Inhibitory Effect of Major Food Components on the Activity of Antimicrobial Active Substance from n-Hexane Fraction of Mallotus japonicus Muell on Listeria monocytogenes (식품의 주요 성분이 예덕나무 헥산추출물의 Listeria monocytogenes에 대한 항균활성에 미치는 영향)

  • Inhibitory action of major food components on antimicrobial active substance (linolenic acid) was tested against Listeria monocytogenes ATCC 19111 for 72 hr at $30^{\circ}C$. Linolenic acid (50, 1000 ppm) and n-hexane fraction of Mallotus japonicus Muell (50, 1000 ppm) were added to the broth culture medium containing casein(1%, 3%), soybean oil(1%, 3%) and soluble starch (1%, 3%), respectively. Linolenic acid 1000 ppm and n-hexane fraction 1000 ppm exhibited strongly antimicrobial actions on L. monocytogenes which were not detected viable cell after 24 hr. But casein (3%) and soybean oil (3%) strongly diminished the antimicrobial action of 1000 ppm of n-hexane fraction. Soluble starch (1%, 3%) did not affect the antimicrobial action of 1000 ppm of linolenic acid and n-hexane fraction.

  • The effects of the essential oil fraction and the fatty acid-fraction of the panax Ginseng on the various lipids (total cholesterol, phospholipid, triglyceride) in serum and liver tissues of the white rats were examined and observed after the administration of the fractions for eight weeks with the following results: 1. Serum total cholesterol level was slightly decreased in the group after giving the essential oil fraction four weeks, whereas the effects in the group after giving itfor eight weeks were increased. Phospholipid level was slightly elevated both in four weeks and in eight weeks, while triglyceride level was elevated after four weeks, whereas it showed nochange at all after eight weeks. In the liver tissues, the cholesterol content was slightly decreased after four weeks and remarkably decreased after the administration for eight week. Phospholipid increased after four week administration and then rather decreased after the eight week administration, while the triglyceride content decreased both in four weeks and in eight weeks. 2. In the group giving the fatty acid-fraction, serum total cholesterol level decreased after the four week administration and then returned to the pre-experiment mean after the eight-week administration, while phospholipid level increased after the four-week administration and then returned to the pre experiment mean. Triglyceride level increased after four week administration of the fraction and rather then decreased after eight weeks. In the liver tissues, total cholesterol content decreased both after four weeks and after eight weeks, while phospholipid level also decreased from four weeks through eight weeks, whereas triglyceride level increased after four weeks, and showed rather decreasing tendency after the eight-week administration.

  • To purify the macrophage phagocytic activity-enhancing component, ethanol-acetic acid (100 : 1, EA) extract of Korean wheat (Gobun wheat) and imported one (Australian Standard White, ASW) were fractionated with ethylacetate : methanol : $H_2O$(65 : 25 : 4, v/v/v), and identified by TLC and column chromatography. At least five fractions were separated from the EA extract of the wheats but amounts of fraction B, C and D were more in Gobun wheat than in ASW. The effects of all fractions on phagocytic activity were tested in macrophage J774 cells. Among the fractions, only fraction b of Gobun wheat showed significant increase of phagocytic activity against yeast.

  • Production of poly($\beta$-hydroxybutyrate-co-$\beta$-hydroxyvalerate)[poly(HB-co-HV) from glucose and propionic acid was studied in a two-stage fed-batch fermentation using Alcaligenes eutrophus NCIMB 11599. When either glucose became sufficient or the feeding rate of propionic acid decreased, production of poly(HB-co-HV) increased but concomitantly resulted in a reduced fraction of HV. During the copolymer accumulation stage, the specific production rate of hydroxyvalerate (HV) increased up to 0.013 (g-HV/g-RCM/h) but it decreased as propionic acid was accumulated. Control of the propionic acid concentration in the medium, therefore, is considered to be one of the most important operating parameters for production of poly(HB-co-HV) with a higher HV fraction. A high titre of poly(HB-co-HV) (85.6 g/I) with HV fraction of 11.4 mol$%$ could be obtained in 50 h by controlling the propionic acid concentration at 1 to 4 g/I.

  • The antioxidant activity of Artemisia iwayomogi was determined by measuring the radical scavenging effect on 1, 1-diphenyl-2-picrylhydrazyl (DPPH) radical. The methanol extract of A. iwayomogi showed strong antioxidant activity, and thus fractionated with several solvents. The antioxidant activity potential of the individual fraction was in the order of ethyl acetate > n-butanol > water > chloroform > n-hexane fraction. The ethyl acetate and n-butanol soluble fractions exhibiting strong antioxidant activity were further purified by repeated sitica get and Sephadex LH-20 column chromatography. Antioxidant chlorogenic acid was isolated as one of the active principles from the n-butanol fraction, together with the inactive components, 1octacosanol, scopoletin, scopolin, apigenin $7, 4^{I}$-di-O-methylether, luteolin $6, 3^{I}$-di-O-methylether (jaceosidin), apigenin methylether (genkwanin), 2, 4-dihydroxy-6-methoxyacetophenone $4-O-{\beta}-$D-glucopyranoside and quebrachitol. The antioxidant activity of chlorogenic acid was comparable to that of L-ascorbic acid, which is a well known antioxidant.

