로부터 오리자놀 분리)

  • To isolate oryzanol from the by-product of rice bran oil refinning, experiment of solvent fractional crystallization was carried out at various conditions with the dark oil obtained by acidifying the soap stock of micella refinning process and the pitch obtained from vacuum distillation of the dark oil. The impurity interfering the crystallization process such as waxes can be removed as precipitates by cooling the 1:1 mixtrue of acetone and dark oil to $0^{\circ}C$, From the dewaxed dark oil, oryzanol concentrate with 51.3% purity was obtained by fractional crystallization at$0^{\circ}C$ with the mixture of 8 part volume of hexane and 1 part of the dewaxed dark oil. The concentrate was recrystallized at room temperature with 20 part volume of methanol to yield oryzanol crystal of 98.3% purity. The optimum condition of vacuum distillation was temperature of $180^{\circ}C\;at\;0.2{\sim}0.4\;torr$ with 2% steam sparging. At this condition, the free fatty acid in the dark oil was removed as distillate without thermal deomposition to yield 82.3% of oryzanol as the pitch of 27.3% purity. After concentration from the pitch with 20 part volume of hexane to yield yellow powder of 75.4% purity, the yellow powder was recrystallized in methanol at room temperature to obtain the crystal containing 99.0% oryzanol. The overall oryzanol yield from the dark oil and the pitch was 9.5 and 28.5%, respectively. The change of the composition of sterols and triterpenoid alcohols in the compounds isolated during fractionation was analyzed by GC-MS.

Pollution Characteristics of Rainwater at Jeju Island during 2009~2010 (2009~2010년 제주지역 강우의 오염 특성 연구)

  • The collection of rainwater samples was made at Jeju area during 2009~2010, and the major ionic species were analyzed. In the comparison of ion balance, conductivity, and acid fraction for the validation of analytical data, the correlation coefficients showed a good linear relationship within the range of 0.966~0.990. The volume-weighted mean pH and electric conductivity were 4.9 and $17.8{\mu}S/cm$, respectively, at the Jeju area. The volume-weighted mean concentrations of ionic species in rainwater were in the order of $Cl^-$ > $Na^+$ > $nss-SO_4{^{2-}}$ > $NH_4{^+}$ > $NO_3{^-}$ > $Mg^{2+}$ > $H^+$ > $nss-Ca^{2+}$ > $HCOO^-$ > $K^+$ > $PO_4{^{3-}}$ > $CH_3COO^-$ > $NO_2{^-}$ > $F^-$ > $HCO_3{^-}$ > $CH_3SO_3{^-}$. The ionic strength of rainwater was $0.26{\pm}0.21$ mM during the study period. The composition ratios of ionic species were such as 50.1% for the marine sources ($Na^+$, $Mg^{2+}$, $Cl^-$), 30.9% for the anthropogenic sources ($NH_4{^+}$, $nss-SO_4{^{2-}}$, $NO_3{^-}$), and 4.7% for the soil source ($nss-Ca^{2+}$), and 3.1% for organic acids ($HCOO^-$, $CH_3COO^-$). From the seasonal comparison, the concentrations of $NO_3{^-}$, $nss-Ca^{2+}$, and $nss-SO_4{^{2-}}$ increased in winter and spring seasons, indicating a reasonable possibility of long range transport from Asia continent. Especially, the acidifying contributions by major inorganic acids ($nss-SO_4{^{2-}}$ and $NO_3{^-}$) and organic acids ($HCOO^-$ and $CH_3COO^-$) were 87.6% and 12.4%, respectively. In comparison by sectional inflow pathway of air mass during the rainy sampling days, the concentrations of $nss-SO_4{^{2-}}$ and $NO_3{^-}$ were relatively high when the air mass was moved from the China continent into Jeju area.

