This study was carried out to evaluate the effect of culture methods on development of embryos with each developmental stage after heat shock in bovine oocytes. The results obtained were as fellows. 1. The culture method after heat shock on development of embryos was better drop-culture than co-culture. 2. The medium without amino acids were not effect of heat sock on development of embryos but it was in need of amino acid during formation of blastocyst.
환경오염원으로서 페녹시계 제초제인 ,2,4-D(2,4-dichlorophenoxyacetic acid)에 노출된 토양으로부터 분리된 세균인 Burkholderia cepacia YK-2에서 2,4-D에 의한 스트레스 충격 단백질의 생성을 조사하였다. 활발하게 생장을 하고 있는 B. cepacia YK-2의 배양은 다양한 농도의 2,4-D에 노출되어 스트레스 충격단백질을 생성하였다. 이 반응은 43kDa의 DnaK와 41kDa의 GroEL 단백질의 생성을 수반하였으며, 이 단백질들은 anti-DnaK 단일 항체와 anti-GroEL 단일 항체를 사용한 SDS-PAGE과 Western blot을 통하여 확인되었다. 생성된 총 스트레스 충격 단백질은 2-D PAGE 에 의하여 분석되었다. 다양한 2,4-D농도와 노출 시간에 따른 B. cepacia YK-2의 생존율을 분석한 결과, 이 세균의 생존율은 스트레스 충격 단백질의 생성과 비례하였다.

Heat shock proteins serve as chaperone by preventing the aggregation of denatured proteins and promote survival of pathogens in harsh environments. In this study, heat shock gene encoding a 84-kDa (p84) protein, which is one of the three major heat shock proteins in S. pneumoniae, was cloned and characterized. PCR with a forward primer derived from N-terminal amino acid sequence of the p84 and a reverse primer derived from the conserved second ATP-binding region of Clp family was used for amplification of the gene encoding the p84 and subsequently the PCR product was used for sequence determination. Sequence analysis of the p84 gene demonstrated that it is a member of ClpL. The deduced amino acid sequence of pneumococcal ClpL shows homology with other members of the Clp family, and particularly, even in variable leader region, with bovine Clp-like protein and L. lactis ClpL. S. pneumoniae clpL is the smallest clop member (701 amono acids) containing the two conserved ATP-binding regions, and hydrophilic N-terminal variable region of pneu-mococcal Clp ATPase is much shorter than any known Clp ATPases. Histidine tagged ClpL was overexpressed and purified from E. coli. Immunoblot analysis employing antisera raised against pneumococcus p84 demonstrated no cross-reactivity with Clp analog in Eschericha coli, Staphylococcus aureus and human HeLa cells. Preimmunization of mice with ClpL extended mice life partially but did not protect them from death.
Genetic responses of the edible seaweed Porphyra yezoensis tissue to acid shock have been compared using differential display technique. The tissue was challenged in seawater containing $0.05{\%}$ hydrogen chloride (pH 3.0) for 5 min, then rehabilitated in normal seawater for 10 min, 30 min, 60 min and 4 hrs. Total RNA extracted by the LiCl-guanidium method was reverse transcribed and amplified by PCR with arbitrary primers. The amplified fragment responded by the acid shock was selectively isolated from agarose gel and sequenced with DNA auto sequencer. Sequence (1056 bp) of the cDNA contained at least two genes for ASP7K (MW 7418) and ASP5K (MW 5512) proteins.