The Metabolism of (2-$^{14}C$) Mevalonic Acid on Photoperiodic Induction in Grafted Solanum Andigena

  • The metabolism of sterol precurosor in leaves of Salanum andigena grafted between photoinduced and noninduced plant was investigated with the use of (2-$^{14}$ C) mevalonic acid. By the technique of the preparative gas-liquid chromatography, radioactive compounds of squalene, 4,4’-dimethylsterols and 4-demethylsterol were isolated and determined quantitatively. When labeled mevalonic acid n as applied to leaves radioactivity was extensively incorporated into non-saponifiable materials of lipid fraction and aqueous fraction (ethanol-water fraction). Radioactivity of 14C derived from (2-$^{14}$ C) mevalonic acid was transmissible from photoinduced plant to non-induced plant across the graft union, as tuberization hormone was, and incorporated into the sterols of the non-induced plant. Inhibitors of sterol biosynthesis, SK & F 7997 and nicotinic acid, are effective suppressors of tuber growing, if applied to leaves during photoinduction period. The experimental results suggest that certain substance containing isoprene unit, or sterol-like compound may participate in tuber growing.

  • 여포액이 여포안에서는 난자의 성숙을 억제하지만 여포액을 일단 여포 밖으로 내어 배양액을 섞거나 혹은 여포액만으로 난자를 배양할 때 난자는 성숙분열을 일으키는 것을 보아 왔다. 저자들은 소의 여포액 내의 성분중 어떤 것 들이 난자 성숙에 유효한 가를 밝히기 위하여 본 실험을 행하였다. 그 결과는 다음과 같다. 첫째, 여포액을 투석하여 얻은 dialysable fraction을 이용하여 난자를 배양하였을 때 높은 성숙율을 얻었다. 반대로 non-dialysable fraction를 포함하는 기본배양액 내에서는 난자의 성숙율은 극히 낮았으며 퇴화하는 경향이 있었다. 둘째, paper chromatography 의 방법으로 dialysable fraction이 포함하는 free amino acid를 조사한 결과 8가지의 amino acid가 동정되었다. 이 amino acid를 기본배양액에 섞어 배양액으로 만들어 난자를 배양했을 때 난자성숙율이 90%이상이었고 제 2차 중기까지 도달한 난자는 30시간 이내에 10% 정도였다. 셋째, 상기의 amino acid가 들어있는 배양액에 다시 biotin를 첨가하여 배양하면 48시간내에 제 2차 분열 중기까지 도달하는 난자가 60%에 이르렀다. 위 결과로 보아 여포액은 in vitro인 경우 난자의 성숙을 유발하며 성숙을 유발하는 주요 성분은 여포액내에 포함되어 있는 몇가지 amino acid와 biotin과 같은 vitamine B가 될 것이라는 결론을 얻었다.

  • Fatty acids and sterols were isolated from petroleum ether extract of seeds of Acanthopanax sessiliflorum Seeman. Fatty acids were obtained from the saponified fraction and sterols from nonsaponified fraction of petroleum ether extract. Fatty acids were identified by gas liquid chromatography, and its composition was myristic acid, palmitic acid, stearic acid, oleic acid, linoleic acid and linolenic acid. The composition of sterols were determined by preparative TLC and gas liquid chromatography. It was confirmed to be a mixture of stigmasterol and ${\beta}-sitosterol$. These fatty acids and sterols were not previously reported from seeds of Acanthopanax sessiliflorum Seemann.

Nutritional Components of Zespri Green Kiwi Fruit (Actinidia delicosa) and Neuronal Cell Protective Effects of the n-hexane fraction (제스프리 그린 키위의 주요 영양성분 및 헥산 분획물의 신경세포 보호효과)

  • The physiological characteristics of kiwi (Actinidia delicosa) fruit were analyzed, which inclued its nutritional composition, in vitro-antioxidant activities, and neuronal cell protective effects. The most abundant components of mineral, amino acid, and fatty acid were found to be potassium (K), glutamic acid, and a-linolenic acid, respectively. The major free sugars were fructose, glucose, and sucrose. In addition, ${\beta}$-carotene and vitamin C contents were $1.35{\mu}g/100mL$ and 29.21 mg/100 g, respectively. The 2,2'-azinobis-(3-ethylbenothiazline-6-sulfonic acid) (ABTS) radical-scavenging activity of the n-hexane fraction obtained from the kiwi extract was 10.52% at a concentration of $1000{\mu}g/mL$. The malondialdehyde (MDA) inhibition of the n-hexane fraction was found to increased in a dose-dependent manner. The intracellular reactive oxygen species (ROS) accumulation after hydrogen peroxide ($H_2O_2$) treatment of PC12 cells was significantly reduced in the presence of the n-hexane fraction compared to PC12 cells treated with $H_2O_2$ only. Moreover, in the a MTT assay, the n-hexane fraction showed in vitro-protective effects against $H_2O_2$-induced neurotoxicity.

  • This study was conducted to develop environment-friendly agricultural products with anti-microbial activity against Acidovorax avenae subsp. citrulli as a pathogen of bacterial fruit blotch in cucurbit. Schlechtendalia chinensis was extracted by MeOH and solvent fraction. The hexane fraction, which showed highest value of anti-microbial activity, was analyzed by GC-MS. Each mass spectra, corresponding to each peak of chromatogram, was compared to MS database of Wiley library. As a result, myristic acid, palmitic acid and 3-n-pentadecylphenol were identified as maine compounds showing antimicrobial activity against A. avenae subsp. citrulli. Bioassay using commercial myristic acid, palmitic acid and 3-n-pentadecylphenol to test for the anti-microbial activity conformed the anti-microbial activity of potential active compounds, and myristic acid and 3-n-pentadecylphenol showed strong activity. In conclusion, myristic acid and 3-n-pentadecylphenol identified from S. chinensis were anti-microbial chemicals.