  • The objectives of this study was to determine whether a new physically modified cornstarch by ultra-fine- or nanoscale pulverizer to reduce particle size offers better bioactive function than native cornstarch in weanling Sprague-Dawley rats. Male weaning Sprague-Dawley rats were fed diets containing native cornstarch (NAC), ultra fine pulverized cornstarch (UFC) or nano-scale pulverized cornstarch (NSC) for 4 weeks. In vitro rate of starch hydrolysis, growth performance, organ weight, intestine length intestinal proliferation and the fermentation by Bifidobacterium of rat cecum were evaluated. The diet with reduced particle size (UFC or NSC) significantly increased body weight gain and organ weight. Feed efficiency was increased in NSC fed rats and was not affected in UFC fed rats. Intestinal proliferation was decreased in NSC group. Reduction of particle size also increased cecal short chain fatty acid concentration and the growth and acidifying activity of Bifidobacterium. It is concluded that a reduction of particle size of starch granules by physically modification may increase growing performance and gut function.

Mineralogy of Precipitates and Geochemisty of Stream Receiving Mine Water in the Sambong Coal Mine (삼봉탄광 주변 수계에 대한 지화학적 특성 및 침전물에 대한 광물학적 연구)

  • One of the most significant environmental issues in abandoned coal mine is acidic drainage which gives rise to the many environmental problems that acidifying streams water, sedimentation of iron/aluminium hydroxide, and pollution of water and soil. Water and precipitate samples for experiments were collected from stream and bottom in the pit mouth of Sambong mine. Mine water shows pH range from 7.24 to 7.94 in winter and 3.87 to 5.73 in summer season. The EC shows range from 432 to $897{\mu}S/cm$ at the stream receiving mine water. The highest concentrations of cations such as Mg, Al, Ca, and Mn are showing 15.50, 4.56, 85.30, 12.76 mg/L in the pit mouth, respectively. The reddish brown precipitates (Munsell color 10R-5YR in winter and 2.5YR-5Y in summer) consist mainly of 2-line ferrihydrite and schwertmannite. The precipitates are characterized by rod or cylindrical forms, and coccus or sphere of 0.1 to $0.5{\mu}m$ in diameter.

Optimizated pH and Mitigated Ammonia Emission in Pig Manure Slurry by Soluble Carbohydrate Supplementation (수용성 탄수화물을 이용한 분뇨슬러리 pH 적정화 및 암모니아 휘산의 저감)

  • In Concentrated Animal Feeding Operations(CAFOs), emission of ammonia from stored manure contributes negatively on the wellness of livestock. In CAFOs facilities, indoor aerial ammonia concentration oftentime surpasses the critical level potentially harmful to livestock's immune system. Understandably, numerous researches to control aerial ammonia have been conducted in countries where CAFOs were practiced for many decades. Some innovative technologies, such as scrubber, bio-filter, and additives emerged, as a result. Among them, microbial additives became popular in Korea, due to an easiness of use and affordability. However, microbial additives still have some weaknesses. Their price is still high enough to discourage farmers who run a small scale farm and their effectiveness are still questioned by many users and researchers. In the present study, we found soluble carbohydrates, such as sugar, glucose, and molasses, when supplemented to pig slurry manure, can mitigate ammonia emission. To be more specific, pig manure slurry(120kg), stored in container(200L), was supplemented with sugar at 0.1%(w/w) and was, subsequently, monitored for pH and aerial ammonia for next 10 days. From this experiment, it was found that the sugar supplementation was effective in mitigating the aerial ammonia concentration (33% in average) when monitored daily. Also, the pH of manure slurry was maintained at relatively low level(8.2) in sugar-supplemented manure slurry while it was elevated to 8.5 in untreated slurry. Conclusively, the obtained data suggest that soluble carbohydrate can mitigate ammonia emission by acidifying manure slurry. Additionally, it can be suggested that soluble carbohydrates, such as sugar, glucose, and molasses, can be reasonable choices for animal farmers who have been looking for an alternative choice to replace expensive microbial additives.