In this study, we examined the expression of heat shock proteins (HSPs) in fish cell line CHSE-2lnl and human HeLa cells exposed to heat shock. In fish CHSE-214 cells HSP70 was the major polvpeptide induced by an elevated temperature or an amino acid analog, while in HeLa cells HSP90 as well as HSP70 were prominently enhanced in response to these stresses. Pretreatment of actinomvcin D prior to heat shock completely inhibited the induction of fish HSP70, indicating the transcriptional regulation of fish HSP70 gene expression. In HeLa and CHSE-214 cells either recovering from heat shock or experiencing prolonged heat shock, attenuation in the HSP90 a'nd HSP70 induction occurred but both induction and repression of HSP70 synthesis appear 19 precede those of HSP90. Moreover, attenuation did not occur in the syntheses of 40 kDa and 42 kOto proteins which were only induced in CHSE-214 cells. The enhanced syntheses of these he proteins continued as long as CHSE-214 cells were Siven heat shock. These results suggest that down-regulation of HSP syntheses during prolonged heat shock may be controlled by several different. as vet undefined, mechanisms.
Changes in the activities of isocitrate dehydrogenase (IDH) and glutamate dehydrogenase (GDH) and changes in free amino acids in the cytoplasm of Saccharomyces cerevisiae have been studied under heat shock condition. Heat shock conditions led to a significant decrease of NAD-IDH and NAD-GDH, It was shown appeared that the meaningful patterns of increase of NADP-IDH and NADP-GDH. It suggested that heat shock in yeast leads to a splitting of the TCA cycle and that glutamate synthesis takes place through the coupling of the NADP-linked isocirate and glutamate dehydrogenase. It was shown that about 14% of total free amino acids of yeast cells was decreased by heat shock. Especially heat shock condition resulted in the marked decreases of serine family amino acids such as serine, glycine and cysteine, and in the considerable increases of the rates of methionine, alanine, glutamin.
The effects of heat shock, or all-trans retinoic acid, on the expression of the C-reactive protein mRNA in the HT-29 human colon carcinoma cells, as well as the functional role of the C-reactive protein as a molecular chaperone, were studied. The expression level of the C-reactive protein mRNA in the HT-29 cells was increased time-dependently when exposed to heat-shock, and dose-dependently when treated with all-trans retinoic acid. The activities of transglutaminase C and K in the HT-29 cells were significantly increased when treated with all-trans retinoic acid. The C-reactive protein prevented thermal aggregation of the citrate synthase and stabilized the target enzyme, citrate synthase. The C-reactive protein promoted functional refolding of the urea-denatured citrate synthase up to 40-70%. These results suggest that the C-reactive protein, which is induced in human colon carcinoma cells, when heated or treated with all-trans retinoic acid has in a part functional activity of the molecular chaperone.
Objective: This study examines the types of hazardous factors in the working environment and the time-trend for their exposure levels over 10 years (2007 to 2016). Study Design and Method: The types of hazardous factors and exposure levels were drawn from the 19 measurement reports on the working environment over 10 years at a shock absorber manufacturing facility. Risk assessment of the types of factors and time-trend of exposure levels were evaluated using the factors and exposure levels. Results: A total of 34 hazardous factors were evaluated. The types were noise, 15 organic compounds, seven kinds of acid sand alkalis, eight kinds of heavy metals, and three other compounds. Special management materials used were nickel, hexavalent chrome, and sulfuric acid. Human carcinogens (1A) used were trichloroethylene, nickel, and sulfuric acid. There were six types of substances belonging to the IARC's 2B (body carcinogens) classification or higher, including, methyl isobutyl ketone, ethyl benzene, and trichloroethylene. No detection was found for 627 out of the 2065 total measurements in 19 exposure survey reports, representing 30.4%. Organic solvents, acid and alkali products, and heavy metals showed continuous low exposure concentrations. Noise, welding fumes, and the evaluation of mixed solvents show a gradual decrease in geometric mean and maximum over the time-trend of 10 years. Conclusions: In the case of a shock absorber manufacturing facility, the hazardous factors of noise and the evaluation of mixed solvents still indicate high concentrations exceeding the exposure limits and necessitate reduction studies. These two factors and welding fumes showed a continuous decrease in their ten-year tendency. Organic compounds, acids/alkalis, and heavy metals were managed smoothly in a work environment of continuous low concentrations.
