본 연구에서는 ursolic acid의 위의 보호효과를 위한 실험을 실시하였다. 위장 질병에 대한 ursolic acid의 효과를 확인하기 위해서 급성, 만성위염은 각각의 HCl ethanol과 indomethacin에 의해 유도된 위염 동물 모델을 사용하여 관찰하였다. 대표적인 공격인자인 위산에 관해서는 PPI activity를 통해서 확인하였고, 위의 손상에 대한 보호인자에 관해서는, $PGE_2$를 정량적으로 분석하였다. 항균활성 실험은 만성위염, 위궤양, 위암에 원인인자로 잘 알려진 H. pylori로 실험하였다. AGS cell를 이용하여 DAPI 염색, Flow cytometry assay를 통하여 ursolic acid가 위암세포의 apoptosis에 관여하는지를 확인하였다. 그 결과 ursolic acid는 HCl ethanol과 indomethacin에 의해 유도된 급성, 만성에 대한 위손상을 억제하였다. Ursolic acid는 위산분비의 마지막 단계인 위염분비효소인 proton pump를 억제시킴으로써 산의 분비를 억제하였다. 그리고 ursolic acid는 위 점막의 보호인자인 $PGE_2$의 농도가 증가함으로써 위 점막 보호 효과를 확인하였다. 또한 ursolic acid는 공격인자인 H. pylori colonization을 억제하였다. DAPI를 이용한 핵 염색에서, 대조군과는 달리, 핵 형상의 변형과 함께 수축 된 세포 또는 염색질의 응축현상이 관찰되었다. Flow cytometry assay에서 ursolic acid에 의해 apoptosis가 증가하는 것을 확인 하였다. 이를 통하여 ursolic acid는 위 손상에 대한 방어 효과가 있음을 확인할 수 있었다.
Antioxidative and scavenging effects were investigated by using two hyaroxycinnamic acids (caffeetannins). such as caffeic acid and chlorogenic acid, on oxidative stress and hepatotoxicity that induced by paraquat. The results are summerized as follows: 1. To assess radical scavenging ability, reduction concentration (IC$\sub$50/) of 1.1 diphenyl-2-dipicrylhydrazine (DPPH) were measured. IC$\sub$50/ values of caffeic acid and chlorogenic acid were 29.7 ${\pm}$0.6 ${\mu}$M and 26.0${\pm}$0.5 ${\mu}$M respectively. Their radical scavenging activities showed concentration-dependent manner. 2. In H$_2$O$_2$-induced hemolysis assay to rat blood, caffeic acid and chlorogenic acid led to different effects, whose hemolysis inhibition ratios at 100 ${\mu}$M were 45.2${\pm}$7.1% and 11.6${\pm}$3.1% respectively 3. In hypoxanthine-xanthine oxidase system producing superoxide anion, caffeic acid and chlorogenic acid showed different inhibitory activities of xanthine oxidase showing 36.8${\pm}$4.3% and 5.4${\pm}$2.3% respectively. 4. To microsomal NADPH dependent cytochrome p-450 reductase in rat liver, paraquat consumed NADPH at a dose-dependent manner from 0 to 1 ${\mu}$M paraquat concentration. Caffeic acid and chlorogenic acid blocked NADPH consumption rates at concentration-dependent manner and inhibition ratios at 100 ${\mu}$M were 67.6% and 59.2% respectively. 5. Administration (30mg/kg, iv) of paraquat to rats caused the marked elevation of glutamic oxaloacetic transaminase (GOT), glutamic pyruvic transaminase (GPT), lactate dehydrogenase (LDH), alkaline phosphatase (ALP) and lipid peroxides (LPO) in the serum and lipid peroxides in the microsome as compared to the control group. Serum GOT, GPT, LDH, ALP and LPO and liver microsomal LPO were reduced significantly by caffeic acid (50mg/kg), chlorogenic acid (25mg/kg) and silymarin (150 mg/kg) as compared to the paraquat group. From these results, caffeic acid and chlorogenic acid exerted their antioxidative agents by removing reactive oxygen substance (ROS) and scavenging effects by inhibiting ROS generating enzyme. As a general, two hydroxyeinnamic acids showed the useful compounds for scavenger and reducer on the paraquat induced hepatotoxicity.
