Studies on the Influence of Vitamin B12 and Acetyl Choline on the Regeneration of Rhodopsin (Vitamin B12 와 Acetyl Choline 이 Rhodopsindml 재생에 미치는 영향에 관하여)

  • The test group and control group, which were grouped with the same weight of five frogs (Rana nigromaculata) respectively, were adapted to light for two hours. Then the anterior lymph heart of the test group were injected with 0.5 cc of 0.1% acetyl choline and 0.5cc of Vitamin B12 (50mc gm/cc). After the frogs of this group were dark-adapted for 90 minutes, their heads were cut off under the dim red light and then their retina removed from eye-balls were extracted with 2% digitonin solution for 20 hours at $0^{\circ}C$. The results of the comparison of these two groups whose optical densities were measured before and after the illumination areas follows : (1) The group which had been injected with 0.5cc of 0.1% acetyl choline solution had the protomotive action on the regeneration of rhodopsin in comparison with the control group. (2) The group which had been injected with 25 mc gm of Vitamin B12 and 0.5cc of 0.1% acetyl choline solution had the controlling action on the regeneration of rhodopsin in comparison with the control group.

  • Acid dissociation constants of 5-acetyl-8-hydroxyquinoline have been determined by spectrophotometric and potentiometric methods at $25^{\circ}C$. $pK_1$, $pK_2$ values obtained by potentiometric method were 4.56, 8.20 respectively and $pK_1$ obtained by spectrophotometric method was 4.21. pK values were lower than those of 8-hydroxyquinoline. The lower basicity of 5-acetyl-8-hydroxyquinoline compared with 8-hydroxyquinoline was considered as the result of the electronic attraction of the acetyl group with aromatic ring.

The Effects on Sperm Parameters and Membrane after Treatment with Progesterone and/or Acetyl-L-Carnitine; Cryopreservation-Thawing (Progesterone 및 Acetyl-L-Carnitine이 정자의 동결-융해에 미치는 영향)

  • Objective : To assess the effects of progesterone and acetyl-L-carnitine used after treated with Isolate�� gradient before semen cryopreservation-thawing on sperm parameters and membrane integrity. Material and Methods : From April 2001 to July 2001, ten normal male partner of couples who were visited in vitro fertilization (IVF) clinics. the semens were treated with $Isolate^{(R)}$ gradient before cryopreservation, spermatozoa was incubated with progesterone (1, 5 and $10{\mu}M$), acetyl-L-carnitine (2.5, 5 and $10{\mu}M$), or both (progesterone, $1{\mu}M$; and acetyl-L-carnitine, $5{\mu}M$) for 30 min. Results: There were no differences in sperm parameters and vital stain among isolate only treated group, progesterone (1, 5 and $10{\mu}M$), acetyl-L-carnitine (2.5, 5 and $10{\mu}M$) and both (progesterone, $1{\mu}M$; and acetyl-L-carnitine, $5{\mu}M$). But, in high concentration of acetyl-L-carnitine ($10{\mu}M$) treated group, sperm parameters and vital stain were decreased. The statistical method was used ANOVA (Kruskal-Wallis test) and p value was <0.01. Conclusions : Neither progesterone nor acetyl-L-carnitine show to be protective effect on the cryodamage assessed by sperm parameters and vital stain (eosin-Y stain) in normal sperm. High concentration of acetyl-L-carnitine ($10{\mu}M$), however, was harmful effect on cryoprevention.