This study was conducted to determine the effects of microorganism inactivation using 3 ppm of aqueous ozone (AO), 1% citric acid, 1% lactic acid, and 1% acetic acid alone, as well as the combinations of AO and organic acid, for washing the raw materials of saengsik (carrot, cabbage, glutinous rice, barley) with or without agitation. The combination of AO and 1% of each organic acid significantly inactivated spoilage bacteria in both the vegetables and the grains (p<0.05). However, in the glutinous rice, no inhibitory effects were shown for total aerobic bacteria by using water, ozone, or the combination of AO with citric acid or lactic acid, without agitation. Microbial inactivation was enhanced with agitation in the grains, whereas dipping (no agitation) treatments showed better inhibitory effects in the vegetables than in the barley, suggesting that washing processes should take into account the type of food material.
It was studied that which components of the ginseng are related to the effects that accelerate Saccharomyces division by comparing the influences of ginseng powder, alcohol extracts of ginseng, and linoleic and stearic acids which are known as the ginseng components on the division. The addition of ginseng powder and alcohol extracts of ginseng to the glucose agar medium marked the conspicuous increases of the division of Saccharomyces: 44% increase by 0.1% powder, 53% increase by 0.05% alcohol extracts. Also, the addition of the fatty acids of less than 0.0008% marked the considerable increases of the division: 22% increase by 0.0008% linoleic acid, 31% increase by 0. 00016% linoleic acid, 12% increase by 0.0008% stearic acid. Therefore it can be concluded that a proper amount of those fatty acids contained in the ginseng has a definite effect on the acceleration of Saccharomyces division.
The experiment was carried out to investigate the effects of sulfuric acid and nitric acid among the main components of simulated acid rain (SAR) on the growth of vegetative organs and seed germination of Arabidopsis thaliana. The Arabidopsis treated with SAR supplemented with sulfuric and nitric acids, respectively, showed 28% and 30% decrease of shoot and root growth compared to the control plants, and also many necrotic spots on leaf surfaces after SAR treatment were observed. The shoot and root length for plants grown with nitric acid rain was 14% and 17% lower, respectively, compared to the control, whereas those grown with sulfuric acid rain was 24% and 25% lower than control plants. When Arabidopsis seeds were sown in distilled water, germination rate was 100% after 7 days. However, 80% in SAR medium supplemented with sulfuric and nitric acids, 88% in sulfuric acid rain medium and 93% in nitric acid rain medium. The germination abilities of seeds harvested from SAR supplemented with sulfuric and nitric acids, sulfuric acid rain, and nitric acid rain were 73%, 73% and 94%, respectively. Consequently, sulfuric acids showed more inhibitory effects than nitric acids on the growth of vegetative organs as well as germination rates in Arabidopsis.
The scavenging effects of two hyaroxycinnamic acids such as caffeic acid and chlorogenic acid on paraquat induced pulmonary toxicity were investigated. The results are summerized as follows: 1. In the 5-lipoxygenase assay, caffeic acid and chlorogenic acid inhibited the enzyme activities whose inhibition concentration (IC$\_$50/) were 4.1 and 9.6 ${\mu}$M respectively. 2. To evaluate the antiinflammatory effects on mediator related to the mechanism of inflammation, ADP-induced platelet aggregation assay and histamine degranulation assay were used. Caffeic acid and chlorogenic acid inhibited on ADP-induced platelet aggregation and histamine release at a concentration dependent manners. 3. Arachidonic acid-induced ear edema were inhibited by administration of caffeic acid and chlorogenic acid. 4. Cytologicad analysis of branchoalveolar lavage fluid (BALF) which was the useful tool for detection of an inflammatory response in the lungs of animals intoxicated with chemicals were used. Alveolar macrophages and neutrophils in BALF, as well as the protein content and the LDH activity in BALF supernatant increased by intoxication of paraquat, but decreased by administration of caffeic acid and chlorogenic acid. Therefore, two hydroxyeinnamic acids tested were the useful candidates for scavenger and antiinflammatory agents on paraquat induced pulmonary toxicity.