Properties of Acetyl-CoA Synthetase from Pseudomonas fluorescens

  • In Pseudomonas fluorescens grown on malonate as sole carbon source, acetyl-CoA synthetase was induced, suggesting that malonate is metabolized through acetate and then acetyl-CoA. Acetyl-CoA synthetase was purified 18.6-fold in 4 steps to apparent homogeneity. The native molecular mass of the enzyme estimated by a native acrylamide gel electrophoresis was 130 kDa. The enzyme was composed of two identical subunits with a molecular mass of 67 kDa. Optimum pH was 70. The acetyl-CoA synthetase showed typical Michaelis-Menten kinetics for the substrates, acetate, ATP and CoA, whose $K_m$ values were calculated to be 33.4, 74.8, and 40.7 mM respectively. Propionate. butyrate and pentanoate were also used as substrates by the enzyme, but the rate of the formation of the CoA derivatives was decreased in the order of the increase in carbon number. The enzyme was inhibited by the group-specific reagents diethylpyro-carbonate, 2,3-butanedione, pyridoxal-5'-phosphate and N-bromosuccinimide. In the presence of substrates the inactivation rate of the enzyme, by all of the group-specific reagents mentioned above decreased, indicating the presence of catalytically essential histidine, arginine, lysine and tryptophan residues at or near the active site. Preincubation of the enzyme with ATP, $Mg^{2+}$ resulted in the increase of its susceptibility to diethylpyrocarbonate, suggesting that ATP, $Mg^{2+}$ may induce a conformational change in the active site exposing the essential histidine residue to diethylpyrocarbonate. The enzyme was acetylated in the presence of acetyl-CoA, indicating that this is one of acyl-enzyme.

  • The crystal structure of N-acetyl-L-cysteine, $C_5H_9NO_3S,$ has been determined from three dimensional photographic intensity data $(CuK{\alpha}$ radiation) by single crystal X-ray diffraction analysis. There is one formula unit in the triclinic unit cell with a = 7.04(3), b = 5.14(2), c = 8.25(3) ${\AA}$, ${\alpha}$ = 106(2), ${\beta}$ = 51(1), ${\gamma}$ = 124(2)$^{\circ}$ and space group P$_1$, The structure was solved by the direct method and refined by the full matrix least-squares method. The final R value is 12.3% for 629 observed reflections. The C-carboxyl group and the N-acetyl group are very neary planar. The molecule appears to form with neighboring molecules a hydrogen bond, $O-H{\cdot}{\cdot}{\cdot}O(3)$ of length 2.59${\AA}$.

  • The structure and physiological function of human skin continuously weaken due to growing older. The reasons of aging from external conditions are long term exposure to sun, wind, heat, cigarette smoke, and etc. This also palmitoyl oligopeptide or ceramide oligopeptide are known asc ingredient stimulating collagens and have the effect of reproducing the upper level of skin. Acetyl hexapeptide is an ingredient that makes the skin and muscle suppler and reduces wrinkles. It is a major high function beauty ingredient that substitutes botox. After dividing 7%, 14%, and 20% Acetyl hexapeptide experimental groups as groups A, B, and C the control group and experiment groups' change of wrinkles, hair follicles, moisture content, and dead skin cells was analyzed. According to the results, Acetyl hexapeptide seems to affect wrinkles, hair follicles and moisture content contrasting to the control group. Experimental groups and control group showed similar change in dead skin cells. In contrast to the control group satisfaction of examines was affected in wrinkles, hair follicles and moisture but removing dead skin cells had similar result in experimental groups and control group.

The Efficient Synthesis of 6-O-(2-Acetamido-2-deoxy-${\beta}$)-D-glucopyranosyl)-D-galactopyranose and Its Derivatives (6-O-(2-Acetamido-2-deoxy-${\beta}$-D-glucopyranosyl)-D-galactopyranose 및 유도체의 합성)

  • Condensation of 3,4,6-tri-O-acetyl-2-deoxy-2-phthalimido-${\beta}$-D-glucopyranosyl bromide (2) with 1,2;3,4-di-O-isopropylidene-${\alpha}$-D-galactopyranose (3) in the presence of silver triflate and syn-collidine gave 1,2;3,4-di-O-isopropylidene-6-O-(3,4,6-tri-O-acetyl-2-deoxy-2-phthalimido-${\beta}$-D-glucopyranosyl)-${\alpha}$-D-galactopyranose (4) in $86{\%}$ yield. Cleavage of phthalimido group and de-O-acetylation with hydrazine, acetylation, and hydrolysis of isopropylidene and O-acetyl groups furnished 6-O-(2-acetamido-2-deoxy-${\beta}$-D-glucopyranosyl)-D-galactopyranose (1) with overall yield of $65.8{\%}$ starting from 3. Some other derivatives of 1 which have free hydroxyl groups at the specific position have also been prepared from 4. These compounds could be used as precursors for further glycosidation reactions.

The Effects of a High-fat or High-sucrose Diet on Serum Lipid Profiles, Hepatic Acyl-CoA Synthetase, Carnitine Palmitoyltransferase-I, and the Acetyl-CoA Carboxylase mRNA Levels in Rats

  • The purpose of this study was to investigate the effects of altering relative intakes of fat and carbohydrates on serum lipid profiles, hepatic acyl-CoA synthetase (ACS), carnitine palmitoyltransferase-I (CPT-I), and the acetyl-CoA carboxlyase (ACC) mRNA level in Sprague-Dawley rats. For four weeks the rats were fed either an AIN-76 diet or one of its modified diets that were supplemented with 20% beef tallow (high-fat diet, HF) and 66.3% sucrose (highsucrose diet, HS). The HS group had significantly higher serum triglyceride and total cholesterol concentrations when compared with the other groups. Serum LDL-cholesterol concentrations in the HS and HF groups were significantly higher when compared to the normal diet (ND) group. Serum HDL-cholesterol levels of the ND and HS groups were significantly higher than those of the HF group. The hepatic total lipid level of the HF group was significantly higher than those of other groups; triglyceride levels of the HS and HF groups were significantly higher than those of the ND group. Hepatic ACS mRNA levels of the HF group were significantly higher than those of the ND group. Hepatic CPT-I mRNA levels were higher in the HF group than other groups. Also, ACC mRNA levels in the liver increased in the HF group. In conclusion, changes in the composition of dietary fat and carbohydrates could affect the hepatic ACS, CPT-I, and ACC mRNA levels. These results facilitate our understanding of the coordinated regulation of the ACS, CPT-I, and ACC mRNA levels and will serve to enhance our understanding of the molecular mechanisms that underlie the regulation of fatty acid metabolism.

Selective Cleavage of 2,2,2-Trichloroethyl Group with Zinc Dust in the Presence of Phthalimido Function (Phthalimido기 존재하에서 Zinc Dust에 의한 2,2,2-Trichloroethyl 기의 선택적 환원분해)

  • In acidic media such as aqueous acetic acid, phthalimide is reduced with zinc dust to give 3-hydroxyphthalimidine while the 2,2,2-trichloroethyl esters or glycosides are reductively cleaved. However, it has been discovered that, by employing a mixture of THF and pH 4.5 buffer solution as a solvent, 2,2,2-trichloroethyl group can be selectively removed with activated zinc dust in the presence of phthalimido function, provided that the reactant or the product does not have any free carboxylic acid function. By applying the above methods, reaction of $2,2,2-trichloroethyl 3,4,6-tri-O-acetyl-2-deoxy-2-phthalimido-{\beta}-D-glucopyranoside$ (1) with activated zinc dust gave a good yield of $3,4, 6-tri-O-acetyl-2-deoxy-2-phthalimido-{\beta}-D-glucopyranose$ (5) in THF-buffer solution, and $3,4,6-tri-O-acetyl-2-deoxy-2-(3-hydroxyphthalimidino)-{\beta}-D-glucopyranose$ (6) in aqueous acetic acid.

  • The synthesis of 5-hyterocyclimethyl-2'-deoxyuridines (4a-f) has been accomplished by displacement reaction of 5-(bromomethyl)-3', 5'-di-O-acetyl-2'-deoxyuridine with heterocyclic compounds, followed by removal of acetyl protecting group with methanolic ammonia. The compoudns synthesized were evaluated the inhibitory effects on L1210 cell probiferation and antiviral activities against Herpes simplex virus type 1 (HSV-1) None of the compounds exhibited sufficient biological activities.