The antibrowning effcts of cysteine, citric acid and ascorbic acid on the browning reaction of enzymatic garlic hydrolyzate were investigated at 37$^{\circ}C$ for 12 days. Cysteine was the most effective antibrowning agent followed by citric acid. The antibrowning effects of cysteine and citric acid were greater as concentrations increased, and the optimal concentration of both cysteine and citric acid as antibrowning agents was 0.3%. Ascorbic acid itself contributed to the browning reaction and showed an accelerating effect as the concentration increased. The addition of 0.1% ascorbic acid as synergist either to 0.3% cysteine or 0.3% citric acid did not enhance significantly the antibrowning effect of cysteine or citric acid. When stored at 3$0^{\circ}C$, 4$0^{\circ}C$ and 5$0^{\circ}C$, the browning reaction was accelerated as the temperature increased, especially at 5$0^{\circ}C$. Even though the effects of citric acid and cysteine as inhibitors on the browning reaction decreased as temperature increased, cysteine was more effective in decreasing browning reaction than citric acid.
The ethanol extract from the root bark of Morus alba showed the strongest antimicrobial activity on the growth of almost all the tested microorganisms which were food-borne pathogens and food-related microorganisms. Therefore, fatty acid composition, amino acid composition and shape change of microorganisms treated with the ethanol extract from the root bark of Morus alba were examined. In effects of treatment with the ethanol extract on the fatty acid compositions of B. subtilis, S. aureus and E. coli, fatty aicd compositions such as hexadecanoic acid (16:0) and octadecanoic acid (18:2) of the tested strains were increased but pentadecanoic acid (15:0) heptadecanoic acid (17:0) and acid (16:1) and octadecenoic acid (18:1) of E. coli were decreased. The ethanol extract did not significantly affect the aminn acid composition of the tested strains. Transmission electron micrographs of microorgani는 treated with the ethanol extract exhibited morphological changes that irregularly contracted cell surface in S. aureus and destructed cell walls in B. subtilis and E. coli.
Free fatty acid (FFA) intake regulates blood pressure and vascular reactivity but its direct effect on contractility of systemic arteries is not well understood. We investigated the effects of saturated fatty acid (SFA, palmitic acid), polyunsaturated fatty acid (PUFA, linoleic acid), and monounsaturated fatty acid (MUFA, oleic acid) on the contractility of isolated mesenteric (MA) and deep femoral arteries (DFA) of Sprague-Dawley rats. Isolated MA and DFA were mounted on a dual wire myograph and phenylephrine (PhE, $1-10{\mu}M$) concentration-dependent contraction was obtained with or without FFAs. Incubation with $100{\mu}M$ of palmitic acid significantly increased PhE-induced contraction in both arteries. In MA, treatment with $100{\mu}M$ of linoleic acid decreased $1{\mu}M$ PhE-induced contraction while increasing the response to higher PhE concentrations. In DFA, linoleic acid slightly decreased PhE-induced contraction while $200{\mu}M$ oleic acid significantly decreased it. In MA, oleic acid reduced contraction at low PhE concentration (1 and $2{\mu}M$) while increasing it at $10{\mu}M$ PhE. Perplexingly, depolarization by 40 mM KCl-induced contraction of MA was commonly enhanced by the three fatty acids. The 40 mM KCl-contraction of DFA was also augmented by linoleic and oleic acids while not affected by palmitic acid. SFA persistently increased alpha-adrenergic contraction of systemic arteries whereas PUFA and MUFA attenuated PhE-induced contraction of skeletal arteries. PUFA and MUFA concentration-dependent dual effects on MA suggest differential mechanisms depending on the types of arteries. Further studies are needed to elucidate underlying mechanisms of the various effects of FFA on systemic arteries.
The effects of several compounds related to signal transduction cascade were determined to induce the production of alizarin and purpurin in the hairy root culture system of Rubia cordifolia var. pratensis. It was found that out of five tested compounds jasmonic acid(1 mg/l) and methyl jasmonate(1 mg/l) stimulated strongly the biosynthesis of the pigments while linolenic acid(1 mg/l) induced no significant increase of the product. Yeast extract(600 mg/l) and arachidonic acid(1 mg/l) showed relatively mild inducing effects on production of alizarin. The effects of jasmonic acid and methyl jasmonate were reduced by treatment with cycloheximide(2.8 mg/l).
